Intervening proteins (inteins) are translated as subdomains within host proteins and removed through an intein-driven splicing reaction where the flanking sequences (exteins) are joined with a peptide bond. Previously, we developed a self-removing translation reporter for labeling Ebola virus (EBOV). In this reporter, an intein (RadA) containing the fluorescent protein ZsGreen (ZsG) is inserted within the EBOV protein VP30. Upon VP30-RadA-ZsG expression from the viral genome, RadA-ZsG is removed from VP30 through the protein splicing activity of RadA, generating functional, non-tagged VP30 and functional ZsGreen. While incorporation of our VP30-RadA-ZsG fusion reporter into recombinant EBOV (rEBOV- RadA-ZsG) resulted in an …

Depth Patterns Of Redear Sunfish Lepomis Microlophus In Kentucky Lake, Justin Graben

Redear Sunfish Lepomis microlophus are a highly sought sportfish known for their size potential, but their ecological behavior in large reservoirs remain poorly understood. In this study, we looked at how Redear Sunfish Lepomis microlophus move vertically in Kentucky Lake. We used acoustic tags with depth sensors to measure the movement of Redear Sunfish both diurnally and seasonally within Kentucky Lake.

During the day, Redear Sunfish occupied significantly deeper waters compared to dawn, dusk, and night (p < 0.0001.). Many fish, including the species in the Lepomis genus, may have a preference for specific light levels for optimal foraging and predation avoidance, even if predation is not the primary concern at larger sizes. This daily pattern was consistent through most seasons, except on average the fish were deeper during the winter season (p < 0.0001).

I have developed an algorithm for finding phase transition in microcanonical data of simulations of homopolymers. These simulations model the behavior of homopolymers in an array of different conditions. This algorithm is generalized and can be applied to a variety of different systems. It works by taking repeated derivatives of the microcanonical entropy of the system and applying several filters, whose parameters are tuned algorithmically as well. Identification of phase transitions allows for the construction of a phase diagram.

The Moderating Role Of Aphantasia In The Link Between Emotion Regulation And Relationship Satisfaction, Brock Borchardt, Dilbur D. Arsiwalla

INSPIRE Student Research and Engagement Conference

The goal of this study was to examine if poor visual imagery (i.e., aphantasia) moderated the link between emotional regulation and relationship satisfaction. We had 314 participants report on a survey about visual imagery, emotional regulation, and relationship satisfaction. Our original hypothesis was supported using the overall emotion regulation scale as well as five of the six different subscales of emotion regulation. However, further research is needed to better understand the rationale behind why aphantasia moderates this relationship.

We investigate the effects of cosmopolitan and traditional county features on the opposition to three successful liberal state-wide initiatives on abortion and recreational marijuana using county-level data. Cosmopolitan/traditional variables include urban/rural living patterns, college graduates, average income and religiosity while control variables include turnout, poverty, race, region, and union support. We observe the results of these three elections to see the relationship between vote shares and factors such as income, education, rural/urban, and religiosity, controlling for other factors. This study investigates the Culex quinquefasciatus is a major vector of West Nile virus, lymphatic filariasis, and arboviruses, which represent public health risk. Widespread application of pyrethroid insecticides has increased resistance in mosquito populations threatening the effectiveness of control strategies. This study aims to (1) assess deltamethrin resistance in Culex quinquefasciatus populations across the Abu Dhabi Emirate, (2) characterize the microbial communities associated with these mosquitoes. In the United States, the clover root curculio, S. hispi­dulus (F.) is the major species associated with lucerne (alfalfa) Medicago sativa L. This insect has until recently remained obscure as a pest of this major forage crop. Field and glasshouse experiments indicate that several nematodes damage ryegrasses and white clover (Cook and York, 1985). Populations of these nematodes under grassland are often at potentially damaging densities. Several nematode species of long-term grassland have co-evolved host relationsh­ips in which host resistance is likely to have been selected. The most important warm-season perennial grass in the southern United States is bermudagrass (Cynodon dactylon [L.]). Fall armyworms (Spodopterafrugiperda [J.E. Smith]) are the most damaging insect pests of bermudagrass. Conse­quently, researchers have evaluated improved bermudagrass varieties for resistance to fall armyworm (Leuck et al., 1968; Lynch et al., 1983; Quisenberry and Wilson, 1985; Jamjanya and Quisenberry, 1988). The UK market for seed of white clover (Trifolium repens L.) cultivars is in the order of 1,000 t within an EEC total of some 3,500 t. Although high yields of around 500 kg ha-1 can be achieved, yields are extremely variable (Hides, Lewis and Mar­shall, 1984) and this may be due in part to pathogenic attack. The results reported here are of a field experiment where dis­eases and pests were controlled by pesticide applications in plots grown under two simulated seed production man­agements.

Fusarium wilt of alfalfa (Medicago sativa L.) was first described in 1927 (Weimer, 1928) in the United States and has since been reported from many other areas of the world. The causal agent is Fusarium oxysporum Sehl. f. sp. medicaginis (Weimer) Snyd. & Hans. Shoots of infected plants become yellow and wilt ; often only one side of the plant is affected. Commonly, white clover stands decline within 2 to 3 years after seeding. Les cultures de ray-grass anglais porte-graine couvrent envi­ron chaque annee en France 2800 hectares, soit un peu mains de 20 % des surfaces nationales toutes graminees confondues. Une enquete realisee sur l'etat sanitaire de ces cultures avait revele qu'elles sont souvent attaquees par des rouilles au debut de l'ete. Breeding Stylosanthes for resistance to anthracnose diseases in northern Australian pastures is complicated by extensive pathogenic variation in the fungal pathogen, Colletotrichum gloeosporiodes. We have put considerable emphasis on epidemiological studies in an attempt to devise optimal strategies for the use of resistance genes in our selection and breeding programs. In addition, techniques of molecular biology are being used to investigate prospects for the use of genes from novel sources and to study the genetics of virulence in the pathogen.

Bacterial wilt of fodder grasses caused by Xanthomonas campestris pv. graminis was first described by Egli et al. (1975), and since then observed all over Europe and in New Zealand. The regular mowing of the crop offers ideal conditions for spreading of the pathogen. Several grass species are affected. The most severe yield losses occur in Lolium multiflorum, particularly in var. westerwoldicum. In extreme cases, all the plants can be killed during hot, dry summers. Des l'epiaison, Jes inflorescences malades sont emprisonnees clans un manchon blanc (stade ase­xue Sphacelia typhina = Acremonium typhinum), qui prend progressivement une couleur orangee, pour donner le symp­t6me typique de quenouille. The rhizoplane and root interior support different popula­tions of microorganisms. A large number of these organisms are microfungi. Studies have shown that fire induced changes are favourable for some groups of microorganisms following grasslands fires. However, most studies have dealt with the effect of burning on surface and subsurface soil organisms but none have examined rhizoplane fungi. This study was conducted to examine little bluestem (Schiz­achyrium scoparium), a native prairie grass species, biomass production and associated vesicular-arbuscular mycorrhizae (V AM) and rhizoplane mycoflora and their response to burning.

Infection of tall fescue (F. arundinacea Schreb.) plants by the endophytic fungus A. coenophialum Morgan-Jones & Garns is associated with substantial economic reduction in grazing ani­mal performance (Bacon and Siegel, 1988 ; Bush and Burrus, 1988 ; Stuedemann and Hoveland, 1988). This problem is serious in the transition zone of the eastern U.S.A., affecting over 12 million ha of pasture land.
