Maternal insulin resistance and hyperglycemia lead to gestational diabetes mellitus (GDM) which can cause fetal hyperinsulinemia, ultimately leading to excessive fetal growth associated with macrosomia and increased adiposity at birth. Increased placental macronutrient transport and epigenetic changes in growth and metabolic genes affected by GDM contribute to fetal overgrowth and can result in complications such as overweight/obesity, insulin resistance/type 2 diabetes, cardiovascular disease risk factors, as well as cognitive impairment later in life. Choline, an essential nutrient, participates in lipid and energy metabolism by providing methyl groups for DNA methylation and thus may overcome the hypomethylation of growth and metabolic …

Indigenous identities in North America are much more complex than generally assumed. No single definition of “Native (American),” “(American) Indian,” or related terms provides a set of necessary and sufficient conditions that accurately or usefully describe all and only the right individuals and communities. I argue that understanding Native identities in the contemporary context requires examining multiple interconnected dimensions of Nativeness, articulated here as Enrollment and Recognition, Tribal Ethnicities, Native Race, Indian Panethnicity, and Indigenous Nationhood.

This dissertation investigates the mechanisms underlying variations in credit assignment to reward cues, with a focus on the lateral orbitofrontal cortex (lOFC). The lOFC has long been implicated in signaling reward expectancies, with controversy as to whether it signals the economic value of future outcomes, their sensory features to guide behavior or both. However, whether the lOFC signals the relative (competition-dependent) or absolute (contiguity-dependent) credit of cues is unknown. To address this, I employed a combination of novel behavioral assays, in-vivo electrophysiological recordings and optogenetic stimulation in rats. Eugenol (EUG) was reacted as a matrix with a mixture of metal oxides (ZnO and CuO) have been prepared from metal acetate, Zn (CH3CO2)2 and Cu (CH3CO2)2 as precursors and using ethylene glycol (EG) as a solvent for synthesis the [Eug/ ZnO: CuO] nanocomposite using Sol-Gel method. The synthesized nanocomposite was characterized using techniques (FT-IR, AFM, SEM, EDX, and XRD), where the average particle diameter was found to be within the nanoscale range. Hydrochlorothiazide drug determination in pharmaceutical and biological urine samples using a novel continuous flow-injection analysis technique that is rapid, simple, economical, and sensitive. The complex formed by the reaction of hydrochlorothiazide and O-phenylenediamine with potassium ferricyanide to produce an orange-colored product at 480 nm was the basis for the proposed method. Other chemical and physical parameters that affect the stability and development of the colored product used in this developed method includes sample volume, flow rate, reagent concentrations, and reaction coil. A straightforward, one-pot, three-component reaction between substituted aromatic aldehydes, 2-naphthol and 4- aminoantipyrine have been used to synthesis a series of new 4-(((2-hydroxynaphthalen-1-yl) (phenyl) methylene) amino)-1,5-dimethyl-2-phenyl-1,2-dihydro-3H-pyrazol-3-one derivatives (A1- A5). This reaction was carried out with zirconyl chloride (ZrOCl2.8H2O) as an efficient catalyst under the condition of ultrasound irradiation. In this study, graphene oxide (GO) flakes were embellished with NiCo2O4 (NC) nanoparticles by in situ deposition, and the produced composite (NC: GO) was utilized as an adsorbent to remove methyl green dye (MG) from aqueous solutions. The successful coating of graphene oxide with nickel cobaltite nanoparticles (NC) was verified using FT-IR, SEM and X-ray diffraction (XRD) studies. E. milii is abundant in phenolic compounds, terpenoids, steroids and alkaloids. The main objective of this study was to investigate the extracted flavonoid and nano-flavonoids against two types of cell lines. Nano-flavonoids were synthesized via chitosan-maleic complex reaction. Glycated hemoglobin (HbA1c) was incorporated as an indicator of dysglycemia, as opposed to fasting glycaemia, for the purpose of classifying patients based on their vascular incidents. The research was conducted to study the effect of different concentrations of nanoparticles (chitosan and titanium dioxide) of Fusarium oxysporum as elicitors to increase the production of active compounds from Salvadora persica callus. Determine the total flavonoids and alkaloids rustle that chitosan in 5 mg/L, titanium dioxide in 1 mg/L, and Fusarium oxysporum in 5 mg/L give 93.10 mg100mg, 128.7 mg/100mg, and 107.61 mg/100mg respectively whereas alkaloids give 2.39%, 3.91%, and 2.20 % respectively. The results showed superiority in the presence of flavonoids in the samples compared with alkaloids. Pregnant women are often prescribed or abuse opioid drugs. The placenta is likely the key to understanding how opioids cause adverse pregnancy outcomes. Maternal oxycodone (OXY) exposure of pregnant mice leads to disturbances in the layer of invasive parietal trophoblast giant cells (pTGC) that forms the interface between the placenta and uterus. These cells are analogous to extravillous trophoblasts of the human placenta. They are crucial to coordinating the metabolic needs of the conceptus with those of the mother and are primary participants in the placenta–brain axis. Opioid use disorder is a major public health crisis that is manifested by persistent drug-seeking behavior and high relapse frequency. Most of the available treatments rely on targeting opioid receptors using small molecules that do not provide sustained symptom alleviation. Psychoplastogens are a novel class of non-opioid drugs that produce rapid and sustained effects on neuronal plasticity, intended to produce therapeutic benefits. Here, we report the complete genome sequence of an Indian strain of chikungunya virus isolated from an infected patient from Hyderabad, Andhra Pradesh, India, during a massive outbreak in 2005–2006. The genome length spans 11,811 nucleotides and has a poly(A) tail of 29 residues at the 3′ end.

This study aimed to study some biological aspects of different stages of Culex pipiens Linnaeus, 1758 in a laboratory that gathered randomly from four regions in Iraq's Kurdistan from Erbil. The results of the current study revealed that the eggs were laid in rafts, and the egg was elongated in shape, which was narrowed in one end. The mean number of laid eggs was 187.4 eggs; the mean hatching percentage was 83.07%, and the mean incubation period was 28.3 hours. Methotrexate is a critical component of curative chemotherapy for pediatric acute lymphoblastic leukemia (ALL), but is associated with neurotoxicity. Information on long-term outcomes following an acute neurotoxic event is limited. Therefore, this report compares neurocognitive performance more than 12 months post diagnosis (mean = 4 years) between ALL patients with (n = 25) and without (n = 146) a history of acute neurotoxicity. Compared to children with no documented on-treatment neurotoxic event, children who experienced a neurotoxic event during treatment exhibited poorer performance on measures of fine motor function (p = .02) and attention (p = .02). Background: In recent years, pragmatic metformin use in pregnancy has stretched to include prediabetes mellitus, type 2 diabetes mellitus, gestational diabetes mellitus, and (most recently) preeclampsia. However, with its expanded use, concerns of unintended harm have been raised.
Objective: This study developed an experimental primate model and applied ultrahigh performance liquid chromatography coupled to triple-quadrupole mass spectrometry for direct quantitation of maternal and fetal tissue metformin levels with detailed fetal biometry and histopathology.

Background: Living donor kidney transplantation is the optimal treatment for end-stage kidney disease; however, few living donor candidates (LDCs) who begin evaluation actually donate. While some LDCs are deemed medically ineligible, others discontinue for potentially modifiable reasons.
Methods: At five transplant centers, we conducted a prospective cohort study measuring LDCs' clinical and psychosocial characteristics, educational preparation, readiness to donate, and social determinants of health. We followed LDCs for 12 months after evaluation to determine whether they donated a kidney, opted to discontinue, had modifiable reasons for discontinuing, were medically ineligible, or had other recipient-related reasons for discontinuing.

Aims: Protozoan and helminth parasitic infections pose significant public health challenges, especially in developing countries with rural populations marked by suboptimal hygiene practices and socio-economic constraints. The parasites are the etiological agents of these infections and have a notably elevated global prevalence. Therefore, this study focuses on estimating the frequency and transmission dynamics of several parasitic species, including Blastocystis, Giardia, Cryptosporidium spp., Entamoeba histolytica, Ascaris lumbricoides, Trichuris trichiura, Taenia spp. and hookworms, within a rural community in southwest Colombia with a particular emphasis on the One Health framework, considering environmental and zoonotic transmission potentials.

Interactions between microbiota and enteric pathogens can promote colonization resistance or enhance pathogenesis. The pathobiont Enterococcus faecalis increases enterohaemorrhagic E. coli (EHEC) virulence by upregulating Type 3 Secretion System (T3SS) expression, effector translocation, and attaching and effacing (AE) lesion formation on enterocytes, but the mechanisms underlying this remain unknown. Using co-infection of organoids, metabolomics, supplementation experiments and bacterial genetics, here we show that co-culture of EHEC with E. faecalis increases the xanthine-hypoxanthine pathway activity and adenine biosynthesis. Background: Black women experience a disproportionate impact of uterine fibroids compared to White women, including earlier diagnosis, higher frequency, and more severe symptoms. The etiology underlying this racial disparity remains elusive.
Objective: The aim of this study was to evaluate the molecular differences in normal myometrium (fibroid-free uteri) and at-risk myometrium (fibroid-containing uteri) tissues in Black and White women.

Background: The brain and spinal cord formation is initiated in the earliest stages of mammalian pregnancy in a highly organized process known as neurulation. Environmental or genetic interferences can impair neurulation, resulting in clinically significant birth defects known collectively as neural tube defects. The Fuz gene encodes a subunit of the CPLANE complex, a macromolecular planar polarity effector required for ciliogenesis. Ablation of Fuz in mouse embryos results in exencephaly and spina bifida, including dysmorphic craniofacial structures due to defective cilia formation and impaired Sonic Hedgehog signaling.

A systematic review of functional neuroimaging studies on drug (self-) administration in rodents is lacking. Here, we summarized effects of acute or chronic drug administration of various classes of drugs on brain function and determined consistency with human literature. We performed a systematic literature search and identified 125 studies on in vivo rodent resting-state functional magnetic resonance imaging (n = 84) or positron emission tomography (n = 41) spanning depressants (n = 27), opioids (n = 23), stimulants (n = 72), and cannabis (n = 3). Coronary heart disease (CHD) is a prevalent cardiac disease that causes over 370,000 deaths annually in the USA. In CHD, occlusion of a coronary artery causes ischemia of the cardiac muscle, which results in myocardial infarction (MI). Junctophilin-2 (JPH2) is a membrane protein that ensures efficient calcium handling and proper excitation-contraction coupling. Studies have identified loss of JPH2 due to calpain-mediated proteolysis as a key pathogenic event in ischemia-induced heart failure (HF). Our findings show that calpain-2-mediated JPH2 cleavage yields increased levels of a C-terminal cleaved peptide (JPH2-CTP) in patients with ischemic cardiomyopathy and mice with experimental MI. BACKGROUND: Dupilumab is a monoclonal antibody that targets the interleukin (IL)-4 receptor alpha subunit, thus blocking the effects of IL-4 and IL-13, and has shown efficacy in treating various conditions including asthma, atopic dermatitis, eosinophilic esophagitis, and others. Because of its immune modulatory effects, clinical trials that studied dupilumab did not allow patients to receive live vaccines during the clinical trials because of an abundance of caution, and thus package inserts recommend that patients who are being treated with dupilumab should avoid live vaccines. Purpose of review: We discuss the sex-based differences in the pharmacokinetics and pharmacodynamics of kidney protective medications and their implications on women's health.
Recent findings: A critical examination of adverse drug reactions highlights the underrepresentation of women in clinical trials for kidney and cardiovascular protective drugs, such as SGLT2 inhibitors, ACE inhibitors, and endothelin receptor antagonists. This underscores the need for sex-specific analyses in clinical studies to accurately assess medication efficacy and safety.
