With age, hematopoietic stem cells can acquire somatic mutations in leukemogenic genes that confer a proliferative advantage in a phenomenon termed CHIP. How these mutations result in increased risk for numerous age-related diseases remains poorly understood. We conduct a multiracial meta-analysis of EWAS of CHIP in the Framingham Heart Study, Jackson Heart Study, Cardiovascular Health Study, and Atherosclerosis Risk in Communities cohorts (N = 8196) to elucidate the molecular mechanisms underlying CHIP and illuminate how these changes influence cardiovascular disease risk. We functionally validate the EWAS findings using human hematopoietic stem cell models of CHIP. We then use expression quantitative …

Acute myeloid leukemia (AML) resides in an immune-rich microenvironment, yet, immune-based therapies have faltered in eliciting durable responses. Bridging this paradox requires a comprehensive understanding of leukemic interactions within the bone marrow microenvironment. We optimized a high-throughput tissue-microarray-based pipeline for high-plex spatial immunofluorescence and mass cytometry imaging on a single slide, capturing immune, tumor, and structural components. Using unbiased clustering on the spatial K function, we unveiled the presence of tertiary lymphoid-like aggregates in bone marrow, which we validated using spatial transcriptomics and an independent proteomics approach. We then found validated TLS signatures predictive of outcomes in AML using an …

Turkish Journal of Zoology

To date, the global introduction of alien amphibians and reptiles has likely been facilitated by the expanding plant nursery trade. Here, we report in detail a singular case of unintentional introduction of Lissotriton vulgaris in Sicily. A newt in the final stages of its larval development was found in a private garden pond in north-western Sicily, where aquatic plants from a nursery in Veneto (northeastern Italy) had been introduced two months earlier. The analysis of the mithochondrial markers ND2 and ND4 revealed a shared haplotype between the Sicilian newt and a sample from Treviso, the province of the putative source. …

Genetic Algorithms From A Biblical Perspective, Richard L. Halterman Phd May 2025

Genetic Algorithms From A Biblical Perspective, Richard L. Halterman Phd

The Journal of Biblical Foundations of Faith and Learning

The dominant opinion expressed by the scientific community is that life on earth as we observe it today evolved over millions of years from more primitive organisms. Its adherents insist that naturalist forces drive this process, and it is unaided by any form of intelligent oversight. In contrast, a biblical worldview upholds the Bible’s account of a six-day special creation by an all-powerful Creator. The competition between these two worldviews appears unresolvable. Complete resolution would require time travel to absolutely certify the winner. Indirect evidence can sway opinions, but individuals do not always apply sound logical reasoning when forming their …

The Business Of Business: A Biblical Approach To The Purpose Of The Firm, Abraham Johannes Oberholster May 2025

The Business Of Business: A Biblical Approach To The Purpose Of The Firm, Abraham Johannes Oberholster

The Journal of Biblical Foundations of Faith and Learning

The debate on the purpose of the business corporation has been raging for more than a century. Inspired by capitalistic thinking, economists, corporate managers and investors have mostly maintained that the predominant goal for corporations is to maximize shareholder wealth. In contrast, society has argued that the corporation also has a social responsibility as a legal entity and community member.

The legal precedent set in 1919 with Dodge v. Ford Motor Co. is that “a business corporation is organized and carried on primarily for the profit of the stockholders”. The oft quoted argument of Milton Friedman (1970) that the social …

A Relational God And Why Worldview Self-Examination Is A Crucial Component Of Christian Education, David R. Nelsen Ph.D May 2025

A Relational God And Why Worldview Self-Examination Is A Crucial Component Of Christian Education, David R. Nelsen Ph.D

The Journal of Biblical Foundations of Faith and Learning

Christian education seeks to transform the lives of its students, preparing them to serve both in their present life and in the life to come. At the center of this process are the students and how they currently think, act, and make sense of the world around them i.e., their worldview. Thus, preparing students for service both here and in the hereafter requires that students become aware of and take control of their worldview systematically and deliberately. Ultimately, this facilitates their ability to form a lasting and transformative relationship with God. Here I present a biblical foundation for why students …

Engineering Hiv-1 Env Protein To Elucidate In Situ Structural Dynamics Of Env, Narendra Kumar Gonepudi May 2025

Engineering Hiv-1 Env Protein To Elucidate In Situ Structural Dynamics Of Env, Narendra Kumar Gonepudi

Biotechnology Theses

The HIV-1 Envelope (Env) glycoprotein trimers undergo structural changes upon interacting with host receptor CD4 and coreceptors (CCR5/CXCR4) for virus entry. As the only surface-exposed viral protein, Env is a key antibody target but evades recognition through structural changes. Studying Env dynamics with respect to the coreceptor binding region (V3 loop) provides insights into virus entry and immune evasion. Fluorescence labeling via genetic code expansion (amber–TAG suppression) offers a minimally invasive approach for fluorescence microscope-based Env dynamics studies. Here, two V3-related dual-amber Env constructs (A135TAG-P308TAG, P308TAG-E395TAG) were generated by incorporating two non-canonical …
