Microbial Population And Fermentation Characteristics Of Total Mixed Ration Prepared With Local Feed Resources In Mozambique, Y. Cai, Z. Du, S. Yamasaki, D. Nguluve, F. Macome, T. Oya

To effectively utilize locally available feed resources for dairy cattle in Africa, fermented total mixed ration (TMR) was prepared using Napier grass, corn meal, wheat bran, formula feed, and mineral and vitamin mix. The fermentation quality of TMR were analyzed after 14 days of ensiling. Before ensiling, aerobic bacteria were the dominant bacteria, after ensiling, lactic acid bacteria counts increased rapidly and became the dominant bacteria in TMR. After 14 days of ensiling, aerobic bacteria, coliform bacteria, yeasts, and molds showed lower (P < 0.05) counts in TMR than those before ensiling. The dry matter (DM) of TMR was adjusted to 45.50%, their crude protein and neutral detergent fiber contents were approximately 11.48, and 55.25% of DM, respectively. After 14 days of ensiling, TMR fermented to a good quality, the pH was reduced to 3.90; the lactic acid and ammonia nitrogen contents were 1.01 and 0.31% of FM, respectively; the propionic acid and butyric acid were too low to detect. The results confirm that fermented TMR prepared with local feed resources can attain a good quality in Africa.

Parkinson’s disease (PD) is a motor disorder mostly affecting aged people, with not known standard therapy. The use of natural products is increasingly gaining attention for the treatment of different diseases. Baphia macrocalyx is a plant previously associated with antioxidant activities in in vitro studies. Thus, the aim of this study was to evaluate the ameliorative potential of methanolic leaf extract of Baphia macrocalyx (MEBM) on rotenone-triggered toxicity in Drosophila melanogaster. Background
Heterochrony, change in the rate or timing of development, is thought to be one of the main drivers of morphological evolution, and allometry, trait scaling patterns imposed by size, is traditionally thought to represent an evolutionary constraint. However, recent studies suggest that the ontogenetic allometric trajectories describing how organisms change as they grow may be labile and adaptive. Here we investigated the role of postnatal ontogenetic development in the morphological diversification of Paleoanguimorpha, the monitor lizards and allies, a clade with extreme body size disparity. Analysis of manganese mineral occurrences and valence states demonstrate oxidation of Earth’s crust through time. Changes in crustal redox state are critical to Earth’s evolution, but few methods exist for evaluating spatially averaged crustal redox state through time. Manganese (Mn) is a redox-sensitive metal whose variable oxidation states and abundance in crustal minerals make it a useful tracer of crustal oxidation. We find that the average oxidation state of crustal Mn occurrences has risen in the last 1 billion years in response to atmospheric oxygenation following a 66 ± 1 million-year time lag. Colonial history exposes Tanzania to triple linguistic heritage, that has at times plunged the nation into linguistic confusion. This heritage involves Kiswahili, ethnic community languages (ECLs) and English. Whereas each of the languages has been limited to some use through policies, some of the languages have pushed their own way, often breaking policy boundaries. It is shown in this paper that, firstly, the exclusion of ECLs by post independence policies has denied the nation important socio-cultural and economic avenues that could foster rapid social and economic development. The 2020 COVID-19 pandemic created a crisis for American society—and the federal courts were not exempt. Court facilities came to a grinding halt, cases were postponed, and judiciary employees adopted work-from-home practices. Having court operations impacted by a pandemic was not a new phenomenon, but the size, scope, and technological lift of the COVID-19 pandemic was certainly unique.

Mesenchymal stromal/stem cells (MSCs) are multipotent cells residing in multiple tissues with the capacity for self-renewal and differentiation into various cell types. Due to the myriad sources of MSCs the International Society for Cellular Therapy (ISCT) proposed certain criteria for their characterization. Here, we isolate adult MSCs from mouse testes and demonstrate that these testis-derived MSCs (tMSCs) follow some of the ISCT’s criteria. tMSCs expressed the MSCs surface markers CD44, CD73, and CD29 and lacked the expression of the hematopoietic cell marker CD45. Moreover, the cells displayed the capacity for self-renewal and in vitro differentiation into adipocytes. Since the isolation …

Noncoding RNAs (ncRNAs) defy the central dogma by representing a family of RNA molecules that are not translated into protein but can convey information encoded in their DNA. Elucidating the exact function of ncRNA has been a focus of discovery in the last decade and remains challenging. Nevertheless, the importance of understanding ncRNA is apparent since these molecules regulate gene expression at the transcriptional and post-transcriptional level exerting pleiotropic effects critical in development, oncogenesis, and immunity. The essay chronicles the early phases of a digital history project on landscape change in the mountains of eastern Tanzania. In collecting sources for a land and culture narrative, the project aims ultimately to create an archive that is locally produced in Tanzania and maintained by Utah State University Library's Special Collections and Archives division. The project draws on more than thirty early twentieth-century landscape photographs from the Usambara Mountains in northeastern Tanzania by Walther Dobbertin, a professional photographer living in German East Africa. The extension of life span driven by 40% caloric restriction (CR) in rodents causes trade-offs in growth, reproduction, and immune defense that make it difficult to identify therapeutically relevant CR-mimetic targets. We report that about 14% CR for 2 years in healthy humans improved thymopoiesis and was correlated with mobilization of intrathymic ectopic lipid. CR-induced transcriptional reprogramming in adipose tissue implicated pathways regulating mitochondrial bioenergetics, anti-inflammatory responses, and longevity. Expression of the gene Pla2g7 encoding platelet activating factor acetyl hydrolase (PLA2G7) is inhibited in humans undergoing CR. Deletion of Pla2g7 in mice showed decreased thymic lipoatrophy, protection against age-related inflammation, …

Successful strategies for the attachment of oligopeptides to mesoporous silica with pores large enough to load biomolecules should utilize the high surface area of pores to provide an accessible, protective environment. A two-step oligopeptide functionalization strategy is examined here using diazirine-based heterobifunctional linkers. Mesoporous silica nanoparticles (MSNPs) with average pore diameter of ~8 nm and surface area of ~730 m2/g were synthesized and amine-functionalized. Tetrapeptides Gly-Gly-Gly-Gly (GGGG) and Arg-Ser-Ser-Val (RSSV), and a peptide comprised of four copies of RSSV (4RSSV), were covalently attached via their N-terminus to the amine groups on the particle surface by a heterobifunctional linker, …

Information on summer distribution and diurnal roosting is lacking for the imperiled, rock-roosting Myotis leibii, a species threatened by habitat loss and White-nose Syndrome. I aimed to 1) identify diurnal, summer roost locations in the greater Ouachita Mountains of Arkansas, and 2) determine roost habitat characteristics at the local, landscape, and roost-level scales. Using acoustic monitoring, rock searches, mist-netting, and temperature monitoring in 2019 and 2020, I found M. leibii in an aggregated distribution across the landscape roosting either solitarily or in small groups in the Rich Mountain-Black Fork Mountain, Mount Magazine, and Mount Nebo areas under sparse canopy cover, …

The Indiana bat (Myotis sodalis) has been listed as a federally endangered species since 1967 due to numerous natural and anthropogenic threats. Research on the species has focused on hibernacula and maternity colonies, but data gaps exist in portions of their range. To prevent further decline, migration resources need protection, but management practices are difficult to implement without having precise data on migration routes. I documented spring migration in Arkansas using aerial and ground telemetry during 2017–2018. Background: Tyrosine kinase inhibitors (TKI) were initially demonstrated as an efficacious treatment for renal cell carcinoma (RCC). However, after a median treatment length of 14 months, a vast majority of patients develop resistance. This study analyzed a combination therapy of tipifarnib (Tipi) + sunitinib that targeted exosome-conferred drug resistance. Methods: 786-O, 786-O-SR (sunitinib resistant), A498, A498-SR, Caki-2, Caki-2-SR, and 293T cells were cultured. Ex-osomes were collected using differential ultracentrifugation. Cell proliferation, Jurkat T cell immune assay, and immunoblot analysis were used for downstream analysis. Results: SR exosomes treatment displayed a cytotoxic effect on immune cells. Research on non-suicidal self-injury (NSSI) has produced mixed findings, resulting in a lack of clarity regarding these behaviors (Klonsky & Meuhlenkamp, 2007). To address this, Hooley and Franklin (2018) developed the Benefits and Barriers Model (BBM) to provide a comprehensive understanding of NSSI, in which they identified the barriers that commonly prevent people from engaging in these behaviors (e.g., self-esteem, shame, and peer-bonding motivations/social norms). They also identified adverse childhood experiences (ACEs) as a distal predictor of NSSI, which aids people in overcoming the barriers to engaging in these behaviors. Since the emergence of SARS-CoV-2, several studies have been published describing neuromuscular manifestations of the disease, as well as management of pre-existing pediatric neuromuscular disorders during the COVID-19 pandemic. These disorders include muscular dystrophies, myasthenic syndromes, peripheral nerve disorders, and spinal muscular atrophy. Such patients are a vulnerable population due to frequent complications such as scoliosis, cardiomyopathy, and restrictive lung disease that put them at risk of severe complications of COVID-19. Macromolecular damage leading to cell, tissue and ultimately organ dysfunction is a major contributor to aging. Intracellular reactive oxygen species (ROS) resulting from normal metabolism cause most damage to macromolecules and the mitochondria play a central role in this process as they are the principle source of ROS. Connexin43 is one of the 21 members of a large protein family that forms intercellular gap junction complexes. It plays a critical role in development by allowing two adjacent cells to exchange cellular content. Mouse deletion studies have demonstrated its critical roles in many organs, including cardiac muscle, blood vessels, bone, adipose tissue and more. However, as the most expressed and most studied connexin in the family, Connexin43 surprisingly shows many gap-junction-independent roles, spanning from the forming of hemichannels, to regulating membrane trafficking, to regulating mitochondrial function. In this work, an energy-resolving thermoelectrically cooled single crystal cadmium telluride (CdTe) detector system upgraded with the latest firmware was optimized for high x-ray flux operations using high bias voltage and fast peaking time. This detector system was deployed into an experimental benchtop x-ray fluorescence (XRF) imaging/computed tomography (XFCT) system developed for quantitative imaging of metal nanoprobes such as gold nanoparticles (GNPs). Using the firmware-upgraded and existing/old CdTe detector systems, the Compton/XRF spectra from small (8 mm diameter) GNP-containing phantoms were acquired. Osteoarthritis is a joint disease characterized by a poorly-defined inflammatory response that does not encompass a massive immune cell infiltration yet contributes to cartilage degradation and loss of joint mobility, suggesting a chondrocyte intrinsic inflammatory response. Using primary chondrocytes from joints of osteoarthritic mice and patients, we first show that these cells express ample pro-inflammatory markers and RANKL in an NF-κB dependent manner. The inflammatory phenotype of chondrocytes was recapitulated by exposure of chondrocytes to IL-1β and bone particles, which were used to model bone matrix breakdown products revealed to be present in synovial fluid of OA patients, albeit their …

Rational decision makers aim to maximize their gains, but humans and other animals often fail to do so, exhibiting biases and distortions in their choice behavior. In a recent study of economic decisions, humans, mice, and rats were reported to succumb to the sunk cost fallacy, making decisions based on irrecoverable past investments to the detriment of expected future returns. We challenge this interpretation because it is subject to a statistical fallacy, a form of attrition bias, and the observed behavior can be explained without invoking a sunk cost-dependent mechanism. Lignocellulosic biomass from the secondary cell walls of plants has a veritable potential to provide some of the most appropriate raw materials for producing second-generation biofuels. Therefore, we must first understand how plants synthesize these complex secondary cell walls that consist of cellulose, hemicellulose, and lignin in order to deconstruct them later on into simple sugars to produce bioethanol via fermentation. Breast cancer (BC) has one of the highest incidences and mortality worldwide. Single nucleotide polymorphisms (SNPs) in TOX3 rs3803662 and MMP7 rs1943779 have been associated with susceptibility to BC. In this case-control study, we evaluated the association of rs3803662 (TOX3)/rs1943779 (MMP7) SNPs with clinical features, immunohistochemical reactivity, and risk association with BC in women from northeastern Mexico. We compared 212 BC cases and 212 controls. DNA was isolated from peripheral blood to perform the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) assay. Nanoparticles (NPs) have been used in drug delivery therapies, medical diagnostic strategies, and as current Covid-19 vaccine carriers. Many microscope-based imaging systems have been introduced to facilitate detection and visualization of NPs. Unfortunately, none can differentiate the core and the shell of NPs. Spectral imaging has been used to distinguish a drug molecule and its metabolite. We have recently integrated this technology to a resolution of 9 nm by using artificial intelligence-driven analyses. Therapeutic use of psilocybin has become a focus of recent international research, with preliminary data showing promise to address a range of treatment-resistant mental health conditions. However, use of psilocybin as a healing entheogen has a long history through traditional consumption of mushrooms from the genus Psilocybe. The forthcoming adoption of new psilocybin-assisted therapeutic practices necessitates identification of preferred sources of psilocybin; consequently, comprehensive understanding of psilocybin-containing fungi is fundamental to consumer safety. Here we examine psilocybin producing fungi, discuss their biology, diversity, and ethnomycological uses.
