Cancer continues to be one of the leading causes of morbidity and mortality globally, influenced by a complex interplay of genetic, environmental, and nutritional factors. Among these, the metabolism of homocysteine (HCY), a sulfur-containing amino acid derived from dietary methionine, has attracted growing attention due to its involvement in one-carbon metabolism and its potential link to cancer. Elevated HCY levels, a condition known as hyperhomocysteinemia (HHCY), have been shown to disrupt crucial cellular processes such as DNA methylation and promote oxidative stress (OS)—both of which contribute to genomic instability and cancer progression. Mindfulness and mindfulness-based interventions have become increasingly popular in recent years. Typically, mindfulness-based interventions and their effects on mental health and/or adult populations have been explored in the literature. Limited research has been completed looking at mindfulness’ impacts on other facets, including chronic pain symptoms. Additionally, some research has been conducted to analyze mindfulness and its effects on school aged children and their day-to-day functioning with promising results. Epitaxial Quantum Dot Scintillators With Monolithic Photodetector Integration: Physics, Fabrication And Characterization Of Materials And Devices, Allan Oreave Minns

This dissertation discloses the physics, fabrication, and characterization of high-performance scintillator detectors based on epitaxial InAs quantum dots (QDs) in GaAs. The detector crystals grown include monolithically integrated In0.35GaAs photodetectors with Al0.92-0.6In0.03-0.35Ga0.05As metamorphic buffer layers (MBL). A device fabrication process was developed for the lithographic patterning, chemical etching, contact metallization and readout integration of materials grown by means of molecular beam epitaxy (MBE) on 3-inch GaAs (001) wafers. Low capacitance integration with readout electronics was achieved through wire bonds to custom printed circuit boards (PCBs) with commercial components.

Psychedelic intentions play an important role in shaping acute psychedelic experiences and influencing long-term outcomes. Limited existing literature suggests that different types of intentions are linked to acute experiences and outcomes in varying ways. However, data on the types of intentions set by recreational users remain scarce, and their effects lack clarity. This study investigates intention-setting practices among recreational psychedelic users, aiming to establish a taxonomy of intentions in naturalistic use contexts and clarify user perspectives on the impact of intention-setting. Exercise can promote health and well-being, and yet a proportion of individuals who exercise develop a maladaptive relationship with physical activity. Problematic exercise is often associated with significant impairment and psychological distress, yet a consensus has not been reached regarding how problematic exercise should be conceptualized. The literature distinguishes between primary exercise dependence, or excessive exercise driven by its intrinsic reinforcement, and secondary exercise dependence, or excessive exercise driven by compensatory motivations related to weight and shape concerns. Links between Type 1 diabetes (T1D) and decreased cognitive functioning in children are well documented, but research on executive functioning (EF) in this population is more limited. Multiple T1D-related risk factors (e.g., hypoglycemia, hyperglycemia, glycemic variability, diabetic ketoacidosis) for EF have been established throughout the literature, but their individual effects have been difficult to determine given their interrelated nature. In result, cumulative risk has been proposed as a more robust predictor of EF differences in children with T1D. RNA plays a critical role in many biological functions, such as normal cellular functions or disease-related processes. Regulating biological processes through RNA-based therapeutics can be employed by the reversible photoregulation of oligonucleotides. Azobenzene, a light-responsive compound that changes conformation when exposed to ultraviolet (UV) or visible light, is attached to cytidine phosphoramidite to be incorporated in a series of RNA oligonucleotides. The reversibility of the nucleoside and oligonucleotide is verified through UV melting point experiments. The modified oligonucleotide can change conformation upon light irradiation to stop reverse transcription (RT) in the presence of many different reverse transcriptase enzymes. This technique …

Duchenne Muscular Dystrophy (DMD) is an X-linked recessive muscle degenerative disease that frequently proves fatal by age 30. It primarily affects adolescent males with symptoms appearing as early as age 6. Patients quickly lose their ability to walk due to muscle weakness and are confined to wheelchairs for the remainder of their lives. DMD is caused by a mutation in the dystrophin gene. Without dystrophin, mechanical stress, injury, or shock takes a drastic toll on muscle tissue and muscle stem cells, rendering them unable to regenerate or be repaired. Insulin has widespread actions within the brain, including energy homeostasis, modulation of feeding, and as a key component of memory processes. Further, brain insulin dysfunction correlates with Alzheimer's disease and other neurodegenerative disorders. To this point, measurement of hippocampal insulin has been limited. Determining in vivo hippocampal insulin levels both at a healthy baseline and after, e.g., induction of a disease state or in response to cognitive challenge, is essential to advance our understanding of both insulin's procognitive actions and to guide potential therapeutic interventions.

Early responsive caregiving hones children’s physiological systems and subsequently supports social interactions (Feldman, 2007; Porges 2001; Steelman et al, 2002). Conversely, mis-attuned interactions with caregivers, such as exposure to maltreatment subtypes, might hinder social engagement capacities at both behavioral and biological levels (Kim & Cicchetti, 2004; Sheridan & McLaughlin et al., 2014). These early adverse influences endure across development and can affect behavioral and physiological interactions with romantic partners (Mayo et al., 2021; River et al., 2020; Zamir, 2022). This dissertation explores the impact of the 2021 New York State Gender Recognition Act (GRA) on mental health outcomes and service utilization among non-binary adults in the public mental health system. The GRA introduced pivotal legal reforms—including the option for an “X” gender marker and self-attestation for gender identity changes on state-issued documents—designed to affirm gender diversity and reduce structural barriers for transgender and non-binary populations. Guided by gender affirmation theory and minority stress frameworks, this study investigates how these legal shifts relate to changes in mental health service engagement and psychological well-being among non-binary individuals.

MicroRNAs (miRNA) are a class of small non-coding RNAs that play a key role in gene silencing. miRNAs form an RNA-induced silencing complex (RISC) with the argonaute 2 (AGO2) protein in order to control gene expression through silencing messenger RNAs (mRNA). The ability for a miRNA-mRNA complex to fit into AGO2 and form the RISC is based on the location and characteristics of a central bulge, but the mechanism and interactions that lead to a functional RISC remain unknown. Despite all the details, each character takes their senses of the world and strings them together as coherent images and stories. Proust shows that perceptions and memories are malleable, as individuals perceive objects in a way that brings entire stories or images to their conscious experience. Increasing numbers of people are fleeing political violence and becoming refugees who resettle all around the world.Compared to the general population in the countries where they resettle, refugees disproportionately suffer from depression and higher trauma-related mental health symptoms including post-traumatic stress disorder (PTSD) due to the experiences that led to their refugee status, as well as post-migration stressors, disadvantages in multiple social determinants of health, and lack of social support. Background: Myalgic encephalomyelitis (ME)/chronic fatigue syndrome (CFS) is a disabling multisystem complex disorder with no known etiology or approved treatment. Our studies aimed to assess the link between ME/CFS and autoimmune disease (AID) and cancer, and to identify potential biomarkers and biological mechanisms to further explore pathophysiology of this diseases.

Conventional computed tomography produces a 2D slice or 3D image from multiple conventional images to avoid overlapping structures and increase visualization of the internal features of the object. A set up, macro controls and processing for computed tomography was created for the Center for X-ray Optics for the first time, and demonstrated in this work. Adding differential phase contrast to conventional x-ray images highlights edges. Two more channels of information, dark field and integrated phase, improve the contrast and increase the signal-to-noise ratios of the image. In the modern digital era, children are increasingly engaging with digital media (i.e., screen time), and the health-related implications of increased screen time are not fully understood. Excessive screen time in early childhood is increasingly recognized as a potential contributing factor to adverse health outcomes, including increased body mass index (BMI) and disrupted sleep patterns. The post-transcriptional regulation of RNA, particularly through alternative splicing (AS) and chemical modifications, plays a critical role in gene expression and is linked to many human diseases. This thesis investigates three interconnected aspects of RNA and DNA dynamics and their regulation, providing new insights into cellular function.
First, we examine how RNA modifications influence the structural stability of transfer RNA Phenylalanine (tRNAPhe) in S. cerevisiae. Using temperature replica exchange molecular dynamics (T-REMD) simulations, we show that specific modifications create a network of interactions essential for maintaining tRNA’s tertiary structure. CRISPR-Cas12a is a rapidly evolving technology for improved gene editing in cells, as well as in vivo. The system, consisting of Cas12a nuclease and approximately 40-nt long guide RNA (gRNA), can efficiently recognize and cut genomic double-stranded DNA (dsDNA). Chemical modifications to the gRNA can potentially improve its dsDNA binding and enhance Cas12a-mediated nuclease activity. In this work, 5-methyl cytidine (m5C) and N6-methyl adenosine (m6A) were explored to improve the binding between gRNA and target DNA. The initial segment elucidates a series of synthetic works involving artificial molecules, specifically featuring the photosensitive azobenzene. These molecules facilitate remote control over the structure and functionality of biomolecules, with subsequent applications in regulating biological processes. The latter segment concentrates on the design and enhancement of organic photocatalysts, utilizing traditional acridine ions as a foundation. Abstract
Background:
Multimorbidity, defined as the co-occurrence of two or more health conditions (physical, mental, or both), has been thought of as a disease of the elderly, with most studies of multimorbidity focusing on adults and the elderly population. However, co-occurring (multimorbid) health conditions are common in adolescence, and it is important to understand the profiles and patterns of multimorbidity during adolescence, a critical developmental period with long-term implications. Social determinants of health, especially lower socioeconomic status (SES), are linked to higher risk of multimorbidity and poor outcomes in adulthood. Cancer cells often exhibit a Warburg-like metabolism, which includes increased fatty acid synthesis and storage that help them survive in hypoxic conditions. This shift is marked by the heightened synthesis and accumulation of fatty acids, acting as a survival mechanism in low-oxygen environments (Baumann et al., 2016). Interestingly, deep-diving whales may utilize a similar metabolic pathway to produce wax esters during their prolonged dives.
Our study focused on exploring the genetic and metabolic similarities between breast cancer cells and deep-diving whales, using computational analyses to investigate lipid metabolism genes. We looked for evolutionarily conserved variations that might reveal adaptive mechanisms …

Post-colonial multiethnic societies still face significant challenges in nation-building today. Not only do countries follow different paths in dealing with ethnic diversity, but many also shift their strategies across time. State policies towards the ethnic Chinese in Southeast Asia, including accommodation, integration, marginalization, and assimilation, vary along two dimensions: the state’s discrimination towards the ethnic Chinese and the room for negotiation between the state and the ethnic Chinese in formulating these policies. Non-tuberculosis mycobacteria (NTM) represent a diverse repertoire of species, most of which are ubiquitous in the environment. Some NTM species, such as Mycobacterium abscessus, can cause life-threatening infections in people with underlying medical conditions. The clinical incidence of such infections is on the rise, posing an emerging threat to public health. M. abscessus is characterized by intrinsic and acquired antibiotic resistance, complicating treatment strategies. Most chemotherapeutic approaches for M. abscessus infection target the bacterial ribosome, necessitating a better understanding of the physiological processes which confer this resistance. Our thoughts, memories, and actions are finely controlled by the coordinated activity of neural circuits in the brain. Prior work has identified a multitude of molecular mechanisms that contribute to the generation of these higher order functions, yet we know very little about how our ability to learn and perform habitual actions change depending on how far neurotransmitters released from neurons can travel away from their point of release. Background: Adequate hand function among older adults is necessary for the successful completion of activities of daily living and can affect whether an older adult is able to live independently. Current research highlights grip strength, the strength people have in their hands, as a robust predictor of morbidity and mortality, and explores the decline in grip strength over time. The transcription factor p63 is critical for development of the epidermis and other epithelial-related structures. Mutations in the DNA-binding domain of p63 lead to a range of human developmental disorders characterized by symptoms such as ectrodactyly, painful skin dysplasia, and cleft lip and palate. The TP63 gene is also amplified in squamous cell carcinomas, contributing to tumor pathogenesis. While the importance of p63 in human health is well established, the mechanisms underlying its control of an essential gene network remain unclear. TP63 encodes multiple isoforms with distinct N- and C-terminal domains. The ∆Np63α isoform is directly linked to epidermal development …
