Expression And Purification Of Cytochrome P450 46a1 For Resonance Raman Spectroscopy, Lauren Jirles Jan 2025

Expression And Purification Of Cytochrome P450 46a1 For Resonance Raman Spectroscopy, Lauren Jirles

Williams Honors College, Honors Research Projects

Researchers are targeting cytochrome P450 46A1 (CYP46A1), an enzyme critical for brain cholesterol homeostasis, as a potential therapeutic approach for neurological diseases. CYP46A1 plays a key role in cholesterol turnover in the brain by catalyzing the conversion of cholesterol to 24S-hydroxycholesterol (24HC) and is linked to Alzheimer’s disease pathophysiology. Given its critical role in brain health, CYP46A1 represents a promising target for therapeutic intervention in neurological disorders. The goal of this project was to express the CYP46A1 protein in GC5 E. coli cells and purify the protein using chromatographic techniques including affinity and ion exchange chromatography. Ultraviolet-Visible (UV-Vis) spectroscopy was …

Effects Of Energy Drinks On Crayfish Physiology, Melyna Sweeney Jan 2025

Effects Of Energy Drinks On Crayfish Physiology, Melyna Sweeney

Williams Honors College, Honors Research Projects

Energy drinks have become the drink of choice among many college students due to the rush of energy it provides via the megadose of caffeine (Vercammen et al. 2019). Because of their newfound popularity, there is much research to be done about the effects on human physiology. Energy drinks are known to increase endurance in muscles of rat subjects (Imagawa et al. 2009) and stimulate enhanced “alertness” (Smit & Rogers, 2002). The purpose of this research was to uncover the potential effects that energy drinks have on metabolic rate and/or stress responses in crayfish (Procambarus clarkii), with the …

Elongate Locomotors Show Consistent Torque Patterns Across Substrates, Spencer Bradford, Henry Astley Jan 2025

Elongate Locomotors Show Consistent Torque Patterns Across Substrates, Spencer Bradford, Henry Astley

Williams Honors College, Honors Research Projects

The four main movements found in snakes are understood mainly due to external study; however, muscular forces and resulting torques are poorly known. The main research objective is to study the torque production on different substrates using a snakebot. The study will give a rudimentary model for torque production on different substrates. It can then be compared against biological systems to understand snake movement at a muscular level. This study used a snakebot, allowing direct assessment of the current being run through each of its motors (proportional to torque). Data was then examined using a Principal Component Analysis test for …

Astrocyte Specific Enhancers In The Cortex And Striatum Of Mice, Mikaela Kottelich Jan 2025

Astrocyte Specific Enhancers In The Cortex And Striatum Of Mice, Mikaela Kottelich

Williams Honors College, Honors Research Projects

Among glial cells, astrocytes have a prevalent influence on supporting neuronal function and overall brain health. Specifically targeting this cell type for disease treatment is an existing issue. Other cell types tend to express the desired treatment, which is not always wanted and can lead to unintended, negative consequences. Astrocytes contribute largely to maintaining the blood brain barrier, which is disrupted in Parkinson’s disease. Certain genes are thought to be the reason for this disruption, so if they can be specifically targeted in astrocytes with a specific enhancer, the disruption of the disease could be reversed. In our experiment, we …

The Impact Of Seasonal Variation On Body Morphology In Temnothorax Curvispinosus, Chelsea Kalb Jan 2025

The Impact Of Seasonal Variation On Body Morphology In Temnothorax Curvispinosus, Chelsea Kalb

Williams Honors College, Honors Research Projects

This study examines how seasonal variations affect the morphology of Temnothorax curvispinosus (acorn ants) by comparing workers hatched in the spring and in the fall. Colonies will be collected from the Bath Nature Preserve in May and September to represent the different seasonal groups. Morphological traits, such as the head, mesosoma, gaster, antennae, and leg, will be measured using a microscope and an image analysis software. The morphological measurements from each group will then be compared using ANOVA. By exploring how environmental variation influences developmental outcomes in acorn ants, this research contributes to our understanding of phenotypic plasticity and insect …

Impact Of Tetrahydrocannabinol (Thc) On Daphnia Magna Heart Rate, Ashley Cosgrave Jan 2025

Impact Of Tetrahydrocannabinol (Thc) On Daphnia Magna Heart Rate, Ashley Cosgrave

Williams Honors College, Honors Research Projects

Marijuana (Cannabis) is currently federally illegal within the United States and classified as a Schedule I drug. This leaves a large research gap on the impacts of Tetrahydrocannabinol (THC) use. Daphnia magna are frequently used as a model organism for the human heart and toxicology screenings due to their myogenic heart, similarities in physiological pathways, and sensitivity to toxins. As such, in order to bridge the research gap and uncover potential use of Daphnia magna as a model organism for humans in regard to impact on heart rate, we hypothesized that if Daphnia magna possess CB1 receptors or an equivalent …

Probiotics are live microorganisms that when administered in adequate amounts confer a health benefit on the host. Beneficial microbes are found in several sites throughout the body such as the oral cavity, skin, respiratory tract, vagina, urinary tract, breast tissue and the gut. In this research, the culture Bifidobacterium longum ITT 13, which was previously isolated from a neonatal faecal sample, was examined for its potential as a probiotic candidate. Probiotics can have numerous desirable characteristics. The pertinent ones to this research are being of human or animal origin, not prone to antibiotic resistance, demonstration of antagonistic activity towards pathogens, …

Statistical Approaches In Evolutionary Biology: Using Threespine Stickleback Fish To Test Fisher's Geometric Model And Evaluate Methods For Handling Missing Data, Aaron Mathew Myrold Jan 2025

Statistical Approaches In Evolutionary Biology: Using Threespine Stickleback Fish To Test Fisher's Geometric Model And Evaluate Methods For Handling Missing Data, Aaron Mathew Myrold

Dissertations

This thesis explores two distinct yet complementary aspects of evolutionary biology. The first chapter tests predictions from Fisher's geometric model of adaptation using a finely resolved fossil sequence of Gasterosteus doryssus spanning approximately 21,250 years. I analyze phenotypic changes in three armor traits during adaptation to a new environment and find that while step sizes follow the predicted exponential distribution—with many small steps and few large ones—they do not adhere to the model's temporal prediction that large-effect mutations should fix early in the adaptive walk. This discrepancy likely reflects adaptation from standing genetic variation rather than de novo mutation. The …

Genetic Architecture Of Complex Traits In African Ancestries Across Diverse Environments, Isadore Gregga Jan 2025

Genetic Architecture Of Complex Traits In African Ancestries Across Diverse Environments, Isadore Gregga

Dissertations

Given genetic and environmental diversity, genetic prediction based on African- ancestries individuals in the US or UK may not transfer well to other African diaspora regions. The Modeling the Epidemiological Transition Study (METS) is a longitudinal study of cardiometabolic health in five African-ancestry populations. We performed genome-wide genotyping in METS participants from the US, Jamaica, South Africa, and Ghana. We aimed to explore how genetic models for the transcriptome and body mass index (BMI) built using US- based African-ancestries populations perform in these four diverse populations. A subset of METS participants also had whole blood RNA-Seq data. For these, we …

Characterizing Niche Specialization In Staphylococcus Epidermidis, Sandra Jablonsk Jan 2025

Characterizing Niche Specialization In Staphylococcus Epidermidis, Sandra Jablonsk

Dissertations

This thesis investigates the microbial diversity of Staphylococcus epidermidis (S. epidermidis) across various anatomical sites in the human body. While traditionally viewed as a benign skin microbe, S. epidermidis also inhabits the urinary, gastrointestinal, and respiratory tracts, raising questions about its genetic variation and adaptability in different environments. The research aims to compare strains of S. epidermidis isolated from the oral cavity, nostrils, urinary tract, and skin of healthy individuals to determine if they share a common ancestor or if they evolved independently in response to specific environmental factors. The study comprises three main aims. Aim 1 focuses on assessing …

Insights On The Cranial Anatomy And Osteohistology Of Cyonosaurus, Alexander Acker Jan 2025

Insights On The Cranial Anatomy And Osteohistology Of Cyonosaurus, Alexander Acker

Dissertations

This study investigates the cranial anatomy and life history of the small-bodied gorgonopsian Cyonosaurus. Previous research on gorgonopsians has disproportionately focused on larger members of the clade, leaving smaller forms relatively understudied. This gap has led to several unresolved questions, the most pressing of which concerns gorgonopsian ontogeny. Specifically, it remains unclear whether small-bodied gorgonopsians represent genuinely diminutive adult individuals or juveniles of larger species. The specimen NHCC LB1087 presents an ideal opportunity to address this issue and gain new insights into the biology of small-bodied gorgonopsians. The first chapter focuses on cranial anatomy. Due to the specimen’s preservation, many …

Queers, Closets, And Corporate Governance, Darren Rosenblum Jan 2025

Queers, Closets, And Corporate Governance, Darren Rosenblum

Faculty Publications

The past few years have seen several corporate diversity initiatives that include Lesbian, Gay, Bisexual, Transgender, Queer (LGBTQ+) people. In 2020, Nasdaq, the world’s second-largest stock exchange, and California, the largest state, both adopted rules to encourage board diversity for underrepresented groups, including LGBTQ+ people. Since that time, the Los Angeles Superior Court struck down California’s law, the Fifth Circuit invalidated Nasdaq’s rule, and the Trump administration initiated a slew of anti-Diversity Equity and Inclusion (DEI) and anti-LGBTQ+ measures. In the context of this sharp ideological turn, this Article attempts to step back and tackle a larger question: how can …

Interactions Between Microplastics, Biofilms, And Antimicrobials In Freshwater Streams, Ryan Thomas Krantz Jan 2025

Interactions Between Microplastics, Biofilms, And Antimicrobials In Freshwater Streams, Ryan Thomas Krantz

Dissertations

Microplastics (MPs) are ubiquitous contaminants in freshwater ecosystems that could be hotspots for the interaction of antimicrobial compounds and surface-attached microbial biofilm communities. MPs and antimicrobials are both common in wastewater and urban freshwaters and MPs can adsorb contaminants like antimicrobials to their surface. Within aquatic habitats, MPs also support colonization by microbial biofilms. Therefore, we hypothesized that the adsorption of antimicrobials to MP surfaces would affect microbial communities colonizing MPs, altering their diversity, composition, and antimicrobial resistance. We investigated the effects of these interactions across two studies. The first study, discussed in Chapter 2, used a microcosm approach to …

A Dual Process Model Perspective On Associations Between Parent And Offspring Problem Drinking, Karallyn E. Fitisone Jan 2025

A Dual Process Model Perspective On Associations Between Parent And Offspring Problem Drinking, Karallyn E. Fitisone

Theses and Dissertations--Psychology

Parental problem drinking (PPD) is a well-established risk factor for adolescent alcohol use, yet the complex mechanisms involved remain underexplored. The present study addresses this critical gap by examining how PPD, parent emotion socialization, and emerging adult problem drinking are associated within the framework of the Dual Process Model of Substance Use. Participants were 2,036 undergraduates who completed an online survey assessing PPD, retrospective reports of parental reactions to their negative emotions in childhood, and students’ current levels of problem drinking, negative urgency, negative emotional reactivity, and emotion-regulation difficulties. Structural equation modeling was used to clarify the roles of emotion-related …

Exploring The Sustainability Of Single Use Plastics In The Biopharmaceuticals Sector: A Scoping Review Of Challenges, Opportunities And Impact, Cormac Mcmahon, Adrienne F. M. Fleming, Philip Owende, Malcolm Goggin Jan 2025

Exploring The Sustainability Of Single Use Plastics In The Biopharmaceuticals Sector: A Scoping Review Of Challenges, Opportunities And Impact, Cormac Mcmahon, Adrienne F. M. Fleming, Philip Owende, Malcolm Goggin

Articles

Single-use plastics (SUPs) are synonymous with the biopharmaceuticals sector, facilitating economies of scale, process efficiency, flexibility and sterility assurance, all with a seemingly negligible environmental footprint. Yet, in ever-tightening regulation, mandated by the Sustainable Development Goals (SDGs) and by concern for large-scale industrial impacts, the sustainability of SUP consumption is increasingly being questioned. Whilst the sector contributes to human welfare, its transition risk is unlikely to remain immune to societal pressure for more sustainable production. This paper aims to present a scoping review of the apparent contradiction between sectoral SUP adoption and the increasing importance of circularity. The approach to …

Language Barriers In Clinical Trials: Exploring Impacts On Continuity And Participants’ Engagement And Effective Mitigation Strategies, Harsha Ani Das Jan 2025

Language Barriers In Clinical Trials: Exploring Impacts On Continuity And Participants’ Engagement And Effective Mitigation Strategies, Harsha Ani Das

Master's Theses and Doctoral Dissertations

Clinical trials are crucial for advancing medical knowledge and developing new treatments. However, language barriers can pose significant challenges for participants, particularly in understanding trial procedures, risks, and benefits. Existing policies, guidelines, and studies from 2020 to 2024 identify key challenges in recruiting and retaining non-English-speaking participants. The lack of data on dropout rates highlights the need for improved language services and culturally sensitive communication methods. Findings demonstrate the importance of addressing linguistic disparities to enhance trial inclusivity and success. Innovative strategies to overcome language-related hurdles include the use of professional interpreters, translated materials, and community engagement initiatives. By addressing …

Neuropsychological Assessment Accuracy In Diagnosing Early-Onset Alzheimer’S Disease, Elise Thomas Jan 2025

Neuropsychological Assessment Accuracy In Diagnosing Early-Onset Alzheimer’S Disease, Elise Thomas

Master's Theses and Doctoral Dissertations

Early-onset Alzheimer’s disease (EOAD), a progressive neurological condition, is often difficult to diagnose before substantial neuronal damage. Neuropsychological assessments offer a more cost-effective and accessible method for detecting early cognitive decline, yet limited research exists on optimal combinations for EOAD identification. This study analyzed data from 24 clinical studies to determine the diagnostic accuracy of FDA-approved EOAD assessments used in combination. No single combination was found to be superior across all diagnostic metrics. MMSE + MoCA + ADAS-Cog showed the highest sensitivity, MMSE + MoCA + RBANS the highest specificity, MMSE + RBANS the highest AUC value, and MMSE + …

English Pronouns: Default Case And Conversion, Olivia Ziemelis Jan 2025

English Pronouns: Default Case And Conversion, Olivia Ziemelis

Master's Theses and Doctoral Dissertations

Case is a well-defined feature of generative syntax. However, the phenomenon of default Case, while widely accepted and discussed, is less defined in its triggers and limitations. The main body of work that seeks to explore default Case is that of Schütze (2001), which presents the main principles of default Case as well as the environments in which it may occur. The purpose of this thesis is to explore default Case at a deeper level, focusing on the interactions of Case, structure, and phi-features to determine how exactly the mechanism works. To achieve this, the data set used consists of …

Standardizing Exoneration Compensation Statutes, Farina Barth Jan 2025

Standardizing Exoneration Compensation Statutes, Farina Barth

Journal Articles

Wrongful convictions plague the American criminal justice system and leave lasting, unimaginable harm on the innocent. Since 1989 through February 2025, the National Registry of Exonerations reported 3,658 exonerations: a total of 32,750 years behind bars that were lost. An exoneree, once labeled a defendant, becomes a victim. To right this wrong, some states aim to make a wrongfully convicted citizen whole through compensation. However, in examining the evidence of wrongful conviction compensation, a pervasive issue is uncovered in the American compensation system: victims of wrongful incarceration are deeply undercompensated due to statutory drafting. For those states with compensation statutes, …

A Quality Improvement Project: Increasing Pre-Term Infant Safety By Enhancing Skin-To-Skin Time In The Nicu, Maureen A. Cassidy

Master's Theses and Capstones

Background: Skin-to-skin (STS) contact, also known as kangaroo care, is a critical intervention for preterm infants, especially for those in the neonatal intensive care unit (NICU). It promotes neurodevelopmental outcomes, physiological stability, and parental bonding. However, STS often falls below recommended daily levels due to a variety of berries. Exploring aids like belly bands may help address these barriers and promote improved outcomes.

Local Problem: STS time in this NICU microsystem falls significantly below nationally recommended levels, with most preterm infants receiving less than one to two hours daily compared to the World Health Organization’s recommendation of 8-24 hours per …
