Global environmental governance is currently encountering serious challenges. The lack of integration between biodiversity conservation and Sustainable Development Goals (SDGs) has imposed significant stress on the Earth system. Ecologically significant regions, particularly those with limited resources, are confronted with issues such as weak governance structures and restricted developmental opportunities. As the core platform of the Man and the Biosphere Programme (MAB), the World Network of Biosphere Reserves (WNBR) serves as a global model for reconciling conservation with sustainable development. Biosphere Reserves are areas recognized by UNESCO that represent key terrestrial, coastal, and marine ecosystems. These areas are strictly protected and designed to fulfill three primary functions: conserving biodiversity, promoting sustainable development, and supporting scientific research and education. As of now, 759 biosphere reserves have been established in 136 countries and regions worldwide, supporting the livelihoods of approximately 236 million people. The total area of biosphere reserves covers 7.67 million km2, accounting for 4.10% of global terrestrial area, 5.08% of freshwater surface, and 0.21% of marine areas. Building human-nature harmonious communities is foundational to effective protected-area governance. Drawing on fifty years of global practice under UNESCO’s Man and the Biosphere (MAB) Programme, this study distills four practice-oriented insights: establishing co-management committees to secure the institutional basis; developing ecological branding to add value to ecological products and raise household incomes; cultivating ecological flagships to enhance international reputation and attract investment; and safeguarding cultural diversity to strengthen residents’ sense of belonging and support for management. Based on a brief review of the Dinghushan National Nature Reserve/Dinghushan World Biosphere Reserve, this study comprehensively reviews its scientific research progress in the past 70 years, including three stages: early ecological investigation and biodiversity monitoring; mid-term research on ecosystem structure, function, and dynamics; and recent comprehensive studies. The study also analyzes the role of scientific research in supporting biodiversity conservation, local community sustainable development, and mitigation and adaptation to global climate change in the reserve. The technologies and methods adopted in ecological monitoring by the Wolong National Nature Reserve, located in Sichuan, China, hold extensive representativeness and influence on a global scale. In September 2025, the Fifth UNESCO World Congress of Biosphere Reserves will be held in Hangzhou, China. The delegates will engage in detailed discussions on key issues, including the harmonious coexistence of humans and nature, as well as balanced conservation and development. This congress will also develop a strategic and action plan for the next phase of the world biosphere reserves. The Tianmushan-Qingliangfeng World Biosphere Reserve represents China’s first successful application for world biosphere reserve achieved through an expansion process. Transboundary conservation, as a pivotal strategy in addressing the global biodiversity crisis, plays a critical role in maintaining ecological connectivity and promoting regional ecological security and sustainable development. Under the initiative of the Xishuangbanna World Biosphere Reserve and with bilateral governmental support from China and Laos, a contiguous transboundary protected area spanning approximately 220 km and encompassing 200 000 hectares was established along their shared border in 2012. The integrated protected area community (IPAC) model is a Chinese innovation in protected area governance, emerging from the convergence of national policy support, local government implementation, and community participation. Rather than relying solely on administrative jurisdiction or geographic proximity, IPAC uses protected areas as institutional hubs to align ecological conservation with community development and regional collaboration. Based on the case of the Gaoligongshan UNESCO biosphere reserve, this study examines how policy coordination, stakeholder integration, and ecological value realization are operationalized under the IPAC approach, highlighting an integrated mechanism that combines state-led planning, multi-entity co-governance, and ecological product valorization. Cancer patients often experience changes in mental health, prompting an exploration into whether nerves infiltrating tumors contribute to these alterations by impacting brain functions. Using a mouse model for head and neck cancer and neuronal tracing, we show that tumor-infiltrating nerves connect to distinct brain areas. The activation of this neuronal circuitry altered behaviors (decreased nest-building, increased latency to eat a cookie, and reduced wheel running). Tumor-infiltrating nociceptor neurons exhibited heightened calcium activity and brain regions receiving these neural projections showed elevated Fos as well as increased calcium responses compared to non-tumor-bearing counterparts. Cancer cachexia mouse models are needed to recapitulate the clinical features of patients with cachexia. Here, we present a protocol for the establishment and evaluation of cancer cachexia mouse models. We delineate the steps in preparing tumor cells for inoculation and surgical procedures. After the establishment of these mouse models, we describe essential techniques to assess cancer cachexia, including grip strength evaluation, tissue collection, and the calculation of cross-sectional areas of muscle tissue. Background: Chronic pain, affecting approximately 20% of the global population, is the leading cause of disability worldwide. Transgender individuals are disproportionately exposed to chronic pain risk factors compared with the cisgender population. This study compares the incidence of chronic pain between transgender and cisgender individuals and examines the impact of gender affirming hormone therapy, anxiety, and depression on chronic pain. Methods: The study analysed medical records data of 56,470 transgender men and 41,882 transgender women in the TrinetX database. Six cohorts were created: transgender women either receiving oestrogen or no intervention, transgender men receiving testosterone or no intervention and cohorts …

Ocean energy extraction is on the rise. While tides are the most predictable amongst marine renewable resources, turbulent and complex flows still challenge reliable tidal stream energy extraction and there is also uncertainty in how devices change the natural environment. To ensure the long-term integrity of emergent floating tidal turbine technologies, advances in field measurements are required to capture multiscale, real-world flow interactions. Manufactured soils, created by combining various organic and inorganic waste materials and byproducts, may be tailored to specific applications, providing an alternative to the extraction of natural soils. It is important for them to be capable of supporting plant growth without the need for significant management or fertiliser applications, the over-application of which can have adverse environmental effects. Clinical trials of active surveillance (AS) for Ductal Carcinoma in Situ (DCIS) are underway. We sought to understand the historical management of biologically favorable DCIS and to determine the outcomes of patients who did not have immediate surgery. Using data from the NCDB from 2004 to 2017, the selected cohort included women >40 years of age, with low or intermediate grade and hormone receptor (HR) positive DCIS. AS was defined as either no surgery or surgery >12 months from diagnosis. Women in the AS group were compared to women who had immediate surgery. A Cochran-Armitage test was used to assess …

Cancer stem cells (CSCs) established from surgical biopsies closely mimic the human context and can be used to investigate disease mechanisms, genetic fitness, and therapeutic evaluation. Here, we present a protocol for the derivation of primary patient-derived CSC lines from ependymal tumors. We describe the necessary steps, from surgical intervention and biopsy to the dissociation of ependymomas to derive cultures. We then detail procedures for cell line propagation and define the characteristics of these primary cancer cell lines. Humans with monogenic inborn errors responsible for extreme disease phenotypes can reveal essential physiological pathways. We investigated germline mutations in GNAI2, which encodes Gαi2, a key component in heterotrimeric G-protein signal transduction usually thought to regulate adenylyl cyclase-mediated cAMP production. Patients with activating Gαi2 mutations had clinical presentations that included impaired immunity. Mutant Gαi2 impaired cell migration and augmented responses to T cell receptor (TCR) stimulation. We found that mutant Gαi2 influenced TCR signaling by sequestering the GTPase-activating protein RASA2, thereby promoting RAS activation and increasing downstream ERK/MAPK and PI3K-AKT S6 signaling to drive cellular growth and proliferation.

BACKGROUND: Hybrids are expected to show greater phenotypic variation than their parental species, yet how hybrid phenotype expression varies with genetic distances in closely-related parental species remains surprisingly understudied. Here, we investigate pelage and morphometric trait variation in anthropogenic hybrids between four species of Brazilian Callithrix marmosets, a relatively recent primate radiation. Marmoset species are distinguishable by pelage phenotype and morphological specializations for eating tree exudates. Over the past three years, new SARS-CoV-2 variants have continuously emerged, evolving to a point where an immune response against the original vaccine no longer provided optimal protection against these new strains. During this time, high-throughput neutralization assays based on pseudoviruses have become a valuable tool for assessing the efficacy of new vaccines, screening updated vaccine candidates against emerging variants, and testing the efficacy of new therapeutics such as monoclonal antibodies. Runoff from rapidly melting mountain glaciers is a dominant source of riverine organic carbon in many high-latitude and high-elevation regions. Glacier dissolved organic carbon is highly bioavailable, and its composition likely reflects internal (e.g., autotrophic production) and external (i.e., atmospheric deposition) sources. However, the balance of these sources across Earth's glaciers is poorly understood, despite implications for the mineralization and assimilation of glacier organic carbon within recipient ecosystems. We assessed the molecular-level composition of dissolved organic matter from 136 mountain glacier outflows from 11 regions covering six continents using ultrahigh resolution 21 T mass spectrometry. Apoptosis is an evolutionarily conserved cell death pathway that plays a crucial role in maintaining tissue homeostasis, orchestrating organismal development, and eliminating damaged cells. Dysregulation of apoptosis can contribute to the pathogenesis of malignant tumors and neurodegenerative diseases. Anticancer drugs typically possess the capacity to induce apoptosis in tumor cells. The Bcl-2 protein family, consisting of 27 members in humans, serves as the key regulator of mitochondrial function. This family can be divided into two functional groups: anti-apoptotic proteins (e.g., Bcl-2, Bcl-xl, Mcl-1) and pro-apoptotic proteins (e.g., Bad, Bax).
