The stress response gene DDR48 has been characterized in Saccharomyces cerevisiae and Candida albicans to be involved in combating various cellular stressors, from oxidative agents to antifungal compounds. Surprisingly, the biological function of DDR48 has yet to be identified, though it is likely an important part of the stress response. To gain insight into its function, we characterized DDR48 in the dimorphic fungal pathogen Histoplasma capsulatum. Transcriptional analyses showed preferential expression of DDR48 in the mycelial phase. Induction of DDR48 in Histoplasma yeasts developed after treatment with various cellular stress compounds. We generated a ddr48∆ deletion mutant to further …

The Use Of Rna Interference To Investigate The Role Of Zmpr10 In Salt Stress Response And Mtl/Nld/Zmpla1 In The Improvement Of Haploid Induction In Maize, Max Carl Miller

LSU Master's Theses

RNA interference (RNAi) is a useful tool in gene characterization and its use is expanding into more practical fields. Two RNAi techniques were used in this case. The studies conducted in the chapter two use transgenic maize lines containing an RNAi construct targeting ZmPR10 to characterize its potential role in growth and development in response to salinity stress. T1 transgenic plants from events A44S1-35-3 and A44S1-3-4, which had significantly reduced ZmPR10 expression compared to the non-transgenic controls, showed no differences compared to the controls in heights after 30 and 60 days, nor did the T2 transgenic from these …

Parental Incarceration And Extracurricular Activities: The Effects On Children's Aggressive Behaviors, Danielle Webb Mcnichol

Student Theses and Dissertations

Previous literature has determined that exposure to parental incarceration increases the chances of children having behavior problems. Using data from the Fragile Families and Childhood Wellbeing Study, the identification of parental incarceration and how it impacts fragile families will be explored. This study focuses on how parental incarceration affects an adolescent’s social, emotional, and behavioral development and how that leads to behavioral problems and delinquency. Further, the current study will examine if involvement in extracurricular activities moderates the relationship between parental incarceration and behavior problems. Identifying how exposure to parental incarceration affects adolescents could help identify new and additional ways …

Toward A More Strategic National Stockpile, Troy Rule Nov 2021

Toward A More Strategic National Stockpile, Troy Rule

Texas A&M Law Review

The COVID–19 pandemic exposed major deficiencies in the United States’ approach to stockpiling for emergencies. States, cities, and hospitals across the country had meager inventories of critical medical items on hand when the pandemic first reached U.S. soil, and the federal government’s Strategic National Stockpile proved far too small to serve the country’s needs in the first several months of the crisis. As nationwide shortages spread, many state governments were compelled to bid against each other to procure scarce medical supplies—a distribution approach that disadvantaged low-income and minority communities and left countless healthcare professionals and staff ill-equipped to protect themselves …

Phenotypic plasticity is one mechanism whereby species may cope with stressful environmental changes associated with climate change. Reef building corals present a good model for studying phenotypic plasticity because they have experienced rapid climate-driven declines in recent decades (within a single generation of many corals), often with differential survival among individuals during heat stress. Underlying differences in thermotolerance may be driven by differences in baseline levels of environmental stress, including pollution stress. To examine this possibility, acute heat stress experiments were conducted on Acropora hyacinthus from 10 sites around Tutuila, American Samoa with differing nutrient pollution impact. A threshold-based heat …

Information on the prevalence of micronutrient deficiencies is needed to determine related disease burden; underpin evidence-based advocacy; and design, deliver, and monitor safe, effective interventions. Assessing the global prevalence of deficiency requires a valid micronutrient status biomarker with an appropriate cutoff to define deficiency and relevant data from representative surveys across multiple locations and years. The Global Burden of Disease Study includes prevalence estimates for iodine, iron, zinc, and vitamin A deficiencies, for which recommended biomarkers and appropriate deficiency cutoffs exist. Because representative survey data are lacking, only retinol concentration is used to model vitamin A deficiency, and proxy indicators …

School of Medicine Publications

Subcortical band heterotopia (SBH), also known as double cortex syndrome, is a malformation of cortical development caused by inherited or somatic gene variants. We present a case of a young adult with posterior SBH and electroclinical features of focal neocortical temporal lobe epilepsy. Genomic blood analysis identified a pathogenic somatic mosaicism duplication variant of the PAFAH1B1 gene. Despite bilateral cortical MRI abnormalities, the interictal and ictal EEG findings indicated a focal epileptogenic region in the left posterior temporal region. Chronic responsive cortical neurostimulation across two four-contact depth electrodes placed 5mm on either side of the maximal interictal spiking identified during …

Cultural Competence In The Baccalaureate Degree Nursing Curriculum, Angela E. Silvestri-Elmore

Theses

Health care providers are members of a helping profession and need to provide quality care to all members of society. As a result of current and projected demographic changes within the United States (U.S.), health care professionals are faced with the challenges of providing culturally competent care and fulfilling the role as the "helping profession." In the past 10 years, minority populations have increased in the U.S. For example, the African American population experienced an approximate 12.3% increase, and the Hispanic population increased by 43%. Just as it is necessary for health care professionals to respond to the increase in …

Deconstructing Sex Differences In C-Reactive Protein Trends Over Time, Ashley W. Kranjac, Dinko Kranjac, Olivia Lounsbury

Sociology Faculty Articles and Research

Objectives

Heightened inflammatory state, as measured by circulating C-reactive protein (CRP) levels, can promote inflammation-mediated disease risk. It is important to account for population fluctuation and sex variation in serum CRP concentrations on overall time trends.

Methods

Using the National Health and Nutrition Examination Survey data, we specify linear and algebraic decomposition models separately by sex to identify the drivers of the changing trends in the distribution of CRP values in the population.

Results

We found a nonsignificant overall increase in CRP, but a significant decrease among women and increase among men, over a 10-year period. We then used linear …

The Contribution Of Racism-Related Stress And Adversity To Disparities In Birth Outcomes: Evidence And Research Recommendations, Sabrina R. Liu, Laura M. Glynn

Psychology Faculty Articles and Research

Currently, racial and ethnic differences in adverse birth outcomes and infant mortality are some of the largest and most persistent health disparities in the United States. This narrative review article synthesizes existing literature to present a conceptual model of how racism-related stress and adversity are critical determinants of such disparities. We describe how historical and ongoing racism has created conditions wherein women of color are disproportionately exposed to chronic, multilayered stress and adversity and how the biological consequences of exposure to these stressors confers risk for adverse birth outcomes. Next, we identify important priorities and considerations for future research, including …

Early theories of efficient coding suggested the visual system could compress the world by learning to represent features where information was concentrated, such as contours. This view was validated by the discovery that neurons in posterior visual cortex respond to edges and curvature. Still, it remains unclear what other information-rich features are encoded by neurons in more anterior cortical regions (e.g., inferotemporal cortex). Here, we use a generative deep neural network to synthesize images guided by neuronal responses from across the visuocortical hierarchy, using floating microelectrode arrays in areas V1, V4 and inferotemporal cortex of two macaque monkeys. We hypothesize …

Tera-Seq: True End-To-End Sequencing Of Native Rna Molecules For Transcriptome Characterization, Fadia Ibrahim, Jan Oppelt, Manolis Maragkakis, Zissimos Mourelatos

Department of Biochemistry and Molecular Biology Faculty Papers

Direct sequencing of single, native RNA molecules through nanopores has a strong potential to transform research in all aspects of RNA biology and clinical diagnostics. The existing platform from Oxford Nanopore Technologies is unable to sequence the very 5′ ends of RNAs and is limited to polyadenylated molecules. Here, we develop True End-to-end RNA Sequencing (TERA-Seq), a platform that addresses these limitations, permitting more thorough transcriptome characterization. TERA-Seq describes both poly-and non-polyadenylated RNA molecules and accurately identifies their native 5′ and 3′ ends by ligating uniquely designed adapters that are sequenced along with the transcript. We find that capped, full-length …

The heterogeneous family of complexes comprising Polycomb repressive complex 1 (PRC1) is instrumental for establishing facultative heterochromatin that is repressive to transcription. However, two PRC1 species, ncPRC1.3 and ncPRC1.5, are known to comprise novel components, AUTS2, P300, and CK2, that convert this repressive function to that of transcription activation. Here, we report that individuals harboring mutations in the HX repeat domain of AUTS2 exhibit defects in AUTS2 and P300 interaction as well as a developmental disorder reflective of Rubinstein-Taybi syndrome, which is mainly associated with a heterozygous pathogenic variant in CREBBP/EP300. Moreover, the absence of AUTS2 or mutation in its …

Development Of A Novel Algorithm To Remove Spurious Edges From Biological Networks Through Functional Enrichment, Jeffrey Page Nov 2021

Development Of A Novel Algorithm To Remove Spurious Edges From Biological Networks Through Functional Enrichment, Jeffrey Page

Theses

The field of systems biology has facilitated the modelling of large and complex biological networks. These networks, generated from prior knowledge contained in the corpus of medical and scientific literature, or from experimental data are being used to model differing macromolecule networks associated with distinct disease states. While these networks are vital in understanding disease pathology and possible treatment options, they are rife with spurious interactions. These interactions arise from the methods used to create such networks, where the ability to discriminate between direct and indirect relationships is a challenge. To combat these spurious interactions an algorithm that leverages functional …

Breeding And Feeding Habitat Selection By An Island Endemic Bird May Increase Its Vulnerability To Climate Change, Paul M. Radley, Eddie J. B. Van Etten, David Blake, Robert A. Davis

Research outputs 2014 to 2021

Characterizing patterns of habitat use is an important first step for effective conservation planning. Species restricted to low-lying islands are at greatest risk from climate change-related sea level rise, and requirements for breeding and foraging habitat may determine their risk from tidal inundation. The endangered Micronesian Scrubfowl (Megapodius laperouse senex) is a model species for understanding these impacts. This species faces the cumulative challenges of tourist visitation, invasive species, and rising sea levels, yet little is understood about its habitat use in the Rock Islands Southern Lagoon Conservation Area (RISL) of Palau. We studied the habitat requirements of this mound-nesting …

Creativity In Science, Engineering, And The Arts: A Study Of Undergraduate Students' Perceptions, Dildora F. Beaulieu

Dissertations and Theses

Creativity is widely recognized as being invaluable for human development and a crucial 21st century talent. Preparing students for an uncertain and complex world requires that higher education promote students' imagination, originality, curiosity, and flexibility and build their capacity to take risks to try new approaches to problem-posing and problem-solving. However, little is known about how undergraduates enrolled in different disciplines view creativity. This quantitative study at a university in the northwestern United States assessed how undergraduate students in different academic disciplines responded to an instrument on creativity measurement developed by Dlouhy (2012). The study asked: How do undergraduates …

Pelagic Tunicate Grazing On Marine Microbes Revealed By Integrative Approaches, Kelly R. Sutherland, Anne W. Thompson

Biology Faculty Publications and Presentations

Marine microorganisms comprise a large fraction of ocean carbon and are central players in global biogeochemical cycling. Significant gaps remain, however, in our understanding of processes that determine the fate, distribution, and community structure of microbial communities. Protists and viruses are accepted as being part of the microbial loop and a source of microbial mortality. However, pelagic tunicates (salps, doliolods, pyrosomes, and appendicularians), which are abundant in oceanic and coastal environments and consume microorganisms with higher individual grazing rates than other common grazers, remain underappreciated in their role controlling microbial communities, distributions, and flux through ecosystems. In spite of sampling …

Characteristics And Causal Factors For The Receptive-Expressive Gap In Typically Developed Spanish-English Adult Bilinguals, Rachael Janelle Javaherian

LSU Master's Theses

A receptive-expressive language gap occurs when a standardized receptive score is statistically greater than an expressive score in either a primary or a second language. In bilingual children, gap studies already exist for determining language impairment. In adults, there are few studies; nevertheless, one needs to distinguish between typical language patterns of bilingualism due to non-pathological loss of L1 skills versus problems resulting from accidents, disease, or age. To this end, the present study attempted to expand findings of studies by Gibson et al. (2012 and 2014) that focused on analysis of the gap and possible factors influencing its existence …
