T cell engagers (TCEs) recruit T cells to the tumor microenvironment (TME) to induce antitumor immune responses. TCEs have demonstrated promising clinical responses in patients with metastatic castration-resistant prostate cancer and have advanced into phase 3 clinical trials. Here we provide an overview of the mechanisms of action of TCEs, including both CD3-targeted and CD28-targeted agents, and review the clinical development of these agents for prostate cancer. We propose a path forward for TCEs in prostate cancer, in which innovative clinical trials will facilitate a biological understanding of mechanisms of efficacy and toxicity to inform: (1) development of predictive biomarkers …

Winner's Curse Free Robust Mendelian Randomization With Summary Data, Zhongming Xie, Wanheng Zhang, Jingshen Wang, Chong Wu

In the past decade, the increased availability of genome-wide association studies summary data has popularized Mendelian Randomization (MR) for conducting causal inference. MR analyses, incorporating genetic variants as instrumental variables, are known for their robustness against reverse causation bias and unmeasured confounders. Nevertheless, classical MR analyses using summary data may still produce biased causal effect estimates due to the winner’s curse and pleiotropy issues. To address these two issues and establish valid causal conclusions, we propose a unified robust Mendelian Randomization framework with summary data, which systematically removes the winner’s curse and screens out invalid genetic instruments with pleiotropic effects. …

The rapid expansion of electrified transportation and renewable energy integration demands advanced lithium-ion (Li-ion) and sodium-ion (Na-ion) batteries with enhanced safety, long-term reliability, and high energy efficiency. However, conventional systems employing liquid electrolytes continue to exhibit persistent failure modes, including thermal instability, parasitic interfacial reactions, transition-metal dissolution, and electrode degradation under high-voltage or fast-charging conditions. These limitations become even more critical in solid-state configurations, where insufficient ion mobility and interfacial incompatibility can severely restrict electrochemical performance. Exonuclease/endonuclease/phosphatase (EEP)-fold hydrolases are canonically monomeric phosphodiesterases exemplified by APE1, DNase I, and TDP2 nucleases. While EEP family domain containing protein 1 (EEPD1) acts in DNA stress responses, its proposed nuclease activities are enigmatic. Here, we integrate hybrid structural methods, evolution, biochemistry, cancer genomics, plus molecular and cell biology to define EEPD1 structure, assembly, and function at stalled DNA replication forks. Results imply EEPD1 surprisingly requires both unique EEP domain dimer and distinctive tandem Helix-hairpin-Helix [(HhH)2] domains to clamp double-stranded (ds) DNA at reversed DNA replication forks for fork protection. Colibactin is a genotoxin produced by Enterobacteriaceae via a polyketide synthase (pks) island cluster. There is less knowledge regarding the distribution of colibactin genes in E. coli isolates in Iraq and its correlation with biofilm and antibiotic susceptibility. Therefore, this study aimed to investigate the frequency of some colibactin genes (CIbA and CIbQ) in uropathogenic E. coli in Iraq and evaluate the correlation with biofilm and antimicrobial resistance. Human head lice are widely dispersed ectoparasites in the world and have been divided into 6 haplogroups (A to F). There are no studies about the louse haplogroup type in Iraq. Thus, the study aimed to determine the species of head lice and their prevalence rate in primary school students in north Basrah and also aimed to determine the genotype and haplogroup of head lice in these regions. The paper addresses the potential air pollution levels resulting from the combustion of natural gas in the Rumaila combined cycle power plant in Basrah city. The study aims to evaluate the concentrations of pollutants (carbon monoxide, sulfur dioxide, nitrogen oxides, and particulate matter) at distances of 100, 500, and 1000 meters from the plant using a Gaussian dispersion model. This includes calculating the emission rates of pollutants from the stacks and estimating atmospheric stability based on data from the Iraqi Meteorological Authority. Once declared biologically dead, the Pasig River now shows signs of aquatic life but remains heavily polluted with heavy metals like lead (Pb), which accumulate in fish and pose health risks. Despite this, many communities still rely on the river for food, particularly tilapia. This study addresses public health concerns by evaluating whether common cooking methods—frying, grilling, and steaming can reduce Pb levels in tilapia (Oreochromis niloticus) from the Pasig River to safe levels for consumption. Background/aim: Brain microvascular endothelial cells (BMECs), which constitute the blood–brain barrier (BBB), are essential for maintaining central nervous system homeostasis. Like BMECs, multipotent mesenchymal stem cells (MSCs) originate from the mesodermal lineage. Thus, MSCs may serve as a direct and efficient cellular source for BMEC-like differentiation. Notably, differentiation of human induced pluripotent stem cells (hiPSCs) into BMECs typically involves a 2-step protocol: inducing mesodermal commitment followed by endothelial specification. In contrast, direct differentiation from MSCs could bypass the initial mesodermal induction step, offering a streamlined alternative. Pancreatic ductal adenocarcinoma (PDAC) is the most lethal form of pancreatic cancer, with poor prognosis driven by late diagnosis, therapeutic resistance, and an immunosuppressive tumor microenvironment. Interactions between tumor cells and immune cells promote immune evasion and tumor progression, limiting the efficacy of immune checkpoint blockade and other immunotherapies. Given the high expression of trophoblast cell surface antigen 2 (TROP2) in PDAC, we developed a TROP2-targeted Toll-like receptor 7 (TLR7) agonist (E104) designed for selective accumulation within the tumor microenvironment to activate antitumor immunity. BACKGROUND. The SARS-CoV-2 pandemic provided a rare opportunity to study how human immune responses develop to a novel viral antigen delivered through different vaccine platforms. However, to date, no study has directly compared immune responses to all 3 FDA-approved COVID-19 vaccines at single-cell multiomic resolution.

METHODS. We longitudinally profiled SARS-CoV-2–naive adults (n = 31) vaccinated with BNT162b2, mRNA-1273, or Ad26.COV2.S, integrating plasma cytokines, antibody titers, and single-cell multiomic data (DOGMA-Seq).

RESULTS. We discovered a distinct, transient IFN program termed ISG-dim, which emerged specifically 1–2 days after the first mRNA dose in approximately 10% of myeloid cells. This state was …

Background/aim: Pyrazole derivatives are of growing interest due to their diverse pharmacological activities. However, their biological activity is often highly sensitive to subtle structural modifications. Existing quantitative structure–activity relationships (QSAR) approaches frequently fail to capture the conformational flexibility and nonlinear structure–activity relationships (SAR) of such heterocyclic scaffolds, creating a gap in the accurate prediction of their biological profiles.
