Central noradrenergic (NA) neurons are key constituents of the respiratory homeostatic network. NA dysfunction is implicated in several developmental respiratory disorders including Congenital Central Hyperventilation Syndrome (CCHS), Sudden Infant Death Syndrome (SIDS), and Rett Syndrome. The current unchallenged paradigm in the field, supported by multiple studies, is that glutamate co-transmission in subsets of central NA neurons plays a role in breathing control. If true, NA-glutamate co-transmission may also be mechanistically important in respiratory disorders. However, the requirement of NA-derived glutamate in breathing has not been directly tested and the extent of glutamate co-transmission in the central NA system remains uncharacterized. …

Tumor-associated neutrophils (TANs) have been shown to promote immunosuppression and tumor progression, and a high TAN frequency predicts poor prognosis in triple-negative breast cancer (TNBC). Dysregulation of CREB-binding protein (CBP)/P300 function has been observed with multiple cancer types. The bromodomain (BRD) of CBP/P300 has been shown to regulate its activity. In this study, we found that IACS-70654, a selective CBP/P300 BRD inhibitor, reduced TANs and inhibited the growth of neutrophil-enriched TNBC models. In the bone marrow, CBP/P300 BRD inhibition reduced the tumor-driven abnormal differentiation and proliferation of neutrophil progenitors. Myofibers are highly specialized contractile cells of skeletal muscles, and dysregulation of myofiber morphogenesis is emerging as a contributing cause of myopathies and structural birth defects. Myotubes are the myofiber precursors and undergo a dramatic morphological transition into long bipolar myofibers that are attached to tendons on two ends. Similar to axon growth cones, myotube leading edges navigate toward target cells and form cell-cell connections. The process of myotube guidance connects myotubes with the correct tendons, orients myofiber morphology with the overall body plan, and generates a functional musculoskeletal system. Adamts12 Promotes Fibrosis By Restructuring Extracellular Matrix To Enable Activation Of Injury-Responsive Fibroblasts, Konrad Hoeft, Benjamin D. Humphreys, Et Al.

Fibrosis represents the uncontrolled replacement of parenchymal tissue with extracellular matrix (ECM) produced by myofibroblasts. While genetic fate-tracing and single-cell RNA-Seq technologies have helped elucidate fibroblast heterogeneity and ontogeny beyond fibroblast to myofibroblast differentiation, newly identified fibroblast populations remain ill defined, with respect to both the molecular cues driving their differentiation and their subsequent role in fibrosis. Using an unbiased approach, we identified the metalloprotease ADAMTS12 as a fibroblast-specific gene that is strongly upregulated during active fibrogenesis in humans and mice. Functional in vivo KO studies in mice confirmed that Adamts12 was critical during fibrogenesis in both heart and kidney. …

Autophagy is an essential self-degradative and recycling mechanism that maintains cellular homeostasis. Estrogen receptor-related orphan receptors (ERRs) are fundamental in regulating cardiac metabolism and function. Previously, we showed that ERR agonists improve cardiac function in models of heart failure and induce autophagy. Here, we characterized a mechanism by which ERRs induce the autophagy pathway in cardiomyocytes. Transcription factor EB (TFEB) is a master regulator of the autophagy-lysosome pathway and has been shown to be crucial regulator of genes that control autophagy. As dozens of new National Immunization Technical Advisory Groups (NITAGs) were established worldwide in the past decade, and as existing NITAGs continued to play an important role in vaccine policy, global NITAG partners recognized a need for a standardized assessment tool to evaluate and strengthen their functions. This article describes the development of the NITAG Maturity Assessment Tool (NMAT), a stepwise evaluation tool that assesses NITAGs on seven key indicators of structure and process. The α-amylase enzyme, sourced from diverse organisms, including plants, animals, and bacteria, plays a crucial role in multiple industries, notably food processing sectors like cakes, fruit juices, and starch syrup. Research identifies thermophilic organisms as prime sources of this enzyme thriving at temperatures ranging from 41°C to 122°C. The enzyme purification was carried out using liquid-liquid extraction, which involved the exchange of substances between two liquid phases that were immiscible or partially soluble. The optimal temperature for α-amylase was 45 to 90°C. The best pH for bacterial and fungal α-amylases ranged from 5.0 to 10.5 and 5.0 to 9.0. Based …

This study assessed the effect of an eight-minute cycling intervention using varying intensities on exercise-induced hypoalgesia (EIH). The main objective of this study was to examine the effect of varying intensities on pressure pain threshold (PPT) and heat pain threshold (HPT) at the thigh and forearm, tested pre- and post-cycling intervention. Healthy male participants (n = 16) performed a graded exercise test on a cycle ergometer to establish their peak power output (PPO). International Journal of Exercise Science 17(3): 1361-1376, 2024. The blood pressure dipping response to acute resistance training exercise (RTE) is scarce in the literature. We determined the inter-individual blood pressure (BP) dipping variability of normotensive (NT) and hypertensive (HT) men completing two modalities of a single session of RTE. Volunteers (NT n = 21, HT n = 20) underwent a non-exercise control (CTRL), RTE high-sets low-repetitions (HSLR), and RTE high-repetitions low-sets (HRLS) conditions. Twenty-four-hour ambulatory BP monitoring recorded diurnal and nocturnal systolic (SBP) and diastolic (DBP) BP. International Journal of Exercise Science 17(6): 1318-1336, 2024. High-incline walking is a relatively new trend with little comparative information. This study compared physiological and psychological differences between high-incline walking at 20% grade (HIW) and level-grade jogging (LGJ) at isocaloric intensities in young adults. Twenty-two participants (M = 11, F = 11) aged 19–31 years completed the study. Participants completed HIW and LGJ on a treadmill for twenty minutes on two separate occasions and matched at isocaloric intensities. International Journal of Exercise Science 17(1): 1306-1317, 2024. Injury of the anterior cruciate ligament (ACL) in the knee is common, with up to 250,000 cases annually in the United States. Such injuries can lead to muscle atrophy, impaired balance, and limited movement. This study aimed to compare the lower limbs of individuals with ACL reconstruction to a Control group. We hypothesized that ACL participants would exhibit greater asymmetry between lower limbs as compared to Controls. Data were collected from 12 ACL participants and 30 Control participants. Alzheimer's disease (AD) is the most common neurodegenerative disease. The trigger for AD is the accumulation of amyloid-beta protein (Aβ) as senile plaques in the brain. Prior to forming the fiber-like structures found in the plaques, Aβ undergoes an oligomerization process that produces intermediate structures called protofibrils. Substantial data from the Nichols laboratory demonstrated that soluble Aβ protofibrils were highly inflammatory compared to other forms of Aβ. Based on these findings, a serum polyclonal antibody, named Antibody St. Louis or AbSL, was developed to target Aβ protofibrils. Economic and Business Review
Risky financial decisions, defined as rational calculations between expected risk and reward, are subject to various psychological and neurobiological mechanisms. In this context, the relationship between testosterone levels and risk propensity has been investigated, but the results are inconsistent. Here, the effects of some personality traits, neuroticism–anxiety and sociability, on the relationship between testosterone levels and risk propensity were examined in decisions under risk (GDT) and under uncertainty (BART). In a mixed-sex sample of 100 graduate students and experienced decision makers, we found that basal testosterone levels were positively correlated with risk propensity for decisions under risk in males with …

INTRODUCTION: Brain magnetic resonance imaging (MRI) and inflammatory biomarkers are crucial for investigating preclinical neurocognitive disorders. Current investigations focus on a few inflammatory markers. The study aims to investigate the associations between inflammatory biomarkers and MRI measures and to examine sex differences among the associations in the Framingham Heart Study. METHODS: Dementia and stroke-free participants underwent OLINK Proteomics profiling and MRI measurements within 5 years. Pairwise cross-sectional analysis assessed 68 biomarkers with 13 brain MRI volumes, adjusting for covariates and familial correlations. Glioblastoma multiforme, a disease of grade IV astrocytoma, accounts for 60-70% of malignant brain tumors.¹ Despite its low survival rate, little is known about GBM and its treatment. The purpose of this study was to assess the comparative effectiveness of the most common drugs (temozolomide, carmustine, bevacizumab) in reducing tumor growth within GBM chemotherapy. It was hypothesized that due to their properties as alkylating agents inhibiting growth genes within GBM, temozolomide and carmustine would provide the most reduction in cellular growth mathematically.² VisualPDE simulations were used to model administration of temozolomide, carmustine, bevacizumab, and none in chemotherapy and provide the …
