Quantifying Inositol Phosphate Dephosphorylation To Understand The Role Of Recalcitrant Organic Phosphorus Forms On Harmful Algal Blooms In Freshwater Systems, Iffat Tasnim
College of Graduate Studies: Theses & Dissertations
Organophosphorus such as phytic acid, a surrogate species of inositol phosphate (IP), may serve as a source for orthophosphate (OP) in freshwater. In absence of OP, competent aquatic microorganisms upregulate the production of specialized enzymes to obtain growth-sustaining OP from organic phosphorus (org-P) forms. The contribution of recalcitrant org-P to the OP pool has been overlooked due to the lack of capable tools to measure OP production from org-P accurately. The objective of this study was to quantify OP production from phytic acid (a surrogate form of recalcitrant org-P) to assess the contribution of recalcitrant org-P forms to the total …

Synthesis And Assessment Of Biobased Acrylated Epoxidized Soybean Oil Resins For Stereolithography 3d Printing Of Microfluidic Devices, Natalie S. Romero Figueroa
College of Graduate Studies: Theses & Dissertations
This research focuses on the development and characterization of a biobased photocurable resin synthesized from soybean oil for use in stereolithography (SLA) 3D printing of microfluidic devices. The study explores a sustainable alternative to petroleum-based resins by performing an acrylation reaction on epoxidized soybean oil (ESO) to produce acrylated epoxidized soybean oil (AESO), which was then formulated into a photocurable resin with the addition of a photo initiator. The fabrication process involved designing microfluidic molds in SolidWorks and printing them using an Elegoo Mars 4 SLA printer under various exposure and layer thickness parameters. The printed samples consisted of microchannels …
Eros And The Sublime: Tools For Reaching Toward An Altered Ecological Consciousness, Aineka Carlson
Eros And The Sublime: Tools For Reaching Toward An Altered Ecological Consciousness, Aineka Carlson
Graduate Student Theses, Dissertations, & Professional Papers
This thesis explores connections between the Kantian sublime and Anne Carson’s ideas regarding “eros.” Since Kant’s Critique of Judgement was published in 1790, the Kantian sublime has grown in popularity and been utilized by theorists and creative writers alike. While the Kantian sublime is often associated with nature in popular culture, there are certain egocentric and phallocentric elements of this mode of thinking that might inhibit a biocentric mindset. Through a character study of Oryx from Margaret Atwood’s novel Oryx and Crake, this thesis draws relations between the subjugation of women, nature, and the ways in which the popularized …

Limited research has examined the possible synergistic interrelationships between serious bacterial infections (SBIs) of the heart (i.e., endocarditis), bone, spine, brain, or joints (e.g., osteomylelitis) and hepatitis C virus (HCV) infections. We examined whether syndemic interactions existed between SBI, HCV, and substance-use-related factors in rural communities, hypothesizing that injection-mediated risks elevated the likelihood for both SBIs and HCV infections, which could be exacerbated by synergistic biological–biological or biological and social interactions. We calculated the prevalence ratios (PRs) of past-year SBI associated with each risk factor in separate models. Effect modification among significant risk factors was assessed using multiplicative interaction. Among …

Human papillomavirus (HPV)-associated head and neck squamous cell carcinoma (HPV-positive HNSCC) has distinct biological characteristics from HPV-negative HNSCC. Using an AI-based analytical platform on meta cohorts, we profiled expression patterns of viral transcripts and HPV viral genome integration, and classified the tumor microenvironment (TME). Unsupervised clustering analysis revealed five distinct and novel TME subtypes across patients (immune-enriched, highly immune and B-cell enriched, fibrotic, immune-desert, and immune-enriched luminal). These TME subtypes were highly correlated with patient prognosis. In order to understand specific factors associated with prognosis, we used the unsupervised clustering of an HPV-positive HNSCC cohort from The Cancer Genome Atlas …

Machine learning algorithms are commonly employed for classification and interpretation of high-dimensional data. The classification task is often broken down into two separate procedures, and different methods are applied to achieve accurate results and produce interpretable outcomes. First, an effective subset of high-dimensional features must be extracted and then the selected subset will be used to train a classifier. Gradient Boosted Trees (GBT) is an ensemble model and, particularly due to their robustness, ability to model complex nonlinear interactions, and feature interpretability, they are well suited for complex applications. XGBoost (eXtreme Gradient Boosting) is a high-performance implementation of GBT that …

Examining The Effects Of Songbird Nest Boxes And Habitat On Avian And Arthropod Communities In A Vineyard Ecosystem, Eleanor Macdonald
Cal Poly Humboldt theses and projects
Agricultural expansion threatens biodiversity, but integrating insectivorous birds into vineyard ecosystems may support both conservation and ecosystem services. We tested how nest boxes and habitat composition influence avian and arthropod communities in Napa Valley vineyards using a before-after, control-impact experiment across 20 sites. During the 2023-2024 breeding seasons we conducted avian point counts, sampled key pest and beneficial arthropods, and collected fecal samples from adult and nestling tree swallows (Tachycineta bicolor) and western bluebirds (Sialia mexicana) to detect vineyard pest DNA. Nest boxes significantly increased swallow and bluebird abundance within one year; swallow responses were strongest …

Environmental And Spatial Drivers Of Quillback Rockfish (Sebastes Maliger) Growth In The Northeast Pacific Ocean From California To Alaska, Claire E. Stuart
Cal Poly Humboldt theses and projects
Quillback rockfish (Sebastes maliger) are a nearshore fishery species found across the Northeast Pacific Ocean, but recent stock assessments have identified a research gap for biological growth parameters, particularly for the southern end of their range. Age-length data compiled from California to Alaska (n=34,396) was used to generate extended von Bertalanffy models with spatial, biological, and environmental covariates, specifically region, sex, depth, and an upwelling index. The objectives of this study were to explore how 1) spatial and 2) environmental covariates affect model parameters L∞, k, and t0. Four total models were developed: …

Schools in Ghana have integrated reproductive health education (RHE) topics into their curricula and School Health Education Programme (SHEP) Coordinators recognize the importance of RHE. However, for RHE to reach its full potential, it needs to be established as a stand-alone subject. This study highlights the challenges and opportunities in delivering in-school RHE, underscoring the interplay between policy frameworks, implementation strategies, and locally accepted approaches.

Combined therapies with Heat Shock Protein 90 (HSP90) inhibitors and Heat Shock Protein 70 (HSP70) inducers are gaining significant interest in cancer and cardiovascular research. Here, we tested the hypothesis that HSP90 inhibitors and HSP70 inducers, together, can block the development of pulmonary fibrosis. We exposed New Zealand White Rabbits to hydrochloric acid (HCl, 0.1 N, 1.5 mL/kg), one of the top five chemicals most commonly involved in accidental exposures and inhalation injuries worldwide, and treated animals with either the orally available HSP90 inhibitor TAS-116 (1.7 mg/kg 5x/week) or TAS-116 combined with the HSP70 inducer, geranylgeranyl acetone (GGA, 50 mg/kg, …

Gelonin is a ribosome-inactivating protein with extreme intracellular toxicity but poor permeation into cells. Targeted disruption of cell membranes to facilitate gelonin entry is explored for cancer and tissue ablation. We demonstrate a hundreds- to thousands-fold enhancement of gelonin cytotoxicity by pulsed electric fields in the T24, U-87, and CT26 cell lines. The effective gelonin concentration to kill 50% of cells (EC₅₀) after electroporation ranged from <1 nM to about 100 nM. For intact cells, the EC₅₀ was unattainable even at the highest gelonin concentration of 1000 nM, which reduced cell survival by only 5–15%. For isoeffective electroporation treatments using 300 ns, 9 µs, and 100 µs pulses, longer pulses were more efficient at lowering gelonin EC₅₀. Increasing the electric field strength of 8, 100 µs pulses from 0.65 to 1.25 kV/cm reduced gelonin EC₅₀ from 128 nM to 0.72 nM. Conversely, the presence of 100 nM gelonin enabled a more than 20-fold reduction in the number of pulses required for equivalent cell killing. Pulsed electric field-mediated delivery of gelonin shows promise for hyperplasia ablation at concentrations sufficiently low to minimize or avoid systemic toxicity.

Surgery has traditionally been a cornerstone in cancer treatment, yet its feasibility can be limited by factors such as tumor location and patient health conditions. When surgery is not viable, thermal and pulsed electric field (PEF)-based ablation technologies offer valuable alternatives. Emerging evidence suggests that these approaches not only target tumors effectively but also stimulate antitumor immune responses. In this review, we begin by examining the distinctive features of hyperthermic treatments (radiofrequency and microwave ablation), cryoablation, and PEF-technologies. Subsequently, we discuss the mechanisms of cell death, stress responses, and release of danger signals triggered by these diverse ablation technologies. Finally, …
Assessment Of The Genetic Population Structure And Extent Of Clonality In A Rare Serpentine Endemic, Lathyrus Biflorus T.W. Nelson & J.P. Nelson (Fabaceae), Using Ddrad Sequencing, Ashley Dickinson
Cal Poly Humboldt theses and projects
Lathyrus biflorus, or the two-flowered pea, is a rare herbaceous perennial restricted to a single population in Northern California. The species is considered critically imperiled but remains unprotected under state or federal endangered species acts. Despite its rarity, little is known about its life history, reproductive biology, or genetic diversity. Using genome-wide reduced representation sequencing, this study aimed to address these gaps by investigating the molecular evidence of clonality, the number and diversity of genets (unique genetic individuals which can be made up of one or many clonal units or ramets), and overall genetic health of the population. My …
“We Are The Future Of This Community”: Exploring Belonging And Rural Healthcare Practice Intent In A High School Medical Sciences Club: A Case Study Of Rural Youth Identity, Purpose, And Place, Sydney Ann Lyons
Cal Poly Humboldt theses and projects
Rural communities across the United States are facing critical healthcare workforce shortages, threatening access to quality care for underserved populations. Health career pathway programs offer a promising solution: a “grow our own” strategy to inspire local youth to pursue healthcare careers. However, little research has explored the role of belonging, identity development, and place-connection in strengthening these programs' outcomes. This case study investigates how participation in the Medical Sciences Club at Del Norte High School— located in rural Northern California— shapes students’, personal growth, career purpose, and rural healthcare practice intent. Through interviews with students, educators, and parents, I found …

Cal Poly Humboldt theses and projects
This research examines changes in Cannabis genetics with a focus on Cannabis genetic bottlenecking. Significant knowledge gaps exist in the study of how and why Cannabis genetics have changed over time. To fill these gaps, a series of semi-structured qualitative interviews were conducted with self-identified Cannabis breeders around the world. Previous qualitative Cannabis research has centered around users, growers, and dealers with few studies looking exclusively at Cannabis breeders. Results include insights into becoming a Cannabis breeder, Cannabis ideotypes, as well as Cannabis genetic bottlenecking due to unanticipated genetic linkages combined with the emphasis on breeding for agronomic factors such …

The Impact Of Plant-Based Diets On Erythrocyte Function, Oxidative Stress, And Vascular Inflammation In Type 2 Diabetes Mellitus, Walter G. Craig
Cal Poly Humboldt theses and projects
Type 2 Diabetes Mellitus (T2DM) is a chronic metabolic disorder with diverse etiological contributors, characterized by insulin resistance, chronic inflammation, oxidative stress, and an increased risk of vascular complications. While much research has focused on plasma and serum markers of metabolic dysfunction, the role of erythrocytes in diabetic pathophysiology remains underexplored. This review examines the impact of dietary macronutrient composition, particularly plant-based diets, on erythrocyte function, oxidative stress, and vascular inflammation in T2DM. Red blood cells (RBCs) are not only oxygen carriers but also dynamic regulators of immune and inflammatory responses, with structural and functional alterations playing a critical role …

Factors Influencing Elk Road Crossing Behavior, Brielle Christine Jaglowski
Cal Poly Humboldt theses and projects
Highways fragment habitat and can impose barriers to wildlife. Installing wildlife crossing systems creates opportunities for safe animal movement, improved human safety, and reduced property damage. Research identifying key zones of connectivity can aid in providing recommendations for locations of these systems and other tools to improve highway safety. Although elk-vehicle collisions occur broadly across northwestern California, factors associated with these highway crossing zones have not been studied in this region. To address this gap in research, the objective of this study was to evaluate how environmental and structural factors influence the probability of elk (Cervus canadensis) crossing US-101 in …

Populations of Nereocystis luetkeana (bull kelp) have experienced localized extinction events in northern California since 2014 due to multiple stressor events, including the area’s most severe marine heatwave (MHW) to-date. Previous surveys in the region observed that bull kelp recovered and persisted at river mouths following the 2014-2015 MHW, suggesting that river mouths may serve as refugia – the refugial river mouth hypothesis. My study aimed to assess this hypothesis and the long-term stability of these refugia by studying how freshwater influences communities at these sites and bull kelp recruitment. Field surveys of salinity fluctuation and kelp and sea urchin …
A Review Of Contact Dependent Interactions Among Encrusting Bryozoans, Sheena Emily Stephens-Norton
A Review Of Contact Dependent Interactions Among Encrusting Bryozoans, Sheena Emily Stephens-Norton
Cal Poly Humboldt theses and projects
This review of published literature is focused on bryozoan interactions and discusses overgrowths, “stalemates,” erect bi-laminate growths and fusions. Overgrowths, where one colony grows zooids atop another, have received the most attention in the scientific literature. Stalemates, where two colonies cease growth at the region of contact, have received relatively little research investigating the possible reasons for its occurrence. Erect, bi-laminate growth, following inter- and/or intraspecific contact between colonies, has also received little research, but could reflect mutualistic behavior of encrusting species to increase feeding and reproduction. Fusion of self-self-contacts appears common, yet non-self-fusions to form natural chimeras may also …
Twice-Exceptionalism And The Complexity Of Supporting Students Who Are Both Gifted And Disabled, River Johnathon Taylor
Twice-Exceptionalism And The Complexity Of Supporting Students Who Are Both Gifted And Disabled, River Johnathon Taylor
Cal Poly Humboldt theses and projects
Drawing on ideas from disabilities studies about compulsory able-bodiedness as well as experiences of disability being fluid and context dependent, this study posits that giftedness, just like disability, is a complex multidimensional form of identity that is both context and temporally dependent. Twice-exceptionalism is defined as an identity in which an individual is gifted in at least one domain of human productivity and manifests at least one disability. Students with unique learning needs (students with disabilities, students with giftedness, and students with twice-exceptionalism) face many complex challenges within Humboldt County, California. Twenty-five educators from Humboldt County working across all grade …
Patterns Of Tree Damage And Fuels Around Powerlines In Northern California, Zoe J. Ziegler
Patterns Of Tree Damage And Fuels Around Powerlines In Northern California, Zoe J. Ziegler
Cal Poly Humboldt theses and projects
Rural powerlines are critical for delivering electricity to communities far from urban areas but present a significant hazard when located in forested regions. During strong wind events, tree failure (i.e., broken branches, snapped stems, and windthrow) can bring combustible material in contact with high-voltage lines, causing electrical outages or setting fire to vegetation below the powerline. Additionally, elevated forest fuel loading can increase wildfire rate of spread and intensity, potentially increasing negative ecosystem impacts and decreasing containment capacity during powerline- ignited wildfires. The risks associated with rural electrical infrastructure, tree failure, and elevated fuel loading emphasize the need for improved …
Physiological Conditions Of Barn Owl (Tyto Furcata) Nestlings As Indicators Of Habitat Quality Within The Wine-Growing Region Of Napa Valley, Christian Cortez
Physiological Conditions Of Barn Owl (Tyto Furcata) Nestlings As Indicators Of Habitat Quality Within The Wine-Growing Region Of Napa Valley, Christian Cortez
Cal Poly Humboldt theses and projects
The global environmental impacts of increasing agricultural demand a transition to sustainable farming practices, including the use of integrated pest management to decrease reliance on harmful pesticides. Vineyard managers in Napa Valley, California, have implemented this strategy by establishing nest boxes that attract barn owls (Tyto furcata) which, in turn, provide an ecosystem service by removing rodent pests. Studies in this system have found uncultivated landcover to be positively associated with distributional and demographic measures of habitat quality for barn owls, but individual measures have not been thoroughly explored. I examined three physiological conditions of barn owl nestlings …

Cal Poly Humboldt theses and projects
The ‘Place-Based Learning Practices Project’ is a graduate project under the Native American Studies Department Rou Dalagurr Food Sovereignty Lab and Traditional Ecological Knowledges Institute (FSL). Making contributions at the intersection of Western academia and Indigenous science, it imagines and realizes content, policies, and protocols surrounding two case studies and the larger cal poly humboldt (cph) institution which respect, honor, and uplift Indigenous sovereignty and resurgence in partnership with the Blue Lake Rancheria Tribe and the greater regional tribal community. Encompassing two projects internal and external to the institution, I aim to address the construction of controlled and protective environments …
