Chulalongkorn University Theses and Dissertations (Chula ETD)
This dissertation is research on the development of biosensors using different types of bioreceptors and nanomaterials for the diagnostic application of various diseases. The research is separated into two parts. The first part is about the development of an enzyme-free impedimetric sensor based on molecularly imprinted polymer (MIP) for the selective determination of L-hydroxyproline (L-hyp), a biomarker of bone diseases. It was found that the use of a mixture containing 3-aminophenylboronic acid (3-APBA) functions as a monomer and o -phenylene diamine (OPD) as a co-monomer can generate the specific MIP through electropolymerization. The detection is based on the increase of …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Vibrio parahaemolyticus causing Acute hepatopancreatic necrosis disease (VPAHPND) has been a major cause of serious shrimp aquaculture economic losses. Moreover, many isolates have exhibited the resistance of various antibiotic drugs. Most recently, bacteriophage or phage, the killer of bacteria, has been gaining interest as an alternative tool to prevent and control pathogens. In this study, we discovered two novel vibriophages - vibriophage Seahorse and vibriophage phiKT1024 – that target various strains of VPAHPND. vibriophage Seahorse and vibriophage phiKT1024 were assigned to the family Siphoviridae and Myoviridae, respectively. Based on the cut-off of genome size that is less than 200 kbp …

Development Of Paper-Based Electrochemical Immunosensor For Sars-Cov-2 Detection, Jutamas Jaewjaroenwatana
Chulalongkorn University Theses and Dissertations (Chula ETD)
This dissertation aimed to develop a simple and low-cost electrochemical sensing platform for detecting SAR-CoV-2 antigen. The proposed sensor combined the innovative disposable paper-based immunosensor and cost-effective plant-based anti-SARS-CoV-2 monoclonal antibody, extracted from Nicotiana benthamiana. The cellulose nanocrystal was modified on screen-printed graphene electrode to provide the COOH functional groups on electrode surface, leading to the high ability for antibody immobilization. The quantification of the presence receptor binding domain (RBD) spike protein of SARS-CoV-2 was performed using differential pulse voltammetry by monitoring the changing current of [Fe(CN)6]3-/4- redox solution. The current change of [Fe(CN)6]3-/4- before and after the presence of …

Delayed Lateral Flow Immunoassay For Determination Of Covid-19 Antigen, Pawanrat Srithong
Chulalongkorn University Theses and Dissertations (Chula ETD)
The lateral flow immunoassay (LFIA) strip is one of the most attractive biosensing platforms for a point-of-care testing (POCT). However, the generally lack of sensitivity and inability to perform quantitative analysis has consequently limited its outreaches for more challenging applications, where detecting low analytic concentrations is required. To overcome such drawbacks, we propose a facile and straightforward alternative method utilizing a delayed hydrophobic barrier fabricated with trimethylsilyl cellulose (TMSC) to improve its sensitivity. The SARS-CoV-2 spike receptor-binding domain (SARS-CoV-2 SP RBD) antigen was chosen as a model analyte to demonstrate the superior detectability of this scheme. The novel device consists …

Development Of Diagnostic Methods And Epidermiology Of Tuberculosis In Free-Ranging Long-Tailed Macaque Macaca Fascicularis And Rhesus Macaque M. Mulatta In Thailand, Suthirote Meesawat
Chulalongkorn University Theses and Dissertations (Chula ETD)
Cross-species infectious disease transmission, especially between humans and wildlife, is increasing during the global warming. Tuberculosis (TB) is an air-borne infectious disease that can be transmitted from humans to non-human primates (NHPs), however, no report of TB prevalence in free ranging NHPs that highly interacted with humans in Thailand was published. Thus, this study was conducted and separated into 3 phases. Phase I, active TB in 1,836 free-ranging macaques including 6 rhesus (Macaca mulatta; Mm), 23 common long-tailed (M. fascicularis fascicularis; Mff), and 3 Burmese long-tailed (M. f. aurea; Mfa) macaque populations throughout Thailand was assesses. Monkeys were captured, anesthetized, …
Glycosylation Of N-Pentenyl Glycoside Activated By Bromodiethylsulfonium Salt, Supanat Buntasana
Glycosylation Of N-Pentenyl Glycoside Activated By Bromodiethylsulfonium Salt, Supanat Buntasana
Chulalongkorn University Theses and Dissertations (Chula ETD)
n-Pentenyl glycosides (NPGs) are a significant class of glycosyl donors with dual functionality, serving as both protecting groups for the anomeric hydroxyl and leaving groups upon activation with halonium activators such as N-bromosuccinimide (NBS) or N-iodosuccinimide (NIS). However, the activation of less reactive glycosyl donors with these reagents has proven to be sluggish and inefficient. Recently, bromodiethylsulfonium bromopentachloroantimonate (BDSB) has emerged as a highly efficient brominating reagent that can be readily prepared and purified through crystallization. In this study, we explored the potential of BDSB as a halonium activator for NPGs in chemical glycosylation reactions. By using BDSB as the …
Seek, Picture Insect, Google Lens: An Analysis Of Popular Insect Identification Apps Using Photos Of Realistic Quality, Morgan Manderfield
Seek, Picture Insect, Google Lens: An Analysis Of Popular Insect Identification Apps Using Photos Of Realistic Quality, Morgan Manderfield
Department of Entomology: Distance Master of Science Projects
Photo-based identification apps for mobile devices have grown in popularity across many fields of organismal biology. While studies have shown that these apps show promise in fields such as botany and ornithology, there is much skepticism surrounding their accuracy and usefulness in entomology. While providing these apps with photos of high quality will undoubtedly lead to the highest accuracy rates, often users submit photos of average quality, e.g., in poor lighting, out of focus, from far away, etc. To quantify realistic accuracy rates of photo-based insect identification apps, this study analyzed three popular apps – Seek, Picture Insect, and Google …

Integrated Pest Management (IPM) is a holistic strategy for the management of pest species. It is a framework that relies on scientifically informed decisions to identify and remediate a pest problem in a way that minimizes harm to the environment, human health, and the economy (Dara, 2019). The term pest is broadly defined not just as a nuisance, but as any organism that has detrimental effects to a resource or public health and that exists at a population level that exceeds a threshold of economic damage. This encompasses invertebrates (such as insects, nematodes, and mites), vertebrates, plant pathogens, diseases, and …

A Guide To Agricultural Insect Pests On The East Coast, Devan Mazza-Mcdonald
Department of Entomology: Distance Master of Science Projects
A Guide to Agricultural Insect Pests on the East Coast is a useful resource comprised of fact sheets of ten invasive pests that have been introduced to the United States. It also includes a fact sheet of three important pollinator species that are of economic and environmental importance. Most of the pests I selected are currently an issue in states on the East coast and in some instances, further west as well. Currently, the khapra beetle is not a problem pest in the United States but does have a possibility of becoming an issue, again. The invasive pests I chose …

The Collegiate Black Space: Black College Students’ Use Of New Counter-Spaces For Support, Knowledge Production, And Organizing For Activism, Heather Streets
Doctoral Dissertations
Black collegians who attend historically white institutions continue to struggle with racism, microaggressions, feelings of alienation, minimal or improper advising, and an undue pressure to prove themselves (Bonner, 2010; Feagin & Sikes, 1995; Strayhorn, 2010). These barriers to success result in part due to a lack of support from the colleges and universities that they attend (Allen, 1992; Parker, Puig, Johnson & Anthony, Jr., 2016). With institutional benefits designed to benefit white students over students of color, Black students must find their own alternatives for collaboration and to provide support for their peers.
Many Black spaces can be defined as …

Gc/Ms And Lc/Ms Approaches To Chemical Communication In Plants And Marine Cyanobacteria, Carolyn Keim
University of the Pacific Theses and Dissertations
Natural products are small organic compounds synthesized by living organisms including plants, animals, and microbes. These fall into one of two categories, primary metabolites are required for survival, but secondary metabolites usually play a more accessory role. Extracts from organisms have proven to be useful to humans throughout history. Prior to evidence-based western medicine practices, natural products were the only source of medicine and remain as a treatment source for underdeveloped nations. Today, natural products have either been approved by or have served as the template for many FDA approved drugs. Based on a survey of all FDA approved drugs …

The Effect Of Acute And Chronic Arsenic Exposure On Malignant Transformation Of The Human Bronchial Epithelial Cell (Beas-2b), Bandar Almutairy
Wayne State University Dissertations
Arsenic (As3+), a metalloid abundant in the environment, is classified as a group I carcinogen associated with several common human cancers, including cancers in the lung, skin, bladder, liver, and prostate (Wei, Zhang & Tao, 2019b). The mechanisms of As3+-induced carcinogenesis had been extensively studied, and different mechanisms might be involved in various types of cancer (Wei, Zhang & Tao, 2019b). Recent studies showed that exposure to a high dose of arsenic is able to induce lung cancer. Moreover, arsenic activates oncogenic signaling pathways such as MAPKs, EGFR/RAS/RAF, PI3K/AKT, and JNK/STAT3 pathways as well as epigenetic alterations such as miRNAs …

Vaginal Microbiota Of Recurrent Bacterial Vaginosis Patients Under Oral Metronidazole Therapy: Prognosis Based On Specific Species Of Gardnerella And Other Bacterial Vaginosis Associated Species In Association With The Vaginal Microbiome, Ashomathi Ashok Sudhamathi Mollin
Wayne State University Theses
Bacterial Vaginosis (BV) is a state of vaginal dysbiosis in which BV-associated species displace the typically dominant species of Lactobacillus. This urogenital disease elevates the risk for complications such as pre-term birth, STDs and HIV. Currently, there is no consensus on the etiology of BV. Standard of care treatment is oral metronidazole; however, therapy has high rates of recurrence. Our previous qPCR study of recurrent BV patients (3+ episodes/year) found strong associations of Gardnerella_Gsp07, G. swidsinksii, and G. leopoldii with refractory and recurrent responses, and a robust recovery of Lactobacillus in remission patients. However, these findings were not applicable across …
Expanding Civil Rights To Combat Digital Discrimination On The Basis Of Poverty, Michele Estrin Gilman
Expanding Civil Rights To Combat Digital Discrimination On The Basis Of Poverty, Michele Estrin Gilman
SMU Law Review
Low-income people suffer from digital discrimination on the basis of their socio-economic status. Automated decision-making systems, often powered by machine learning and artificial intelligence, shape the opportunities of those experiencing poverty because they serve as gatekeepers to the necessities of modern life. Yet in the existing legal regime, it is perfectly legal to discriminate against people because they are poor. Poverty is not a protected characteristic, unlike race, gender, disability, religion or certain other identities. This lack of legal protection has accelerated digital discrimination against the poor, fueled by the scope, speed, and scale of big data networks. This Article …

Addressing Gender-Based Violence Using Evidence-Based Practices During Covid-19: The Case Of Puerto Rico, Ayorkor Gaba, Roseanne Flores, María Rebecca Ward, Bailey Pridgen
Publications and Research
As gender-based violence (GBV) surged during the COVID-19 pandemic, the 65th session of the Commission on the Status of Women (CSW65) called for member states, civil, and other stakeholders to consider the specific needs of women and girls in COVID-19 response and recovery efforts. Psychology provides scientific knowledge to help answer this call. Despite existing global guidance and psychological research to mitigate GBV, COVID-19 presents new challenges for consideration. This article summarizes existing GBV guidance/research and COVID-19 considerations, uses an illustrative case study to describe Puerto Rico’s application of GBV guidance/research during COVID-19, and provides preliminary policy and practice recommendations.

Distribution Of Toxicity Of Plant Species In The Plant Family Sapindaceae (Sub-Family Hippocastanoideae), Vishvam Mehta
Undergraduate Honors Thesis Projects
Maple leaf toxicosis is a lethal condition in which wilted leaves from the plant genus Acer are ingested by equines, leading to hemolytic anemia and methemoglobinemia. In prior studies, the toxicity of various Acer species was quantified, revealing a wide array of responses in terms of hemolysis and methemoglobin production. The object of this study, therefore, is to determine the toxicity of Acer’s closest relatives within the family Sapindaceae, specifically the four additional genera in the subfamily Hippocastanoideae. The information gained from this study will be used to understand the evolution of the toxins responsible for hemolysis and methemoglobin production. …

In this study, a quick microwave-based treatment was developed as a front end for DNA analysis of forensic samples. The effect of microwave treatment is to cause cell disruption which can improve the release of DNA during direct PCR as well as with extraction methods. Exposure to microwave preprocessing improved the quality of rapid genotyping, particularly when used with low level samples. Optimal results were obtained when samples were microwaved at 300W for 40 s, resulting in improved allele detection. Overall, the addition of this simple preprocessing step improves sensitivity and allele recovery for low level DNA samples when combined …

Background: We sought to address the paucity of data to support the evidence-based management of hypertension to achieve optimal blood pressure (BP) control on a sex-specific basis in Africa.
Methods: We undertook a post hoc analysis of the multicenter, randomized CREOLE (Comparison of Three Combination Therapies in Lowering Blood Pressure in Black Africans) Trial to test the hypothesis that there would be clinically important differences in office BP control between African men and women. We compared the BP levels of 397 and 238 hypertensive women (63%, 50.9 ± 10.5 years) and men (51.2 ± 11.3 years) from 10 sites across …

There are very few studies in Africans investigating the association between early life exposure to malnutrition and subsequent hypertension in adulthood. We set out to investigate this potential association within an adult cohort who were born around the time of the Biafran War (1968–1970) and subsequent famine in Nigeria. This was a retrospective analysis of Abia State NonCommunicable Diseases and Cardiovascular Risk Factors (AS-NCD-CRF) Survey, a community-based, cross-sectional study that profiled 386 adults (47.4% men) of Igbo ethnicity born in the decade between January 1965 and December 1974. Based on their date of birth and the timing of the famine, …

This editorial summarizes collections on the Research Topic “Insights in Vector Biology’’. Vectors of four major tropical diseases including malaria, Chagas, dengue and West Nile were considered. The editorial discusses vectors contributing to residual malaria transmission that reside mainly outdoors and not targeted by indoor vector control interventions, notably, long lasting insecticidal nets and indoor residual spraying; the concept of paratransgenesis- a parasite transmission blocking strategy which uses genetically-modified microbes made to produce antimalarial effectors inside the vector for malaria control; assessment of insecticide resistance in malaria vectors; insecticide resistance in Chagas disease vectors; reproductive biology of the dengue vector; …

Waste accumulation and microbial pollution are key environmental challenges in most developing countries. Land filling, the commonest waste disposal method, is not sustainable due to land scarcity and point source pollution. Thus, a large fraction of organic wastes is not properly disposed, yet the conventional methods of waste recycling are less effective. The degradation of waste using insect life stages (entomocomposting) has emerged as a novel tool that is fast, affordable, and sustainable in recycling organic wastes with maximum pathogen suppression. This bioconversion process also generates an organic fertilizer with potential to enhance soil remediation and provide additional benefits on …

Long-lasting insecticidal nets (LLINs) and indoor residual spraying (IRS) have resulted in a major decrease in malaria transmission. However, it has become apparent that malaria can be effectively transmitted despite high coverage of LLINs/IRS. Residual transmission can occur due to Plasmodium-carrying Anopheles mosquitoes that are insecticide resistant and have feeding and resting behavior that reduces their chance of encountering the currently deployed indoor malaria control tools. Residual malaria transmission is likely to be the most significant hurdle to achieving the goal of malaria eradication and research and development towards new tools and strategies that can control residual malaria transmission is …

We evaluated the effect of dietary inclusion of different ratios of Desmodium intortum (DI) and black soldier fly larvae meal (BSFLM) on sensory traits and physicochemical qualities of broiler chicks meat. Three formulations: T1:75%BSFL:25%DI; T2:50%BSFL:50DI, and T3:25BSFL:75% DI as a protein source were tested. A conventional feed was used as the control diet. On day 42, the chickens were slaughtered, and thigh muscles were used for sensory and physicochemical evaluations. T1 was preferred for its flavour and tenderness, T2 for its juiciness and T3 for its colour, flavour, taste and overall acceptability scores. In terms of physical properties, all three …

Although the black soldier fly frass fertiliser (BSFFF) is globally recognised as a promising and potential high-quality organic fertiliser, there is inadequate information on its impact on the growth performance and nutritional status of maize. Furthermore, no information exist on their combination with mineral fertiliser (NPK) as well as economic impact on maize production. This study evaluated the comparative impact of BSFFF, NPK and commercial organic fertiliser (Evergrow®) on growth, nitrogen use efficiency, yield, nutritional quality, and profitability of maize under greenhouse conditions. The treatments included: (1) sole application of BSFFF, conventional compost of brewers’ spent grain (BSG), Evergrow, and …

A water main break that contaminated the Boston area's water distribution system prompted a four-day “boil water” order. To understand risk communication during this incident, 600 randomly sampled residents were mailed questionnaires, yielding 110 valid responses. This article describes how perceptions of different social stakeholders influenced whether respondents complied with the Protective Action Recommendation—PAR (i.e., drank boiled water), took alternative protective actions (i.e., drank bottled water or/and self-chlorinated water), or ignored the threat (i.e., continued to drink untreated tap water). Respondents perceived technical authorities (i.e., water utility, public health, and emergency management) to be higher on three social influence attributes …

The current global platelet supply is often insufficient to meet all the transfusion needs of patients, in particular for those with alloimmune thrombocytopenia. To address this issue, we have developed a strategy employing a combination of approaches to achieve more efficient production of functional megakaryocytes (MKs) and platelets collected from cord blood (CB)-derived CD34+ hematopoietic cells. This strategy is based on ex-vivo expansion and differentiation of MKs in the presence of bone marrow niche-mimicking mesenchymal stem cells (MSCs), together with two other key components: (1) To enhance MK polyploidization, we used the potent pharmacological Rho-associated coiled-coil kinase (ROCK) inhibitor, KD045, …
