The purpose of this study was to determine whether a periodized resistance training program would influence self-reported depression and anxiety scores in college-aged females. Methods: Eight participants participated in a six-week periodized resistance training program. The participants completed a 3–5 repetition maximum (3–5 RM) for the sumo deadlift (SDL), bench press (BP), barbell back squat (BBS), and standing shoulder press (SSP). These data were used to estimate the 1 repetition maximum (1RM), which in turn was used to develop the periodization program. Following baseline testing, participants participated in two full-body workouts per week for six weeks. Each individual was retested …

Abstract Adipose tissue plays a multifaceted role in regulating metabolic health and contributing to the pathophysiology of various diseases. Brown Adipose Tissue (BAT), a vital organ that senses different environmental cues such as cold stress and diet and then undergoes non-shivering thermogenesis in mammalian animals, enhancing energy expenditure to combat obesity. In our recent published study, we found that the mechanistic target of rapamycin complex 1 (mTORC1) signaling pathway exhibits doubleedge effects in regulating thermogenic function, with opposite role in BAT and white adipose tissue (WAT). Purpose: Performing self-care and domestic life tasks are key aspects of functional independence for older adults. While both depression and anxiety symptoms are correlated with increased functional limitation, these disorders are often studied separately, despite frequent co-occurrence and plausible interaction, and without the consideration of social health. This study examined the impact of comorbid depressive and anxiety symptoms, controlling for social participation and loneliness, on severity of functional limitation in a nationally representative sample of older adults to improve patient-centered care.

BACKGROUND/OBJECTIVES: Cystic fibrosis (CF) is a life-limiting genetic disorder affecting multiple organ systems. This study compared clinical outcomes, hospitalization rates, and survival between children and adolescents with CF who received CFTR modulator therapies (ivacaftor, lumacaftor, tezacaftor, and elexacaftor). METHODS: A retrospective cohort study was conducted using data from the TriNetX global collaborative network. Patients with CF aged 2-12 years (children) and 13-18 years (adolescents) who received CFTR modulator therapies were included. The propensity score matching balanced baseline characteristics between the two age groups. RESULTS: After propensity score matching, 946 patients per group were analyzed. New research on rural law enforcement stress levels reveals concerning implications for police reform and community safety, particularly in western Colorado where officers face unique challenges. This quantitative study examined age group differences in operational and organizational stress among rural law enforcement officers using one-way ANOVAs with post-hoc Tukey's HSD tests. The study of 191 rural officers found significant differences across age groups for both operational stress (F(2, 188) = 4.08, p = .018, η2 = .042) and organizational stress (F(2, 188) = 4.15, p = .017, η2 = .043), with officers aged 24-27 experiencing the highest stress levels. This qualitative narrative study explored the lived experiences of African American women aged 50 and older who self-reported eating pathology and body image dissatisfaction. Disordered eating behaviors, including starvation, binge eating, excessive dieting, and purging, often function as maladaptive coping mechanisms for managing stress, regulating emotions, or responding to traumatic events. The study contributed to the literature by capturing detailed narratives that traced the development of disordered eating behaviors and body image dissatisfaction from early life through adulthood and across the lifespan. Immune thrombotic thrombocytopenic purpura (iTTP) is a chronically relapsing disorder caused by autoantibody-mediated deficiency of ADAMTS13. Rituximab is frequently administered to prevent relapses, but whether the durability of rituximab effect is maintained with subsequent treatment courses has not been studied. Using the United States Thrombotic Microangiopathy Consortium (USTMA) retrospective iTTP registry, we evaluated clinical relapse-free survival (RFS) with subsequent courses of rituximab treatment in multiply relapsing patients. Separately, we evaluated overall RFS (composite of time to clinical relapse, ADAMTS13 relapse, or preemptive rituximab) in a prospective iTTP cohort from the Johns Hopkins University and the University of Minnesota. Ductal carcinoma in situ (DCIS) may progress to ipsilateral invasive breast cancer (iIBC), but often never will. Because DCIS is treated as early breast cancer, many women with harmless DCIS face overtreatment. To identify features associated with progression, we developed an artificial intelligence-based DCIS morphometric analysis pipeline (AIDmap) on hematoxylin-eosin-stained (H&E) tissue sections. We analyzed 689 digitized H&Es of pure primary DCIS of which 226 were diagnosed with subsequent iIBC and 463 were not. ERK activity oscillates between sustained activation during oocyte formation and transient inactivation during oocyte maturation, fertilization, and early embryogenesis. Consequences of ectopic ERK activity upon oocyte maturation and in early embryogenesis are unknown. We show, in Caenorhabditis elegans, that ectopic ERK activity upon oocyte maturation (metaphase I oocytes) results in embryos with abnormalities in nuclear divisions leading to embryonic death. We uncover that ERK directly phosphorylates Polo-like kinase I (PLK-1), on Serine 404, to inhibit nuclear envelope breakdown (NEBD) in early embryogenesis. The RAS/ERK/PLK-1 pathway poisons zygotic NEBD and inhibits the merging of parental genomes, underlining the importance of turning …

Recently, wood modification with environmentally friendly modification agents has received special attention. To this end, this study was conducted to use humin fractions, in combination with citric acid (CA) and succinic acid (SA), as reaction catalysts for the modification of Scots pine (Pinus sylvestris L.) sapwood. The effects of humination modification were evaluated by means of dimensional stability, static and dynamic mechanical properties, thermal stability, crystalline structure, and biological durability tests on modified samples and compared with the unmodified reference ones. tRNA modifications are critical for several aspects of their functions, including decoding, folding, and stability. Using a multifaceted approach encompassing eCLIP-seq and nanopore tRNA-seq, we show that the human tRNA methyltransferase TRMT1L interacts with the component of the Rix1 ribosome biogenesis complex and binds to the 28S rRNA as well as to a subset of tRNAs. Mechanistically, we demonstrate that TRMT1L is responsible for catalyzing N2,N2-dimethylguanosine (m22G) solely at position 27 of tRNA-Tyr-GUA. Surprisingly, TRMT1L depletion also impaired the deposition of 3-(3-amino-3-carboxypropyl) uridine (acp3U) and dihydrouridine on tRNA-Tyr-GUA, Cys-GCA, and Ala-CGC. BACKGROUND: Previous studies have described sex-specific patient subtyping in glioblastoma. The cluster labels associated with these "legacy data" were used to train a predictive model capable of recapitulating this clustering in contemporary contexts.
METHODS: We used robust ensemble machine learning to train a model using gene microarray data to perform multi-platform predictions including RNA-seq and potentially scRNA-seq.
RESULTS: The engineered feature set was composed of many previously reported genes that are associated with patient prognosis. Interestingly, these well-known genes formed a predictive signature only for female patients, and the application of the predictive signature to male patients produced unexpected results. …

CD226 plays a vital role in natural killer (NK) cell cytotoxicity, interacting with its ligands CD112 and CD155 to initiate immune synapse formation, primarily through leukocyte function-associated-1 (LFA-1). Our study examined the role of CD226 in NK cell surveillance of acute myeloid leukemia (AML). NK cells in patients with AML had lower expression of CD226. CRISPR-Cas9 deletion of CD226 led to reduced LFA-1 recruitment, poor synapse formation, and decreased NK cell anti-leukemic activity. Engineering NK cells to express a chimeric antigen receptor targeting the AML antigen CD38 (CAR38) could overcome the need for CD226 to establish strong immune synapses. LFA-1 …

Cytotoxic immune cells mediate precise attacks against diseased cells to maintain organismal health. Their operational unit of killing and host defense is lytic granules (LGs), which are specialized lysosomal-related organelles. Precision in cytotoxicity is achieved by converging the many LGs to the microtubule-organizing center (MTOC) and polarizing these to the diseased cell for secretion. We identify unappreciated intimate relationships between the Golgi, MTOC, and LGs after cytotoxic cell activation, as well as the trans-Golgin protein GCC2 on the LG surface. The poor electronic conductivity of metal-organic framework (MOF) materials hinders their direct application in the field of electrocatalysis in fuel cells. Herein, we proposed a strategy of embedding carbon nanotubes (CNTs) during the growth process of MOF crystals, synthesizing a metalloporphyrin-based MOF catalyst TCPPCo-MOF-CNT with a unique CNT-intercalated MOF structure. Physical characterization revealed that the CNTs enhance the overall conductivity while retaining the original characteristics of the MOF and metalloporphyrin. Simultaneously, the insertion of CNTs generated adequate mesopores and created a hierarchical porous structure that enhances mass transfer efficiency. Graphitic carbon nitride (g-C3N4) exhibits great mechanical as well as thermal characteristics, making it a valuable material for use in photoelectric conversion devices, an accelerator for synthesis of organic compounds, an electrolyte for fuel cell applications or power sources, and a hydrogen storage substance and a fluorescence detector. Astrocytes exhibit diverse cellular and molecular properties across the central nervous system (CNS). Recent studies identified region-specific transcription factors (TF) that oversee these diverse properties; how sex differences intersect with region-specific transcriptional programs to regulate astrocyte function is unknown. Here, we show that the TF Nkx6.1 is specifically expressed in ventral astrocytes of the spinal cord and that its deletion results in sex-specific effects on astrocyte morphology. Astrocytes from males exhibit enhanced morphological complexity, accompanied by increased motor function and cholinergic synapses. Background: Previous studies have described sex-specific patient subtyping in glioblastoma. The cluster labels associated with these "legacy data" were used to train a predictive model capable of recapitulating this clustering in contemporary contexts.
Methods: We used robust ensemble machine learning to train a model using gene microarray data to perform multi-platform predictions including RNA-seq and potentially scRNA-seq.
Results: The engineered feature set was composed of many previously reported genes that are associated with patient prognosis. Interestingly, these well-known genes formed a predictive signature only for female patients, and the application of the predictive signature to male patients produced unexpected results. …

BACKGROUND AND OBJECTIVES: Safety and efficacy of IV onasemnogene abeparvovec has been demonstrated for patients with spinal muscular atrophy (SMA) weighing < 8.5 kg. SMART was the first clinical trial to evaluate onasemnogene abeparvovec for participants weighing 8.5-21 kg.
METHODS: SMART was an open-label, multicenter, phase 3b study conducted across 13 sites in 9 countries (NCT04851873). Symptomatic pediatric participants with SMA (any type; treatment-naïve or had discontinued prior treatment) were stratified into 3 weight cohorts (≥8.5-13, >13-17, and >17-21 kg), administered onasemnogene abeparvovec, and followed for 52 weeks. Corticosteroids were initiated 24 hours before infusion with dose increases in response to adverse events (AEs) and subsequent tapering at investigator discretion. The primary objective was safety. This article analyzes two types of arguments for mathematical platonism, paying close attention to the deductive argument advanced by Proclus. In the first section of the article, I lay out two arguments for mathematical platonism: a contemporary indispensability argument and Proclus’s more ancient Neoplatonic argument. The former takes an approach based on ‘inference to the best explanation’ and scientific theories, but as a result, faces serious objections. For example, scientific theories change, and because the argument is dependent on the latest scientific theories, it seems that the argument cannot give one epistemic certainty in the conclusion. The Great Salt Lake (GSL) is the largest saline lake in the Western Hemisphere. It supports billion-dollar industries and recreational activities, and is a vital stopping point for migratory birds. However, little is known about the spatiotemporal variation of phytoplankton biomass in the lake that supports these resources. Spectral reflectance provided by three remote sensing products was compared relative to their relationship with field measurements of chlorophyll a (Chl a). The MODIS product MCD43A4 with a 500 m spatial resolution provided the best overall ability to map the daily distribution of Chl a. The imagery indicated significant spatial …

The STEM workforce is large and fast-growing. However, underrepresented minorities- URMs (African Americans, Hispanics, and Native Americans) remain poorly represented within the STEM workforce. This is partly due to the lack of retention of URMs in STEM majors. It has been shown by various studies that immersion in undergraduate research enhances the educational experience of science undergraduates and attracts and retains talented students to careers in science. Services at the local disciplinary alternative education program (DAEP) include counseling and behavioral services to referred secondary students, aiming to improve their behavior. The problem addressed in this quantitative study was that a DAEP was in place in the local district since 2009, but there had been no formal assessment to determine whether differences in student discipline occurred in the semester before and the semester after students attended the DAEP.
