Patient responses to immune checkpoint inhibitors can be influenced by the gastrointestinal microbiome. Mouse models can be used to study microbiome-host crosstalk, yet their utility is constrained by substantial anatomical, functional, immunological and microbial differences between mice and humans. Here we show that a gut-on-a-chip system mimicking the architecture and functionality of the human intestine by including faecal microbiome and peristaltic-like movements recapitulates microbiome-host interactions and predicts responses to immune checkpoint inhibitors in patients with melanoma. The gut microbiota has emerged as a critical determinant of immune checkpoint inhibitor (ICI) efficacy, resistance, and toxicity. Retrospective and prospective studies profiling the taxonomic composition of intestinal microbes of patients treated with ICI have revealed specific gut microbial signatures associated with response. By contrast, dysbiosis, which can be caused by chronic inflammatory processes (such as cancer) or comedications, is a risk factor of resistance to ICI. Recent large-scale meta-analyses have confirmed that antibiotic (ATB) use before or during ICI therapy alters the microbiota repertoire and significantly shortens overall survival, even after adjusting for prognostic factors. Bispecific antibodies (bsAbs) have emerged as a novel class of therapeutics, offering a dual-targeting strategy to enhance the therapeutic efficacy of monoclonal antibodies, which is often limited by tumor heterogeneity and the occurrence of resistance mechanisms. By simultaneously engaging two distinct antigens or pathways, bsAbs disrupt multiple signaling cascades simultaneously, preventing escape mechanisms and offering a more durable response. Furthermore, they can optimize immune activation, improving immune cell recruitment strategies. In particular, T-cell engager bsAbs facilitate immune cell-mediated tumor destruction by linking T cells to tumor antigens. Instead, dual immune checkpoint inhibitors (CPIs) enhance immune activation by blocking inhibitory signals. …

The origins of immunosuppression, neutropenia, and anemia in patients with chronic lymphocytic leukemia (CLL) are not fully understood. Because in patients with CLL, circulating exosomes, which participate in cell-to-cell interactions, are CLL cell-derived, we examined whether those exosomes contribute to abnormal features of this disease. Our data revealed that CLL cell-derived exosomes engulfed by healthy donors’ monocytes, fibrocytes, and lymphocytes altered target-cell gene and protein expression and suppressed normal hematopoiesis. CLL cell-derived exosomes increased normal monocytes’ CD14 and CD16 expression such that it mimicked the accessory cell profile and upregulated T cells’ checkpoint PD-1 and CD160 protein levels, potentially reducing …

Ovarian cancer remains one of the leading causes of cancer-related mortality among
women, highlighting the need for novel therapeutic agents with improved selectivity and
reduced toxicity. Anticancer peptides (ACPs) have emerged as promising candidates
due to their ability to selectively target cancer cells. In this study, the anticancer activity
of a computationally designed ACP was evaluated in vitro using the SKOV-3 ovarian
cancer cell line, with HEK cells employed as a non-cancerous control.
Cell viability was assessed using the MTT assay, and cytotoxicity was further confirmed

When land that was once covered with vegetation and soil becomes dominated by impervious concrete and rooftops, the amount of water infiltrated by the ground plumets. As a result, most suburban areas contribute to the impairment of streams by adding excess water, nutrients, and pollutants to nearby waterways each time it rains. One harmful result of this is the erosion of stream banks. The rapid influx of water that occurs after rain events can destabilize and erode stream banks, resulting in excess sediment in the water. Parents’ etiological explanations for youth mental health problems are relatively understudied; most studies focus on adult beliefs about the causes of mental disorders experienced by adults. This is concerning given research demonstrating that parents’ causal beliefs may influence several aspects of youth mental health service utilization including whether parents seek help for their youth and how they engage in treatment. Post-germinative growth in plants is controlled by sucrose mobilization from the endosperm. Regulation of this process, however, is not fully understood. Transcriptional suppression of the rice vacuolar H+ translocating pyrophosphatase (V-PPase) referred to as VPP5 led to enhanced starch content in mature grains. However, vpp5 suppressed lines displayed a slower post-germinative growth rescuable by sucrose, indicating inefficient sucrose mobilization from the endosperm. Biochemical and gene expression analyses showed a slower rate of starch to sucrose conversion and altered expression of the key starch utilization pathway genes. The Density Of Virgin, Consumer And Environmental Plastics: An Investigation Using Gas Displacement Pycnometry, Andrew Turner, Katie Jones, Montserrat Filella

Density is a fundamental property of plastics and is particularly significant in determining the transport and fate of waste plastics that enter aquatic systems. However, densities are rarely determined in the environmental literature and values employed for modelling or impacts are often unsourced or derived from secondary databases. In this study, we employ helium displacement pycnometry to determine the skeletal densities of non-porous plastics whose polymer composition had been established from manufacturer's data, resin codes or Fourier transform infrared spectrometry. Two independent, collaborative laboratories, providing measurements within 3.5 % of each other and with precisions of <1 % (as relative standard deviation), analysed a total of 42 virgin, consumer and environmental plastics consisting of ten common polymer types. Measured densities of plastics in all categories, and most notably for polybutylene terephthalate, polyethylene terephthalate, polypropylene and polyvinyl chloride, were often outside the ranges reported by a comprehensive online resource. Possible reasons for discrepancies include the occurrence of dense additives (evaluated by X-ray fluorescence analysis), the presence of inaccessible microscopic pores below a laminated surface, contamination of the main polymer by a secondary one, and structural changes on weathering. Regardless of precise causes, most results suggest that individual polymers have a broader range of densities than is generally published or considered in the literature. Accordingly, and in particular where buoyancy is critical, more precise, sample-specific measurements are recommended.

Since the development of the three-dimensional (3D) "mini-gut" culture system, adult stem cell-derived organoid technology has rapidly advanced, providing in vitro models that replicate key cellular, molecular, and physiological properties of multiple organs. Background
Multiple myeloma (MM) is virtually always preceded by monoclonal gammopathy of undetermined significance (MGUS). Elevated serum markers are used to classify MGUS patients into clinical risk categories. Previous research has indicated that the absence of a normal plasma cell signature in MGUS is linked to early progression.
Objective
To confirm that the presence of a “polyclonal plasma cell (PolyPC) signature” serves as a robust negative predictor of MGUS progression.

DNA methylation is a crucial epigenetic marker used in various clocks to predict epigenetic age. However, many existing clocks fail to account for crucial information about CpG sites and their interrelationships, such as co-methylation patterns. We present a novel approach to represent methylation data as a graph, using methylation values and relevant information about CpG sites as nodes, and relationships like co-methylation, same gene, and same chromosome as edges. We then use a graph neural network (GNN) to predict age. Thus our model, GraphAge leverages both the structural and positional information for prediction as well as better interpretation. AIMS: Patients hospitalized with acute heart failure (AHF) treated with a 7 day prednisone course in the CORTAHF pilot trial had a greater improvement in health-related quality of life (QoL) at Day 7 in both the overall population and in patients with baseline interleukin 6 > 13 pg/mL. This post-hoc analysis examines the specific QoL domains and the relationship between clinical signs of congestion and QoL.
METHODS: In the CORTAHF pilot trial, patients with AHF and high-sensitivity C-reactive protein (hsCRP) > 20 mg/L were randomized 1:1 to once-daily oral 40 mg prednisone for 7 days plus usual care or usual care alone. …

OBJECTIVE: The claustrum is a small, thin structure of predominantly gray matter with broad connectivity and enigmatic function. Little is known regarding the impact of claustrum pathology in multiple sclerosis (MS).
METHODS: This study assessed whether claustrum volume was reduced in MS and whether reductions were associated with specific disability domains. We compared the impact of claustrum volume reductions on neurological disability versus the well-studied thalamus. Data from the 15,793 participants in the MS PATHS study were used for primary analyses. A second cohort of 71 people with MS and healthy controls from our center was used for validation.

BACKGROUND: World Health Organization Grade 2 meningiomas (G2Ms) often recur and resist therapies. Grade 2 meningiomas with histopathological necrosis have been associated with worse local control (LC) after radiation therapy, but the drivers and biomarkers of radiation resistance in G2Ms remain unknown.
METHODS: We performed genetic sequencing and histopathological analysis of 113 G2Ms and investigated the role of genetic and microenvironmental factors on clonogenic survival after ionizing radiation. We performed transcriptional profiling of our in vitro model and 18 human G2M tumors by bulk RNA sequencing as well as 8 G2Ms by single nuclei RNA sequencing.

Objective
This study aimed to compare the quality and readability of the responses generated by 3 publicly available artificial intelligence (AI) chatbots in answering frequently asked questions (FAQs) related to Oral and Maxillofacial Radiology (OMR) to assess their suitability for patient education.
Study Design
Fifteen OMR-related questions were selected from professional patient information websites. These questions were posed to ChatGPT-3.5 by OpenAI, Gemini 1.5 Pro by Google, and Copilot by Microsoft to generate responses. Three board-certified OMR specialists evaluated the responses regarding scientific adequacy, ease of understanding, and overall reader satisfaction. Arab, Middle Eastern, and North African Americans experience various forms of racial–ethnic trauma in the United States, including historical and intergenerational trauma (IGT). The population has been migrating to the United States since the 1800s, and the number of migrants has risen in recent decades due to war, persecution, and family reunification. As a result, the children of these migrants may be exposed to IGT, with research suggesting that descendants of individuals who have experienced trauma are more susceptible to poor mental health outcomes. Habitat is an important driver in shaping the distribution of a range of species. For demersal fish species, benthic habitat characteristics can drive distribution and community structure. Understanding the relationship between habitat and species distribution in important, especially for exploited species such as rockfishes (Sebastes spp.). Baby kale (Brassica oleracea and other species) is a vital commodity as a salad green in spring mix and is widely produced on the Central Coast of California. Downy mildew, caused by Hyaloperonospora brassicae, is a devastating disease infecting economically important crops like cabbage, broccoli, cauliflower, horseradish, rapeseed, kohlrabi, Brussels sprouts, and kale. There is a growing need to predict the complex hydrologic and hydraulic effects of beaver-maintained wetlands. This study uses relatively simple inputs and a replicable methodology to predict flood flow impacts of beaver habitat restoration at the site design scale using the engineering industry’s customary hydrology and hydraulic modeling approach. This study focuses on Toro Creek, the primary drainage for a 15.2 square mile watershed on the California Central Coast.

Marine heatwaves (MHWs) are increasing in frequency and intensity globally, with grave effects on marine ecosystems and their dependent industries. There are significant gaps in knowledge of the subsurface behavior and geographic distribution of MHWs, especially in eastern boundary upwelling systems like the California Current. Here, two decades of full water-column temperature observations from a shallow autonomous profiler in San Luis Obispo Bay (~10m) revealed that while MHWs have similar average durations and intensities across all depths, one-third of bottom MHWs occur without a concurrent surface signal. Northern elephant seals (Mirounga angustirostris) are large, ocean migrating carnivores that have a wide distribution along the west coast of North America. Like other pinniped species, northern elephant seals spend considerable time out at sea foraging and return to land to molt and breed. Northern elephant seals were hunted to near extinction in the late 1800s. Due to conservation efforts they have since rebounded, and they currently occupy 27 known terrestrial breeding colonies. Current computational tools for analyzing chromatin organization are mainly focused on intrachromosomal interactions, despite growing evidence that suggests long-range interactions across chromosomes contribute to transcriptional regulation and disease development. This thesis aims to address this gap in interchromosomal genome analysis, presenting a robust computational pipeline that identifies a clique (i.e., a subgraph) of highly interacting trans-chromosomal regions anchored at a user-specified seed genomic locus. A weighted interaction network is constructed from an input Hi-C contact matrix, a widely used experimental assay for measuring genome-wide chromatin interactions. Since the industrial entrance of polymer plastic materials, plastic has become ubiquitous in both everyday use and waste.
