As global temperature shifts due to anthropogenic impacts, the frequency and magnitude of extreme temperatures in shallow aquatic ecosystems are expected to increase. Previous studies performed on fish under fluctuating temperature regimes have reported increased physiological stresses, compared to stable thermal conditions, leading to morphological, behavioral and biochemical adaptations. Moreover, many studies have reported the effects of warmer temperatures on numerous aquatic species, but seasonal temperature changes experienced in aquatic systems are understudied. We surveyed changes in the bioenergetic machinery of a native fish population ( Lepomis macrochirus) in the temperate zone of North America and observed season-dependent thresholds in …

Brown carbon (BrC) formed from glyoxal+ammonium sulfate (AS) and methylglyoxal+AS reactions photobleaches quickly, leading to the assumption that BrC formed overnight by Maillard reactions will be rapidly destroyed at sunrise. Here, we tested this assumption by reacting glyoxal, methylglyoxal, glycolaldehyde, or hydroxyacetone in aqueous mixtures with reduced nitrogen species at pH 4–5 in the dark and in sunlight (>350 nm) for at least 10 h. The absorption of fresh carbonyl+AS mixtures decreased when exposed to sunlight, and no BrC formed, as expected from previous work. The tripartite AcrAB-TolC assembly, which spans both the inner and outer membranes in Gram-negative bacteria, is an efflux pump that contributes to multidrug resistance. Here, we present the in situ structure of full-length Escherichia coli AcrAB-TolC determined at 7 Å resolution by electron cryo-tomography. The TolC channel penetrates the outer membrane bilayer through to the outer leaflet and exhibits two different configurations that differ by a 60° rotation relative to the AcrB position in the pump assembly. AcrA protomers interact directly with the inner membrane and with AcrB via an interface located in proximity to the AcrB ligand-binding pocket. Our …

Background Coronavirus disease 2019 (COVID-19) has had a disproportionate impact on people with intellectual disability (PwID). PwID are at higher risk of mental illness and receive psychotropic prescribing 'off licence' also, to manage distress behaviour. The lockdown and reduction of multidisciplinary face-to-face appointments had an impact on care delivery, the recourse possibly being psychotropic prescribing. It is imperative to comprehend the influence the pandemic had on psychotropic prescribing patterns to enable future planning. BACKGROUND: In response to injury, neurons activate a program of organized axon self-destruction initiated by the NAD
METHODS: To investigate whether naturally occurring human variants might disrupt SARM1 autoinhibition and potentially contribute to risk for neurodegenerative disease, we assayed the enzymatic activity of all 42 rare SARM1 alleles identified among 8507 amyotrophic lateral sclerosis (ALS) patients and 9671 controls. We then intrathecally injected mice with virus expressing SARM1 constructs to test the capacity of an ALS-associated constitutively active SARM1 variant to promote neurodegeneration in vivo.

Racial disparities in breast cancer present a vexing and complex challenge for public health. A diverse array of factors contributes to disparities in breast cancer incidence and outcomes, and, thus far, efforts to improve racial equity have yielded mixed results. Systems theory offers a model that is well-suited to addressing complex issues. In particular, the concept of a systemic leverage point offers a clue that may assist researchers, policymakers, and interventionists in formulating innovative and comprehensive approaches to eliminating racial disparities in breast cancer. There is now high-level recognition that the UN Sustainable Development Goals can only be achieved if the decline of ecosystems and biodiversity can be halted and reversed. This will require effective control of ongoing pressures, meaningful protection and enforcement of protected areas, and significant investments in ecosystem restoration. The sustainable management of Blue Growth is an urgent issue for coastal states. Marine industries have rapidly expanded over the last two decades and this is projected to continue with the European Green Deal and post-COVID economic recovery policies. The intensification of Blue Growth could have adverse socio-ecological implications and must, therefore, be managed in terms of sustainability, natural resource boundaries, and coastal community well-being. The EU Atlantic Action Plan (AAP) has recently been updated and revised to support ‘blue growth’ along Europe’s western coastal regions. The revisions reflect recent challenges facing the Atlantic Arc maritime economies including the Covid-19 crisis, Brexit and the new requirements of the European Green Deal. This new revision, termed AAP 2.0, also addresses some of the weaknesses highlighted in the original Atlantic Action Plan particularly regarding identifying indicators that may be used to measure progress in the achievement of the Plan’s objectives. The marine and coastal environment provides crucial ecosystem services to sustain human activities and wellbeing. Advancing measurement frameworks to record the contribution of marine and coastal ecosystems to national wealth and economies and assess the sustainability of their use is therefore paramount. The recently proposed Ocean Accounting Framework outlined within the System of Environmental Economic Accounting represents a decisive step in this direction and provides a foundation for developing an integrated and standardised set of ecosystem accounts to support decision making and use of the seas. Alfalfa has played an important role in providing farmers of Kentucky with a high yielding, high quality, persistent forage legume for many years. In 1980, a group of individuals under the leadership of University of Kentucky Forage Extension Specialists organized and implemented the first statewide educational event on alfalfa. This initial meeting gave rise to the Kentucky Alfalfa Conference that celebrated it’s twentieth anniversary in February 2000. Using state-of-the-art telecommunication technologies, this project is developing a comprehensive knowledge resource for alfalfa (Medicago sativa L.); the National Alfalfa Information System (NAIS). This project will serve as an improved model for Extension educational programs. Background
Hundreds of millions of people in poor countries continue to suffer from disease caused by bloodfeeding hookworms. While mice and rats are not reliably permissive hosts for any human hookworm species, adult Golden Syrian hamsters are fully permissive for the human and animal pathogen Ancylostoma ceylanicum. Similar to humans, hamsters may be infected with A. ceylanicum third-stage larvae orally or percutaneously. Oral infection typically leads to consistent worm yields in hamsters but may not accurately reflect the clinical and immunological manifestations of human infection resulting from skin penetration.

Going beyond H. Richard Neibuhr’s famous paradox model on “Christ and Culture,” Dr. Paul Raabe contends for a biblical, dynamic tension that exists between the culture and the faith. Using the image of a house, Raabe explores four “rooms” : (1) Christianity affirms culture in some respects; (2) Christianity opposes culture in some respects; (3) Christianity leavens culture in some ways; and (4) Christianity uses cultural forms to communicate its message. Each of the four rooms needs to be lived in for maintaining the tension presented in the Scriptures as a faithful witness to the lordship of Christ.

The current study aimed to develop pH-responsive cisplatin-loaded liposomes (CDDP@PLs) via the thin film hydration method. Formulations with varied ratios of dioleoyl phosphatidylethanolamine (DOPE) to cholesteryl hemisuccinate (CHEMS) were investigated to obtain the optimal particle size, zeta potential, entrapment efficiency, in vitro release profile, and stability. The particle size of the CDDP@PLs was in the range of 153.2 ± 3.08-206.4 ± 2.26 nm, zeta potential was -17.8 ± 1.26 to -24.6 ± 1.72, and PDI displayed an acceptable size distribution. Sperm quantity and quality are key features explaining intra- and interspecific variation in male reproductive success. Spermatogenesis is sensitive to ionizing radiation and laboratory studies investigating acute effects of ionizing radiation have indeed found negative effects of radiation on sperm quantity and quality. In nature, levels of natural background radiation vary dramatically, and chronic effects of low-level background radiation exposure on spermatogenesis are poorly understood. The Chernobyl region offers a unique research opportunity for investigating effects of chronic low-level ionizing radiation on reproductive properties of wild organisms. The aim of this study was to determine the effects of a pin‐to‐plate cold atmospheric plasma (CAP) on U‐251 MG three‐dimensional (3D) glioblastoma spheroids under different conditions. 3D tumorspheres showed higher resistance to the CAP treatment compared to 2D monolayer cells. A single CAP treatment was able to induce cytotoxicity, while multiple CAP treatments augmented this effect. CAP was also able to induce cytotoxicity throughout the tumoursphere, and we identified that reactive oxygen species(ROS) plays a major role, while H2O2plays a partial role in CAP‐induced cytotoxicity in tumour-spheres. Altered fatty acid metabolism continues to be an attractive target for therapeutic intervention in cancer. We previously found that colorectal cancer (CRC) cells with a higher metastatic potential express a higher level of fatty acid translocase (CD36). However, the role of CD36 in CRC metastasis has not been studied. Here, we demonstrate that high expression of CD36 promotes invasion of CRC cells. Consistently, CD36 promoted lung metastasis in the tail vein model and GI metastasis in the cecum injection model. The protozoan parasite, Trypanosoma cruzi, causes severe morbidity and mortality in afflicted individuals. Approximately 30% of T. cruzi infected individuals present with cardiac pathology. The invasive forms of the parasite are carried in the vascular system to infect other cells of the body. During transportation, the molecular mechanisms by which the parasite signals and interact with host endothelial cells (EC) especially heart endothelium is currently unknown. The parasite increases host thrombospondin-1 (TSP1) expression and activates the Wnt/β-catenin and hippo signaling pathways during the early phase of infection. Conventional anti-cancer therapies based on chemo- and/or radiotherapy represent highly effective means to kill cancer cells but lack tumor specificity and, therefore, result in a wide range of iatrogenic effects. A promising approach to overcome this obstacle is spliceosome-mediated RNA trans-splicing (SMaRT), which can be leveraged to target tumor cells while leaving normal cells unharmed. Notably, a previously established RNA trans-splicing molecule (RTM44) showed efficacy and specificity in exchanging the coding sequence of a cancer target gene (Ct-SLCO1B3) with the suicide gene HSV1-thymidine kinase in a colorectal cancer model, thereby rendering tumor cells sensitive to the prodrug ganciclovir (GCV). Background: The evolutionary stages of manufacturing have led us to conceptualize the use of Industry 4.0 for COVID-19 (coronavirus disease 2019), powered by Industry 4.0 technologies. Using applications of integrated process optimizations reliant on digitized data, we propose novel intelligent networks along the vaccine value chain. Vaccine 4.0 may enable maintenance processes, streamline logistics, and enable optimal production of COVID-19 vaccines.

Genome editing in the lung has the potential to provide long-term expression of therapeutic protein to treat lung genetic diseases. Yet efficient delivery of CRISPR to the lung remains a challenge. The NIH Somatic Cell Genome Editing (SCGE) Consortium is developing safe and effective methods for genome editing in disease tissues. Methods developed by consortium members are independently validated by the SCGE small animal testing center to establish rigor and reproducibility. Artificial coastal structures (ACSs) are primarily designed to provide services for human use, such as flood defence or shipping, and are generally poor for marine biodiversity. Consequently, there has been significant research effort to enhance these hard structures to increase biodiversity and habitat availability via eco-engineering. On seawalls and breakwaters, this has included the creation of habitats for benthic species found on natural rocky shores, including the provision of cracks, crevices and water retaining features, such as artificial rockpools. When sediment retention in these features has occurred, it has often been deemed detrimental to the overarching aim of the intervention. …
