Pleurotus ostreatus is a common cultivated edible mushroom worldwide. The fruiting bodies of P. ostreatus is a rich source of a β-glucans polysaccharide. The current study aimed to investigate the effectiveness of β-glucans as a natural polysaccharide produced by P. ostreatus as an antioxidant, antimicrobial, and anticancer. The molecular identification of P. ostreatus isolate was confirmed by Internal Transcribed Spacer (ITS) sequence. The sequence alignment and phylogenetic evolutionary relationship of studied ITS sequence were performed against some deposited sequences in GenBank. This study examines the analysis of natural vegetation cover, the spatial distribution and identification of the most important existing plants and trees species in Ajloun Governorate by choosing spatial samples on Quadrat way which based on the frequency account of grass plants and apply it in (the Runkaier Method). Cow-calf beef operations are the primary users of the 8.75 million hectares of rangelands in the U.S. Pacific Northwest and will have to adapt to ongoing and future climate change. Management directed toward current rangeland stressors which may be amplified under a changing climate—including fire risk, invasive plants, and droughts—is likely to improve future resilience. And although climate change discussions have become highly politicized, our experience suggests that Pacific Northwest ranchers support "no-regrets” strategies that provide ecological and economic benefits under a variety of future conditions. Our goal is to foster adoption of these strategies by sharing individual success stories. …

Wilted (35.9 % DM and 2.6% WSC) moderately difficult to ensile grass-legume mixture (red clover, alfalfa and timothy 50:20:30) was ensiled in cylindrical big bales (1.2 m hight and 1.2 m diameter) with a weight about 700 kg. The crop was ensiled for 8, 32 and 120 days with or without a microbial inoculation (SiloSolve® FC containing Lactococcus lactis DSM 11037/1k2081 and Lactobacillus buchneri DSM 22501/1k20738 applied at 1.5×105 CFU g-1 forage). Species and biotypes of the genus Tripsacum include: eastern gamagrass (T. dactyloides) in North and Central America; Guatemala grass (T. fasciculatum syn. T. andersonii) in Latin America and the West Indies; and wide-leaf gamagrass (T. latifolium) throughout Mesoamerica. Of these, Guatemala grass has become widely popular throughout the tropics as a fodder plant and for its benefits related to environmental and ecosystem services. Guatemala grass is a hearty, long-lived perennial warm-season bunchgrass. Mature plants are somewhat shallow rooted, with short rhizomes, and produce robust foliage with leaf blades 10cm wide and 120cm long. The …

The debate on appropriate stocking rates (SR) increasingly recognizes the importance of variable, targeted and adaptive stocking in response to a temporally variable and spatially heterogeneous resource distribution, particularly in challenging environments such as the Namibian savannah. In common extensive ranching systems, a farmer’s scope of action is often limited to flexibly adjusting the number and concentration of livestock to the available forage in space and time. SRs are commonly set according to carrying capacity recommendations which usually have a low spatio-temporal resolution. This may lead to punctual under- and overutilization. Aminoacyl-tRNA synthetases (aaRSs) are enzymes that synthesize aminoacyl-tRNAs to facilitate translation of the genetic code. Quality control by aaRS proofreading and other mechanisms maintains translational accuracy, which promotes cellular viability. Systematic disruption of proofreading, as recently demonstrated for alanyl-tRNA synthetase (AlaRS), leads to dysregulation of the proteome and reduced viability. Recent studies showed that environmental challenges such as exposure to reactive oxygen species can also alter aaRS synthetic and proofreading functions, prompting us to investigate if oxidation might positively or negatively affect AlaRS activity. DNA-dependent protein kinase catalytic subunit (DNA-PKcs) is a pleiotropic protein kinase that plays critical roles in cellular processes fundamental to cancer. DNA-PKcs expression and activity are frequently deregulated in multiple hematologic and solid tumors and have been tightly linked to poor outcome. Given the potentially influential role of DNA-PKcs in cancer development and progression, therapeutic targeting of this kinase is being tested in preclinical and clinical settings. This review summarizes the latest advances in the field, providing a comprehensive discussion of DNA-PKcs functions in cancer and an update on the clinical assessment of DNA-PK inhibitors in cancer therapy.

Social agents associated with cheerleading environments are increasingly linked to body image dissatisfaction (BID) and eating disorders (ED). This study examined ED risk across team type, squad type, and position. An additional purpose determined BID in clothing type (daily clothing, midriff uniform, and full uniform), and meta-perceptions from the perspective of peers (MP peers), parents (MP parents), and coaches (MP coaches). Female cheerleaders ( = 268) completed an online survey which included demographics, the Eating Attitudes Test-26, and pathogenic behavior questions. Body image perceptions were assessed by using the Sex-Specific Figural Stimuli Silhouettes. Overall, 34.4% of cheerleaders ( = 268; …

Coastal dune habitats provide valuable ecosystem services but are declining globally due to the interacting effects of multiple stressors, including global climate change (GCC). To explore the potential severity of the effects of GCC on dune plant communities of the Northern Gulf of Mexico and surrounding areas, we modeled the fundamental bioclimatic niche of a dune plant species, largeleaf pennywort (Hydrocotyle bonariensis Comm. ex Lam.), in this region. We then projected its niche to estimate habitat suitability under current and future environmental conditions. Two distinct species of New World vultures are native to North America and inhabit Alabama, black vultures (Coragyps atratus) and turkey vultures (Cathartes aura). They are scavengers that consume decaying carcasses or carrion, which helps reduce the spread of disease. Vultures are susceptible to declining population numbers as they have been trapped and killed due to the belief that they spread disease, additionally contact with poisons and habitat destruction have also led to their decline. Type 2 diabetes is a complex metabolic disorder with interactions between genetic and environmental factors, making it more complicated to understand the clinical implications of identified genetic variants and disease pathology. Many of the diabetes-associated variants are currently classified as VUS and occur within the genes involved in insulin signaling and regulation, so we turned our focus on the human INSR gene. INSR encodes the insulin receptor protein that plays a major role in insulin signaling. Marine algae are a rich source of biologically active compounds that can be utilized in various food and pharmaceutical applications. In this study, ultrasound-assisted extraction (UAE) was optimized to maximize yield and total carbohydrate content extracted from the red algae, Pterocladia capillacea. The extract was shown to possess potent antioxidant activity of up to ~70%, and was successfully used as a reducing and capping agent in the green synthesis of copper nanoparticles, which were characterized by UV-spectroscopy, Fourier transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD), transmission electron microscopy (TEM), and dynamic light scattering (DLS). The polar bear (Ursus maritimus) is a popular and easily recognisable species and has a long history of being housed in zoological collections. However, due to concerns regarding space requirements and welfare, many zoos have since removed this species from their collection plans. Polar bears require spacious enclosures with considerable amounts of enrichment and are susceptible to stereotypy if their needs are not met. Yorkshire Wildlife Park, UK, has set up a large enclosure to demonstrate best-practice care of polar bears. We performed a database mining on 102 transcriptomic datasets for the expressions of 29 m6A-RNA methylation (epitranscriptomic) regulators (m6A-RMRs) in 41 diseases and cancers and made significant findings: (1) a few m6A-RMRs were upregulated; and most m6A-RMRs were downregulated in sepsis, acute respiratory distress syndrome, shock, and trauma; (2) half of 29 m6A-RMRs were downregulated in atherosclerosis; (3) inflammatory bowel disease and rheumatoid arthritis modulated m6A-RMRs more than lupus and psoriasis; (4) some organ failures shared eight upregulated m6A-RMRs; end-stage renal failure (ESRF) downregulated 85% of m6A-RMRs; (5) Middle-East respiratory syndrome coronavirus infections modulated m6A-RMRs the most among viral infections; …

Nonylphenol ethoxylates (NEPOs) are widely used as nonionic surfactant in the industry, which pose a threat to environment and human health. In this study, we conducted kinetic experiments and chemical analysis to investigate the effects of NEPOs micelles on degradation kinetics and electro-oxidation degradation mechanism. The electro-oxidation degradation of NEPOs followed pseudo-first order kinetics and more than 92% of NEPOs was degraded. Furthermore, the hydroxyl radicals generated by the Ti4O7 anode contributed to 41% of degradation performance. Animal abuse is an understudied problem in Egypt with possible ramifications on both non-human and human animals and links to interpersonal violence and conduct problems. This study aims to explore the attitudes and behaviors of Egyptian university students and graduates towards the treatment and use of animals in society and to test if exposure to animal abuse is associated with abusive behavior. The study also aims to identify possible protective factors against animal abuse among a group of positive deviants. Atypical protein kinase C (aPKCs) serve key functions in embryonic development by regulating apical-basal polarity. Previous studies have shed light on their roles during adulthood, especially in the development of Alzheimer's disease (AD). Although the crystal structure of PKCι has been resolved, an agonist of aPKCs remains to be discovered. In the present study, by using the Discovery Studio program and LibDock methodology, a small molecule library (K66-X4436 KINA Set) of compounds were screened for potential binding to PKCι. BACKGROUND: Cryptosporidium parvum is an apicomplexan parasite commonly found across many host species with a global infection prevalence in human populations of 7.6%. Understanding its diversity and genomic makeup can help in fighting established infections and prohibiting further transmission. The basis of every genomic study is a high-quality reference genome that has continuity and completeness, thus enabling comprehensive comparative studies.
FINDINGS: Here, we provide a highly accurate and complete reference genome of Cryptosporidium parvum. The assembly is based on Oxford Nanopore reads and was improved using Illumina reads for error correction. We also outline how to evaluate and choose from …

SARS-CoV-2 triggers a complex systemic immune response in circulating blood mononuclear cells. The relationship between immune cell activation of the peripheral compartment and survival in critical COVID-19 remains to be established. Here we use single-cell RNA sequencing and Cellular Indexing of Transcriptomes and Epitomes by sequence mapping to elucidate cell type specific transcriptional signatures that associate with and predict survival in critical COVID-19. Patients who survive infection display activation of antibody processing, early activation response, and cell cycle regulation pathways most prominent within B-, T-, and NK-cell subsets. The embryonic mouse brain undergoes drastic changes in establishing basic anatomical compartments and laying out major axonal connections of the developing brain. Correlating anatomical changes with gene-expression patterns is an essential step toward understanding the mechanisms regulating brain development. Traditionally, this is done in a cross-sectional manner, but the dynamic nature of development calls for probing gene-neuroanatomy interactions in a combined spatiotemporal domain. Here, we present a four-dimensional (4D) spatiotemporal continuum of the embryonic mouse brain from E10.5 to E15.5 reconstructed from diffusion magnetic resonance microscopy (dMRM) data. Butterfly eyespots are beautiful novel traits with an unknown developmental origin. Here we show that eyespots likely originated via cooption of parts of an ancestral appendage gene-regulatory network (GRN) to novel locations on the wing. Using comparative transcriptome analysis, we show that eyespots cluster most closely with antennae, relative to multiple other tissues. Furthermore, three genes essential for eyespot development, (), (), and (), share similar regulatory connections as those observed in the antennal GRN.
