In United States v. Skrmetti, the Supreme Court upheld Tennessee Senate Bill 1 (SB 1), a state law that prohibits transgender minors from accessing gender-affirming care. For the first time, the Court considered a fundamental question in civil rights law: How does the Fourteenth Amendment regard transgender people? The Court gave little guidance. It did not sort out principles for deciding when anti-transgender discrimination classifies by sex, nor whether transgender people are a suspect class. Frass-the main by-product of insect rearing for animal feed-is emerging as a promising soil amendment and plant growth promoter. However, basic agronomic information is lacking and prevents frass' widespread use as a biofertilizer. This study assessed impacts of black soldier fly (Hermetia illucens L. [Diptera: Stratiomyidae]) frass on soil fertility, crop growth, and quality compared to poultry litter (PL). Irrigated and non-irrigated soybean (Glycine max L. Merr.) and non-irrigated switchgrass (Panicum virgatum L.) plots received either PL (5.6 Mg ha(-1)), low frass rate (LF; 5.6 Mg ha(-1)), high frass rate (HF; 11.2 Mg ha(-1)), besides the unamended control (CT). Anthropogenic microfibres (AMFs) and microplastics (MPs) are pervasive and ubiquitous environmental contaminants of growing concern. However, their study in geosolids, like sediments, is often conducted in the absence of other important geochemical and anthropogenic markers. In the present study, surface and subsurface sediments from the Plym Estuary, southwest England, have been analysed for AMFs and MPs along with organic matter (as loss on ignition; LOI), calcium, iron, manganese and potentially toxic metals (PTMs). The latter were heterogeneously distributed throughout the estuary because of multiple inputs and variations in sediment granulometry. The capacity of crops to accumulate nanoplastics has remained relatively unexplored largely due to analytical constraints. To address this challenge, the present study employed a radiolabelling approach to demonstrate, for the first time, that polystyrene nanoplastics (14C-PS NPs) accumulate and move into the edible tissues of radishes (Raphanus sativus). Radishes were exposed hydroponically for five days, with only their non-fleshy roots in contact with the 14C-PS NPs, while accumulation in the fleshy (edible) roots and shoots was subsequently measured. In this study, we developed a piezoelectric-polydimethylsiloxane (pz-PDMS) composite by blending poly(vinylidene fluoride-co-trifluoroethylene) (P(VDF-TrFE)) with PDMS to create a biocompatible, mechanoelectrical responsive material. The pz-PDMS was synthesized with varying piezoelectric concentrations (0%, 1%, 3%, and 5%) and characterized for visual properties, mechanical properties, mechanoelectrical sensitivity, and biocompatibility. Compression testing showed no significant change in mechanical strength with the addition of piezoelectric particles, while mechanolectrical sensitivity testing revealed a non-linear increase in voltage response, with 5% pz-PDMS producing the highest sensitivity. Fatigue testing demonstrated no change in sensitivity after 7 days of cyclic displacement. Additionally, microcantilever experiments demonstrated the high fidelity …

A multi-modal anion exchange ligand 2-methyl-phenylimidazole has been successfully functionalized on highly porous electrospun regenerated cellulose membrane substrate for the separation of full and empty adeno-associated viral (AAV) capsids. The functionalized membrane has an average pore size of 0.5 mu m and exhibits both charge and hydrophobic functionality. The ligand is able to distinguish the subtle physio-chemical differences between the full and empty viral capsids. The multi-modal anion exchange membrane is able to separate the full capsids from affinity-captured mammalian AAV2 using both gradient and two-step elution methods with superior performance. Introduction: Higher cognitive reserve (CR) is associated with reduced dementia risk. We hypothesized that higher CR is associated with less baseline Alzheimer's disease (AD) pathology in the U.S. Study to Protect Brain Health Through Lifestyle Intervention to Reduce Risk (U.S. POINTER) cohort.
Methods: A subsample of participants underwent amyloid beta and tau positron emission tomography imaging. Regression analysis was used to model the association between educational attainment (EA) as a CR proxy measure, amyloid positivity, and entorhinal cortex (ERC) and meta-temporal region of interest (meta-ROI) tau standardized uptake value ratio (SUVR).

Purpose: XPO1 functions in key cellular processes, including nucleo-cytoplasmic export and mitosis. The gene is deleted in a subset of patients with the 2p15p16.1 microdeletion syndrome; however, no monogenic XPO1-related disorder has been described to date.
Methods: We collected clinical data of individuals with de novo XPO1 variants through online matchmaking. We used Drosophila to study XPO1 function in development and habituation learning.
Results: A total of 22 individuals met the criteria to be included in the main study cohort. Of these, half have putative loss-of-function variants, and half have coding variants (10 missense and 1 in-frame deletion variant). We …

Citrin deficiency (CD) is caused by the inactivation of SLC25A13, a mitochondrial membrane protein required to move electrons from cytosolic NADH to the mitochondrial matrix in hepatocytes. People with CD do not like sweets. Here we show that SLC25A13 loss causes the accumulation of glycerol-3-phosphate (G3P), which activates the carbohydrate response element-binding protein (ChREBP) to transcribe FGF21, which acts in the brain to restrain intake of sweets and alcohol and to transcribe key genes driving lipogenesis. Microbiota-derived metabolites have emerged as key regulators of longevity. The metabolic activity of the gut microbiota, influenced by dietary components and ingested chemical compounds, profoundly impacts host fitness. While the benefits of dietary prebiotics are well-known, chemically targeting the gut microbiota to enhance host fitness remains largely unexplored. Here, we report a novel chemical approach to induce a pro-longevity bacterial metabolite in the host gut. We discovered that wild-type Escherichia coli strains overproduce colanic acids (CAs) when exposed to a low dose of cephaloridine, leading to an increased life span in the host organism Caenorhabditis elegans.
