Purpose: Lynch syndrome (LS) is a cancer predisposition syndrome caused by a germline loss-of-function mutation in a mismatch repair (MMR) gene. While sarcomas are not classically considered LS cancers, we investigated the MMR status and clinical features of sarcomas in LS patients to help inform optimal treatment strategies.
Methods: A prospectively maintained institutional clinical cancer genetics database was queried for LS patients (defined by pathogenic germline mutation in a MMR gene) with a documented diagnosis of sarcoma between 1998-2022. Cancer driver mutations often show distinct temporal acquisition patterns, but the biological basis for this, if any, remains unknown. RAS mutations occur invariably late in the course of acute myeloid leukaemia, upon progression or relapsed/refractory disease1-6. Here, by using human leukaemogenesis models, we first show that RAS mutations are obligatory late events that need to succeed earlier cooperating mutations. We provide the mechanistic explanation for this in a requirement for mutant RAS to specifically transform committed progenitors of the myelomonocytic lineage (granulocyte-monocyte progenitors) harbouring previously acquired driver mutations, showing that advanced leukaemic clones can originate from a different cell type …

Objectives: Chemotherapy-induced peripheral neuropathy (CIPN) is a complication that may occur after treatment with various anticancer drugs. In refractory CIPN cases, spinal cord stimulation (SCS) has garnered increased attention. The use of gait analysis and psychophysical quantitative sensory testing (QST) as an objective measurement of CIPN-related damage has burgeoned; however, these changes have not been reported for patients with CIPN after SCS implantation using either burst or tonic stimulation.

OBJECTIVE: Investigation of explainable deep learning methods for graph neural networks to predict HIV infections with social network information and performing domain adaptation to evaluate model transferability across different datasets.
METHODS: Network data from two cohorts of younger sexual minority men (SMM) from two U.S. cities (Chicago, IL, and Houston, TX) were collected between 2014 and 2016. Feature importance from graph attention network (GAT) models were determined using GNNExplainer. Introduction: Letters of recommendation (LOR) are an integral component of physical therapy residency applications. Identifying the influence of applicant and writer gender in LOR will help identify whether potential implicit gender bias exists in physical therapy residency application processes.
Review of literature: Several medical and surgical residency education programs have reported positive, neutral, or negative LOR female gender bias among applicants and writers. Little research exists on gender differences in LOR to physical therapy education programs or physical therapy residency programs.

Background: Uveal melanoma (UM) is a highly aggressive disease with very few treatment options. We previously demonstrated that mUM is characterized by high oxidative phosphorylation (OXPHOS). Here we tested the anti-tumor, signaling and metabolic effects of imipridones, which are CLPP activators, which inhibit OXPHOS indirectly and have demonstrated safety in patients.
Methods: We assessed CLPP expression in UM patient samples. We tested the effects of imipridones (ONC201 and ONC212) on the growth, survival, signaling and metabolism of UM cell lines in vitro, and for therapeutic efficacy in vivo in UM liver metastasis models.

Sexually predatory behavior has long been an issue within many institutions and organizations. The Catholic Church has been scrutinized for the involvement of Popes, priests, nuns, and others as perpetrators and enablers of child sexual abuse. Reports of child sexual abuse dating back as far as the 1940s have plagued the Catholic Church in both the US and Europe, with estimates in the US across all dioceses suggesting over four thousand priest/deacon perpetrators and over ten thousand victims affected between the 1950s and 2002. This thesis explores the implementation of a fully 3D Richards’ equation solution into MODFLOW 6 to solve for saturated/unsaturated flow. Currently, MODFLOW 6 has limited capability for solving unsaturated zone flow problems by using the Unsaturated Zone Flow (UZF) package, which neglects capillarity and assumes vertical 1-dimensional flow. Various formulations of the Richards’ equation were first evaluated for their strengths and weaknesses. Various numerical solution approaches to Richards’ equation were also studied to understand the robustness and efficiency of different approaches and to select one that would best fit into the MODFLOW 6 framework. The relationship between dietary fiber intake and human health has gained significant attention due to its potential in preventing chronic diseases and promoting gut health. Grains are a primary source of dietary fiber; however, the health impact of grain fibers is often limited because they are incompletely metabolized by gut bacteria. This dissertation explores innovative strategies to enhance the concentration and of dietary fiber from grains and its utilization by gut bacteria. By investigating various treatments, this research aims to maximize the health benefits of grain-derived dietary fibers, thereby contributing to a healthier society.

This study introduces a simulation of biological reinforcement learning to explore the behavior of natural enemies in the presence of host pests, aiming to analyze the population dynamics between natural enemies and insect pests within an ecological context. The simulation leverages on Q-learning, a reinforcement learning algorithm, to model the decision-making processes of both parasitoids/predators and pests, thereby assessing the impact of varying parasitism and predation rates on pest population growth. The climate crisis is upon us, whether we like it or not, and its undeniable effects are evident. Take, for example, Hurricane Helene, which caused unimaginable devastation to what was once considered a “climate haven,” Asheville, NC (Andreoni, 2024; Medina & Fausset, 2024). The Florida Keys reef tract has rapidly shifted from a structurally complex, hard coral-dominated reef to a less rugose, soft coral-dominated reef. Persistent anthropogenic stressors facilitate ecological transitions by including recreational and commercial fishing and increased anthropogenic marine debris. These artificial structures are becoming habitats for small marine taxa known as cryptofauna. Cryptofauna are among the most essential yet understudied reef taxa where little is known about their fate considering degrading coral reefs and increasing anthropogenic disturbance. In higher education, there has been a growing interest in identifying and addressing the systemic factors that impact the educational experiences and outcomes of underrepresented students pursuing degrees in engineering. This dissertation examines the resilience of former foster youth in higher education, focusing on the perspectives of Independent Living Coordinators (ILCs) in Arkansas. The study explores the support systems and challenges that foster youth encounter, assessing factors that contribute to or hinder their educational success. Using a qualitative methodology with semi-structured interviews, it identifies key protective factors such as emotional support, stable mentorship, and institutional programs that promote resilience among foster youth. The findings suggest that tailored interventions and a strengths-based approach can significantly improve outcomes for this vulnerable student population. Rice blending or mixing of different rice cultivars, a common practice in the industry, significantly impacts processing efficiency and end-product quality, yet its effects on end products remain understudied. This research investigated the implications of blending contemporary Arkansas rice cultivars through two comprehensive studies focusing on milling yields and physicochemical properties, instantization, and parboiling processes. Five long-grain cultivars, including hybrids (RT 7521 FP, RT 7321 FP, XP 753) and purelines (Ozark, CLL 16), were blended in various combinations and different drying.

Since its inception in 1973, Project Tiger, which laid the groundwork for India's tiger conservation program, has grown from 9 to 53 intensively monitored and managed tiger reserves. With an increase in tigers' permanent occupancy outside of designated tiger reserves, one of the many challenges confronting India's tiger conservation has been managing its success, with a particular emphasis on this spillover population that ranges extensively in human-dominated, multi-use forest landscapes.
