Latanoprostene bunod ophthalmic solution (LBN) 0.024% is a topical nitric oxide (NO)-donating prostaglandin F2α (PGF2α) analog first approved in November 2017 for reduction of intraocular pressure (IOP) in patients with ocular hypertension (OHT) or open-angle glaucoma (OAG). This narrative review describes the unique mechanism of action of LBN and summarizes available real-world data. Upon instillation, LBN is metabolized into latanoprost acid and butanediol mononitrate, which is further reduced to NO and an inactive metabolite. Latanoprost acid increases aqueous humor outflow primarily through the uveoscleral (unconventional) pathway, whereas NO increases outflow through the trabecular (conventional) pathway. Eight studies were identified: 2 …

Emerging evidence has shown that epigenetic regulation plays a fundamental role in cancer metastasis, the major cause of cancer-related deaths. Here, we conducted an in vivo screen for vulnerabilities of brain metastasis and identified N-acetyltransferase 10 (NAT10) as a driver of brain metastasis. Knockdown of NAT10 restrains cancer cell proliferation and migration in vitro and tumor growth and brain metastasis in vivo. The poorly characterized RNA helicase domain of NAT10 is critical for cell growth in vitro, while both RNA helicase and NAT domains are essential for primary tumor growth and brain metastasis in vivo. Mechanically, NAT10 promotes the …

Background: Acute liver failure (ALF) is a loss of liver function due to a severe hepatic insult. Studies utilizing the azoxymethane (AOM) mouse model of ALF, which also generates hepatic encephalopathy, have primarily focused on development of neurological deficits. However, the molecular processes that generate liver damage have not been fully characterized. Therefore, a more comprehensive characterization of the hepatic consequences of AOM toxicity is needed to better understand this disease model.

Aim: To identify molecular pathology contributing to hepatic injury during the progression of AOM-induced ALF.

Methods: C57BL/6 mice were injected with AOM to produce ALF and hepatic encephalopathy. …

The life-threatening coronavirus disease 2019 (COVID-19) affects about 1 in 1,000 healthy people under 50 without underlying conditions. Among patients with critical COVID-19 pneumonia, rare germline variants at genes controlling type I IFN immunity have been reported in up to 5% of patients. Causal etiologies in 80-85% of cases are still unknown. We analyzed two families with hypoxemic COVID-19 pneumonia for known single-gene inborn errors of immunity. In Family 1, two siblings with critical COVID-19 were homozygous for a DOCK2 variant, c.3624+5G>A. DOCK2 deficiency is a known T-cell disorder underlying severe viral diseases. The variant resulted in skipping exon …

Journal of Electrochemistry

Detection of target analytes at low concentrations is significant in various fields, including pharmaceuticals, healthcare, and environmental protection. Theophylline (TP), a natural alkaloid used as a bronchodilator to treat respiratory disorders such as asthma, bronchitis, and emphysema, has a narrow therapeutic window with a safe plasma concentration ranging from 55.5–111.0 μmol·L–1 in adults. Accurate monitoring of TP levels is essential because too low or too high can cause serious side effects. In this regard, non-enzymatic electrochemical sensors offer a practical solution with rapidity, portability, and high sensitivity. This article aims to provide a comprehensive review of the recent developments …

Studies On Cell Cycle Regulation And Ecm Modification During Tubular Organ Formation In Drosophila Embryos, Jeffrey Matthew Mar 2025

Studies On Cell Cycle Regulation And Ecm Modification During Tubular Organ Formation In Drosophila Embryos, Jeffrey Matthew

LSU Doctoral Dissertations

The formation of tubular organs—such as the heart and kidneys—is a complex developmental process that requires the precise coordination of tissue remodeling with dynamic changes in cellular behavior. Key processes including cell proliferation, apoptosis, and extracellular matrix (ECM) formation must be tightly synchronized with mechanisms that generate and transmit physical forces, transforming a flat epithelial sheet into a three-dimensional organ. Disruptions in the homeostasis of these processes during organogenesis can lead to congenital defects, such as pulmonary atresia and renal hypoplasia.

To investigate the mechanisms underlying epithelial morphogenesis, the Chung laboratory employs the Drosophila embryonic salivary gland (SG) as a …

Dnajb6: A Potent Dnajb Chaperone In Regulating Tau Proteostasis, Abigail R. Esquivel Mar 2025

Dnajb6: A Potent Dnajb Chaperone In Regulating Tau Proteostasis, Abigail R. Esquivel

USF Tampa Graduate Theses and Dissertations

Neurodegenerative diseases often feature the pathological buildup of misfolded proteins, and tau is a principal culprit in multiple tauopathies. One chaperone of emerging interest is DnaJB6b, a member of the Hsp40/DnaJ family, which exhibits potent anti-aggregation activity against several disease-related proteins. Recent investigations highlight its unique capacity to recognize and neutralize aggregation-prone forms of tau. DnaJB6b appears potent against multiple variants of tau and may be acting against tau, in part, by directing misfolded tau toward proteasomal clearance. The first lines of evidence come from systematic chaperone screens, where DnaJB6b repeatedly emerged as a strong suppressor of tau seeding. Further …

The field of printed electronics (PEs) is expanding rapidly, driven by increasing market demand. According to reports, the global market for PEs is expected to reach $23 billion by 2026, with a compound annual growth rate of 18.3% from 2023 to 2028. This growth necessitates the development of innovative printing technologies that can meet these demands efficiently, delivering high-quality, high-resolution PEs suitable for applications in health, environmental monitoring, sports, aerospace, and more.

Currently, PEs manufacturing relies on two main groups of printing technologies. Contact printing methods offer high-speed production but struggle with precision, while non-contact methods provide high resolution but …

Refinement Of Megakaryocyte Lineage Commitment In Hematopoietic Stem And Progenitor Cells During Postnatal And Adult Hematopoiesis, Ryan Michael Mack Mar 2025

Refinement Of Megakaryocyte Lineage Commitment In Hematopoietic Stem And Progenitor Cells During Postnatal And Adult Hematopoiesis, Ryan Michael Mack

Dissertations

Mammalian hematopoiesis ensures the continuous production of specialized blood cells, yet the mechanisms that govern megakaryocyte/erythroid (MK/E) bifurcation remain incompletely understood. While HSPCs sustain all blood lineages, how transient waves of progenitors arise and adapt from fetal to adult stages is less clear. This gap in knowledge limits our capacity to precisely manipulate platelet and red blood cell outputs in both developmental and therapeutic contexts. Using refined gating strategies and various bulk- and single-cell transcriptomics, we dissected the traditional “common myeloid progenitor” and Multipotent Progenitor (MPP) compartment into multiple functionally distinct subsets (C1, C2, C3). Within C1, an ESAM⁺ fraction …

Gestational diabetes mellitus (GD)-induced gut dysbiosis in pregnant mothers may increase the risk of cognitive impairment and neurological disorders in both the mother and offspring as they age. Restoring gut balance could improve cognitive outcomes for both. Despite advancements in GD treatment, side effects have increased, and long-term neurocognitive impacts on offspring born to GD mothers remain underexplored. This study uses a GD mouse model, inducing pancreatic dysfunction in 3-month-old pregnant C57BL/6J mice with Streptozotocin. The efficacy and mechanism of the prebiotic phytocompound green leaf extract (

Metabolic imaging produces powerful visual assessments of organ function in vivo. Current techniques can be improved by safely increasing metabolic contrast. The gold standard, 2-[18F]fluorodeoxyglucose-positron emission tomography (FDG-PET) imaging, is limited by radioactive exposure and sparse assessment of metabolism beyond glucose uptake and retention. Deuterium magnetic resonance imaging (DMRI) with [6,6-2H2]glucose is nonradioactive, achieves tumor metabolic contrast, but can be improved by enriched contrast from deuterated water (HDO) based imaging. Here, we developed a DMRI protocol employing [2H7]glucose. Imaging 2H-signal and measuring HDO production in tumor-bearing mice detected differential glucose utilization across baseline tumors, tumors treated with vehicle control or …

Cancer immunotherapy has improved the survival of a subset of patients by harnessing the power of the immune system to find and destroy malignant cells. The immune system also protects the host by destroying developing premalignant and malignant tumors. Advancing our knowledge of premalignant immunity and immune changes seen in lesions that develop into invasive cancer versus those that regress offers an exciting opportunity to leverage the immune system for immune prevention and immune interception of premalignancy. Understanding the immune environment of premalignant lesions and how chronic inflammation plays a central role in the evolution of premalignancy is essential for …

Cigarette smoke (CS) creates a "cancer field" in the lung that promotes malignant transformation. The molecular changes within this field are not fully characterized. We examined the significance of microRNA-1 (miR-1) downregulation as one of these changes. We found that tumor miR-1 levels in three non-small cell lung cancer cohorts show inverse correlations with the smoking burden. Lung MiR-1 levels follow a spatial gradient, have prognostic significance, and correlate inversely with the molecular markers of injury. In CS-exposed lungs, miR-1 is specifically downregulated in the endothelium. Exposure to CS induces angiogenesis by selectively degrading mature miR-1 via a vascular endothelial …

Monitoring Population Pressure In The Semiarid Zone Of Kenya, Isaac Sindiga Mar 2025

Monitoring Population Pressure In The Semiarid Zone Of Kenya, Isaac Sindiga

The Geographical Bulletin

Population pressure is a maladjustment between resources of a community and its population. This maladjustment worsens when the population increases at a faster rate than the development of the economy. The pressure leads to the deterioration of land resources and low, risky income opportunities and widespread poverty. Kenya's population growth rate of 4 percent per year is one of the highest in the world. Its populationresource relations are deteriorating and foreshadow the country's future. In the semi-arid and arid parts of the country, especially those occupied by traditional pastoral people, population pressures are particularly serious. Here, the pressures of people …

The Syrian Tabqa Dam: Its Development And Impact, Monib Ei-Khatib

The Geographical Bulletin

The Euphrates River has its sources in eastern Turkey's Anatolian Plateau and extends 3,000 kilometers through Syria and Iraq before it discharges into the river's delta at Shat AI-Arab in the Arabian Gulf. It is an exotic river, a center of past civilization and population, and a managed river with an extensive delta. It floods in early summer but its flooded area dries-out very quickly in the semiarid climate. The Euphrates River can maintain an annual flow throughout its course because of relatively high rainfall and snow-melt at higher elevations. Utilization of its drainage basin has been intensified from its …

The Nowruz Oil Spill In The Arabian Gulf: Case Study Of Saudi Arabia, Abdullah S. Ai-Amirah Mar 2025

The Nowruz Oil Spill In The Arabian Gulf: Case Study Of Saudi Arabia, Abdullah S. Ai-Amirah

The Geographical Bulletin

The Arabian Gulf has suffered from hundreds of oil spills, some of them reported but most unreported. In many cases the amounts of the spills and their causes are usually undetermined. During 1978-1979, tanker captains reported to the Japan Oceanographic Data Center about 412 oil slicks in the Gulf and its approaches.' Table 1 shows major oil spills exceeding 3,000 barrels which occurred between August, 1980, and August, 1982. Although these are relatively current, it should be noted that other significant oil spills have occurred since oil was discovered in the region by Iran in 1908.

Web Application For Simulation Of An Agent-Based Model In Netlogo3d, Chris Davis Perumal, Abraham Nofal, Benedict J. Kolber, Rachael Miller Neilan

Spora: A Journal of Biomathematics

Agent-based models (ABMs) are computer simulation models for studying systems of autonomous agents. Modelers often use specialized software like NetLogo to develop ABMs, but this software poses barriers to researchers in other disciplines with no prior programming experience. To address this issue, we developed a web application that allows users to simulate our ABM via a web browser, eliminating the need for the user to download and use specialized software. While presented in the context of a specific ABM, our approach can be applied to other ABMs to enhance accessibility. The ABM presented here was developed in NetLogo3D. The model …
