Toward Measurable Explainable Ethical Ai: An Llm-Driven Data Analytics Study, Greg Bacon Jan 2025

Toward Measurable Explainable Ethical Ai: An Llm-Driven Data Analytics Study, Greg Bacon

Dissertations

Large Language Models (LLMs) have been a hot area for popular and research audiences. Users marvel subjectively at Artificial Intelligence (AI) outputs — so much that OpenAI delayed public release of Generative Pre-trained Transformer 2 (GPT-2) back in 2019, citing ``safety and security concerns.'' This research advances ethical and transparent AI through qualitative and quantitative measures between user intent and LLM responses in a way that aims to assist users with clear indications of good and poor alignment and of where to backtrack to minimize bias and hallucinations. The use case is recommending grant topics in a United States Department …

Novel Ai/Ml-Based Frameworks For Protein Conformation Selection In Drug Discovery Applications, Shivangi Gupta Jan 2025

Novel Ai/Ml-Based Frameworks For Protein Conformation Selection In Drug Discovery Applications, Shivangi Gupta

Dissertations

Drug development is a lengthy, expensive process with a high failure rate. The urgency brought on by the COVID-19 pandemic has accelerated the integration of artificial intelligence (AI) and machine learning (ML) to enhance drug discovery by increasing target specificity, reducing toxicity, and optimizing formulation strategies. Building on this momentum, this dissertation introduces novel AI/ML-driven frameworks for protein conformation selection and classification, addressing critical challenges in modern drug discovery. However, designing such a novel AI/ML data-driven framework is a difficult task because most real-world biomedical datasets suffer from class imbalance issues, which can significantly skew AI/ML model training, resulting in …

Manganism And The Development Of A Green Sustainable Low-Threshold Spectrophotometric Test For The Monitoring Of Manganese In Consumable Water, Anna Merritt Holmes Jan 2025

Manganism And The Development Of A Green Sustainable Low-Threshold Spectrophotometric Test For The Monitoring Of Manganese In Consumable Water, Anna Merritt Holmes

Dissertations

Manganism, a neurotoxic disease from the hyperaccumulation of manganese, follows an etiology that parallels Parkinsonism. Non-occupational Manganism is acquired by the lifelong accumulation of manganese through water contaminated by natural and industrial sources. Biologically active manganese (II) eludes flocculation and filtration removal methods at water utilities. The U.S. Standard Methods For the Examination of Water and Wastewater persulfate testing method uses mercury sulfate, concentrated nitric and phosphoric acids, silver nitrate, and ammonium persulfate. The low detection limit of 210 µg/L is an order of magnitude greater than the World Health Organization (WHO) allowable limit of 10-50 µg/L. In 2004, the …

In Vitro Characterization And Evolution Of Sylvatic And Endemic Dengue-4 Viruses, Andrew Peric Jan 2025

In Vitro Characterization And Evolution Of Sylvatic And Endemic Dengue-4 Viruses, Andrew Peric

Yale Graduate School of Arts and Sciences Dissertations

Dengue virus is the world’s preeminent arthropod-borne virus that infects hundreds of millions of people each year, and circulates in four distinct serotypes named 1, 2, 3, and 4. Dengue virus cycles between mosquito and human hosts in a transmission cycle called the endemic cycle, which is responsible for the majority of recorded dengue infections worldwide. However, dengue also exists in a second transmission cycle called the sylvatic cycle, which mirrors the endemic cycle with wildlife, including non-human primates, replacing humans in this transmission cycle. Phylogenetic analyses support the hypothesis that spillover events from the sylvatic cycle into human populations …

Unionid mussel populations in a section of the Clinch River in Virginia, USA, has declined substantially, but the causes of the decline remain unknown. To investigate this zone of decline (ZOD), we deployed juvenile freshwater mussels (Villosa iris in 2012 and Lampsilis fasciola in 2013) in both cages and silos at sites within the Clinch River System. We analyzed mussel tissues for trace element and organic contaminant concentrations, shells for trace elements, and environmental media (total water, dissolved water, particulate sediment, and bedload sediment) for both inorganic and organic contaminants. We found a few differences between mussels deployed in cages …

Disposal Of Nanoplastics By The Brain: Plastic Persistence And Upregulation Of A Putative Plastic-Degrading Enzyme After Exposure To Polystyrene-Infused Food, Callie C. Mauersberg Jan 2025

Disposal Of Nanoplastics By The Brain: Plastic Persistence And Upregulation Of A Putative Plastic-Degrading Enzyme After Exposure To Polystyrene-Infused Food, Callie C. Mauersberg

College of Graduate Studies: Theses & Dissertations

Plastic is a ubiquitous material used in many domestic and commercial applications. Widespread use of plastic has led to environmental accumulation of plastic waste, and microplastics have inundated human food and water sources. Once consumed, it is well established that microplastics translocate from the gut to other tissues, including the brain, via the circulatory system. However, it is unknown if nanoplastics persist permanently in the brain. Additionally, it is not known if microplastics can be enzymatically degraded by phagocytic cells in the CNS, such as microglia and astrocytes. Alkane monooxygenase-homolog 7 (Alkbh7) is a mitochondrial enzyme that may …

Organizations play significant roles in society by providing goods and services and contributing to the economies of communities, globally. Amid the predicted increase in the frequency and impact of disasters, which will invariably lead to higher disaster costs, there is an urgent need to ameliorate disaster impact on society and organizations. Disaster mitigation and preparedness are strategies for stemming disaster impact and losses within organizations. Cognizant of the importance of disaster mitigation and preparedness, researchers have conducted a myriad of studies on both concepts at the organizational level. To advance knowledge on this topic, this study conducted a comprehensive systematic …

Development And Application Of Gpu Based Monte Carlo Simulation Techniques In Radiation Medicine, Satzhan Sitmukhambetov Jan 2025

Development And Application Of Gpu Based Monte Carlo Simulation Techniques In Radiation Medicine, Satzhan Sitmukhambetov

Physics Dissertations - Archive

Cancer remains a significant public health challenge, with treatments like radiation therapy forming a cornerstone of modern care. To address this challenge, advanced computer simulations can improve radiotherapy in two primary ways. From one hand, mechanistic simulations help elucidate the fundamental radiobiological working principles, which could contribute to the clinical advancements from a bottom-up framework. On the other hand, modality simulations could help design better clinical systems with better detection and therapeutic capabilities. This research enhances these critical simulation tools by introducing two significant developments.

First, this work introduces a metaphase DNA model into a microscopic Monte Carlo simulation framework …

On The Orbital Dynamics Of Exoplanets, Exomoons, And Submoons, Shaan Dharmesh Patel Jan 2025

On The Orbital Dynamics Of Exoplanets, Exomoons, And Submoons, Shaan Dharmesh Patel

Physics Dissertations - Archive

Although exoplanet detection has long been a central focus in astronomy, growing interest in exomoons has highlighted the increasing need for theoretical frameworks to interpret emerging candidates. Furthermore, while submoons, natural satellites orbiting moons, are not presently observed in the Solar System and remain speculative (e.g., the hypothesized past submoon of Saturn’s Iapetus), their existence is plausible within the context of the complex and diverse architectures now identified across planetary and stellar systems. We develop a generalized framework to assess the orbital stability of exomoons and submoons through comprehensive N-body simulations incorporating both three-body dynamics and tidal interactions, with the …

West From West Point: The U.S. Military Academy And The Romantic Frontier, Ryan Shaw Jan 2025

West From West Point: The U.S. Military Academy And The Romantic Frontier, Ryan Shaw

Yale Graduate School of Arts and Sciences Dissertations

This dissertation examines the influence of Romanticism on the culture and curriculum of the United States Military Academy in the early nineteenth century, and demonstrates how it shaped the behavior of U.S. Army officers on the far western frontier. While historians have long associated Romanticism with the frontier experience, it has traditionally been framed as an output, rather than an input. This study identifies West Point as an unexpected but significant source of Romantic ideals.

The history of the Academy is presented uniquely here as a contest between three competing visions: A republican ideal of a modest military …

Dusp8 Gene Expression Is Increased Upon The Stimulation Of Hek293 Cells, Oanh Vo, Lukas Macha Jan 2025

Dusp8 Gene Expression Is Increased Upon The Stimulation Of Hek293 Cells, Oanh Vo, Lukas Macha

Student Summer Scholars Manuscripts

Dual specificity phosphatase 8 (DUSP8) is one of the phosphatases that oppose the activity of mitogen-activated protein kinases. Previous mouse studies have demonstrated the functional role of DUSP8 in regulating cardiomyocyte growth. However, it is still unclear how the activity of DUSP8 is regulated, and little is known about the regulation of DUSP8 at the gene level. The goal of this study is to determine whether the DUSP8 gene is immediately transcribed upon MAPK activation in the cells. Here, we found that the expression of the DUSP8 gene was significantly increased after stimulation of cells for 30, 60, 90, and …

Dna Methylation Variation In Eastern Diamondback Rattlesnakes (Crotalus Adamanteus) On A Georgia Barrier Island, Megan Hoog Jan 2025

Dna Methylation Variation In Eastern Diamondback Rattlesnakes (Crotalus Adamanteus) On A Georgia Barrier Island, Megan Hoog

College of Graduate Studies: Theses & Dissertations

The Eastern Diamondback Rattlesnake (Crotalus adamanteus, EDB) is the largest rattlesnake in the world and is thought to be declining in many portions of its range. Genetic venom studies in the Southeast United States have shown evidence of fine-scale differences within two main geographic groups on Jekyll Island. My objective is to expand on this finding to determine if epigenetic mechanisms, specifically DNA methylation, varies among different populations of EDB in similar environments, age classes, and sexes. I collected EDB blood samples through a partnership with the Jekyll Island Authority and screened 44 individuals for epigenetic variation from …

Effects Of Perinatal Exposure To Lipopolysaccharide And Acetaminophen On Learning In Adult Long-Evans Rats, Lucy A. Sivils Jan 2025

Effects Of Perinatal Exposure To Lipopolysaccharide And Acetaminophen On Learning In Adult Long-Evans Rats, Lucy A. Sivils

College of Graduate Studies: Theses & Dissertations

The present study investigated the individual and combined effects of elevated perinatal inflammation, oxidative stress caused by acetaminophen (APAP) exposure, or elevated inflammation and oxidative stress on adult learning behaviors in Long-Evans rats. While prior studies have examined these factors independently, the combined effects of inflammation and oxidative stress via APAP exposure during early development on learning-related behaviors remain unexplored. The present research aimed to address this gap by the analysis of potential long-term neurodevelopmental consequences of lipopolysaccharide (LPS)-induced inflammation and APAP-induced oxidative stress. LPS, APAP, or LPS + APAP was administered during the perinatal period: postnatal days (PD) 9, …

Sex-Specific Regulation Of Mash And Hepatocellular Carcinoma: A Role Of Sphk2 In Females, Bria Pierre Jan 2025

Sex-Specific Regulation Of Mash And Hepatocellular Carcinoma: A Role Of Sphk2 In Females, Bria Pierre

Theses and Dissertations

Metabolic Dysfunction-Associated Fatty Liver Disease (MAFLD) is characterized by excessive fat accumulation in the liver, with metabolic dysfunction-associated steatohepatitis (MASH) representing a more severe form that can progress to cirrhosis or hepatocellular carcinoma (HCC). The sphingosine kinases and the S1P signaling pathway are key regulators of MASH and HCC progression. While much is known about the mechanisms contributing to MASH-HCC in males, less is known for females. Nevertheless, we previously identified sphingosine Kinase 2 (SphK2) to protect females from MASH-driven HCC. This study first performed differential gene expression analysis on human liver cohorts to examine sex-specific differences in gene expression …

The Diseased Extracellular Matrix In Chronic Obstructive Pulmonary Disease Modeling, Leigh-Ann M. Antczak Jan 2025

The Diseased Extracellular Matrix In Chronic Obstructive Pulmonary Disease Modeling, Leigh-Ann M. Antczak

Theses and Dissertations

Chronic obstructive pulmonary disease (COPD) is currently the fourth leading cause of death in the world, with an increasing prevalence as the world’s population ages. In the distal regions of the lungs, COPD is characterized by progressive emphysema due to an imbalance of proteases that continuously degrade essential extracellular matrix (ECM) proteins within the alveolar walls, causing fatal tissue impairment. This dissertation examines the unique properties of this diseased lung ECM in two parts: (1) As a key constituent in inflammatory signaling that leads to increased inflammation and progression of COPD, and (2) as a tool in three-dimensional (3D) in …

Breaking Ground: The Impact Of Construction On Nutrient Loads In An Urban Stream, Brody N. Wilson, Shannon Speir, Alana G. Strauss, Caroline G.T. Anscombe Jan 2025

Breaking Ground: The Impact Of Construction On Nutrient Loads In An Urban Stream, Brody N. Wilson, Shannon Speir, Alana G. Strauss, Caroline G.T. Anscombe

2025 Research Poster Competition

Degradation of water quality is primarily driven by human activities, such as urbanization. Urban Stream Syndrome (USS) can be characterized by the negative physical, chemical, and biological changes to water quality as a result of urbanization. One key effect of USS is the input of excess nutrients, which can drive unwanted macrophyte and algal growth. New construction is prevalent in urbanizing areas, resulting in the influx of nutrients, sediments, and other materials into stream ecosystems. However, few studies have examined the impacts of active construction on water quality. As such, we do not understand how construction affects nutrient transport downstream. …

Intercellular Stress Generation During 1d Collective Migration Of Cancer Cells, Logan Waddle, Jian Zhang Jan 2025

Intercellular Stress Generation During 1d Collective Migration Of Cancer Cells, Logan Waddle, Jian Zhang

2025 Research Poster Competition

Physical forces drive many cellular processes such as migration and proliferation. A metastatic tumor will have invasive strands/chains that extend from the main tumor. These chains of cells collectively migrate away from the main tumor to establish new colonies elsewhere in the body. However, how physical forces drive this collective invasion and the required energy for this process is not well understood. Invasion of a metastatic tumor to surrounding tissues leads to a majority of cancer-associated death and is often a collective effort among cells, it is therefore important to fully understand the process behind collective cancer invasion.

The goal …

Effect Of Ventilation And Wearing A Facemask In Reducing Indoor Aerosol Transmission, Clothilde Brochot, Ali Bahloul Jan 2025

Effect Of Ventilation And Wearing A Facemask In Reducing Indoor Aerosol Transmission, Clothilde Brochot, Ali Bahloul

Articles dans des actes de congrès

The COVID-19 pandemic has significantly disrupted research in respiratory protection and transmission routes. Indeed, the pandemic has highlighted a number of issues, including those related to the performance and use of respiratory protective equipment such as masks and ventilation. A plethora of commercial and homemade masks have widely appeared during the pandemic, although they are not yet fully regulated in performance and fit test. However, with regard to the source reduction process, testing facemasks not at the inhalation, but at the source (exhalation) offers a new perspective on how to prevent particle emissions. Different means of transmission reduction are measured …

Assessing Arbuscular Mycorrhizal Fungal Community Composition And Diversity Across A Land-Use Gradient In Eastern Washington Prairies, Katherine I. Cole Jan 2025

Assessing Arbuscular Mycorrhizal Fungal Community Composition And Diversity Across A Land-Use Gradient In Eastern Washington Prairies, Katherine I. Cole

EWU Masters Thesis Collection

Arbuscular mycorrhizal fungi (AMF) are key to healthy, functioning terrestrial ecosystems worldwide. Associating with over 80% of all extant land plants, they play a pivotal role in nutrient cycling, and contribute to overall plant health. Given their significance to terrestrial ecosystems, AMF have a crucial role to play in the successful restoration of Eastern Washington’s native prairies. However, AMF community composition is significantly altered by the commercial agricultural practices used extensively in this region for over 100 years. The Eastern Washington University (EWU) Prairie Restoration Project aims to restore ~120 acres of farmland to native prairie. This research provides insight …

Wiring Attachment, Alexander K. Bacon Jan 2025

Wiring Attachment, Alexander K. Bacon

Theses and Dissertations

This thesis began with a question: can diagrams create understanding, or only reflect it? Through paintings that layer automotive wiring schematics with family photographs, I explore how the human attachment system resists easy mapping. Using digital tools like Photoshop and scanning, alongside analog methods such as papier-collé and photo transfer, I examine how systems built for clarity—like wiring diagrams—contrast with the layered, often messy realities of human relationships. Drawing from attachment theory’s empirical foundation, rather than the metaphorical leanings of psychoanalysis, my work investigates how emotional bonds form, falter, and repair over time. Material loss and layering are not flaws, …

The Bacteriostatic, Regenerative, And Immunomodulatory Properties Of Extracellular Matrix Particles For Lung Injury, Keera P. Rhoads Jan 2025

The Bacteriostatic, Regenerative, And Immunomodulatory Properties Of Extracellular Matrix Particles For Lung Injury, Keera P. Rhoads

Theses and Dissertations

Acute respiratory distress syndrome (ARDS) is a prevalent, life-threatening lung condition, affecting nearly 200,000 Americans annually, with a 40% international mortality rate. There is no cure for ARDS, and current pharmacological treatments have limited effectiveness. Symptoms can be mitigated with mechanical ventilation, though this often leads to ventilator-induced lung injuries (VILI) and puts critically ill patients at risk of infections, including ventilator-associated pneumonia (VAP). A promising therapeutic is the extracellular matrix (ECM), a complex network of structural proteins and bioactive molecules that has been shown to have anti-inflammatory properties and prevent fibrosis. We aim to utilize the regenerative and immunomodulatory …

Ecological Interactions Of Large Mammals In Luangwa, Zambia, Jahzara Osborne, Amelia Villaseñor Jan 2025

Ecological Interactions Of Large Mammals In Luangwa, Zambia, Jahzara Osborne, Amelia Villaseñor

2025 Research Poster Competition

"The connection between mammalian behavior in national parks and human-driven environmental change is tenuous. National parks are often considered the last vestiges of natural space, yet they are created and maintained by humans. South Luangwa National Park (SLNP) within the southern East African Rift is a microcosm of this global conundrum and is also a region where stable isotope data is sparce. Understanding the mammalian diets of mammals in the SLNP via carbon and oxygen stable isotopes in teeth will provide a means of understanding variation in one aspect of mammal behavior that may be affected by changing animal-human relationships. …
