The present thesis is a compilation of a literature review, journal manuscript, and two standard operating procedures (SOP) on the qualitative determination of zinc in fortified cereal grain products and matrices. The literature review addresses the premise of the research as well as potential solutions for the development of a rapid qualitative spot assay for zinc determination. Quality assurance assays affirm the presence of micronutrients in food fortification initiatives, but no rapid qualitative spot methods for the assessment of zinc currently exist. Chromogenic reagents that may perform a key role in the rapid determination of zinc are identified. The journal …

Background: Mycobacterium avium (M. avium) is a nontuberculous mycobacterium (NTM) that can cause pulmonary and extrapulmonary infections mostly in immunocompromised individuals, such as those with HIV and diabetes. Traditionally, rifampicin (RIF) and azithromycin (AZ) have been used for a 12-month duration as first-line antibiotics against M. avium. Due to the increased multidrug resistance, novel ways, such as enhancement of macrophages response, are needed to provide adequate immune response required to clear M. avium infection.

Incubation and parental care are essential behaviors for the healthy development of offspring. Stressors such as migration and disease can affect parental care resulting in altered egg survival and hatch rates. Therefore, varying individual fitness and life history events may lead to changes in reproductive fitness across a population. We examined the relationship between incubation behavior and blood parasite prevalence, migratory strategy (migrant or resident), mass, and first egg date in a population of American Kestrels (Falco sparverius) in southwest Idaho to determine influences on incubation behavior. Artificial food dye is added to foods to enhance their color and appearance for consumers. This systematic review aims to review the literature on Red Dye No. 40 and its effects on non-human animals. Studies were included if (a) exposure to Red Dye No. 40 was the independent variable, (b) nonhuman animals were the subjects, and (c) at least one behavioral, neurological, or physiological dependent variable was reported. The macroinvertebrate microbiome controls various aspects of the host's physiology, from regulation of environmental contaminants to reproductive output. Aquatic insects provide critical nutritional subsidies linking aquatic and riparian food webs while simultaneously serving as a contaminant pathway for riparian insectivores in polluted ecosystems. Previous studies have characterized the transport and transfer of contaminants from aquatic to riparian ecosystems through insect metamorphosis, but both contaminant exposure and metamorphosis are energetically intensive processes that may cause host microbiomes to undergo radical transformation in structure and function, potentially affecting the host's physiology. Aqueous two-phase systems (ATPS) are a liquid–liquid extraction method that offers low-cost, continuous-adaptable virus purification. A two-step ATPS using polyethylene glycol (PEG) and sodium citrate that recovered 66% of infectious porcine parvovirus with 2.0 logs of protein removal and 1.0 logs of DNA removal in batch has now been run continuously. The continuous system output of < 10 ng/mL DNA regardless of starting DNA titer agreed with batch studies. However, the continuous system had a five-fold higher contaminating protein titer than batch studies, likely because of incomplete mixing or settling. Turbidity was tested as a measure of mixing and settling efficiency. Monitoring in-line absorbance at 880 nm directly after mixing and before collection in the settling reservoir could track both mixing and settling during operation. Settling time was reduced by changing the settling line material from PVC to PTFE, which is more hydrophobic. A flow-through AEX filter tested to make impurity removal more robust recovered 90% of PPV and removed an additional 87% of host cell DNA. The filter did not add any additional protein removal. In the future, in-line absorbance sensors could be implemented along with conductivity sensors to measure salt concentration, refractive index sensors to track the PEG-citrate interface, and scales to track mixer and reservoir volumes to enable automated, continuous ATPS. Our vision is to integrate continuous ATPS into a fully continuous end-to-end production for viral vectors.

Tree mortality can have cascading effects on the composition and resilience of interacting fungal communities. Yet, little is known about the impact tree mortality, distinct from abiotic drivers (i.e., drought), has on these communities. This dissertation focuses on the resilience of fungal communities to experimental tree mortality in a piñon-juniper woodland. In chapter one, I found that piñon mortality reduced piñon fine root biomass and piñon mycorrhizal fungal diversity, whereas juniper mycorrhizal fungi were unaffected by juniper mortality. In chapter two, I revealed that piñon mortality altered the composition and spatial structure of mycorrhizal and saprotrophic fungi in the soil, …

Regenerating conifers after harvest through planting and postharvest broadcast application of herbicide is effective in ensuring the survival and growth of seedlings, but faces challenges in meeting broad social and ecological objectives of forest management. This study reports the effectiveness of alternative options in regenerating jack pine (Pinus banksiana Lamb.), 21 years after harvest of trembling aspen (Populus tremuloides Michx.)-dominated boreal mixedwood stands. Lynch Syndrome and Constitutional Mismatch Repair Deficiency are human diseases implicated in mutations of DNA mismatch repair (MMR) genes. This experiment tested a mutation of an MMR gene, MSH6, and evaluated how the mutation affected overall MMR effectiveness. Using the yeast Saccharomyces cerevisiae, we performed the CAN1 forward mutation assay to study msh6-L696F and its implications in the MMR process. We hypothesized that there would be a significant change in molecular function in the Msh6 protein in the presence of this mutation. Carbohydrate active enzymes, or CAZymes, are enzymes that are important for the breakdown of carbohydrates. Fungi produce many of these enzymes as they cycle essential nutrients. For this study, we compared the abundance of CAZymes families among fungal genomes from the taxonomic orders Capnodiales, Dothideales, Eurotiales, Filobasidiales, Pleosporales, Tremellales, and Ustilaginales that were isolated from different sources including plant, soil, and food, among others. Microbes play an essential role in plant ecology, supporting growth, pathogen protection, and drought tolerance. Big sagebrush (Artemisia tridentata), a foundational plant species, hosts a diverse microbial community, but the benefits of its leaf microbiome remain unclear. We conducted an experiment to determine if the bacterial species, Bacillus amyloliquefaciens, may promote sagebrush growth and survival in a restoration setting. Seeds from Hillside to Hollow (HH) and Polecat Gulch (PC) were grown in a greenhouse, and once leaves emerged, the seedlings were randomly assigned to receive an inoculum containing B. amyloliquefaciens or a sterile water control. Fine-scale environmental features display variation between close proximity habitats. These differences can impact the behavior of species in these environments. One local example is the Piute Ground Squirrel (Urocitellus mollis). They live in sagebrush steppe environments that vary in amount of ground cover. The ground squirrel population has endured a steady decline in recent years and, to understand why they may not be recovering from this decline, we evaluated ground squirrel phenology in open and closed habitats by observing ground squirrel activity. Purpose/Background Perinatal Mood and Anxiety Disorders (PMADs), including postpartum depression, anxiety, and psychosis, significantly impact maternal and infant health. This scoping review consolidates current literature on PMADs, emphasizing prevalence, risk factors, screening, and treatment. The primary aim is to identify research gaps and inform clinical guidelines to enhance perinatal care.
Methods A literature search followed PRISMA guidelines from August 2022 to November 2024. Hard-bodied ticks are known to carry a wide range of viruses, some of which can cause human diseases. In the U.S., the tick Amblyomma americanum is an important disease vector. The tick has recently expanded its distribution range and its abundance is rising in many areas of the U.S. Its virome is, however, largely underexplored. In this preliminary study, we used a Nanopore MinION sequencer to detect and identify the viral fauna in nymphal ticks (n=43) collected in Statesboro, GA. Science fiction (SF) films are typically some of the most popular movies that are released each year. They tend to be blockbuster phenomena, raking in millions or even billions of dollars. To determine the source of transfer in third language acquisition (L3A), we tested the effects of grammatical feature typology and L2 Spanish proficiency on the comprehension and production of grammatical [gender] and [number] in the early stages of L3A of Portuguese. We distinguish between the two features based on their participation in the lexical-conceptual structure of the lemma and its interaction with the morpho-syntactic derivation. L1 English speakers were tested on their knowledge of the features in both their L2 and their L3 through a grammaticality judgment task and an elicited production task. Women of reproductive age (18-44 years) are at an increased risk of developing obesity due to pregnancy, stressors during this stage in the life course (e.g., leaving home, new jobs, parenthood), and historical and ongoing discrimination and marginalization. Preventing obesity for this population needs to be a public health priority. Obesity negatively affects women’s health (e.g., chronic disease, cancer); leads to poor pregnancy outcomes (e.g., gestational diabetes); and increases the risk children will develop obesity through intergenerational transmission of obesity, further entrenching health disparities for future generations. Advancing personalized medicine depends on effectively integrating and interpreting the vast, heterogeneous landscape of biological data, from genomic sequences and transcriptomics to the insights embedded in scientific literature. Current machine learning models often focus on single data modalities, limiting their capacity to capture the multifaceted nature of biological systems. We address this gap by developing three attention-based machine-learning models integrating diverse data modalities. Firstly, DeepVul is a multi-task model that leverages cancer transcriptome data to predict genes critical for cancer survival and their corresponding drugs. Ontologies have gained popularity in the scientific community as a way to standardize terminologies in organizations’ data. Although certain cohorts have created frameworks with rules and guidelines on creating ontologies, there exist significant variations in how Materials Science ontologies are currently developed. We seek to provide guidance in the form of a unified automated framework for developing interoperable and modular ontologies for Materials Data Science that simplifies the ontology terms matching by establishing a semantic bridge up to the Basic Formal Ontology(BFO). Neotropical nectar bats in cloud forest ecosystems must navigate and forage in dark, complex landscapes containing an incredible diversity of flowering plants. Despite these challenges, nectar bats are extremely successful at exploiting nectar resources from plants. Much of the sensory ecology behind these bat-plant interactions has been investigated and we know from previous studies that echolocation and olfaction are critical sensory modalities to nectar bats. Echo-acoustically conspicuous floral and vegetative structures as well as sulfur-containing volatile compounds function as signals, allowing bats to sense plants. Nitrogen (N) is an essential macronutrient required by all plants to complete their life cycles. Inefficient N uptake contributes to over fertilization and subsequent environmental and sustainability issues resulting in the need for improved nitrogen use efficiency (NUE). Chenopodium quinoa is a resilient pseudocereal crop that is relatively adapted to N-deficient soils. Previous studies have uncovered the unique genetic structure of quinoa and its physiological responses to N availability; however, gaps remain in understanding its molecular mechanisms for NUE. Epidemiological models delineate the spread of diseases within a population. In this research project, the Susceptible-Exposed-Infected-Recovered (SEIR) Model was examined numerically for comparison between several methods. Approximations were obtained through Euler’s Method, Taylor’s Method, Runge-Kutta Methods, and Multi-step Methods. Hypothetical situations with parameter alterations were considered in order to better understand the effects the parameters have on the model. The goal of this project was to portray the usefulness of numerical approximations for predicting the behavior of the SEIR model and thus the course of a pandemic.

Rising atmospheric CO₂ and CH₄ drive climate change, prompting research into their electrochemical conversion to valuable products. This study investigates electrodeposited nanostructured transition metal oxides (TMOs), unary (CuO, Co₃O₄) and dual (Cu-Co oxide) as catalysts for CO₂ reduction and CH₄ oxidation under ambient conditions. Fabricated on various substrates (Cu foil, SS316, carbon paper) and characterized by SEM, their electrochemical performance was evaluated via cyclic voltammetry.
Cu-Co oxide showed superior CO₂ reduction, leveraging the strengths of individual oxides. Co₃O₄ exhibited higher current densities for CH₄ oxidation, with SS316 proving the best substrate. Catalyst composition, morphology, and substrate significantly influenced efficiency. While …

Metastatic uveal melanoma is an aggressive disease with poor outcome, which is refractory to immune checkpoint inhibitors. A T cell receptor (TCR)-based CD3 bispecific, tebentafusp, delivers clinical benefit in patients with metastatic uveal melanoma. Understanding the molecular basis for the anti-tumor activity of tebentafusp in an indication where checkpoint inhibitors are ineffective could aid in identification of other solid tumor indications where CD3 bispecifics may serve an unmet need. Applications of isoscapes in terrestrial and marine ecosystems have grown notably as a tool for studying animal movement. However, isoscapes are rarely used in freshwater ecosystem because current iterations of isoscapes commonly model baseline distributions of bulk stable isotope analysis (SIA) data, which may be inadequate for providing the necessary resolution for fine spatial scale stream environments. This issue could potentially be addressed using compound specific stable isotope analysis, a new method that measures the isotopic composition of individual compounds to provide more comprehensive information than bulk SIA.
