The Impact Of Diet-Induced Obesity On Cerebrovasculature Structure And Function, Riley M. Feagle
The Impact Of Diet-Induced Obesity On Cerebrovasculature Structure And Function, Riley M. Feagle
MUSC Theses and Dissertations
Adequate cerebral perfusion is necessary to help the brain maintain a constant and steady state of nutrient delivery. Since the brain is highly metabolic, changes in the vasculature structure can impact perfusion levels and, therefore, reduce the ability of the brain to receive the support necessary for full functionality. Vascular dysfunction is a common comorbidity among obese individuals. Given the strong association between obesity and cognitive dysfunction, understanding the correlation between reduced cerebral perfusion and altered cerebrovascular structure is crucial. The overall hypothesis of this thesis originated after ample preliminary data, and states that diet-induced obesity reduces cerebral perfusion and …

Background and Purpose: Peripheral nerve stimulation is approved by the US Food and Drug Administration for treating various disorders, but it is often limited by side effects, highlighting the need for a clear understanding of fascicular and fiber organization to design selective therapies. Micro-CT imaging of contrast-stained nerves enables the visualization of tissue microstructures, such as the fascicular perineurium and vasculature. In this work, we evaluated phosphotungstic acid (PTA) as a contrast agent and assessed its compatibility with downstream histology. Methods: Human vagus nerve samples were collected from three embalmed cadavers and subjected to three different staining methods, followed by …
A Computational Framework For Investigating Novel Bacterial Conjugation Inhibitors, Serena Tuytschaevers
A Computational Framework For Investigating Novel Bacterial Conjugation Inhibitors, Serena Tuytschaevers
Theses
Antimicrobial resistance (AMR) is a major global threat, contributing to an estimated 700,000 deaths annually. One of the main ways that AMR genes spread among bacteria is through conjugation. Therefore, it is crucial to develop conjugation inhibitors (COINs) to combat AMR. Shared characteristics between known COIN molecules are a hydrophobic tail, unsaturations, and a polar head. Considering these characteristics and accounting for the efficacy of 2-hexadecynoic acid and Tanzawaic Acids (TZAs) low toxicity, we designed a new set of TZA analogs. The predicted protein target of previously identified COINs is TrwD, an ATPase in the type IV secretion system (T4SS). …

This Article demonstrates that nested tiers of interchangeable parts serve as the foundation for the regulatory programs administered by the U.S. Food and Drug Administration. Core interchangeable parts used in FDA-administered programs include product categories, prohibitions on adulteration and misbranding, agency-registration requirements, current good manufacturing practices, product standards, marketing authorization requirements, postmarket requirements, and user fees. In each of these areas, Congress or the FDA has engaged in interchangeable-part lawmaking (IPL). IPL occurs when a government takes a portion of its law in one subject area and uses it as a model for its own law in another subject area. …

Current strategies for active targeting in the brain are entirely based on the effective interaction of the ligand with the orthosteric sites of specific receptors on the blood-brain barrier (BBB), which is highly susceptible to various pathophysiological factors and limits the efficacy of drug delivery. Here, we propose an allosteric targeted drug delivery strategy that targets classical BBB transmembrane receptors by designing peptide ligands that specifically bind to their transmembrane domains. This strategy prevents competitive interference from endogenous ligands and antibodies by using the insulin receptor and integrin α

Dukunolide D From The Root Of Lansium Domesticum Corr. Cv Kokosan, Angela Suryana, Andi Hairil Alimuddin, Rudiyansyah Rudiyansyah
Chimica et Natura Acta
Lansium domesticum has three varieties and one of the varieties is kokosan. These plants contain limonoids as the main constituents. Phytochemical investigation of L. domesticum cv kokosan has identified onoceranoids-type triterpenoids and tetranortriterpenoids from the seeds, fruit peels, and bark except for the root. This research described dukunolide D (1) that has been obtained from the methanol extract of the root of kokosan. Methanol root extract was fractioned by n-hexane and dichloromethane (DCM). All fractions including the crude extract were screened for phytochemical test. The DCM fraction was chromatographed in order to obtain compound 1. DART-HRMS of compound 1 showed …

Two-cell-like cells (2CLCs), a rare population (∼0.5%) in mouse embryonic stem cell (mESC) cultures, are in a transient totipotent-like state resembling that of 2C-stage embryos, and their discovery and characterization have greatly facilitated the study of early developmental events, such as zygotic genome activation. However, the molecular determinants governing 2C-like reprogramming remain to be elucidated. Here, we show that ZBTB24, CDCA7, and HELLS, components of a molecular pathway that is involved in the pathogenesis of immunodeficiency, centromeric instability, and facial anomalies (ICF) syndrome, function as negative regulators of 2C-like reprogramming by maintaining DNA methylation of the Dux cluster, a master …

Hierarchical Lineage Tracing To Unravel Mechanisms Of Cancer Treatment Resistance, Rachel Danielle Saxe
Hierarchical Lineage Tracing To Unravel Mechanisms Of Cancer Treatment Resistance, Rachel Danielle Saxe
Dartmouth College Ph.D Dissertations
Cancer cells adapt to treatment, leading to the emergence of clones that are more aggressive and resistant to anti-cancer therapies. We have a limited understanding of the development of treatment resistance as we lack technologies to map the evolution of cancer under the selective pressure of treatment. To address this, we developed a hierarchical, dynamic lineage tracing method called FLARE (Following Lineage Adaptation and Resistance Evolution). We use this technique to track the progression of acute myeloid leukemia (AML) cell lines through exposure to Cytarabine (AraC), a front-line treatment in AML, in vitro and in vivo. We map distinct cellular …

Soybean meal (SBM) is commonly used in aquafeeds due to its wide availability, reasonable protein content, and cost-effectiveness. However, high SBM inclusion levels in the diets of carnivorous fish, such as Atlantic salmon (Salmo salar), can cause soybean meal-induced enteritis (SBMIE), resulting in compromised gut health, reduced nutrient absorption, and impaired growth. An 8-week study was conducted to evaluate the potential of brewers’ yeast-derived functional feed additives (FFAs), specifically yeast cell wall β-glucans (PβG [purified β-glucan]) and yeast cytosolic extracts (YEs), to mitigate the adverse effects of SBMIE in Atlantic salmon parr. Fish were fed diets containing 30% SBM (30-SBM) …

While ECS provides prospective parents with insight into their genetic risk, it also raises serious ethical questions for many.
Impacts Of Reef Fish Length On Foraging Radius: Exploring Behavioral Patterns Within A Patch Reef Ecosystem, Kilia Brawand
Impacts Of Reef Fish Length On Foraging Radius: Exploring Behavioral Patterns Within A Patch Reef Ecosystem, Kilia Brawand
Student Research Symposium
Introduction
Coral reefs support high levels of marine biodiversity
- Patch reefs serve as vital refuges for small fish.
- Predation risk shapes how far fish stray from their reef, creating a “foraging radius”— the maximum distance traveled before retreating.
Absolute swimming speed in fish increases with body size (1).
-
Therefore, larger fish may travel farther away from their refuge to forage over greater areas
This study tested the following question: Does a reef fish’s length influence how far it travels from its patch reef refuge?
Investigation Of Immune/Performance Trade-Offs In Neonate Children's Pythons, Ellie Hartman
Investigation Of Immune/Performance Trade-Offs In Neonate Children's Pythons, Ellie Hartman
Student Research Symposium
Introduction
Energetic trade-offs are a key consideration when understanding an organism’s life history. One of these tradeoffs occurs between investment in immunity and performance.
- Individuals that invest more energy into immunity may be less equipped to avoid predation or find food, while those prioritizing performance may be more susceptible to infection
- Hypothesis: In response to a novel antigen, neonate snakes will have an immune response we can measure by counting immune cells. Additionally, we predict that strike frequency, swim speed, and endurance will be reduced in snakes undergoing an immune response and that with greater immune response we will see …

Evaluating The Effects Of Oxidative Stress On Primary Retinal Pigment Epithelial Cells, Sophia Hessami
Student Research Symposium
Introduction
Age-Related Macular Degeneration (AMD) is the leading cause of vision loss worldwide, affecting 200 million adults over the age of 40
- Affects subretinal layers: retinal pigment epithelium (RPE), Bruch’s membrane, and choroid (Fig. 1)
Problem: Early mechanisms of AMD are not well understood; oxidative stress is often cited as a key contributing factor to the development of AMD.3 Current studies neglect to use primary RPE cells when investigating oxidative stress.
Oxidative Stress occurs when there is an imbalance between reactive oxygen species (ROS) production and cellular antioxidant defenses
- Overabundance of ROS results in damage to tissue and …
