Impulsivity In Cerebellar Ataxia: An Online, Multidimensional Assessment, Brooke Chasalow, Yakov Flaumenhaft, Yael De Picciotto, Chi-Ying R Lin, Leila Montaser-Kouhsari, William Saban

Faculty, Staff and Students Publications

While considered a motor control structure, the cerebellum contributes to non-motor functions, including impulsivity. However, whether it contributes to impulsivity in a domain-specific manner remains unknown. Studies on cerebellar ataxia (CA), a common model for cerebellar dysfunction, typically have small sample sizes, limiting robustness. In a multicenter cross-sectional study, we investigated the cerebellum's role in various forms of impulsivity by comparing large cohorts of CA to age- and education-matched neurotypical healthy (NH) controls. Additionally, to examine the ability to identify individuals with CA using impulsivity features alone, we developed supervised machine learning (ML) models. In experiment 1 (CA = 140, …

Does Genetic Testing Influence Outcomes: Early Experience In Melanoma Case Series, Ryan Reyes, George Skenteris, Stella Coker Watson Self Ph.D., Ms, Christine Marie-Gilligan Schammel, Steven D. Trocha

Faculty Publications

Background:

The treatment and prognosis of melanoma have historically been based on histologic stage and Breslow depth; however, due to the increase in surveillance, melanomas are being identified at an earlier stage and lower Breslow depth. Advances in genetic testing, such as Decision Dx®, mean that melanoma diagnostic decisions and prognosis can now be directed by genetics. The purpose of this project was to assess the influence of a Decision Dx® high-grade (Class 2A/B) classification on the treatment of melanoma patients at a regional medical center, particularly those not deemed high risk by conventional classification methods, including Breslow depth and …

The Role Of Top-Down Appetite Self-Regulation In The Development Of Healthy Eating Behaviors Among Children: A Narrative Review And Socialization Framework, David J Bridgett, Sheryl O Hughes, Matthew Broussard, Daniela Mccourt, Christina M Croce, Jennifer O Fisher Mar 2026

The Role Of Top-Down Appetite Self-Regulation In The Development Of Healthy Eating Behaviors Among Children: A Narrative Review And Socialization Framework, David J Bridgett, Sheryl O Hughes, Matthew Broussard, Daniela Mccourt, Christina M Croce, Jennifer O Fisher

Children’s Nutrition Research Center Staff Publications

Appetite self-regulation (ASR) among children is thought to have a fundamental role in shaping the development of healthy eating behaviors, dietary intake, and growth during childhood. Parallel to developmental frameworks for understanding "general" self-regulation among children, ASR has been described as involving children's use of "top-down" cognitive processes to moderate "bottom-up" biological drives around food approach and avoidance in the interest of achieving desired eating behaviors or outcomes. Whereas bottom-up ASR processes during early childhood are well characterized, particularly in the context of dysregulation and obesity risk, the role of top-down ASR processes in the development of healthy eating behaviors …

The New York State Dental Journal

Background: Dens evaginatus cusps typically have pulpal tissue, and fracturing of the cusp leads to necrosis. Immature permanent teeth with pulp necrosis have traditionally been treated with apexification, which eliminates infection but does not promote root development.

Methods: Regenerative endodontics utilizes revascularization by infiltrating stem cells from the apical papilla and using them to treat an immature necrotic tooth with periapical infection.

Results: Periapical radiolucency completely resolved, root length and thickness increased, and vitality returned at the one-year follow-up.

Conclusion: Regenerative protocols promote root development, reducing fracture risk. This treatment offers improved outcomes with fewer appointments and shorter treatment duration.

A modular synthetic route has been developed to prepare a new series of cationic ruthenium(II) complexes with electron-withdrawing 1,2-diacylcyclopentadienyl ligands. The 2-acyl-6-hydroxyfulvenes were synthesized from cyclopentadienide and acyl chlorides and converted to Tl(I) cyclopentadienyl salts using Tl2SO4/KOH. Transmetalation with [Ru(η6-p-cymene)(μ-Cl)Cl]2 followed by PF6 metathesis gives the complexes [Ru{η5-1,2-C5H3(CO–R)2}(η6-p-cymene)][PF6] (R = t-Bu, p-Tol, p-ClC6H4, p-IC6H4) in moderate to high yields. The new compounds were characterized by NMR and IR spectroscopy; mass spectrometry …

Polygyny Between Sharia, Custom And State Law As A Persistent Dilemma In Post-Soviet Kyrgyzstan, Zharkynbai Sebetov, Mairambek Tagaev

Occasional Papers on Religion in Eastern Europe

This article presents a comprehensive study of the century-long historical and legal evolution of state-imposed prohibitions on the institution of polygyny in Kyrgyzstan. The primary objective of the research is to evaluate the outcomes of a hundred years of repressive policies – originally classified as “crimes of daily life” (bytovye prestupleniya) during the early Soviet era–and to uncover the transformed legal paradoxes of this phenomenon today. Drawing on historical archival materials, Communist Party reports, and contemporary regulatory acts, the authors investigate the fundamental reasons why polygyny persists as a resilient “shadow” institution in Kyrgyz society despite its criminalization. Within the …

Population stratification is one of the source of inflation in epigenome-wide association studies (EWAS) when not properly accounted for. To address this, we developed methylation population scores (MPSs) to predict genetic principal components (GPCs) using a feature selection approach. We used multi-ethnic DNA methylation data from Illumina EPIC arrays across five cohorts, including MESA (n = 929), CARDIA (n = 1123), JHS (n = 1365), ARIC (n = 2338), and HCHS/SOL (n = 1475), randomly splitting participants into training (85%) and test (15%) sets. Within each cohort, associations between GPCs and CpG sites were estimated using linear regression adjusting for …

The Brown Foundation: Institute of Molecular Medicine

The choroid plexus forms a key barrier and signaling interface between the brain and peripheral circulation, yet its immune landscape remains incompletely understood. Using single-cell transcriptomics combined with lineage and spatial tracing methods, we identified three biologically distinct populations of choroid plexus macrophages, defined by differential expression of CD163, MHCII or CD9. These subsets arise from separate hematopoietic waves, occupy distinct anatomical niches and differentially rely on CSF1 and IL-34 for survival. We found that TGFβ signaling is essential to maintain their tissue-specific identities, and deletion of Tgfbr2 in these cells induces broad phenotypic reprogramming. During neuroinflammation, choroid plexus macrophages …

Glutamatergic Dysfunction Of Astrocytes In Paraventricular Nucleus Of Thalamus Contributes To Adult Anxiety Susceptibility In Adolescent Ethanol Exposed Mice, Aubrey Bennett, Hyunjung Kim, David Thomas, Peter Biggs, Roxan Ara, Asamoah Bosomtwi, Seungwoo Kang Mar 2026

Glutamatergic Dysfunction Of Astrocytes In Paraventricular Nucleus Of Thalamus Contributes To Adult Anxiety Susceptibility In Adolescent Ethanol Exposed Mice, Aubrey Bennett, Hyunjung Kim, David Thomas, Peter Biggs, Roxan Ara, Asamoah Bosomtwi, Seungwoo Kang

The Brown Foundation: Institute of Molecular Medicine

Repeated ethanol exposure during adolescence increases adult anxiety risk, but the underlying mechanisms remain unclear. The paraventricular nucleus of the thalamus (PVT) has been considered a hub for controlling anxiety and is affected by experiences from early life. Thus, this study investigated how adolescent intermittent repeated ethanol exposure (AIE) affects the PVT activities and anxiety-related behaviors in adulthood. We found that AIE triggers anxiety-like behaviors and parallelly exhibited elevated firing rates and increased calcium signaling in the PVT neurons compared to control counterpart mice. Chemogenetic inhibition of PVT neurons reduced anxiety-like behaviors in AIE-treated animals, confirming PVT's role in adolescent …

Lipids Grease The Chain Of Cancer Progression, Joseph Rupert, Pham Hong Anh Cao, Daniel E Frigo, Mikhail G Kolonin Mar 2026

Lipids Grease The Chain Of Cancer Progression, Joseph Rupert, Pham Hong Anh Cao, Daniel E Frigo, Mikhail G Kolonin

The Brown Foundation: Institute of Molecular Medicine

The role of lipids in cancer progression has become a fervent area of exploration. The crosstalk of tumors with adipose tissue is a complex but well-regulated orchestration of signaling pathways, lipid transporters, and enzymes. They regulate fatty acid synthesis, their deposition into lipid droplets (LDs) as triglycerides, induction of lipolysis, shuttling lipids across cells, and their systemic trafficking, modification, and catabolism. For the latter, lipid oxidation has emerged as a metabolic process of particular clinical importance. Products of lipid processing can become secondary messengers, contribute to reactive oxygen species (ROS) generation, stimulate the production of antioxidants, and, if left unchecked, …

The Brown Foundation: Institute of Molecular Medicine

Engineered immunoglobulin M (IgM) antibodies typically exhibit superior neutralization potency and avidity compared to their parental IgG counterparts, primarily due to multivalent binding to repeated epitopes on a targeting antigen. In this study, we characterize the neutralization breadth and mechanism of action of IgM-14, a previously reported intranasally deliverable antibody targeting SARS-CoV-2. IgM-14 demonstrates remarkably potent antiviral activity against all pre-Omicron variants but significantly reduced efficacy against Omicron BA.1, and complete loss of activity against the later subvariant JN.1. Resistance selection identified two key mutations in the receptor-binding domain (RBD), G476D and F486P, which disrupt IgM-14 binding and confer strong …

SR12813 is an experimental cholesterol-lowering drug that reduces intracellular cholesterol through accelerated proteasomal degradation of 3-hydroxy-3-methylglutaryl-coenzyme A reductase and is also recognized as a prototypical activator of the pregnane X receptor (PXR, NR1I2). Rifampicin, a clinically used antibiotic, likewise functions as a human PXR agonist. Although PXR-mediated induction of drug metabolism genes has been extensively characterized in hepatocytes and humanized mouse liver, comparatively little is known about the transcriptional effects of these ligands in intestinal and colon cancer cells. Here, we used RNA-sequencing in LS180 colon adenocarcinoma cells to compare transcriptional responses elicited by SR12813 and rifampicin. Both compounds induced …

Comparing Methods For Measuring 210po In Seawater: Fe(Oh)3 Can Extract Less 210po Than Co-Apdc And Thus Overestimate Poc Export Fluxes, Harold Carrasco, J. Kirk Cochran, Beat Gasser, Inés Sanz-Alvarez, Montserrat Roca-Martí, Christina Heilbrun, Nicholas S. Fisher, Jana Friedrich, Pere Masqué

Research outputs 2022 to 2026

Over six decades of research, two methods have been used to extract the natural radionuclides 210Pb and 210Po from seawater- the precipitation of iron hydroxide (Fe(OH)3) and coagulation of cobalt ammonium pyrrolidine dithiocarbamate (Co-APDC). Although the two methods had been intercompared for 210Pb and shown to agree, they were never intercompared for 210Po until Roca-Martí et al. (2021) , in which only unfiltered samples were used. Here we document that Fe(OH)3 may be less effective in extracting 210Po from a seawater sample than Co-APDC. We hypothesize that cycling of 210Po through the marine food …

Broadhurst, Thomas Dignan, Jamie Whitfield, Lisa Murray-Segal, Naomi X.Y. Ling, Jonathan S. Oakhill, Bruce E. Kemp, John A. Hawley, Stacey N. Reinke, Nolan J. Hoffman

Research outputs 2022 to 2026

Background/Objectives: Acute exercise remodels many interconnected biochemical pathways in metabolically active tissues. This remodeling involves the activation of the energy-sensing AMP-activated protein kinase (AMPK) to maintain cellular energy homeostasis. Critical energy reserves of glycogen, primarily stored in liver and skeletal muscle and known to interact with AMPK, are utilized to help meet increased energy demands with exercise. However, the breadth of metabolic pathways regulated by acute exercise and AMPK’s interactive roles with glycogen remain incompletely understood. This study therefore aimed to map mouse liver and skeletal muscle metabolite responses to continuous acute exercise and disruption of AMPK-glycogen interactions. Methods: Liquid …

Generative AI (GenAI) is increasingly integrated into users’ daily work as a collaborative partner. Drawing on the symbiotic agency theory, this study investigates the quality of human-GenAI collaboration using a mixed-methods approach, including qualitative interviews and quantitative surveys. Study 1 identified three dimensions of human-GenAI collaboration quality that comprise outcome quality, comfort, and efficiency; and six driving factors which can be categorized under human agency (domain knowledge, desire for control, and domestication ability), and GenAI agency (communication ability, working memory, and long-term memory). Study 2 applied fuzzy-set qualitative comparative analysis (fsQCA) to explore configurations of the driving factors that lead …

Congenital heart disease (CHD) remains the most common birth defect and a leading contributor to global infant morbidity and mortality. Early diagnosis improves outcomes, yet current screening methods frequently miss CHD during routine prenatal screening. The placenta, which develops in parallel with the fetal heart and shares critical molecular pathways, represents an underexplored diagnostic tissue. We highlight clinical, epidemiological, and mechanistic insights that link placental abnormalities to CHD and explore how imaging, in vitro models, and multiomics can improve our understanding of the placenta–heart axis. Leveraging these tools will offer opportunities for earlier CHD detection and illuminate avenues for intervention …
