Protein aggregates are emerging therapeutic targets in rare monogenic causes of cardiomyopathy and amyloid heart disease, but their role in more prevalent heart-failure syndromes remains mechanistically unexamined. We observed mislocalization of desmin and sarcomeric proteins to aggregates in human myocardium with ischemic cardiomyopathy and in mouse hearts with post-myocardial infarction ventricular remodeling, mimicking findings of autosomal-dominant cardiomyopathy induced by the R120G mutation in the cognate chaperone protein CRYAB. In both syndromes, we demonstrate increased partitioning of CRYAB phosphorylated on serine 59 to NP40-insoluble aggregate-rich biochemical fraction. Objective: The current work was undertaken to evaluate in vitro antibacterial, antifungal and antioxidant properties, together with total phenolic contents of ethyl acetate extracts from wild edible mushrooms collected in September 2017 from the rehabilitated forest area in the northeast of Thailand. Results: All the wild edible mushrooms collected could be classified into 6 families and 13 species namely Macrolepiota rhacodes, Heimiella japonica, Boletus persoonii, Mycoamaranthus cambodgensis, Cantharellus cibarius, Amanita princeps, Amanita hemibapha, Russula cyanoxantha, Russula eburneureolata, Russula virescens, Russula emetica, Russula violeipes, and Termitomyces clypeatus. Background: Patient access to anti-cancer medicines is challenging in Thailand and worldwide. Objective: This study aimed to explain and quantify access to anti-cancer medicines in Thailand. Methods: Mixed methods of literature review and in-depth interview were conducted. Access to anticancer medicines was compared to the World Health Organization-Model List of Essential Medicines (WHO-EML), and recommendations of the National Comprehensive Cancer Network (NCCN). Results: Access to National List of Essential Medicines (NLEM) is indifferent among the three public schemes. Universal coverage scheme (UCS) and Social security scheme (SSS) limit access to non-NLEM anticancer medicines by reimbursement caps. Civil Servant Medical Benefit …

Alcohol is associated with increased mortality and morbidity. Pulmonary infections with opportunistic pathogens can occur in healthy humans; however, hazardous alcohol exposure like binge alcohol intoxication (≥ 0.08% BAC) is a major risk factor. We’ve previously investigated the efficacy of Sulforaphane (SFN) prophylaxis in protecting human and murine alveolar macrophage function against opportunistic infections when exposed to binge alcohol conditions in vitro. However, an in vivo model investigating SFN and hazardous alcohol exposure in the context of infection is not well understood. 1,8-Cineole is a naturally occurring chemical molecule predominantly found in fragrant plants, particularly Myrtle. Its aroma is distinctive and has been the subject of numerous investigations due to its various biological actions. This study examines the characterization of 1,8-Cineole derived from Myrtle and investigates its antibacterial and antioxidant properties. This study seeks to compare 1,8-Cineole with antibiotics like Amoxicillin and Tetracycline, and moreover, to investigate its antioxidant capabilities against diverse bacterial strains (both Gram-positive and Gram-negative). 1,8-Cineole exhibits the most effective antibacterial properties, demonstrating an inhibition zone of 12.00 mm against Staphylococcus aureus and 10.00 mm against Pseudomonas aeruginosa. …

The science of embodied cognition has established that thinking involves dynamic exchanges between the body and the brain and webs of interaction between humans and their environments. While learning draws on these connected ecosystems, persistent notions of the brain as separate from and hierarchically managing the body—an enduring legacy of Cartesian dualism—perpetuate static teaching and learning mindsets. Disregard for the body’s role in learning fuels disengagement, isolation, and inequity. This chapter weaves key findings from embodied learning research together with scenes from the author’s own academic journey to illustrate how leveraging brain–body connections can foster educational well-being. Incivility among nursing students is well documented in the academic, clinical, and leadership literature. Student incivility towards faculty was less studied, and merited investigation because incivility can lead to faculty discontentment, stress, and attrition, exacerbating the scarcity of nursing faculty. The purpose of this qualitative phenomenological study, which was underpinned by the health belief model, was to explore the perceptions of nursing faculty who experience student incivility in the Western region of the United States. Open-ended, semistructured interviews were conducted with 11 nursing faculty who shared perceptions of their experience of student incivility behavior. Open coding and thematic analysis yielded …

AXL, a member of the TAM receptor family, has emerged as a potential target for advanced-stage human malignancies. It is frequently overexpressed in different cancers and plays a significant role in various tumor-promoting pathways, including cancer cell proliferation, invasion, metastasis, epithelial-mesenchymal transition (EMT), angiogenesis, stemness, DNA damage response, acquired therapeutic resistance, immunosuppression, and inflammatory responses. Beyond oncology, AXL also facilitates viral infections, including SARS-CoV-2 and Zika highlighting its importance in both cancer and virology. In preclinical models, small-molecule kinase inhibitors targeting AXL have shown promising anti-tumorigenic potential. The decision-making autonomy of female intimate partner violence (IPV) survivors to make financial transactions is a stronger predictor of help-seeking behavior that is not yet fully explored. Despite the plausibility of using the mobile phone for financial transactions, it is yet to be explored as a predictor of help-seeking behavior after IPV events through digital financial inclusion. The dataset from the 2018 Nigerian Demographic Health Survey was used to explore if digital financial inclusion could predict help-seeking behavior instead of the severity of abuse among ever-partnered Nigerian female IPV survivors (aged 15 to 49 years) who use their mobile phones …

This article presents a paper developed by the AAG Organizing Committee on Locational Information and the Public Interest through a summit held in Santa Barbara, California in June 2022. The summit resulted in goals and ideas for addressing the issues that arise from the present environment for geodata, whereby public, private, and third-sector entities can tap into publicly available locational information with relatively little regulation on its access or use. Inspired by the diverse set of technologies used in underground object detection and imaging, we introduce a novel multimodal linear search problem whereby a single searcher starts at the origin and must find a target that can only be detected when the searcher moves through its location using the correct of p possible search modes. The target’s location, its distance d from the origin, and the correct search mode are all initially unknown to the searcher. Quantifying emesis in Suncus murinus (S. murinus) has traditionally relied on direct observation or reviewing recorded behaviour, which are laborious, time-consuming processes that are susceptible to operator error. With rapid advancements in deep learning, automated animal behaviour quantification tools with high accuracy have emerged. In this study, we pioneere the use of both three-dimensional convolutional neural networks and self-attention mechanisms to develop the Automatic Emesis Detection (AED) tool for the quantification of emesis in S. murinus, achieving an overall accuracy of 98.92%. Hermansky-Pudlak syndrome (HPS) is a genetic disorder of endosomal protein trafficking associated with pulmonary fibrosis in specific subtypes, including HPS-1 and HPS-2. Single-mutant HPS1 and HPS2 mice display increased fibrotic sensitivity while double-mutant HPS1/2 mice exhibit spontaneous fibrosis with aging, which has been attributed to HPS mutations in alveolar epithelial type II (AT2) cells. We utilized HPS mouse models and human lung tissue to investigate mechanisms of AT2 cell dysfunction driving fibrotic remodeling in HPS. Background: Despite reassuring clinical and histological features, low-grade meningiomas can recur after surgery. Targeted gene expression profiling improves risk stratification of meningiomas, but the utility of this approach for clinical low-risk meningiomas is incompletely understood.
Methods: This was a multicenter retrospective cohort study of meningiomas from patients who were treated at 4 institutions from 1992 to 2023. Molecular subtypes, such as defined by The Cancer Genome Atlas (TCGA), delineate a cancer's underlying biology, bringing hope to inform a patient's prognosis and treatment plan. However, most approaches used in the discovery of subtypes are not suitable for assigning subtype labels to new cancer specimens from other studies or clinical trials. Background: M6223 is an intravenous (IV), Fc-competent, fully human, antagonistic, anti-T cell immunoreceptor with immunoglobulin and immunoreceptor tyrosine-based inhibitory motif domains (TIGIT) antibody. Bintrafusp alfa (BA) is a bifunctional fusion protein that simultaneously blocks nonredundant immunosuppressive TGF-β and PD-(L)1 pathways.

Background: Immune checkpoint inhibitors (ICIs) are the gold standard therapy in patients with deficient mismatch repair (dMMR)/microsatellite instability-high (MSI-H) metastatic colorectal cancer (mCRC). A significant proportion of patients show resistance, making the identification of determinants of response crucial. Growing evidence supports the role of sex in determining susceptibility to anticancer therapies, but data is lacking for patients with MSI-H CRC.

Growing environmental concerns have facilitated an increased need for renewable energy. Solar energy is an excellent source of renewable energy, however current dye-sensitized solar cells are not efficient enough for commercial use. This thesis attempts to create a novel dye-sensitized cathode to increase the efficiency of dye-sensitized solar cells.
Dye-sensitized cathodes could lower the energy level on the cathode and thereby increase the overall efficiency of the solar cell, however until now it has not been attempted. Genomic structural variants (SVs) are a major source of genetic diversity in humans. Here, through long-read sequencing of 945 Han Chinese genomes, we identify 111,288 SVs, including 24.56% unreported variants, many with predicted functional importance. By integrating human population-level phenotypic and multi-omics data as well as two humanized mouse models, we demonstrate the causal roles of two SVs: one SV that emerges at the common ancestor of modern humans, Neanderthals, and Denisovans in GSDMD for bone mineral density and one modern-human-specific SV in WWP2 impacting height, weight, fat, craniofacial phenotypes and immunity. Meningiomas are the most frequent primary intracranial tumors. Hence, they constitute a major share of diagnostic specimens in neuropathology practice. The 2021 WHO Classification of Central Nervous System Tumors ("CNS5") has introduced the first molecular grading parameters for meningioma with oncogenic variants in the TERT promoter and homozygous deletion of CDKN2A/B as markers for CNS WHO grade 3. However, after the publication of the new classification volume, clarifications were requested, not only on novel but also on long-standing questions in meningioma grading that were beyond the scope of the WHO "blue book."
