The rapid growth of multi-omics datasets and the wealth of biological knowledge necessitates the development of effective methods for their integration. Such methods are essential for building predictive models and identifying drug targets based on a limited number of samples. We propose a framework called GNNRAI for the supervised integration of multi-omics data with biological priors represented as knowledge graphs. Our framework leverages graph neural networks (GNNs) to model the correlation structures among features from high-dimensional 'omics data, which reduces the effective dimensions in data and enables us to analyze thousands of genes simultaneously using hundreds of samples. Primary cilia are hair-like structured cell organelles functioning as cellular antennae with chemosensory and mechanosensory roles. Dysfunction of cilia results in ciliopathies, including renovascular diseases (polycystic kidneys, hypertension, aneurysm), mental impedances (Alzheimer's disease, Parkinson's disease, dementia), metabolic diseases (obesity, developmental skeleton defects), blindness, and so on. With a diameter of 150 nm, cilia can be cleaved, released and circulated in the body as injury biomarkers. We here assessed the potential use of isolated cilia as a novel delivery vehicle. Despite growing recognition of persons with disabilities (PWDs) as an underserved population, healthcare disparities persist, often exacerbated by provider misconceptions and lack of training. This study evaluated the impact of a two-part educational module consisting of a didactic session and a panel discussion with individuals with disabilities focused on healthcare students’ knowledge and comfort in caring for patients with intellectual and developmental disabilities (IDDs). Conducted at the University of South Dakota, the sessions included pre- and post-surveys assessing attitudes, confidence, and perceived competencies. Freshwater ecosystems in East Texas contain a rich freshwater biodiversity, with the Neches and Sabine River basins supporting some of the greatest freshwater fish diversity in the state. Given the heterogeneity of habitats and species diversity, these rivers serve as excellent natural systems for examining the ecological processes shaping biodiversity patterns and species distributions. In 2023-2024, 60 streams in the Neches and Sabine River basins were surveyed across three seasons to investigate the patterns and ecological drivers of taxonomic (TDβ), functional (FDβ), and phylogenetic β-diversity (PDβ) facets of freshwater fish assemblages. Taxonomic identification of closely related plants can be challenging due to convergent evolution, hybridization, and overlapping geographic distribution. To derive taxonomic relationships among planted and wild plants across a large geographic range, we complemented three standard plastid barcodes , , and with soil and fruit chemistry, soil microbiome, and plant morphology analyses. Soil and plant sampling included planted from manicured sites in Southern California, USA, wild plants from Southern and Northern California, and wild populations from Mediterranean island of Hvar, Croatia. This data analysis examined associations between cardiovascular and muscular fitness indicators and the presence of cardiovascular disease (CVD) in older adult athletes. Using cross-sectional data from 1,110 participants aged 50 and older who completed the Sustained Athlete Fitness Exam (SAFE) at state-level Senior Games events, we evaluated both objective performance metrics and self-reported exercise behaviors. Cardiovascular measures included systolic blood pressure (SBP), waist circumference (WC), waist-to-hip ratio (WHR), and body mass index (BMI). Patients with visible skin diseases, including acne, psoriasis, and atopic dermatitis, face heightened risks of psychological distress, depression, and suicidal ideation, often exacerbated by social stigma and reduced quality of life. This review explores the intersection between dermatology and suicide, highlighting the psychosocial impact of visible skin conditions and the controversies surrounding treatment-related risks, including isotretinoin and biologic exposure. Current evidence suggests that while isotretinoin has been historically linked to mood changes, robust studies do not support a direct association with suicide. Phages are the most abundant life-forms on the planet. Due to their unique ability to infect and lyse specific bacterial strains, phages have been proposed as a potential therapeutic agent to fight antibiotic resistance bacterial infections. Consequently, more research is required to further our understanding of phages. We were provided with an isolated, novel Mycobacterium bacteriophage: Bellis. Utilizing a bioinformatic approach, we successfully annotated Bellis’ genome using a variety of computer programs like GeneMark, Glimmer, HHpred, and PECAAN. We discovered that Bellis has 122 genes, including a RecB-like endonuclease and an AAA-ATPase, as well as 10 tRNAs. Through the genomic …

Several previously unknown phages have been discovered including Stargaze, whose bacterial host is Mycobacterium Smegmatis. Adverse Childhood Experiences (ACEs) are strongly linked to long-term health outcomes, yet discussions around ACEs in clinical settings remain inconsistent. This study investigates the comfort levels of healthcare providers in addressing ACEs with their patients, identifying barriers and facilitators to such conversations. Using a Quasi-experimental, single group, pretest-post test experimental design, we surveyed a diverse group of physicians, nurse practitioners, and physician assistants working in a primary care setting, providing ACE training, and an opportunity for learning reflection. Purpose: Postmenopausal osteoporosis (PMOP) is the most prevalent metabolic bone disease among women, characterized by significant bone density loss and increased fracture risk. With a genetic component, a systematic review was conducted on the association between genetic polymorphisms and PMOP risk. Methods: A comprehensive review of PubMed literature examined genetic polymorphisms linked to PMOP risk. The primary outcome was to identify the most frequently studied genes linked to PMOP. There are 129 red-flowered species in western North America that fit the hummingbird pollination syndrome. Theory predicts that these flowers should produce a specific red color that is conspicuous to hummingbirds but not to bees. In a few well-studied cases, red flowers can be produced with pelargonidin, cyanidin, or both—sometimes complemented with the addition of yellow carotenoids and sometimes not. Which pigment combinations are most common in California and whether they affect conspicuousness to hummingbirds and bees remain unknown. The present thesis aims to determine the effects, if any, of six potential moderators on the effects of acute moderate-intensity exercise on cognitive functioning. The modes of exercise undertaken by the participants included treadmill (Experiment 1) and stationary bike (Experiment 2) exercise. Participants in each experiment either exercised or rested for twenty minutes and then participated in either a memory recall task or cognitive functioning (“Stroop”) task. In Drosophila melanogaster, the mechanism influencing the differentiation of glia and neurons is directed by a key transcription factor known as glial cells missing (gcm). gcm is a crucial regulator of cell fate decision, functioning as a binary switch to direct glial fate in gcm- positive cells and neuronal fate in gcm-negative cells (Jones, et.al, 1995; Hosoya, et.al, 1995; and Vincent, et.al, 1996). Total knee arthroplasty (TKA) is a common orthopaedic procedure used to alleviate pain and restore mobility in patients with knee pain. Substantial resources are committed to updating and refining new knee implant designs to improve outcomes. The aim of this study was to compare revision rates and failure modes between newer implants and their older counterparts.

Methods

A systematic literature review of national registries was conducted to identify predecessor and successor implant designs from four manufacturers. Deciphering the “genotype to phenotype” map is a crucial goal of biology. We will not improve our ability to predict phenotypes from DNA sequences until we better incorporate epigenetic and environmental variation into our models of human health and disease. Genes are typically presumed to express both parental alleles equally, but genetic and epigenetic factors can cause one allele to function differently than the other (e.g. expressed or silenced more). These allele-specific imbalances can alter the final composition of a gene’s transcriptional product and ultimately shape phenotypes. Cancer remains a leading cause of mortality worldwide, with treatment resistance and recurrence posing major barriers to long-term survival. Despite advancements in targeted therapies, many cancers develop resistance to chemotherapy, radiation, and immunotherapy, significantly limiting their effectiveness. One key driver of therapeutic resistance is nuclear factor erythroid 2-related factor 2 (NFE2L2/NRF2), a transcription factor that regulates antioxidant responses and drug detoxification. Transposable elements (TEs) contribute to nearly half of the human genome yet have been overlooked in genomic medicines due to the stereotypic impression as “junk DNA” and technical challenges. However, studies in the last two decades demonstrated that dysregulation of the epigenome in cancer cells resurrects the cis-regulatory functions of TEs, including the promoter activities. Of the protein structures deposited in the Protein Data Bank, less than half have pockets suitable for the binding of drugs. Even when proteins contain pockets in their ground state structures (e.g., the nucleotide-binding active site in myosin motors), achieving specificity remains a central challenge in drug design as many protein families share common structural motifs. Cryptic pockets are cavities absent in ligand-free experimental structures that form due to protein fluctuations in solution. Tens of millions of Americans suffer from metabolic disorders, predisposing patients to cardiovascular disease, kidney disease, liver disease, infertility, and cancer. Unfortunately, the etiology of many of these diseases remains a mystery, limiting our ability to identify targets for pharmacologic therapies. Due to the relationship between altered lipid dynamics and metabolic dysfunction, we decided to study the effect of the posttranslational lipid modification, S-palmitoylation, on metabolism. Infant leukemias arise as either B cell acute lymphoblastic leukemia (B-ALL) or acute myeloid leukemia (AML) and are primarily driven by MLL/KMT2A-rearrangements (MLLr). Prior work has shown that MLLr transform fetal/neonatal hematopoietic progenitors more efficiently than adult progenitors. However, the genetics and epidemiology of MLLr leukemias present an important paradox. MLLr arise during late gestation under conditions that would seemingly favor leukemic transformation (i.e., fetal/neonatal progenitors have high proliferation rates and self-renewal). Furthermore, MLLr infant leukemias require very few cooperating mutations for transformation. Mayer and Salovey, seminal researchers of emotional intelligence (EI), indicated that the ability measures of EI are still evolving, and the mental abilities involved in EI remain to be determined.
