In the early 2000s, fish kills of different species, including smallmouth bass Micropterus dolomieu, were reported in areas of the Potomac River drainage, including the Shenandoah River in Virginia. Since the first reports of fish kills and lesions affecting smallmouth bass, they have continued to show signs of adverse health effects. Additionally, in some areas of the Potomac River drainage, population declines have occurred, and lethal sampling is discouraged. Severe acute respiratory syndrome corona virus 2 (SARS-CoV-2) enters human respiratory cells through binding of its spike (S) protein to the ACE-2 receptor, which is highly expressed in lung cells, but is also found in over 150 other cell types encompassing major organs including GI enterocytes. SARS-CoV-2 causes gut dysbiosis in infected individuals and patients with COVID-19 and also coincides with lower alpha diversity of the individual’s gut microbiome. Certain gut microbiome-associated genera are lower in abundance in COVID-19 patients including Lachnospira, Prevetolla, Roseburia, Bifidobacterium, and Faecalibacterium while genera such as Bacteroides, Enterococcus and Proteobacteria are increased during a SARS-CoV-2 …

Force feedback is a type of robotic control where forces are measured, fed back into the control system, and actuation is adjusted to compensate for environmental forces, with the goal of quick and precise limb positioning in walking. Robots of today use single force sensors (e.g. pressure sensitive pads, load cells) per leg which are often bulky, expensive, and susceptible to crosstalk. Walking robots often employ model predictive controllers or policy controllers trained through reinforcement learning (RL) to actuate their limbs, but these methods are time and energy consuming. This dissertation questions how human and non-human actors share power. By exploring Confederate logistics within the Subsistence Department, the collaboration between bureaucrats and the material world is revealed. Contributing to scholarship aimed at expanding the field of Civil War studies, this dissertation not only provides a more complex and interesting view of the conflict, but also an understanding that human beings are not solely responsible for the war’s results. The first month of life is a critical and vulnerable developmental stage, characterized by a heightened risk of life-threatening infections. Each year, 1.3 million newborns succumb to neonatal bacterial sepsis. The immune system during the neonatal period has unique features that lead to inefficiencies at clearing infection. Early life immunity is characterized by a Th2-biased profile with decreased IL‑12, IFN-γ, and TNF-α, along with increased IL‑6 and IL‑23. Reactive oxygen species play a crucial role in many cellular processes. Despite being a natural byproduct of cellular metabolism, overexpression of these species causes damage to the cell, leading to cell dysfunction. Central to the regulation of these highly reactive molecule is the mitochondria. Impairment in its function has been associated with oxidative stress within the cell. Herein, a ligand-based and structure-based approach to design new small molecules to inhibit mitochondrial redox proteins associated with oxidative stress and reactive oxygen species. Primary cilia are highly specialized organelles that regulate signaling, trafficking, and sensory perception across multiple tissues, including the retina. The small GTPase ADP Ribosylation Factor-Like Protein 13B (ARL13B) is an atypical ARF-family protein enriched in cilia. ARL13B acts as a guanine exchange factor (GEF) for another small GTPase ARF-like Protein 3 (ARL3). Mutations in ARL13B cause Joubert syndrome, a ciliopathy characterized by brain malformation, developmental delays, and retinal degeneration. Despite ARL13B’s known ciliary localization and clinical relevance, ARL13B’s precise functions in cilia remain poorly defined. Sleep plays a crucial role in memory consolidation, neuronal plasticity, and cognitive function. Individuals with autism spectrum disorder (ASD) often experience sleep disturbances, which are positively correlated with symptom severity. However, on the surface, sleep architecture is remarkably normal under baseline conditions in many rodent models of ASD. The goals if this dissertation were to investigate whether regulation of sleep, biochemical signaling during sleep, and downstream consequences of sleep on behavior are altered in two mouse models of ASD. Weakly reversible, deficiency zero (WR0) systems form a large class of polynomial ODEs modeling chemical reaction networks. Their behavior is exceptionally stable: they have unique positive steady states, which are locally asymptotically stable (they are also conjectured to be globally asymptotically stable). This powerful result (the Deficiency Zero Theorem) applies to a large class of high dimensional, nonlinear polynomial dynamics, and is independent of the choice of parameters in the model. Per- and polyfluoroalkyl substances (PFAS) are a group of over 15,000 synthetic compounds widely used for their resistance to heat, stains, grease, and water. While central to many modern conveniences, ingestion of PFAS is increasingly linked to adverse human health outcomes and are now detected globally in drinking water systems. This dissertation investigates three interrelated aspects of PFAS contamination in U.S. drinking water: (1) socioeconomic determinants and spatial patterns of contamination; (2) the economic impacts of contamination on residential property values; and (3) the effect of inconsistent PFAS regulations on public trust in environmental governance.

The work necessary for land managers to survey for and treat invasive species takes up limited time, labor, and financial resources. The emergence of antimicrobial resistance (AMR) has quickly become one of the largest global health and development threats. Research related to environmental modes of transmission of AMR has accelerated in recent years, and the US Environmental Protection Agency is particularly interested in this emerging contaminant. However, several critical factors, including precision drug delivery at the site of interest, long-term storage stability, and controlling release profiles of drugs in the systemic circulation, need to be addressed for successful clinical translation of nanomedicine. The rise of antibiotic-resistant microbes and concerns over non-biodegradable waste highlight the need for innovative materials with integrated functionalities that address these critical issues. In this study, we synthesized poly(3hydroxybutyrate) (PHB) electrospun nanofibers blended with gelatin (Ge) and loaded with photoactive AgTiO2 nanoparticles with improved mechanical and biological properties. Biological tests revealed excellent antibacterial activity of the prepared membrane, with efficiency exceeding 99% against Escherichia coli and 95% against Staphylococcus epidermidis after 90 and 60 min of exposure to low-power commercial LED lights, respectively. Interoceptive sensibility involves the detection, integration, and interpretation of internal bodily signals, primarily from cardiovascular, pulmonary, or gastrointestinal systems. Dysfunction in interoceptive processing has been increasingly recognized as a clinically relevant factor in numerous neurological, psychiatric, and behavioral disorders. This study aimed to elucidate the relationship between interoceptive sensibility and distress tolerance, hypothesizing that individuals with heightened interoceptive sensibility may report poorer distress tolerance. This relationship could help identify patterns between interoceptive sensibility and tendencies toward maladaptive coping strategies or emotional dysregulation. Humans prefer kind romantic partners. Worldwide surveys have identified kindness as the defining feature of an ideal long-term partner, often explained as a signal for evolutionary fitness, demonstrating the ability to provide resources and ensure survival. Instead, we propose an alternative explanation: human preference for prosociality stems from an innate desire to be treated well. To test this, we asked participants whether they preferred kindness directed towards them or kindness exhibited more generally. Results indicate that participants were significantly more attracted to people who were kind to them personally than to people who were broadly kind to others. Interestingly, participants …

Background/Purpose:
Hemophilic arthropathy (HA) is a progressive joint disease caused by recurrent hemarthroses in patients with hemophilia. Early radiographic identification of HA is critical for timely intervention and long-term joint preservation. This study aimed to develop and evaluate deep learning models for automated detection of HA in knee radiographs.
Methods:
A retrospective dataset of 849 knee radiographs (AP and lateral views) was used to train three convolutional neural networks: ResNet-34, DenseNet-121, and EfficientNet-B0. Model performance was assessed using standard classification metrics, including accuracy, precision, recall, F1-score, and AUC. Interpretability was evaluated using Grad-CAM heatmaps.

Individuals with diabetes mellitus frequently develop severe skin and soft tissue infections (SSTIs) that are recalcitrant to antibiotic treatment. We examined how diabetes affects the emergence of antibiotic resistance in a Staphylococcus aureus SSTI. We determined that S. aureus evolves antibiotic resistance rapidly in diabetic mice, while resistance did not occur in nondiabetic mice over the course of infection. Diabetes-associated immune cell dysfunction plays a minor role in the emergence of resistance, while hyperglycemia plays a dominant role facilitating the expansion and takeover of resistant mutants in diabetic infections. In the U.S., following colonization and agricultural expansion, giant cane (Arundinaria gigantea) has been reduced to approximately 2% of its original extent, potentially causing shifts in the abundance and distribution of over 70 associated animal species. Among those species is the Swainson’s Warbler (Limnothlypis swainsonii), which has been known to occupy canebrakes during the breeding season. This study utilizes anonymous survey data collected from 26 mental health professionals in various regions of Canada to understand which psychological disturbances are the most prevalent among victims of violent crime as well as which psychological interventions are most beneficial for treating violent crime victims. The data revealed that Post-Traumatic Stress Disorder (PTSD), Major Depressive Disorder (MDD), Generalized Anxiety Disorder (GAD), anger, guilt, self-blame, and shame are each highly prevalent. Dams have been constructed throughout the world and provide several resources for humans (e.g., flood control, hydropower). These structures heavily alter the upstream and downstream environment. One consequence of these alterations is the creation of “tailwaters”, which can create cold-water habitat via hypolimnetic releases. In the southeast United States these tailwater environments are often managed for the recreational use of nonnative salmonids. The unnatural flow regimes of tailwaters affect the biotic and abiotic conditions and ultimately the dynamic rate functions (i.e., recruitment, growth, mortality) of the fish inhabiting them. Hypochlorous acid (HOCl), a naturally occurring compound in neutrophils, has been used as an antimicrobial in wound treatment. Osteoblasts, bone-forming cells, within the bone tissue play a large role in bone fracture healing. Orthopedic infections are challenging to treat and highly morbid, including surgical site infection of arthroplasty procedures or infection in the setting of compound fractures. HOCl is being considered as a surgical antiseptic to decrease infections in orthopedic surgical procedures. This study therefore seeks to determine the effects of HOCl on osteoblasts and E. coli. Characterization Of The Overexpression Of Reca Homologs Rad51 And Dmc1 In Tetrahymena Thermophila, Jianna M. Cox

RecA homologs, Dmc1 and Rad51, work to repair DNA double-strand breaks (DSBs) within the cell through the recombination of homologous sections of DNA. Dmc1 works to repair programmed DSBs through meiotic recombination, while Rad51 functions to repair both meiotic and non-meiotic DSBs, the latter repaired through the process of homologous recombination repair (HHR). Chemotherapeutics, exogenous agents, work to form DSBs in cancer cells, attempting to inhibit the cell’s growth. A hyper recombinant phenotype is often seen in cancer cells due to the overexpression of RAD51, leading to drug resistance, the persistence of cancers, and an overall poor patient outcome. …

The Northern Giant Musk Turtle (Staurotypus triporcatus), a near-threatened species native to southern Mexico, Guatemala, Belize, and western Honduras, faces multiple environmental challenges, yet its activity patterns, movement patterns, and reproductive biology remain poorly understood. This thesis investigates both seasonal activity and reproductive cycles of female S. triporcatus in Belize. To assess diel and seasonal activity, I tracked 12 female turtles and collected data using activity loggers. I also collected movement data through radio-telemetry and acoustic tracking methods. Results indicated that the species is primarily nocturnal, with increased activity during the wet season, particularly during flooding events. Turtles …

This thesis examines whether cyber warfare now poses a more immediate threat to U.S. national security than weapons of mass destruction (WMD). Cyber operations have become a dominant instrument of contemporary conflict, with malicious actors operating in a persistent “grey zone” that blurs traditional boundaries between war and peace. These operations target military and civilian entities, both directly and often as collateral damage due to the uncontrollable nature of cyber threats. WMDs, despite their catastrophic destructive potential, remain largely constrained by established deterrence frameworks.

Spring-fed aquatic ecosystems in the Ozark Highlands are characterized by stable water depths, clear water, and diverse aquatic vegetation (filamentous algae, submerged, floating, and emergent plants). Sudden changes in water depth can impact aquatic vegetation dynamics but the impacts of prolonged changes are poorly understood. I investigated seasonal and annual changes to aquatic vegetation abundance in a spring impoundment during and after a two-year period of low water depth that was caused by a broken outlet below an artificial dam. Water depth was significantly lower in 2020 and 2021 (low water years), compared to 2022 and 2023 (high water years). …

Introduction: Lupus nephritis (LN) leads to end stage renal disease (ESRD), and early diagnosis and disease monitoring of LN could significantly reduce the risk. however, there is not such a system clinically. In this study we aim to develop a biomarker-panel based point-of-care system for LN.

The Asian tiger mosquito, Aedes albopictus, is a significant vector for diseases such as Dengue, Chikungunya, and Zika. Its increasing resistance to insecticides poses a critical challenge for vector control efforts. This study investigates knockdown resistance (kdr) mutations in the voltage-gated sodium channel (VGSC) genes, specifically V1016G and F1534C, which are associated with pyrethroid resistance.
