The Association Between Symptoms Of Adhd And Alcohol Use And Related Consequences In College Students: Social And Environmental Protective And Risk Factors As Moderators Of This Relation, Cameron Shepard Massey

Theses and Dissertations

Alcohol use, while common in college student populations, can bring about significant mental health and educational problems that have negative consequences (Harris et al., 2017; White & Hingson, 2013). Attention-deficit/hyperactivity disorder (ADHD) can also result in significant impairment in academic settings (Kuriyan et al., 2012; Blase et al., 2009, Barbaresi et al., 2007), and those with ADHD often show increased rates of alcohol consumption and related problems, which can greatly increase the risk for students to struggle in college (Barbaresi et al., 2013; Baker et al., 2012). Examining the impact that specific risk and protective factors play in this relation …

The Use Of Project-Based Learning To Scaffold Student Social And Emotional Learning Skill Development, Science Identity, And Science Self-Efficacy, Michelle Sutton Spigner Oct 2023

The Use Of Project-Based Learning To Scaffold Student Social And Emotional Learning Skill Development, Science Identity, And Science Self-Efficacy, Michelle Sutton Spigner

Theses and Dissertations

Project-based learning (PBL) offers an ideal approach for cultivating a robust STEM workforce, empowering students to actively construct their knowledge. This process has the potential to enhance science identity and self-efficacy. Furthermore, because PBL is inherently collaborative, it fosters the development of 21st-century skills, many of which align with social and emotional learning (SEL). While past studies have explored the relationship between PBL, SEL, science identity, and self-efficacy, there is a research gap in examining all three variables collectively within a high school context. Thus, this mixed-methods action research study aimed to determine if the implementation of a PBL unit, …

From Oceans To Public Health: Impact Of Climate Change And Socioecological Factors On Non-Cholera Vibrio Spp., Karlen Enid Correa Vélez Oct 2023

From Oceans To Public Health: Impact Of Climate Change And Socioecological Factors On Non-Cholera Vibrio Spp., Karlen Enid Correa Vélez

Theses and Dissertations

Climate change-induced stressors, such as temperature, salinity, pH, and wildfire, are contributing factors to the increase in frequency of Vibrio infections, altering Vibrio geographical constraints. At the same time, the expansion of the coastal population is resulting in increased input of treated municipal wastewater into areas that are also experiencing Vibrio proliferation. Given that increases in ocean temperature and sea level coupled with increases in human activity in coastal waters support greater Vibrio-human interactions, it is important to examine factors and mechanisms within this socio-ecological coupled system that may further strengthen this interaction. This study aimed to better understand the …

Theses and Dissertations

The two-spotted spider mite, Tetranychus urticae, is a major cosmopolitan pest that feeds on more than 1,100 plant species, including 150 crops. Such a wide host range indicates this mite’s unparalleled xenobiotic responsiveness that is also coincident with their ability to rapidly develop resistance to pesticides. An unprecedently large detoxifying machinery is one of the main biological traits responsible for their extraordinary adaptation potential and devastating effect on crops. Availability of the whole genome sequence revealed expansions in all major detoxifying families and demonstrated that this genome has been enriched by horizontally transferred genes from bacteria and fungi. Motivated by …

Biology: Faculty Publications and Other Works

Phytoliths are opal silica particles formed within plant tissues. Diatoms are aquatic, single-celled photosynthetic algae with silica skeletons. Phytolith and diatom morphotypes vary depending on local environmental and climatic conditions and because their silicate structures preserve well, the study of phytolith and diatom morphotypes can be used to better understand paleoclimatic and paleoenvironmental dynamics and changes. This article presents original data from an 820cm-deep stratigraphy excavated at the Hazen diatomite deposits, a high-elevation desert paleolake in the Fernley District, Northern Nevada, USA. The site has been studied for an assemblage of fossilized threespine stickleback, Gasterosteus doryssus, that reveal adaptive …

Theses and Dissertations

Paddle stamped pottery has a long history in what we now call the southeastern United States. From 100-800 CE, intricate curvilinear designs were carved into paddles and impressed in ceramic vessels in Georgia, Florida, Alabama, Tennessee, and South Carolina. Called Swift Creek Complicated Stamped, this type has been long recognized by archaeologists for its uniqueness. Artists Bettye J. Broyles and Frankie Snow reconstructed hundreds of paddle designs from sherds and modern archaeologists use these designs to study the movements and connections of the people who made the pottery. The Snowvision team has developed a machine learning computer vision algorithm to …

Broken Bottles: Understanding The Relationship Between Alcohol Outlet Availability And The Proximity To Alcohol Outlets With Binge Drinking Behaviors In College Fraternity Men At Southeastern Public Universities, Jarod Alan Holt

Theses and Dissertations

Alcohol use among fraternity affiliated men presents a concerning trend with harmful consequences (DeSimone, 2009; Hingson et al., 2017; McCreary et al., 2021; Nuwer, 2001; Patrick et al., 2022; Ranker & Lipson, 2022). The availability theory of alcohol-related problems suggests alcohol use could be influenced based on the availability and proximity of alcohol outlets in the environment (Dimova, 2023; Kypri et al., 2008; Scribner et al., 2008; Single, 1984; Weitzmann et al., 2003). However, no existing studies of fraternity-affiliated students have examined the relationship of binge drinking and access to alcohol through alcohol outlet availability or proximity. This study examined …

Design And Synthesis Of N-Succinyl-L,L-2.6-Diaminopimelic Acid Desuccinylase Inhibitors As Potential Novel Antibiotics, Thomas Dipuma Oct 2023

Design And Synthesis Of N-Succinyl-L,L-2.6-Diaminopimelic Acid Desuccinylase Inhibitors As Potential Novel Antibiotics, Thomas Dipuma

Master's Theses

N-Succinyl-L,L-2,6-Diaminopimelic Acid Desuccinylase (DapE) is a bacterial enzyme located in the lysine biosynthetic pathway of all Gram-negative and most Gram-positive species of bacteria including the notorious ESKAPE pathogens, or pathogenic strains of bacteria that have developed significant levels of resistance to currently available antibiotics. As a means to combat this challenge of growing antimicrobial resistance, we have identified the bacterial enzyme DapE as a conserved, novel enzymate target within these resistant bacterial strains,as DapE is responsible for the production of m-DAP and lysine which are ultimately employed in bacterial cell wall synthesis. The overall aim of this research is to …

Histopathologic whole-slide images (WSI) are generally considered the gold standard for cancer diagnosis and prognosis. Survival prediction based on WSI has recently attracted substantial attention. Nevertheless, it remains a central challenge owing to the inherent difficulties of predicting patient prognosis and effectively extracting informative survival-specific representations from WSI with highly compounded gigapixels. In this study, we present a fully automated cellular-level dual global fusion pipeline for survival prediction. Specifically, the proposed method first describes the composition of different cell populations on WSI. Then, it generates dimension-reduced WSI-embedded maps, allowing for efficient investigation of the tumor microenvironment. In addition, we introduce …

Faculty, Staff and Student Publications

Glioblastoma (GBM) tumors consist of multiple cell populations, including self-renewing glioblastoma stem cells (GSCs) and immunosuppressive microglia. Here we identified Kunitz-type protease inhibitor TFPI2 as a critical factor connecting these cell populations and their associated GBM hallmarks of stemness and immunosuppression. TFPI2 promotes GSC self-renewal and tumor growth via activation of the c-Jun N-terminal kinase-signal transducer and activator of transcription (STAT)3 pathway. Secreted TFPI2 interacts with its functional receptor CD51 on microglia to trigger the infiltration and immunosuppressive polarization of microglia through activation of STAT6 signaling. Inhibition of the TFPI2-CD51-STAT6 signaling axis activates T cells and synergizes with anti-PD1 therapy …

Advances In Vaccine Development For Cancer Prevention And Treatment In Lynch Syndrome, Ana M Bolivar, Fahriye Duzagac, Krishna M Sinha, Eduardo Vilar

Faculty, Staff and Student Publications

Lynch Syndrome (LS) is one of the most common hereditary cancer syndromes, and is caused by mutations in one of the four DNA mismatch repair (MMR) genes, namely MLH1, MSH2, MSH6 and PMS2. Tumors developed by LS carriers display high levels of microsatellite instability, which leads to the accumulation of large numbers of mutations, among which frameshift insertion/deletions (indels) within microsatellite (MS) loci are the most common. As a result, MMR-deficient (MMRd) cells generate increased rates of tumor-specific neoantigens (neoAgs) that can be recognized by the immune system to activate cancer cell killing. In this context, LS is an ideal …

KRAS is the most commonly mutated oncogene in advanced, non-squamous, non-small cell lung cancer (NSCLC) in Western countries. Of the various KRAS mutants, KRAS G12C is the most common variant (~40%), representing 10-13% of advanced non-squamous NSCLC. Recent regulatory approvals of the KRASG12C-selective inhibitors sotorasib and adagrasib for patients with advanced or metastatic NSCLC harboring KRASG12C have transformed KRAS into a druggable target. In this review, we explore the evolving role of KRAS from a prognostic to a predictive biomarker in advanced NSCLC, discussing KRAS G12C biology, real-world prevalence, clinical relevance of co-mutations, and approaches to molecular testing. Real-world evidence …

Faculty, Staff and Student Publications

The term variant histology renal cell carcinomas (vhRCCs), also known as non-clear cell RCCs, refers to a diverse group of malignancies with distinct biologic and therapeutic considerations. The management of vhRCC subtypes is often based on extrapolating results from the more common clear cell RCC studies or basket trials that are not specific to each histology. The unique management of each vhRCC subtype necessitates accurate pathologic diagnosis and dedicated research efforts. Herein, we discuss tailored recommendations for each vhRCC histology informed by ongoing research and clinical experience.

Cytosolic Dna Accumulation Promotes Breast Cancer Immunogenicity Via A Sting-Independent Pathway, Jing Zhang, Hui Dai, Lei Huo, Jared K Burks, Daniel J Mcgrail, Shiaw-Yih Lin

Faculty, Staff and Student Publications

BACKGROUND: Immune checkpoint blockade (ICB) has revolutionized cancer treatment. However, ICB alone has demonstrated only benefit in a small subset of patients with breast cancer. Recent studies have shown that agents targeting DNA damage response improve the efficacy of ICB and promote cytosolic DNA accumulation. However, recent clinical trials have shown that these agents are associated with hematological toxicities. More effective therapeutic strategies are urgently needed.

METHODS: Primary triple negative breast cancer tumors were stained for cytosolic single-stranded DNA (ssDNA) using multiplex immunohistochemical staining. To increase cytosolic ssDNA, we genetically silenced TREX1. The role of tumor cytosolic ssDNA in promoting …

Anchors: A Way For Ftsz Filaments To Stay Membrane Bound, Arindam Naha, Daniel P Haeusser, William Margolin

Faculty, Staff and Student Publications

Most bacteria use the tubulin homolog FtsZ to organize their cell division. FtsZ polymers initially assemble into mobile complexes that circle around a ring-like structure at the cell midpoint, followed by the recruitment of other proteins that will constrict the cytoplasmic membrane and synthesize septal peptidoglycan to divide the cell. Despite the need for FtsZ polymers to associate with the membrane, FtsZ lacks intrinsic membrane binding ability. Consequently, FtsZ polymers have evolved to interact with the membrane through adaptor proteins that both bind FtsZ and the membrane. Here, we discuss recent progress in understanding the functions of these FtsZ membrane …
