The extent of walking impairment varies among individuals with peripheral artery disease (PAD), which may reflect differences in the adaptability of lower extremity muscles to ischemia-reperfusion injury characteristic of the disease. Analyses of gastrocnemius muscle biopsies from 113 individuals with PAD [mean ankle-brachial index (ABI) = 0.65 ± 0.13, 38 (33.6%) women, 76 (67.2%) Black] showed a wide range of myofiber type distributions (9.6%-82.6% type 1 myofibers). The abundance of oxidative type 1 myofibers negatively correlated with ABI (r = −0.22, P = 0.02), a measure of PAD severity. The abundance of type 1 myofibers also negatively correlated to 2a/x …

Early diagnosis of muscle wasting in critically ill patients with acute kidney injury requiring continuous kidney replacement therapy (AKI-CKRT) may improve outcomes via timely rehabilitation and nutrition. Muscle ultrasound (MUS) has recently gained traction for assessing muscle atrophy in the intensive care unit (ICU) but requires training to achieve reproducibility. We evaluated the inter-rater reliability of MUS in patients with AKI-CKRT performed by multidisciplinary raters including nephrologists. Two blinded independent raters used portable ultrasound to acquire images of the rectus femoris (RF). Efficient interrogation of neurobiology remains bottlenecked by obtaining mature neurons. Immortalized cell lines still require lengthy differentiation periods to obtain neuron-like cells, which may not efficiently differentiate and are challenging to transfect with plasmids relative to other cell lines such as HEK-293's. To overcome challenges with limited access to cells that express mature neuronal proteins, we knocked out the RE1-silencing transcription factor (REST) from HEK-293's to create a novel neuron-like cell, which we name Neuro293. Background: Most tumors initially respond to treatment, yet refractory clones subsequently develop owing to resistance mechanisms associated with cancer cell plasticity and heterogeneity. Methods: We used a chemical biology approach to identify protein targets in cancer cells exhibiting diverse driver mutations and representing models of tumor lineage plasticity and therapy resistance. An unbiased screen of a drug library was performed against cancer cells followed by synthesis of chemical analogs of the most effective drug. The fusion of skeletal muscle stem cell (MuSC) to myofibers during hypertrophy has exclusively focused on the transfer of the MuSC nucleus, leaving the fate of other MuSC organelles, such as mitochondria, largely unexplored. The objective of this study was to determine whether MuSCs transfer their mitochondria upon myofiber fusion in response to a hypertrophic stimulus. To achieve this goal, we specifically labeled MuSC mitochondria with Dendra2 fluorescence by crossing the MuSC-specific CreER (Pax7CreER/CreER) mouse with the Rosa26-Dendra2 mouse to generate the Pax7-Dendra2 mouse. Sex estimation of human skeletal remains is a facet of the biological profile. The pelvis has been suggested as the best source to accurately predicting the biological sex, but not always available after the recovery of human remains. At times, only long bones of human skeletal remains are recovered for identification. It has been shown that metric measurements using long can accurately predict biological sex. The most common methods use discriminant functions to predict the likelihood an individual falls within a certain population. Dominant notions of pregnancy frame gestation as a solely female process. However, the experiences of transmasculine birthers challenge this assumption, demonstrating that pregnancy is not inherently tied to womanhood but is instead shaped by social and cultural narratives and representations. This paper explores the unique pregnancy and parental exercises of transmasculine birthers by examining public social media posts under the hashtag #seahorsedad on Instagram. Utilizing metaphorical criticism, I aim to uncover how transmasculine birthers, known within the community as “seahorse dads,” both uphold and resist traditional notions of gender, pregnancy, and parenting through a combination of textual and visual metaphor. …

Fire is a major ecological force in most terrestrial systems, but the responses of animal communities to fire vary markedly across ecosystems due to variability in fire regimes and vegetation communities. Quaking aspen (Populus tremuloides) is unique within coniferous forests of western North America for its ability to regenerate following wildfire via vegetative resprouting. Enhanced loading of nitrogen (N) and phosphorus (P) is pervasive in rivers around the world. As a consequence of nutrient-rich conditions, many well-lit rivers experience severe algal blooms, which threaten water quality and aquatic habitats. Nutrient enrichment in the upper Clark Fork River, located in western Montana, has increased the productivity of benthic algae, Cladophora glomerata, which routinely forms extensive blooms during summer growing seasons (July – September). Addressing eutrophication in rivers begins with identifying the nutrients that contribute toward algal productivity. Extended fire seasons and increased fire severity in and around protected ecosystems and the wildland urban interface (WUI) will increasingly challenge fire suppression, impact communities and threaten property and resources. Addressing this growing threat through proactive management solutions that protect values at risk requires improved fire behavior prediction capabilities.
Here I explore the radiant and convective energy transfer mechanisms of fire ignition or extinction associated with fuel discontinuities. This project investigates how Indigenous Data Sovereignty protocols apply to environmental data collected on Tribal lands by federal and state entities. The 2011 and 2014 flooding of the Missouri River and a need for further soil moisture data led the United States Army Corps of Engineers Omaha District to implement a Mesonet weather station monitoring network across the Upper Missouri River Basin (UMRB). Post-mortem interval (PMI) estimation is a central component of forensic anthropological investigation, but traditional methods for estimating time since death lose accuracy after ~72 hours. Recent research indicates that some steroid hormones may undergo predictable degradation patterns after death, offering potential use as biochemical clocks for PMI estimation beyond this threshold. Dehydroepiandrosterone (DHEA), and its sulfate form (DHEA-S) are of particular interest due to their metabolic stability, systemic influence, and association with trauma. High-order harmonic generation, or HHG, is a highly nonlinear quantum optical process capable of generating X-ray/ultraviolet radiation. An atom or molecule is irradiated with a mid-IR laser at high intensity, ionizes, and then emits high-frequency light at integer multiples of the driving laser frequency when an ionized electron recollides with the molecular ion. A convenient tabletop source of coherent XUV light and a common seed for free-electron lasers, HHG can also serve as a self-probe of the source atom or molecule. Healthcare-acquired infections (HAIs) are a significant global challenge driven by biofilm-forming pathogens. Polymicrobial biofilms, involving interactions between multiple microbial species, exacerbate treatment difficulties due to their enhanced resistance to antimicrobial therapies. Borate bioactive glasses (BBGs) are an emerging class of biomaterials that have attracted significant interest in infection control. The incorporation of copper and zinc into the BBG matrix can effectively disrupt biofilm formation and bacterial colonization. The academic excellence gap, which refers to the disproportionalities of minority groups in advanced academic programs, presents challenges for educational leaders striving to provide access to equitable educational opportunities for all students. This study, guided by Charles Tilly’s opportunity hoarding, examines the systematic underrepresentation of African American students in advanced academic programs. Amyotrophic Lateral Sclerosis (ALS), also known as Lou Gehrig’s disease, is a progressive neurodegenerative disorder that leads to the degeneration of motor neurons, which control voluntary muscle movements such as eating, speaking, and breathing. As these neurons are damaged, voluntary movement ceases, leading to a fatal outcome. The survival time of ALS patients varies, ranging from a few months to up to 10 years. Preptin, a 34-amino-acid peptide derived from proinsulin-like growth factor II, has emerged as a promising therapeutic candidate for both Type 2 diabetes mellitus and osteoporosis. Its dual functionality—stimulating insulin secretion without inducing hypoglycemia and promoting osteoblast proliferation—positions it as a unique alternative to traditional sulfonylureas and osteogenic agents. In this study, we developed a purification protocol for green fluorescent-preptin fusion proteins. Compared to other high-income countries, women in the United States are more likely to die from problems related to pregnancy and childbirth. In Georgia, maternal mortality is particularly acute. Midwifery models of maternal care have been shown to produce positive pregnancy and childbirth outcomes for patients, but these services are underutilized in Georgia. This study sought to identify women’s attitudes toward midwifery, what contributed to those attitudes, and what influenced women in Middle Georgia to seek midwifery services using a qualitative, Grounded Theory approach. Nutrition plays a critical role in women’s reproductive health, yet many women face barriers to maintaining a balanced diet. This study aimed to examine the relationship between nutritional intake and reproductive health outcomes among women of reproductive age, as well as to assess their knowledge and challenges related to healthy eating. Using a cross-sectional survey design, data were collected from 50 women at a community health center and analyzed using SPSS to generate descriptive statistics and explore patterns through chi-square and regression analyses. This thesis develops and experimentally validates an online adaptive Linear Quadratic Regulator (LQR) control method for prosthetic joint systems using Recursive Least Squares (RLS)-based real-time parameter estimation on the Quanser QUBE-Servo 2 platform. Traditional LQR controllers assume a fxed system model, which limits adaptability and results in reduced tracking accuracy, poor robustness, and loss of optimal performance when applied to dynamically changing prosthetic joints, infuenced by load variations, user gait changes, and mechanical wear. Limited experimental validation exists for combining RLS with online LQR adaptation in prosthetic-like systems.

In the mid-nineteenth century, as the United States reached the Pacific through westward expansion, Asia emerged as a new stage for commerce and power. Japan, long isolated under the Tokugawa shogunate, became an object of American fascination when forced into relations. While early depictions cast Japan as an exotic yet uncivilized nation in need of Western guidance, Japan’s rapid modernization and the growing number of Japanese immigrants on the West Coast transformed curiosity into anxiety. Single-walled carbon nanotubes (SWCNTs) possess unique physicochemical and optical properties that make them ideal candidates for biomedical imaging, biosensing, and disease diagnostics. This thesis explores the potential of SWCNT-based spectral fingerprinting combined with ML (Machine Learning) algorithms to optimize bioimaging, disease detection and prediction, with a specific focus on differentiating between healthy and amyotrophic lateral sclerosis (ALS) lymphoblastic patient samples. By functionalizing SWCNTs with single-stranded DNA, we enhance their stability and target specificity, enabling their application in serum patient samples.

Attention-Deficit/Hyperactivity Disorder (ADHD) is a widespread psychological disorder in children, poorly impacting executive functioning domains of inhibitory control and working memory. Event-related potentials (ERPs) provide neuroelectric insight into these cognitive processes. However, most ERP research focuses on one task type, limiting direct comparisons of cognitive demands across different task paradigms. This study examined P3 amplitude and latency in ADHD children using a visual NoGo task (response inhibition) and an auditory oddball task (working memory), and associations between the P3 ERP and behavioral measures of executive functions.
