Evaluating Multiparametric Imaging Biomarkers For Guiding Targeted Therapies In Her2+ Breast Cancer, Ameer Mansur
Evaluating Multiparametric Imaging Biomarkers For Guiding Targeted Therapies In Her2+ Breast Cancer, Ameer Mansur
All ETDs from UAB
Human epidermal growth factor receptor 2 positive (HER2+) breast cancer is a biologically diverse disease, in which intratumoral heterogeneity plays a significant role in tumor progression. Advanced medical imaging and image processing techniques offer the potential to noninvasively characterize the underlying intratumoral biology. Capturing these complex changes can allow for assessing therapeutic response and eventually guiding personalized therapeutic strategies. This work evaluates single and multiparametric imaging biomarkers to characterize tumor biology, identify treatment-induced changes, and predict response to targeted therapies. First, the utility of 3’-Deoxy-3’-[18F]Fluorothymidine positron emission tomography ([18F]FLT-PET) was assessed as a biomarker of proliferation for monitoring response to …
Investigating Potential Therapeutic Vulnerabilities And Elucidating Molecular Mechanisms For The Tumor Suppressors Nfkbia And Anxa7 In Diffuse Gliomas, Aran Merati
All ETDs from UAB
Gliomas, and especially diffuse gliomas, can be a devastating disease when attempting treatment in the clinic. Diffuse gliomas are typically malignant and pose definitive problems with respect to patient care predominantly due to their ability to exist outside of the bulk tumor by invading into the surrounding brain parenchyma. Consequently, not every cancerous cell can be fully removed by surgical resection and therapeutic resistance, so the stem-like population left behind can repopulate a recurrent tumor from a single cell. There is a desperate need for pinpointing novel therapeutic vulnerabilities in diffuse gliomas, which could potentially involve the targeting of key …
Maternal Immune Activation Causes Oligodendrocyte Deficits In The Hypothalamus, Kate Miller
Maternal Immune Activation Causes Oligodendrocyte Deficits In The Hypothalamus, Kate Miller
All ETDs from UAB
Maternal immune activation (MIA) refers to the occurrence of an inflammatory immune response during any stage of pregnancy. This manifests as the upregulation of proinflammatory cytokines and can be caused by air pollution, obesity, or viral infection. When this occurs during the first trimester in humans, it increases the offspring’s susceptibility to developing neuropsychiatric disorders such as autism (ASD) and schizophrenia (SZ), in males more commonly than females. In addition to behavioral manifestations of neuropsychiatric disorders, metabolic syndrome (MetS) is a common comorbidity, defined as the co-occurrence of three or more symptoms including but not limited to hyperlipidemia, hypertension, and …
Reducing Stress: The Role Of The Rcl Operon In Bacterial Stress Response To Hypothiocyanite, Julia Meredith
Reducing Stress: The Role Of The Rcl Operon In Bacterial Stress Response To Hypothiocyanite, Julia Meredith
All ETDs from UAB
The human immune system utilizes a variety of weapons against bacterial communities within the body, which allow it to control the overall microbial population. Understanding how some bacterial species are able to better deal with and survive damage by the immune system while others are not, can help us elucidate the complex relationship between us and our microbiome. One tool that immune cells use for microbial control is the potent oxidant hypothiocyanite/hypothiocyanous acid (-OSCN/HOSCN). The response mounted by Escherichia coli and other bacterial species in order to survive damage caused by HOSCN is the focus of this dissertation. In the …

The master's thesis presents research on enhancing plant resilience and growth through the study of the microbiome of submerged aquatic plants and the application of plasma technology in agriculture. We isolated 177 endophytic microbial strains from submerged plants collected from various locations. Screening revealed 50 strains producing ACC deaminase, an enzyme that mitigates plant stress by reducing ethylene levels. Predominant genera included Pseudomonas, Bacillus, and Acinetobacter. Metagenomic analysis characterized the diverse bacterial communities associated with these plants. Different plasma treatments were tested to optimize their effects on plant growth. Atmospheric jet and globe plasma treatments significantly improved root length and …
An Investigation Into The Effects Of Preservatives On Ss-Hydroxybutyrate Concentrations Measured By Commercial Ketone Meter And Gas Chromatography-Mass Spectrometry, Kerry F. Murr
All ETDs from UAB
Ketoacidosis occurs when fatty acids are metabolized into ketone bodies in the liver and released into the bloodstream. This condition, which can be fatal, is commonly observed in diabetics, alcoholics, and individuals who experienced starvation or hypothermia. Although the ketone ß-hydroxybutyrate (BHB) is an accepted biomarker of ketoacidosis, the effects of preservatives on BHB concentrations in postmortem blood are unaddressed. The first aim of this research was to determine whether preservatives affect ketone levels as measured by a point-of-care device. Paired preserved and unpreserved blood samples were obtained from the Jefferson County morgue: preserved samples in grey top tubes and …
Therapeutic Implications Of Fractionated Radiotherapy And Marcks’ Regulation Of Tunneling Nanotubes In Glioblastoma, Lauren Catherine Nassour-Caswell
Therapeutic Implications Of Fractionated Radiotherapy And Marcks’ Regulation Of Tunneling Nanotubes In Glioblastoma, Lauren Catherine Nassour-Caswell
All ETDs from UAB
Glioblastoma (GBM) is the most common primary brain malignancy in adults and the gradual development of treatment resistance prevents patients from achieving remission. A primary objective in the field is to devise strategies to overcome this. We predicted this could be achieved by improving existing therapies, creating better models to study GBM, and by identifying a potential therapeutic target. Fractionated radiotherapy (RT) has been used in GBM standard of care for over a century but does not account for GBM heterogeneity. We first hypothesized that in silico mathematical modeling would be a useful tool for cytotoxicity prediction. While this model …

ABSTRACT As scientific understanding of environmental determinants of health advances, growing attention has turned to the role of toxic metal exposures in exacerbating neurodevelopmental outcomes, particularly in vulnerable populations such as children with autism spectrum disorder (ASD). This dissertation investigates the intersection of environmental metal exposure, biomonitoring innovation, and social factors shaping ASD severity in Alabama. Using a multidisciplinary framework, four interrelated studies explore biological, environmental, and participatory dimensions of ASD research and public health. The first study evaluates the utility of Volumetric Absorptive Microsampling (VAMS) for measuring exposure to cadmium (Cd), chromium (Cr), lead (Pb), manganese (Mn), and mercury …
Sex Differences In The Development Of T-Bet+ B Cells In Lupus, Kathryn A. Sullivan
Sex Differences In The Development Of T-Bet+ B Cells In Lupus, Kathryn A. Sullivan
All ETDs from UAB
Cardinal features of lupus include elevated B cell activation and autoantibody production with a female sex preponderance. Development of T-bet+ B cells, driven by aberrant TLR7 and interferon (IFN) signaling, is a key pathogenic feature of systemic lupus erythematosus (SLE). In the present study, we developed two novel approaches to elucidate the mechanisms of female sex predisposition to lupus. We (i) quantified interactions of sex and genetic variation on the development of autoimmune B-cell phenotypes and autoantibodies in the BXD2 murine model of lupus using a cohort of backcrossed progeny (BXD2 x C57BL/6J) x BXD2; (ii) used a combination approach …
Parsing Clinical And Neurobiological Heterogeneity In First-Episode Psychosis Patients With And Without Features Of The Deficit Syndrome Of Schizophrenia, Matheus Teles Gomes De Araujo
Parsing Clinical And Neurobiological Heterogeneity In First-Episode Psychosis Patients With And Without Features Of The Deficit Syndrome Of Schizophrenia, Matheus Teles Gomes De Araujo
All ETDs from UAB
Schizophrenia is highly heterogeneous in its neurobiological and clinical presentations, which hinders attempts to understand its pathophysiology, limits efforts to identify biomarkers, and deters optimal patient care. A way to parse clinical heterogeneity is to subtype patients based on clinical presentations. The deficit syndrome (DS) is a possible subtype, characterized by primary and enduring negative symptoms. Evidence for neurobiological abnormalities dissociating DS from other forms of schizophrenia is inconsistent as most studies do not account for heterogeneity within patient groups due to limitations of group-level comparisons. The question remains whether DS presents a unique and less heterogeneous neurobiological phenotype. The …

The Prize Biter Of Modernity? Investigating Killer Whale Bite Mechanics, Daniel Hong
West Chester University Graduate Theses, Dissertations, and Final Projects
Although the killer whale (Orcinus orca) is a cosmopolitan apex predator, very little work has been done on its feeding biomechanics. Here I use finite element analysis to investigate the mandibular stiffness of a juvenile male Bigg’s killer whale, and compare it to that of the false killer whale (Pseudorca crassidens) and the saltwater crocodile (Crocodylus porosus). Full mandibles and left and right halves of mandibles are simulated under FEA. I also estimate the total jaw muscle force produced by the Bigg’s killer whale using the dry skull method. The killer whale lacks an …

Avoiding Racial Equity Detours: Racial Equity Trainers’ Visions Of Racially Equitable Residential Environmental Education, Anna Mooney Ph.D.
Avoiding Racial Equity Detours: Racial Equity Trainers’ Visions Of Racially Equitable Residential Environmental Education, Anna Mooney Ph.D.
Antioch University Dissertations & Theses
This study investigated how racial equity trainers envision racially equitable residential environmental education (REE). While addressing racial inequities has been central to environmental scholarship, equity detours undergirded by systemic power structures continue to pose significant barriers in addressing inequities in practice. The research explored the question: “How do racial equity trainers envision racially equitable residential environmental education?” Using constructivist grounded theory and drawing on portraiture’s “search for goodness,” the study conducted 1-hour virtual interviews with six racial equity trainers, employing iterative coding through the constant comparative method. A 2-hour virtual focus group and subsequent 30-minute “dissertation reality check” provided additional …

A Pedagogy Of Self: A Critical Autoethnography Evaluating The Self And The Lessons Of Exposure, Experience, Action, And Inaction, Jeremy Grisham
Antioch University Dissertations & Theses
This dissertation is a critical autoethnography, which evaluates a lifetime of experience through the lens of an educational theory in critical pedagogy, and two psychological theories in liberation psychology and feminist therapy. These theoretical frameworks will help me evaluate how the lessons of my life have acted in both oppressive ways, and ways in which I have supported and benefited from systemic oppression. This research demonstrates that the existence of my own diagnosed psychopathologies, which have provided symptomatic pressures, have ultimately stemmed from systemic pressures, even those systems of which I have inherently benefited. Through this research, I conclude that …

The researchers aim to explore the effects of gender (male and female) and socioeconomic status (low, moderate & high) on resilience and further investigate whether the effect of gender is independent of different levels of SES. The quantitative survey study included 490 first-year undergraduates (M = 18.97, SD = .83) from three districts of West Bengal. The questionnaire comprised socio-demographic data and the adapted Bengali version of the Connor-Davidson Resilience Scale (CD-RISC). The …
Cultivating Counter-Stories To Deficit Narrative Of Students: A Literacy Problem Of Practice, Amanda Brown
Cultivating Counter-Stories To Deficit Narrative Of Students: A Literacy Problem Of Practice, Amanda Brown
Theses and Dissertations
In urban schools across the United States, literacy outcomes remain inequitable, especially for BIPOC students and those living in poverty. In District 17, the mandate to rigidly implement a purchased curriculum resource reinforces deficit-oriented views of student ability and limits teachers’ professional agency to adjust instruction based on formative data. As a result, instruction becomes disconnected from student needs and identities, further exacerbating literacy inequities.
Guided by Mintrop’s cycle of inquiry, this qualitative study examined how participation in a 90-day inquiry cycle influenced the narratives teachers constructed about their students’ literacy abilities. Kindergarten through third grade teachers collaboratively planned and …
Raft-Based Polymer And Nanoparticle Materials For Traumatic Brain Injury Treatment And Diagnostics, Aaron Priester
Raft-Based Polymer And Nanoparticle Materials For Traumatic Brain Injury Treatment And Diagnostics, Aaron Priester
Doctoral Dissertations
"Traumatic brain injury (TBI) is a leading cause of death and disability worldwide. Neurodegenerative diseases that develop post-TBI can be, in part, attributed to DNA and cell-damaging reactive oxygen species (ROS) and lipid peroxidation products (LPOx). This thesis focused on overcoming the limits of current TBI material treatment approaches by employing a RAFT (reversible-addition fragmentation chain transfer) polymer approach that incorporated novel therapeutic, diagnostic and peptide-targeting monomers. An improved nanoparticle synthesis approach was also developed. Thiol and thioether-containing monomers neutralize both ROS and LPOx while Gd-containing monomers with enhanced magnetic resonance imaging (MRI) contrast provide diagnostics and material tracking in …
Metal-Free Photoredox Catalysis For The S-Trifluoromethylation Of Thiols And Investigation Of Ph-Dependent Decontamination Of Dimethyl Chlorophosphate, A Nerve Agent Simulant, Via Hydrolysis With Azoles, Raheemat Rafiu
Doctoral Dissertations
"Organofluorine chemistry plays a critical role in synthetic chemistry due to the unique properties of fluorine. However, the S-trifluoromethylation of thiols remains underexplored, with existing methods often relying on toxic and expensive organometallic catalysts. Given the advantages of S-trifluoromethylated compounds, including enhanced pharmacological activity and improved metabolic stability, we developed a green and cost-effective method for the synthesis of S-CF3 compounds using diacetyl as the organic catalyst and Langloi's reagent as the trifluoromethylating reagent. This reaction, which proceeds under mild conditions using visible light photoredox catalysis, works across various substrate scopes, including aromatic, heteroaromatic, and aliphatic thiols.

Applications Of Tetramethyl-Guanidine Supported Cationic Transition Metal Sites Towards C─N Bond Construction Methodologies And [3+2] Cycloaddition Reactions, Suraj Kumar Sahoo
Doctoral Dissertations
"We present a family of divalent cationic bipodal and tripodal M(II) reagents (M = Mn, Fe, Co) supported by superbasic tetramethyl-guanidinyl (TMG) residue framework with applications towards C─N bond construction methodologies. These cationic reagents possess unique characteristics since their enhanced Lewis acidity facilitates in situ elaboration of the primary nitrene-transfer product (aziridine). Indeed, further aziridine-ring opening and cycloaddition of several dipolarophiles (nitriles, alkenes, ketones) permits the three-component synthesis of valuable five-membered N-heterocycles, commonly found in pharmaceuticals and agrochemicals.
We undertake a more systematic study to unravel parameters that make certain divalent metal sites more suitable as catalysts for the in-situ …

Use Of Light Traps To Assess The Taxonomy And Seasonality Of Early Life Stages Of Fishes Recruiting To Upper Matagorda Bay In Texas, Polly Hajovsky, Joel Anderson, Damon Williford
Gulf and Caribbean Research
Detecting the ingress of larval and early juvenile fishes into estuaries is crucial for understanding the early life stages and population dynamics of species—specific recruitment seasonality. However, monitoring these early life stages presents challenges due to their small size, seasonal variability, spatial patchiness, and their use of structurally complex habitats. Although plankton nets have been widely used to sample larval fishes, light traps have been found to have more flexibility, durability, and cost—effective. This study aimed to assess the effectiveness of light traps for sampling larval and early juvenile fishes in a Texas estuary. The objectives of this study were …
