The DyGAF model, which stands for Dynamic Gene Attention Focus, is specifically designed and tailored to address the challenges in biomarker detection, progression reporting of pathogen infection, and disease diagnostics. The DyGAF model introduced a novel dual-model attention-based mechanism within neural networks, combined with machine learning algorithms to enhance the process of biomarker identification. The model transcended traditional diagnostic approaches by meticulously analyzing gene expression data. DyGAF not only identified but also ranked genes based on their significance, revealing a comprehensive list of the top genes essential for disease detection and prognosis. This study investigates the perceptions of artificial intelligence (AI) among undergraduate construction management students who are poised to become future leaders in the construction industry. As the industry increasingly adopts AI technologies to enhance project planning, design, site management, and safety, understanding students’ attitudes toward these innovations becomes crucial. This research employs a mixed-methods approach, combining quantitative survey data with qualitative data to capture the students’ insights on AI’s potential applications, benefits, and challenges within the construction sector. Background:Becium grandiflorum is a fragrant perennial shrub of the Lamiaceae family. Objectives: The current study aimed to explore the cytotoxic potential of the nhexane fraction from Becium grandiflorum aerial parts and, further, isolate its major diterpene and conduct in vitro and in vivo anticancer activities along with its molecular mechanism and synergy with doxorubicin. Methods: The hydroalcoholic extract of Becium grandiflorum aerial parts was fractionated, and the n-hexane fraction was analyzed via GC-MS. The major isolated diterpene, 18-epoxy-pimara-8(14),15-diene (epoxy-pimaradiene), was quantified using UPLC-PDA. West Nile Virus (WNV), a member of the Flaviviridae family and introduced to the Western hemisphere in 1999, presents a unique and emerging threat to human health. Infection in humans is often mild but can occasionally result in severe neurological impairment and even death. West Nile Virus non-structural protein 3 (NS3) plays an important role in the process of viral genome amplification. More specifically, the C-terminal domain of the protein folds into a helicase enzyme, which is responsible for the “unwinding” of double-stranded RNA intermediates and RNA secondary structure on the WNV genome. Most walking organisms tend to have relatively light limbs and heavy bodies in order to facilitate rapid limb motion. However, the limbs of brittle stars (Class Ophiuroidea) are primarily comprised of dense skeletal elements, with potentially much higher mass and density compared to the body disk. To date, little is understood about how the relatively unique distribution of mass in these animals influences their locomotion. In this work, we use a brittle star inspired soft robot and computational modeling to examine how the distribution of mass and density in brittle stars affects their movement. Naturalistic neuroimaging has significantly advanced our understanding of the brain in everyday situations. Stimulus designs that previously targeted individual brain regions are now being replaced by activities such as watching movies, engaging in conversations, and interacting with virtual reality environments. However, traditional functional neuroimaging techniques like functional magnetic resonance imaging (fMRI) are inherently unnaturalistic, as participants must lay down and remain still within the magnet's bore. Consequently, these naturalistic paradigms require an equally naturalistic neuroimaging modality. High-density diffuse optical tomography (HD-DOT) offers a wearable alternative to fMRI by utilizing overlapping optical measurements to densely sample cortical brain activity. HD-DOT has …

INTRODUCTION: The past decade has seen a paradigm shift in the evaluation and management of Parkinson's disease psychosis (PDP), with the first approval of an antipsychotic in the US in 2016. An evidence-based review by the Movement Disorder Society found pimavanserin and clozapine to be clinically useful, (low-dose) quetiapine to be possibly useful, and all other antipsychotics to be avoided due to motor worsening. Clozapine and quetiapine use can be limited by provoking Parkinson's disease (PD) nonmotor symptoms of somnolence and hypotension. Quetiapine may also be limited by its risk in cognitive impairment. Pimavanserin is not associated with these symptoms. …

Buckwheat (Fagopyrum esculentum) is a fast-growing plant that quickly produces a dense groundcover. The utilization of buckwheat as a cover crop in vegetable production could be beneficial to Georgia producers, however for successful adoption, weed control in the cover crop coupled with control of buckwheat volunteers must be achievable. A preemergence (PRE) and a postemergence (POST) experiment were each conducted three times in Ty Ty, GA (2020-2021) addressing these objectives. In the PRE study, acetochlor at 0.56 lb ai acre(-1) injured buckwheat up to 16% without negatively influencing growth, suggesting potential for use in buckwheat for weed control. Maackia amurensis lectins, including MASL, MAA, and MAL2, are widely utilized in biochemical and medicinal research. However, the structural and functional differences between these lectins have not been defined. Here, we present a high-resolution cryo-EM structure of MASL revealing that its tetrameric assembly is directed by two intersubunit disulfide bridges. These bridges, formed by C272 residues, are central to the dimer-of-dimers assembly of a MASL tetramer. This cryo-EM structure also identifies residues involved in stabilizing the dimer interface, multiple glycosylation sites, and calcium and manganese atoms in the sugar-binding pockets of MASL. Ancient discourse about the beneficial yet predominantly dangerous role of curiosity inattaining knowledge was prevalent in the Roman Empire around the 2nd century C.E. when the North African author, Apuleius, composed his Latin novel, the Golden Ass. Blockbuster dystopian sci-fi features stories of environmental collapse, authoritarian governments, out of control corporations, unbounded consumerism, and other awful possibilities. Scholars of science fiction broadly agree that this genre explores collective fears, anxieties, and speculations about the present and future. At the same time, film scholars have established that dystopian sci-fi cinema is compelling, and audiences respond to their effects-driven visuals, photorealistic mise-en-scene, and dramatic narratives. This creates an apparent paradox: Blockbuster cinematic aesthetics portray undesirable worlds desirably. I resolve this paradox by arguing that the worldbuilding of spectacle-driven, dystopian sci-fi cinema is Romanticized. That is, the provocative style of …

Objective: To compare left ventricular (LV) and aortic (AO) pressures obtained using fluid-filled and high-fidelity solid-state pressure catheters in subjects undergoing left heart catheterization.

Materials and Methods: Twenty subjects scheduled for a left heart catheterization were enrolled and 18 subjects completed this IRB-approved study. LV and AO pressures were obtained using fluid-filled pressure catheter (standard-of-care) and high-fidelity solid-state pressure catheter synchronously. Pressure tracings were analyzed to measure LV systolic (LVSP), LV minimum-diastolic (LVMDP), LV end-diastolic (LVEDP), AO systolic (AOSP), and AO diastolic (AODP) pressures. 4-1BB is a transmembrane glycoprotein found on T lymphocytes, and signaling through 4-1BB activates and differentiates CD3+ and CD8+ T cells. The ability of 4-1BB to stimulate cytotoxic T-cell responses makes it a promising target for therapeutic cancer immunotherapy development. 4-1BB antibodies have shown promising antitumor activity in preclinical studies and clinical trials. Common side effects include transaminase elevation, cytopenias, fatigue, and nausea. This clinical review summarizes past and current 4-1BB antibodies in oncology clinical trials.

Cirrhosis, a marker of severe liver diseases, limits future liver remnant (FLR) growth, preventing many cancer patients from undergoing surgery. While portal vein blockade (PVB) techniques are used to stimulate liver regeneration, 20-30% of patients still fail to achieve the required growth. Although mesenchymal stem cell (MSC) therapy improves PVB, its efficacy is limited by poor cell retention. To address this, we utilized alginate hydrogels to deliver MSCs and improve their retention. MSCs were loaded in the hydrogel and injected intraportally in cirrhotic rats. Liver volume, weights, enzyme levels, and histology were monitored. Results showed that the hydrogel maintained 89.0 …

Clostridium perfringens is a prevalent gut bacterial pathogen in humans and animals. This study investigated the role of the mechanistic targets of rapamycin (mTOR) and deoxycholic acid (DCA) on C. perfringens intestinal infection. Chickens were sequentially infected with Eimeria maxima and received the mTOR inhibitor rapamycin and DCA. C. perfringens-induced necrotic enteritis (NE) was evaluated using body weight gain (BWG), histopathology, bile acids, pathogen colonization, cell infiltration and death, and gene expression. The significant difference of p < 0.05 was analyzed by one-way ANOVA and multiple comparisons. Notably, rapamycin strongly reduced the subclinical and clinical NE histopathologies. DCA and DCA combined with rapamycin alleviated clinical NE and BWG loss. Rapamycin, DCA, and DCA + rapamycin attenuated bile acid reduction in NE birds, and they also reduced immune cell infiltration into the intestinal lamina propria as well as immune cell migration in vitro. At molecular levels, DCA and DCA + rapamycin reduced proinflammatory IFN gamma, MMP9, IL23, and IL17 gene expression. Rapamycin, DCA, and DCA + rapamycin reduced NE-induced intestinal cell apoptosis. Together, these results suggest that mTOR signaling mediates C. perfringens-induced ileitis, and combining mTOR inhibition and DCA improves the intervention efficacy against NE ileitis and BWG loss.

The Consider, Construct, Confirm Learning cycle is a framework for teachers to organize their planning—be it individual lessons, units of study, or whole courses. The CCC Learning Cycle framework shifts the format of a typical lesson from didactic instruction leading to short-term renting of knowledge through memorization, to student-centered, lived experiences resulting in students owning their new knowledge.
