Self-Replicating Rna For Cellular Reprogramming In Domestic Cat, Woranop Sukparangsi, Nattakorn Suban Na Ayuthaya, Ratchapon Prakongkaew, Santhita Karoon, Ampika Thongphakdee

The Thai Journal of Veterinary Medicine

Cat Induced Pluripotent Stem Cells have been previously established using viral vectors integrating into the host genome to deliver reprogramming factors. To aid wild felid conservation using stem cell technology, in this study we utilized non-integrative Venezuelan Equine Encephalitis-based self-replicating RNA (srRNA) to carry the reprogramming factors. This approach induced cat somatic cells towards pluripotency without genomic modification, supporting the future use of srRNA in wild felid cells. The srRNA expressing human OCT4, SOX2, KLF4, CMYC, and GLIS1 (OSKMG) could induce the formation of cat iPSC colonies after 4 rounds of transfections. Different species-specific Leukemia Inhibitory Factor (LIF) could capture …

Alternative Approaches To Live Attenuated Oral Rotavirus Vaccines, Makoye Mhozya Kanyema, Junhong Wang, Gui-Lian Yang, Xin Cao, Chunfeng Wang

The Thai Journal of Veterinary Medicine

Oral attenuated rotavirus vaccine is safe and effective in developed countries but, however, it is less effective in low income countries such as African and Asian countries. Low vaccine effectiveness in low-income countries is arising from a variety of sources such as maternal antibodies, chronic enteropathy and other enteric diseases such as cholera, typhoid, and parasitic diseases, as well as the microbiota. There is increasingly evidence related to the risk of intussusception for the oral vaccine. The development of effective and safer rotavirus vaccines, particularly, non-living candidates, is critical. The aim of this review therefore is to explore some of …

Protective Immunity Of The Pore-Forming Domains Of Actinobacillus Pleuropneumoniae Apx Toxins In A Mouse Model, Kamonpun Chuekwon, Li-Ting Cheng, Chun-Yen Chu

The Thai Journal of Veterinary Medicine

The hemolytic/leukolytic toxins ApxI, II, and III play a central role in the pathogenicity of Actinobacillus pleuropneumoniae and may serve as effective vaccine antigens. However, since these toxins are large in size, we aimed to determine whether only the pore-forming domain of the toxins may be sufficient for protective immunity. Mice were vaccinated with the pore-forming domains for immune response analysis and challenge test. Significant antibody response was observed for the ApxI and II vaccine groups. For cellular immune response, CD4+ and CD8+ T cell expansion was observed for ApxI. Pro-inflammatory cytokine (IL-1b, IL-6) and TH2-type …

Discovery Of Chicken Proventricular Necrosis Virus From Transmissible Viral Proventriculitis Outbreak In Broilers, Thailand, ์Nantaporn Wandee, Preeda Lertwatchrasarakul, Thaweesak Songserm

The Thai Journal of Veterinary Medicine

Transmissible viral proventriculitis (TVP) is an infectious disease-causing retarded growth in chickens. In Thailand, many stunted chickens have been found on commercial broiler farms since 2017. Significant post-mortem lesions are found only in the proventriculus. This study aims to investigate the causative agents of TVP outbreaks focusing on chicken proventricular necrosis virus (CPNV), the possible significant cause of TVP. CPNV was reproduced in specific pathogen-free (SPF) chickens. Affected chickens that have undergone proventriculitis are CPNV-positive by RT-PCR and nucleotide sequencing followed by phylogenetic analysis. The result showed that proventriculitis could be experimentally reproduced by inoculation of proventricular homogenate obtained from …

Green algae usually assigned to the genus Oophila are known to colonize egg capsules of amphibian egg masses across the Nearctic and Palearctic regions. We study the phylogenetic relationships of these algae using a phylotranscriptomic data set of 76 protein-coding single-copy nuclear genes. Our data set includes novel RNAseq data for six amphibian-associated and five free-living green algae, and draft genomes of two of the latter. Within the Oophila clade (nested within Moewusinia), we find samples from two European frogs (Rana dalmatina and R. temporaria) closely related to those of the North American frog R. aurora (Oophila …

Optimal Limb Regeneration Strategies In Hemigrapsus Sanguineus, Laura S. Fletcher Aug 2024

Optimal Limb Regeneration Strategies In Hemigrapsus Sanguineus, Laura S. Fletcher

Theses and Dissertations

Diet quality and energy allocation influence an individual’s life-history traits by determining how much energy is available for processes such as growth and reproduction. One common method of assessing diet quality involves measuring gut length or size, which is typically inversely related to diet quality. However, this often requires animal sacrifice and time- consuming dissections. Similarly, many traditional bioenergetics models fail to capture the context-dependence of energy allocation, as these models rely on fixed energy allocation rules. One such rule is the κ-rule of dynamic energy budget theory, which assumes a fixed proportion (κ) of assimilated energy is always allocated …

Exploring Multilevel Factors Associated With Dual-Method Contraceptive Use Among Adolescent And Young Adult Women, Lauren Holt Aug 2024

Exploring Multilevel Factors Associated With Dual-Method Contraceptive Use Among Adolescent And Young Adult Women, Lauren Holt

Dissertations

Sexually transmitted infections (STIs) continue to rise in the US, and adolescent and young adult women (AYAW) between ages 18 and 24 have some of the highest STI rates in the country. Further, AYAW who use prescribed contraceptives (commonly referred to as birth control) are less likely to use condoms and more likely to be diagnosed with an STI than non-prescribed contraceptive users. The purpose of this dissertation is to understand multilevel factors contributing to condom use decision-making and STI risk perception analysis among AYAW who use prescribed contraceptives. Data was using qualitative research methods in two separate studies. In …

A Comparison Of Diploid And Triploid Eastern Oysters For Aquaculture Production Under Extreme Temperatures And Salinities, Joshua H. Kim Aug 2024

A Comparison Of Diploid And Triploid Eastern Oysters For Aquaculture Production Under Extreme Temperatures And Salinities, Joshua H. Kim

LSU Master's Theses

Off-bottom aquaculture of Eastern oysters (Crassostrea virginica) is a nascent industry that is increasingly supported by the use of triploid oysters, which grow faster than diploids. Despite their growth advantage, elevated triploid mortality compared to diploids under high temperature, low salinity, or a combination of these conditions challenge consistent triploid production. Identifying the environmental thresholds at which differential triploid mortality occurs and predicting its economic impact are important to informing decision-making in oyster aquaculture. The goal of this thesis was to compare the biological and economic performance of diploid and triploid oysters under high temperature and low salinity conditions. …

The Basic Dualism In The World: Object-Oriented Ontology And Systems Theory, Martin Zwick Aug 2024

The Basic Dualism In The World: Object-Oriented Ontology And Systems Theory, Martin Zwick

Complex Systems Faculty Publications and Presentations

Graham Harman writes that the “basic dualism in the world lies…between things in their intimate reality and things as confronted by other things.” However, dualism implies irreconcilable difference; what Harman points to is better expressed as a dyad, where the two components imply one another and interact. This article shows that systems theory has long asserted the fundamental character of Harman’s dyad, expressing it as the union of internal structure and external function, which correspond exactly to what Levi Bryant, characterizing Harman’s views, refers to as the intra-ontic and the inter-ontic, respectively. After interpreting Harman’s dyad in terms of the …

School of Medicine Publications

Background

Major depressive disorder (MDD) is considerably heterogeneous in terms of comorbidities, which may hamper the disentanglement of its biological mechanism. In a previous study, we classified the lifetime trajectories of MDD-related multimorbidities into seven distinct clusters, each characterized by unique genetic and environmental risk-factor profiles. The current objective was to investigate genome-wide gene-by-environment (G × E) interactions with childhood trauma burden, within the context of these clusters.

Methods

We analyzed 77,519 participants and 6,266,189 single-nucleotide polymorphisms (SNPs) of the UK Biobank database. Childhood trauma burden was assessed using the Childhood Trauma Screener (CTS). For each cluster, Plink 2.0 was …

The Association Between Long Non-Coding Rnas And Alzheimer’S Disease, Carson M. Black, Anneliesse A. Braden, Samia Nasim, Manish Tripathi, Jianfeng Xiao, Mohammad Moshahid Khan

School of Medicine Publications

Neurodegeneration occurs naturally as humans age, but the presence of additional pathogenic mechanisms yields harmful and consequential effects on the brain. Alzheimer’s disease (AD), the most common form of dementia, is a composite of such factors. Despite extensive research to identify the exact causes of AD, therapeutic approaches for treating the disease continue to be ineffective, indicating important gaps in our understanding of disease mechanisms. Long non-coding RNAs (lncRNAs) are an endogenous class of regulatory RNA transcripts longer than 200 nucleotides, involved in various regulatory networks, whose dysregulation is evident in several neural and extraneural diseases. LncRNAs are ubiquitously expressed …
