The new Schiff base ligand (LB) was synthesized by reacting precursor (P) with 4-bromobenzaldehyde. This ligand formed three new complexes: [Pd(LB)(H2O)2]Cl2 (square planar) and [M(LB)(H2O)2Cl2].mCl2 (octahedral), where m=0 for Co+2 and 1 for Pt+4, upon reaction with the respective metal ions. Characterization of the ligand (LB) and its metal complexes involved various techniques including 1H- and 13C-NMR, FT-IR, UV-Vis spectroscopy, magnetic susceptibility measurements, CHNS analysis, and molar conductivity. Breast cancer treatment has many options directed at therapeutic targets, but some remain incurable, so new therapies are needed. Pyrazoline has shown various biological activities, one of which is anticancer effectiveness. The purpose of this study was to conduct an in silico analysis of Pyrazoline inhibition of PI3K and PR proteins using docking and MD simulation methods. Predictions of toxicity and pharmacokinetics, including absorption, distribution, metabolism, excretion, and toxicity, were also conducted computationally. The Maillard mechanism frequently gives numerous varieties of food a distinctive flavor and visually appealing hue. The Maillard reaction (MR) is additionally referred to as the non-enzymatic glycosylation, and MR contain a variety of chemical compounds, including precursors for the brown hue generated in the Maillard reaction and compounds that eventually form melanoidins, which are brown that develop into dark-colored hue nitrogenous polymer molecules. Melanoidin polymer was synthesized by integrating L-glutamine and D-fructose and then scorching the resulting mixture at 130 °C for 20 minutes. An analysis was conducted on the variation in the counts of seven meristic traits of the eastern mosquito fish, Gambusia holbrooki, using samples collected from ten different sites in the Middle East. These sites are located along the Tigris, Euphrates, and Shatt al-Arab Rivers, spanning across Iraq, Iran,Türkiye, and Syria. The study revealed a northward increase in the average values of these meristic traits, with the highest recorded in Türkiye and the lowest in the southern region of Iraq. The statistical analysis using Boxplots demonstrated that the seven meristic features were distinct across different rivers, with no overlap observed. …

BACKGROUNDThe SARS-CoV-2 pandemic provided a rare opportunity to study how human immune responses develop to a novel viral antigen delivered through different vaccine platforms. However, to date, no study has directly compared immune responses to all 3 FDA-approved COVID-19 vaccines at single-cell multiomic resolution.METHODSWe longitudinally profiled SARS-CoV-2-naive adults (n = 31) vaccinated with BNT162b2, mRNA-1273, or Ad26.COV2.S, integrating plasma cytokines, antibody titers, and single-cell multiomic data (DOGMA-Seq).RESULTSWe discovered a distinct, transient IFN program termed ISG-dim, which emerged specifically 1-2 days after the first mRNA dose in approximately 10% of myeloid cells. Mechanosensitive Piezo1 channels participate in regulating pain sensitivity, insulin secretion, and vascular tension; however, their expression in the autonomic paraventricular nucleus (PVN) and role in modulating sympathetic outflow and cardiovascular function remain unstudied. In this study, unilateral PVN microinjection of the Piezo1 channel blocker Dooku1 (0.1, 1, 10, 100, and 200 pmol) administered to anesthetized male rats increased renal sympathetic nerve activity (RSNA) and mean artery pressure (MAP) in a dose-dependent manner, with maximum increases of 93 ± 30% (p < 0.0001) and 21 ± 5 mmHg (p < 0.0001), respectively, elicited by Dooku1 at 100 pmol. Similarly, PVN microinjection of the peptide Piezo1 channel blocker GsMTx4 (1 nmol) significantly increased RSNA (p < 0.001) and MAP (p < 0.0001). Conversely, PVN-microinjected Piezo1 channel activators Yoda1 (5 nmol) and Jedi2 (5 nmol) did not significantly alter RSNA or MAP. Western blot and qRT-PCR analyses of the hypothalamic PVN showed abundant Piezo1 mRNA and protein expression. Immunofluorescence detection showed that Piezo1 was expressed in pre-sympathetic PVN neurons with axons projecting to the rostral ventrolateral medulla. We conclude that Piezo1 channels expressed in the autonomic PVN neurons play an important role in regulating sympathetic outflow and cardiovascular function.

Background/aim: Chlorpyrifos (CPF) is an organophosphate pesticide extensively used in agriculture, posing significant threats to human health due to its neurotoxic effects. Its presence in food products has raised considerable public health concerns, necessitating the development of rapid, sensitive, and reliable detection methods. This study aims to design and validate an electrochemiluminescence (ECL) sensor platform operating via a quenching-based mechanism to enable ultrasensitive detection of CPF in food matrices.

Background/aim: Limited drug development and increased antibiotic tolerance among Gram-negative bacteria have led researchers to consider combination therapy. Actinomycin derivatives, particularly actinomycin D, exhibit a wide range of bioactivities, including antibacterial effects. However, actinomycin D is less effective against Gram-negative pathogens. Therefore, it is essential to demonstrate the synergy of actinomycin D and its derivatives with clinically known antibiotics against Escherichia coli ATCC 25922 and investigate the effects on cellular morphology.

Background/aim: Neuregulin 3 (NRG3) is an epidermal growth factor-like ligand that is involved in neuronal circuit formation in mammals. However, the expression profile of its zebrafish ortholog, nrg3b, remains poorly defined. The transcriptomic analyses of zebrafish sco-spondin mutants, which display alterations in their neurodevelopmental pathways, have indicated potential dysregulation of Nrg3b. Background/aim: High-grade serous ovarian cancer (HGSOC) remains one of the most aggressive forms of ovarian malignancy and frequently shows resistance to conventional therapies. This study aimed to synthesize a novel series of purine analogues, 6-[(4-substituted benzyl amine)/(4-substituted aniline)]-9-cyclopentyl purines, and evaluate their anticancer efficacy against HGSOC cell lines.
Materials and methods: We assessed the biological effects of the synthesized purine analogues on the OVCAR3, OVSAHO, and KURAMOCHI HGSOC cell lines using the sulforhodamine B assay. Synthesis of six new curcumin analogues was performed through condensation reaction of 3-methyl-2, 4-pentandione with substituted aromatic aldehyde derivatives and boron oxide in the presence of trimethylborate and butylamine. The reaction was carried out under ultrasonic irradiation. The synthesized compounds were identified using 1H-, 13C-NMR, FT-IR spectroscopy, and electron impact mass spectrometry. The Swiss ADME website was used to predict the physicochemical properties of the new compounds using computational techniques like ADME studies. Allergies are a common health issue where the immune system reacts to substances in the environment that are normally harmless. This study further investigates the effects of vascular endothelial growth factor (VEGF), neutrophil gelatinase-associated lipocalin (NGAL), interleukin-38 (IL-38), immunoglobulin E (IgE), and the neutrophil-lymphocyte ratio (NLR) in these individuals, offering insight into the probable processes behind allergic responses following the pandemic. The Schiff base ligand (L)- 1-((7-((Z), was recently synthesized 2–(2-((Z)–5,5-diethyl2-(methoxyimino)acetamido)-2-(methoxycarbonyl)-2,6-dioxotetrahydropyrimidin-4(1H)-ylideneamino) thiazol-4-yl8-oxo-5-thia , the next mineral ions, Ni2+, Cu2+, Zn2+, and Co2+, are permitted to react with 1-azabicyclo[4.2.0]oct-2-en-3-yl)methyl)-1-methylpyrrolidinium, leading to the formation of new metal complexes with distinct geometric shapes. By detecting the shifting in the azomethines band and the appearance of M-N and M-O bands, FT-IR confirmed the occurrence of coordination through N of azobenzene and two O atoms of beta lactam and ester, indicating the development of Schiff base complexes as well as the original Schiff base. Lung cancer remains the leading cause of cancer-related deaths worldwide, partly because many patients are diagnosed at late stages, highlighting the urgent need for effective early detection and intervention strategies. Low-dose computed tomography (LDCT)-based screening reduces lung cancer mortality in high-risk populations defined by age and smoking history; however, the uptake of LDCT remains low among eligible individuals. Compounding this issue, modelling studies estimate that nearly half of lung cancers occur in individuals who do not meet eligibility criteria for LDCT-based lung cancer screening under current guidelines. SUPT16H encodes a subunit of the FACT (FAcilitates Chromatin Transcription) complex, a histone chaperone essential for maintaining chromatin integrity during transcription, replication, and DNA repair. Pathogenic de novo SUPT16H missense variants have previously been linked to neurodevelopmental disorders in eight individuals. Here, we expand the genotypic and phenotypic spectrum by identifying 24 additional individuals harboring ultrarare heterozygous missense or truncating variants, who share overlapping clinical features including intellectual disability, autism spectrum disorder, hypotonia, and characteristic craniofacial dysmorphism. To elucidate the underlying mechanisms, we generated a supt16h knockout zebrafish model using CRISPR/Cas9. The supt16h loss-of-function (LOF) model recapitulated key patient phenotypes …

Objectives: In higher education institutions (HEIs), building a strong quality culture (QC) is essential to ensure the quality of education provided. QC integrates organizational, managerial, and psychological elements that shape how quality is understood and improved in HEIs. This study explores how leaders at the departmental and faculty levels navigate and contribute to quality culture development (QCD).
Methods: We employed a sequential explanatory mixed-methods design. A total of 1,029 academics completed questionnaires, and 33 leaders at the departmental and faculty levels were interviewed at three public universities in Ethiopia.

Cytokines are immune elements that have been found as marker either for tumor-promoting or inhibiting effects on breast tumor development, depending evidently on their exact quantities as well as the existence of additional modifying variables. The study aims to evaluate the relationship between breast cancer tissue and the expression level of IL-19 and IL-33. This study includes using breast cancer tissue (30 specimens) and normal breast tissue (20 specimens) to analyze the expression level of IL-19 and IL-30.
