Voltage-gated sodium channels are essential for initiating action potentials in neurons. Mutations in the human brain sodium channel hNaV1.1 ion channel are associated with a large number of neurological disorders, such as epilepsy. Despite its clinical relevance, there is still much to characterize about the specific effects of how these mutations affect sodium channel behaviour, including the role of the smaller beta subunits. To investigate this, hNaV1.1 channels were expressed with varying beta subunits (beta 1,beta 2, or none) in Xenopus oocytes. The Columbian Sharp-tailed Grouse (Tympanuchus phasianellus; hereafter CSTG) formerly occupied shrub-steppe ecosystems across western North America but now inhabit less than five percent of their former range due to habitat loss and other anthropogenic factors. Despite their social and ecological significance, research on CTSG diet has been limited, with the last comprehensive study taking place over 20 years ago. Irrigated agricultural lands can provide important habitat and forage locations for wild birds. The costs and benefits of irrigated agricultural areas compared to wildlands remain unclear. Green vegetation attracts insects and small mammals, creating an abundant food source for predators. In addition, the use of chemicals on crops may reduce vectors of disease. American Kestrels (Falco sparverius) are a small species of falcon that make extensive use of both irrigated and non-irrigated landscapes. Emerging evidence suggests that cellular senescence contributes to the progression of neurodegenerative diseases, including Parkinson’s Disease (PD). Cellular senescence is a state in which cells cease to proliferate in response to damage and contribute to tissue dysfunction through the release of toxic factors. While dopaminergic neurons are the primary cells lost in PD, senescent glia: astrocytes and microglia, are considered to play a vital role in regulating neuronal health. Using the pre-formed fibril (PFF) of alpha-synuclein to induce PD-like pathology, we examined the impact of senolytic treatment on glial cell senescence. Transition metal PCO (P=C+O) complexes have potential uses to produce organophosphorus complexes in a much more efficient and sustainable manner. This project shows the synthesis, identification, and characterization of a Cu(I) PCO complex which is supported by a bulky N-heterocyclic carbene ligand (NHC*). This compound has been characterized via NMR (1H, 31P, 13C) and IR spectroscopy. Thermolysis of this compound shows the expulsion of carbon monoxide to generate a reactive Cu-P bond which appears to transfer the P-atom to the carbene ligand as gauged by NMR spectroscopy, as well as mass spectroscopy.

Purpose: GPKOW, a gene on the X-chromosome, encodes a nuclear RNA-binding protein important in messenger RNA (mRNA) processing as a spliceosome subunit. This work aims to establish GPKOW as a disease-associated gene.

Methods: We describe 3 males from 2 unrelated families with hemizygous frameshift variants affecting the last exon of GPKOW p.(Arg441SerfsTer30) and p.(Ser444GlufsTer28). The effect of p.(Ser444GlufsTer28) on gene expression was evaluated in patient's fibroblasts. In vivo studies in Drosophila melanogaster targeting the sole GPKOW fly ortholog, CG10324 (Gpkow) were performed.

Results: Clinical presentations included intrauterine growth restriction, microcephaly/microencephaly, and eye, brain, skin, and skeletal abnormalities. Heterozygote females presented …

A 28-GHz multibeam joint communication and sensing system called SideSense is presented, in which a line-of-sight (LoS) beam is used to maintain reliable communication, while other sensing beams are used to enhance physiological motion detection. SideSense decodes the motion frequency and shape from the channel state information (CSI) by first tuning the gain ratio and phase differences between the LoS communication beam and non-LoS (NLoS) beam to maximize the sensing signal-to-noise ratio (SSNR) without significantly degrading the communication channel capacity (CCC). Targeting drugs to cancer cells via overexpressed cell-surface receptors has emerged as an effective therapeutic strategy for several cancers. However, identifying cell-surface receptors that allow selective uptake of targeting ligands by cancer cells—while sparing normal cells—remains a challenge, especially for triple-negative breast cancer (TNBC), which lacks a well-defined receptor for targeted delivery. In this study, immunohistochemical (IHC) analysis revealed that human TNBC patient tissues have significantly higher levels of keratin 1 (K1) compared to normal breast tissues. Although many genes are subject to local regulation, recent evidence suggests that complex distal regulation may be more important in mediating phenotypic variability. To assess the role of distal gene regulation in complex traits, we combine multi-tissue transcriptomes with physiological outcomes to model diet-induced obesity and metabolic disease in a population of Diversity Outbred mice. Using a novel high-dimensional mediation analysis, we identify a composite transcriptome signature that summarizes genetic effects on gene expression and explains 30% of the variation across all metabolic traits. The signature is heritable, interpretable in biological terms, and predicts obesity status from gene expression in …

Introduction

Pharmacogenomics (PGx) is an emerging discipline with the potential to revolutionize personalized medicine, but its successful implementation requires interprofessional collaboration. To address this need, a virtual interprofessional education (IPE) session was designed for student pharmacists and medical students to engage in a case-based learning experience.

Objective

The primary objective was to develop and implement an IPE activity focused on a patient case requiring PGx-guided dual antiplatelet therapy and to assess students' perceptions of two Interprofessional Education Collaborative (IPEC) Version 3 Core Competencies. Procedural learning difficulties are commonly reported in children with developmental coordination disorder (DCD), yet the neural basis of this impairment remains unclear. This study addressed this gap by examining the correlation between cortical oscillatory activity and procedural learning of a sequence of finger movements in children with and without DCD. Participants were 19 children with DCD and 38 typically developing (TD) children, with a mean age of 10 years and 3 months. Children completed the Serial Reaction Time task, a standard measure of procedural learning, during which they unintentionally learned a sequence of finger movements. Background: Early childhood caries (ECC) is the most common chronic health condition among Australian preschool-aged children. Untreated, ECC can affect children's physical and emotional health and future social opportunities. ECC is largely preventable through primary caregivers' correct understanding of oral health promoting diets, oral hygiene practices and dental service engagement. The study aim is to explore Western Australian (WA) primary caregivers' child oral health knowledge, experience and practices. ZnO/SiC nanocomposite materials possess significant potential for various technological fields due to their extraordinary optical, electrical, thermal, and mechanical properties. The synthesis methods, material properties, and diverse applications of ZnO/SiC composites have been systematically explored in this study. The potential application areas of this nanocomposite include their roles in photocatalysis, optoelectronic devices, gas sensors, and photovoltaic systems. The synergetic effects of ZnO and SiC are analyzed to highlight their advantages over their individual components. Future research directions must focus on the remaining challenges to optimize these nanoscale composite materials for industrial and emerging applications.

Vapour pressure deficit (VPD) and blue light (BL) are import signals for stomatal regulation and highly relevant during cultivation of economically important crops such as cucumber in controlled environments. However, the information about combined effects of VPD and BL and the regulation of abscisic acid (ABA) in this respect, is limited and seems to vary between species. This study investigated how moderate (1.12 kPa) or low (0.28 kPa) VPD combined with 5% or 30% BL affect the foliar ABA metabolism, transpiration rate, nutrient uptake, and growth in well-watered cucumber plants (Cucumis sativus ‘Quatro’).
