A 3-Week Pause Versus Continued Bruton Tyrosine Kinase Inhibitor Use During Covid-19 Vaccination In Individuals With Chronic Lymphocytic Leukaemia (Improve Trial): A Randomised, Open-Label, Superiority Trial, Helen M. Parry, Claire Hutchinson
Peninsula Medical School
Background: Chronic lymphocytic leukaemia is the commonest leukaemia and is associated with profound immunosuppression. Bruton tyrosine kinase inhibitors (BTKi) have revolutionised chronic lymphocytic leukaemia management; however, therapy impairs vaccine-induced immunity. We evaluated whether a 3-week pause of BTKi treatment improved spike protein receptor binding domain (RBD) immunity to SARS-CoV-2 booster vaccination while maintaining disease control. Methods: We performed an open-label, two-arm, parallel-group, randomised trial in secondary-care haematology clinics in 11 UK hospitals. Participants aged 18 years or older, diagnosed with chronic lymphocytic leukaemia, and currently taking BTKi therapy (frontline or relapsed setting) for at least 12 months were eligible. Participants …

Neurofibromatosis type 1 (NF1) is an inherited disorder that predisposes individuals to malignant peripheral nerve sheath tumors (MPNSTs), a highly aggressive sarcoma with limited treatment options and poor prognosis. This study explores the potential of targeting the interaction between Galectin-1 and Ras as a novel therapeutic strategy for MPNSTs. Through molecular docking, we identified critical residues involved in the Galectin-1 and H-Ras interaction. We developed LLS30, a compound designed to target this Ras-binding pocket on Galectin-1, and tested its efficacy. LLS30 effectively disrupted the Galectin-1/Ras interaction, causing Ras delocalization from the plasma membrane and inhibiting Ras signaling. In vitro experiments …

Bi-Level Energy Dispatch Optimisation For Peak Load Shaving In Microgrids With Battery Storage, Mohd Fakhizan Romlie, Abid Ali, Gm Shafiullah
Platform: A Journal of Engineering (PAJE)
Peak load periods in electrical networks result in significant power losses, straining generation capacity, and increasing operational costs. Strategically optimising electricity supply from battery storage-based distributed generation (DG) during these peak times can mitigate these losses and enhance grid efficiency through peak load shaving (PLS). However, electrical loads vary throughout the day and night. Thus, identifying peak and off-peak loads would be the most challenging task to complete before scheduling operations for batteries. This study presents a bi-level energy optimisation framework to identify the peak and off-peak loads and plan the energy dispatching operations for battery storage. The bi-level energy …

Poor Cancer Prevention And Detection In Zimbabwe, Kenyon C. Chipman
Journal of Nonprofit Innovation
Abstract
In the aftermath of the HIV/AIDS epidemic, Sub-Saharan Africa, particularly Zimbabwe, has witnessed a surge in noncommunicable diseases such as cancer. Despite the decline in HIV/AIDS and other cancer-associated illnesses like HPV, these diseases have set the stage for a growing cancer epidemic. Key contributing factors to this rise include a lack of knowledge about cancer among both patients and health professionals, the prevalence of cancer-associated communicable diseases, and limited accessibility to early detection and prevention services and resources. These factors have led to late diagnoses, increased burden on the healthcare system, and adverse effects on individuals and families. …

Modern Sport
Jumping ability is a fundamental variable in many sports, as its execution requires an integration of muscular strength Q1 and certain biomechanical variables. This is particularly evident in gymnastics jumping events and jump shots in ball games, both of which rely on a high level of vertical resistance. Vertical resistance serves as an indicator of an athlete’s ability to overcome their body weight while counteracting gravitational force to achieve optimal performance. As such, it is considered one of the key factors in movements that demand explosive power and speed. The researchers believe that despite the significant relationship between vertical resistance, …

Although periodontal disease (PD) is reported to be associated with changes in various genes and proteins in both invading bacteria and the host, its molecular mechanism of pathogenesis remains unclear. Changes in immune and inflammatory genes play a significant role in PD pathogenesis. Some reports relate alterations in cellular epigenetic patterns to PD characteristics, while several high-throughput analyses indicate thousands of differentially methylated genes in both PD patients and controls. Furthermore, changes in DNA methylation patterns in inflammation-related genes have been linked to the efficacy of periodontal therapy, as demonstrated by findings related to the cytochrome C oxidase II gene. …

Coastal Community Perspectives On Marine Resource Conservation And Access In Rote Island, Indonesia, Nara Wisesa
Oral Presentations
Indonesia’s coastal and marine environment supports high marine biodiversity, with conservation efforts aimed at sustainable management. Marine Protected Areas (MPAs), such as the Savu Sea MPA, play a key role in these efforts. Rote Island, located on the eastern side of this MPA, has coastal communities heavily reliant on marine resources, primarily fishing and seaweed farming. While conservation measures impact local resource access, there is limited research on their socio-cultural dimensions, particularly from the perspective of local stakeholders. Although conservation should prioritize local communities, studies indicate minimal involvement in planning and implementation. This study examines how marine conservation affects Rote’s …

Coal is a major fuel source but raises environmental concerns. This study investigated the feasibility of utilizing a composite briquette sustainable fuel produced from subbituminous coal and Jatropha curcas biomass. Briquettes were prepared with varying weight ratios of coal to J. curcas biomass (90:0–0:90) using corn starch as a binder. The influence of this ratio on the briquettes’ properties was comprehensively evaluated through proximate analysis, ultimate analysis, scanning electron microscopy (SEM), and X-ray fluorescence (XRF) analysis. The results revealed that increasing the J. curcas content in the briquettes gradually decreased the moisture content, ash content, and fixed carbon content while …

Summary: Cells differentiate to their final fates along unique trajectories, often involving multi-potent progenitors that can produce multiple terminally differentiated cell types. Recent developments in single-cell transcriptomic and epigenomic measurement provide tremendous opportunities for mapping these trajectories. The visualization of single-cell data often relies on dimension reduction methods such as UMAP to simplify high-dimensional single-cell data down to an understandable 2D form. However, these dimension reduction methods are not constructed to allow direct interpretation of the reduced dimensions in terms of cell differentiation. To address these limitations, we developed a new approach that places each cell from a single-cell dataset …
