"Recent advances in analytical methods have furthered the quantitative insights that can be gleaned from cellular analyses, with applications in cancer and nanoparticle research. In the first part of the research presented, a recently developed HPLC-MS/MS method was advanced to elucidate pteridine metabolism in an isogenic progressive MCF10A breast cancer cell model. The folate-derived pteridine pathway in breast cells was established by individually dosing cell cultures with folic acid and 15 pteridines. Eight potential pteridine biomarkers in breast cancer cells were identified including pterin and isoxanthopterin, which yielded the first in vitro evidence for the cellular metabolisms behind previously reported …

"This research focuses on the processing of novel feedstocks for and during direct-write additive manufacturing (AM), specifically the direct ink writing (DIW) and Ceramic On-Demand Extrusion (CODE) manufacturing processes, in order to produce ceramic and ceramic-based composite components. Strongly dispersed, concentrated (φ = 0.42), nanoparticle (d50 ~0.3 µm), zirconia (ZrO2) pastes were used to print densely filled, large continuous volume (≳ 1 cm3) ceramic components. An elastic shear modulus (G’) of 56,000 Pa and yield stresses between 6 and 10 Pa allowed for printed components of 34.5 mm in height over 115 layers without slumping …

"Purine-derived compounds are widely investigated as cyclin-dependent kinase inhibitors that have broad applications in the design of pharmaceuticals for treating diseases, such as diabetes, atherosclerosis, and cancers. Towards the goal of effective AGE-inhibitors, and neuroprotector compounds we have synthesized a series of novel purine-based triazoles and investigated their neuroprotective effects, using SHSY-3Y human neuroblastoma cell line. Through these studies, we have identified purine-based neuroprotector compounds that favorably modulate oxidative stress induced by the Fenton reaction-generated reactive oxygen species (ROS).

"Seismic damage assessment is a critical step to enhance community resilience in the wake of an earthquake. This study aims to develop deep learning-based surrogate models for widely used fragility curves to achieve more accurate and rapid assessment in practice. These surrogate models are based on artificial neural networks trained from the labelled ground motions whose resulting damage classes on targeted structures are determined by nonlinear time history analyses. The development of various surrogate models is progressed in four phases. Bordetella pertussis is a Gram-negative respiratory pathogen that can infect individuals of all ages and is notably responsible for infant fatalities. Pertussis (whooping cough) is a vaccine-preventable respiratory disease for which vaccines have been available in the United States since the 1940s. Whole cell pertussis vaccines kept incidence of pertussis under 5,000 cases per year, but were associated with reactogenicity issues. Following the introduction of less reactogenic acellular pertussis vaccines in the late 1990s, there have been noteworthy outbreaks of pertussis. For example, in 2012 there were over 50,000 reported cases of pertussis and 20 deaths in the U.S. The …

Vegetation data were collected from eight maple syrup farms (sugarbushes) and eight undeveloped maple-dominated sites to examine potential differences in understory plant communities due to disturbance effects. Understory plants were identified to the species level and percent cover of aboveground leaf-area for each species was estimated within 600 total quadrats. Overstory data and environmental data were collected to help determine if they had any effect on herbaceous-layer plant communities. Species richness, Shannon diversity, and Pielou’s evenness were calculated, with the analysis showing no significant differences between the site types. Wetland ecosystem spatial extent and integrity have declined due to conversion to anthropogenic land uses. Wetland mitigation offsets the impact on wetlands encountered during development by restoring a previously degraded wetland or creating new wetlands. This activity creates an ecosystem service market where wetland impacts (debits) are offset by wetland restoration/creation (credits). Mitigation aims to replace functions provided by aquatic resources lost during development. Post-restoration monitoring criteria known as performance standards are used to evaluate and verify restored wetland functionality and regulate mitigation credit release. In this dissertation, I complete three studies to evaluate potential reactions to target role congruity, especially gender role congruity, through an examination of Student Evaluations of Instructors (SEIs). Target role congruity refers to assessments an observer makes of whether or not the various roles a target person fills “fit” with one another. For example, a woman surgeon may be perceived as being in an incongruent role due to the masculine characteristics associated with the occupation and the continued dominance of men in the field. Given the increasingly apparent consequences of climate warming, interest in how to better predict and manage Earth’s carbon sinks has never been greater. Soil, the largest terrestrial carbon sink, harbors an incredibly taxonomically and functionally diverse microbial community. These soil-dwelling microbes govern the fate of soil carbon and nutrients by cycling organic matter as they live, grow, and die. We consider a one-dimensional Chemotaxis model describing the dynamics of the cell density n, cell velocity u, chemoattractant c1, and chemorepellent c2, respectively. The model is related to angiogenesis in cancer cells. n represents the cell density of the vessel, the chemoattractant c1 is vascular endothelial growth factor (VEGF) and the chemorepellent c2 is the antiangiogenic drug. We study the existence of a nonconstant steady-state solution using the singular perturbation method. Incarceration is a rapidly increasing occurrence in the United States with more than 1.4 million people incarcerated and more than 2 million on some form of restricted confinement. While men are incarcerated at a much higher rate than women, women are still at a steady incline of incarceration as well. Also, people of color disproportionately make up a large number of those incarcerated while accounting for a small percentage of the general population. The Musashi (Msi) family of RNA binding proteins consists of two paralogs, Msi1 and Msi2, that are highly conserved across species. The two paralogs have emerged as factors that promote stem cell proliferation by post-transcriptionally regulating translation. In addition to their expression in stem cells, the Musashi proteins are also expressed in postmitotic neurons, including the photoreceptor cells. The Musashi proteins have been observed to maintain high expression levels in the postmitotic photoreceptors within the eye of both invertebrates and vertebrates. These observations suggest an additional role in the maintenance of terminally differentiated neurons.

Virtually all cellular processes are precisely regulated by the proteasome which is the primary enzyme responsible for the degradation of misfolded, damaged, or no longer necessary soluble proteins. To prevent any untimely degradation of these target protein substrates and protect the cell, the proteasome is tightly regulated via adaptor proteins, known as proteasomal regulators. There are many classes of proteasomal regulators each with their own unique structures, functions, and effects on protein degradation through the proteasome. Berger's theory of religion and world construction fails to consider the critical issue of power and who is allowed to construct their own reality and thus, does not adequately capture the experience of Black Christians and the Black religious experience. I use White Jesus as a case study to analyze this process. Though the White Jesus phenomenon has been more readily explored in theological and historical fields, very little sociological research discusses this phenomenon. I argue that a Europeanized Jesus has had harmful sociocultural effects on Black Christians and Black people, in the form of cultural trauma. Yet, the White …

This project explores a topic in Chemical Reaction Network Theory. We analyze networks with one dimensional stoichiometric subspace using mass-action kinetics. For these types of networks, we study how the capacity for multiple positive equilibria and multiple positive nondegenerate equilibria can be determined using Euclidian embedded graphs. Our work adds to the catalog of the class of reaction networks with one-dimensional stoichiometric subspace answering in the affirmative a conjecture posed by Joshi and Shiu: Conjecture 0.1 (Question 6.1 [26]). Hemophilia is a rare genetic disorder that requires specialty care and treatment. Pediatric patients with hemophilia have unique medical issues that may lead to permanent disability or death if not properly diagnosed and treated in a timely manner. Due to lack of resources and proper training of staff, rural hospitals are not equipped to properly treat pediatric hemophilia patients. Utilizing the Healthcare Cost and Utilization Project (HCUP) Kids´ Inpatient Database (KID) of the Agency for Healthcare Research and Quality. The bobcat (Lynx rufus) is an important furbearer in South Dakota. However, management of bobcats can be difficult because of their elusive nature and lack of demographic information. Head and neck squamous cell carcinoma (HNSCC) is a cancer originating from the epithelial lining of the oral cavity, larynx, and pharynx. HNSCC is associated with extensive locoregional invasion and tissue destruction. Invasive processes underlying HNSCC contribute to poor prognosis and decreased quality of life. Amplification of chromosome 11q13 is the most common DNA aberration in human papilloma virus negative (HPV-) HNSCC. This region includes various genes known to drive tumor progression. This applied dissertation was designed to evaluate the effects of social skills training programs as a method to improve social skills in children diagnosed with autism spectrum disorder. The study sought to advance the social skills training options for children with autism spectrum disorder by comparing various evidence-based social skills training programs in hope to provide meaningful, significant, and sustainable social-emotional changes that are explicitly taught and later generalized across individuals, settings, and environments. Little information is available concerning the distribution of genetic diversity in non-salmonid, non-imperiled, freshwater fish. In order to fill in this knowledge gap, I conducted a population genomics survey in Redbreast Sunfish (Lepomis auritus; RBS), a widespread, generalist species distributed along the Atlantic slope rivers of eastern North America. I sampled four basins (ACF, Savannah, Roanoke, and James) at eight sites each with a factorial experimental design. The Reticulated flatwoods salamander (Ambystoma bishopi) is a federally endangered amphibian endemic to the longleaf-pine ecosystem of the southeastern U.S. This study used analyses of single-nucleotide polymorphism (SNP) data, collected from 2,255 unique individuals across 5 breeding seasons, spread across the known extant range of A. bishopi, to characterize the genetic diversity and demographics of populations, genetic relationships among populations, and patterns and spatial extents of gene flow, and to evaluate potential effects of management on A. bishopi’s resiliency. Argentine ants, Linepithema humile, are a highly successful invasive species around the globe and are especially prominent in states such as California and the southeastern United States. L.humile have a unique form of unicoloniality, called “supercolonies”. L. humile can detect colonymates through scent markers in their outer cuticle. With these chemical markers, ants will exhibit high aggression if they smell different from one another. In our study, we performed aggression assays among ten different nest sites and analyzed their CHCs through gas chromatography mass spectrometry, or GC-MS, analysis. Phosphatase and tensin homolog deleted on chromosome 10 (PTEN) protein is a negative regulator of the AKT serine threonine kinases (Protein Kinase B) in the mechanistic target of rapamycin (mTOR) signaling pathway. Mutations in PTEN are found in patients with autism, epilepsy, or macrocephaly. In mouse models, Pten-loss results in neuronal hypertrophy, hyperexcitability, seizures, and social deficits. The underlying molecular mechanisms of these phenotypes are not well-delineated. Background: New specific and sensitive spectrofluorimetric fluorotag method has been developed and validated for the quantification of metformin hydrochloride. Materials and Methods: The derivatization reaction was carried out using 1,2-naphthoquinone-4-sulfonic sulfonate as (NQS) fluorotag in the presence of strong alkaline medium (potassium hydroxide) to form the highly fluorescent product. The fluorescence was observed at emission wavelength 453 nm after excitation at 359 nm. The different experimental conditions affecting the reaction product formation and stability were carefully studied and optimized as 2% w/v of potassium hydroxide solution, 0.5% w/v NQS reagent.
