Context.—: Blasts in myelodysplastic syndromes (MDSs) typically have a primitive myeloid immunophenotype (CD34+CD117+CD13+CD33+HLA-DR+). On rare occasions, blasts were found to be CD34 negative or minimally expressed in a presumptive MDS.
Objective.—: To investigate the occurrence of these cases, and to examine any unique molecular genetic features, and clinical relevance.
Design.—: More than 2000 flow cytometry immunophenotyping tests for MDS performed during a 5-year period were retrospectively reviewed. Chronic myelomonocytic leukemia and overt acute myeloid leukemia (AML) (≥20% blasts) were excluded.
Results.—: Approximately 800 cases had abnormal myeloblasts consistent with myeloid neoplasms; 96% of cases showed a typical primitive phenotype, but …

One common approach for dose optimization is a two-stage design, which initially conducts dose escalation to identify the maximum tolerated dose, followed by a randomization stage where patients are assigned to two or more doses to further assess and compare their risk-benefit profiles to identify the optimal dose. A limitation of this approach is its requirement for a relatively large sample size. To address this challenge, we propose a seamless two-stage design, BARD (Backfill and Adaptive Randomization for Dose Optimization), which incorporates two key features to reduce sample size and shorten trial duration. The first feature is the integration of …

Background: Blinding mitigates bias in randomized trials and may be especially crucial for surrogate endpoints, such as progression-free survival (PFS). Here, we characterize utilization of and factors associated with blinding in Phase III oncology trials with PFS primary endpoints.
Methods: Two-arm, superiority-design trials investigating systemic therapy were identified in May 2024 from ClinicalTrials.gov with no date limitation. Trials were required to have a PFS primary endpoint. The study outcomes were the presence of double-blind designs and blinded independent central review (BICR) for the primary endpoint. Ninety-five percent credible intervals for binary outcomes were estimated from beta distributions, and multivariable logistic …

Immune checkpoint blockade (ICB) therapy is effective against many cancers, although resistance remains a major issue and new strategies are needed to improve clinical outcomes1-5. Here we studied ICB response in a cohort of patients with ovarian clear cell carcinoma-a cancer type that poses considerable clinical challenges and lacks effective therapies6-8. We observed significantly prolonged overall survival and progression-free survival in patients with tumours with PPP2R1A mutations. Importantly, our findings were validated in additional ICB-treated patient cohorts across multiple cancer types. Translational analyses from tumour biopsies demonstrated enhanced IFNγ signalling, and the presence of tertiary lymphoid structures at the baseline, …

Metaheuristics are commonly used in computer science and engineering to solve optimization problems, but their potential applications in clinical trial design have remained largely unexplored. This article provides a brief overview of metaheuristics and reviews their limited use in clinical trial settings. We focus on nature-inspired metaheuristics and apply one of its exemplary algorithms, the particle swarm optimization (PSO) algorithm, to find phase I/II designs that jointly consider toxicity and efficacy. As a specific application, we demonstrate the utility of PSO in designing optimal dose-finding studies to estimate the optimal biological dose (OBD) for a continuation-ratio model with four parameters …

Objectives: Kilovoltage x-ray therapy, which includes superficial x-rays and orthovoltage x-rays, is particularly suited for treatments near the skin surface. Radiation dose deposition at depth is typically characterized by percentage depth dose (PDD) curves. However, PDD data are difficult to obtain in kilovoltage x-ray therapy, hence clinics often rely on British Journal of Radiology Supplement 25 (BJR-25) archival data. This study measured PDD for different energies and field sizes on the Xstrahl 300 kilovoltage x-ray therapy unit for comparison with BJR-25 data.
Methods: We irradiated EBT3 film in water using different energy and field size combinations, then, compared the acquired …

Developmental stuttering is a highly heritable, common speech condition characterized by prolongations, blocks and repetitions of speech. Although stuttering is highly heritable and enriched within families, the genetic architecture is largely understudied. We reasoned that there are both shared and distinct genetic variants impacting stuttering risk within sex and ancestry groups. To test this idea, we performed eight primary genome-wide association analyses of self-reported stuttering that were stratified by sex and ancestry, as well as secondary meta-analyses of more than one million individuals (99,776 cases and 1,023,243 controls), identifying 57 unique loci. We validated the genetic risk of self-reported stuttering …
Bystanders Intervening During Acts Of Cyberbullying: Examining The Moderating Influence Of “Likes” And Pleas For Help, Joseph Kallman
Bystanders Intervening During Acts Of Cyberbullying: Examining The Moderating Influence Of “Likes” And Pleas For Help, Joseph Kallman
Student Theses
Abstract
Cyberbullying research has only recently begun exploring “cyberstanders,” also known as bystanders, and their role in cyberbullying, as well as testing the bystander effect (Barlinska et al., 2013; Machackova et al., 2013; Obermaier et al., 2016; Shultz et al., 2014). In the current study, we aim to deepen our understanding of the bystander effect by manipulating the number of likes, which will indicate the number of bystanders present and the plea for help on social media via Twitter among college students, by examining how participants respond to fictitious cyberbullying incidents. The results of this study did not support our …

The population of multilingual learners is struggling to adapt to mainstream classes because the educational system has failed to support them on the same level as native English speakers. The gap in achievement and lack of teacher preparation demonstrates a clear need to support teachers who educate them. This study, a nonequivalent (pretest and posttest) control-group design combined with a cross-sectional survey design, addressed the effectiveness of a professional development series focused on best practices for teaching secondary math to multilingual learners. The professional development intervention consisted of one 45-minute initial session followed by twelve 15-minute mini lessons throughout the …
Characterization Of Search Spaces And Effects On Machine Learning, Leo Ghelarducci
Characterization Of Search Spaces And Effects On Machine Learning, Leo Ghelarducci
Doctoral Dissertations and Master's Theses
The present status of the field of Machine Learning (ML) focuses on optimization of popular models. Rarely are the effects of the problem characteristics upon the solution algorithm studied. There exists no standard for knowing when to apply ML algorithms to a given problem or how to estimate the effectiveness of results. Focusing on the search space of problems, a rigorous study was conducted to generate an in-depth understanding of the impact of search space characteristics to the performance of a ML algorithm, specifically a Genetic Algorithm (GA). The effects of specific problem characteristics, represented via solution space characteristics, on …

Glenn Korff School of Music: Dissertations, Theses, Student Creative Work, and Performances
The Corfu Philharmonic Society band was founded in 1840 and is still active today. It is considered the one of the oldest continuously operating music education organizations in Greece. The people, repertoire, and instrumentation of this band influenced all other wind bands in Greece. There are important resources available on the topic, but dedicated research to the organization is in its infancy. Focusing on the changes in instrumentation through original manuscripts from the inaugural band (1840) through the end of the First World War reveals significant multinational influences that continue to be felt to this day. This is the first …
Non-Blackbox Robust Design Of Machine Learning In Networks, Venkat Sai Suman Lamba Karanam
Non-Blackbox Robust Design Of Machine Learning In Networks, Venkat Sai Suman Lamba Karanam
Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–
Networked systems have become increasingly complex, with newer communication technologies and standards being added every day. Machine Learning (ML) and Artificial Intelligence (AI) paradigms have been adopted in networks to not only solve many fundamental problems, but also to allow seamless integration of components comprising them. The saying “let’s not reinvent the wheel” in ML/AI adoption implies that model architecture design be left for pure ML/AI researchers, while network researchers focus on input preprocessing (e.g. formatting the packet data to be fed to a model), hyperparameter fine-tuning and a trial-and-error approach to find the “best” result. …

Carrying Light In The Shadows: Four Portraits Of Critical Literacy Teacher Agency In Unlikely Contexts, Cassandra Schroeder
Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–
This qualitative study utilizes portraiture methodology to explore how educators in politically conservative and predominantly White schools enact critical literacy teacher agency. Using a theoretical lens of critical literacy and second-wave Whiteness studies, this research examines the lived experiences and instructional moves of four veteran English/language arts teachers. Through cross-case analysis, the study traces how teacher agency is contextually responsive and dependent on their professional identity, awareness of sociopolitical issues, and traditional school curricular expectations. The findings highlight how educators assess risk, navigate community norms, and maintain critical literacy through dialogic pedagogy and strategic adaptation. The four teachers in the …

The Significance Of Snow In Hydrometeorological Extremes, Sinan Rasiya Koya
Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–
Snow is the largest natural reservoir on Earth. It accumulates water during the cold and dry months and releases it during warm and dry months. Societies, especially in the extratropical zone, heavily depend on snow as their freshwater resources, resulting in the massive economic value of snow. The recent changes in snowpacks and associated processes are concerning. We are susceptible to disruptions in snow processes, which can have disastrous implications. Yet the importance of snow is often not realized by a wider population. There are limited studies quantifying snow-related extremes, partly stemming from the limitations of existing methodologies.

Blood-Red And Flaming And True, Kate Gaskin
Blood-Red And Flaming And True, Kate Gaskin
Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–
Blood-Red and Flaming and True contains poems that explore the immense feelings of love and loss tied to the relationship between parent and child. They chronicle my experience raising a neurodivergent child, as well as undergoing infant loss and navigating a U.S. medical system that is renowned for putting women and people capable of giving birth—especially BIPOC—at greater risk than other countries with similar resources. Through poems that inhabit a variety of forms, I sift through grief and joy while attempting to tap into the sublime state that underlies extreme loss and love. Throughout, I investigate how lyric poetry is …
Rhetorics Of Encounter: Toward A Biocentric Theory Of Discourse, Matthew D. Whitaker
Rhetorics Of Encounter: Toward A Biocentric Theory Of Discourse, Matthew D. Whitaker
Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–
Encounters with more-than-human identities are transformative moments that stand to affect dispositional and bodily change in individuals. From close brushes with wild animals in National Parks to everyday interactions with local ecologies, encounters invite us to feel and think differently about the nature of our realities. In Rhetorics of Encounter: Toward a Biocentric Theory of Discourse, I draw from interdisciplinary perspectives to disrupt the idea of rhetoric as a representational medium, arguing that rhetoric occurs not just through textual exchanges of words and symbols, but through embodied moments of contact with other responsive beings. Invoking George Kennedy’s theory …

We introduced couple different novel approaches to incorporate latent variable information to multivariate mixture regression models with both Gaussian and count data. We also evaluated the performance of these models with existing best approaches with simulated data from various sampling structures and also evaluated one of the models performance with rice metabolite data that provided some novel insights as well as validating existing literature about performance and behavior of these metabolites. We validated the method using extensive simulations and a real-world application. In both quantitative covariate designs and complex treatment design simulations, our method consistently outperformed established tools like limma, …

In a commentary, SMU Professor of Organisational Behaviour & Human Resources (Education) Thomas Menkhoff stressed the need for educators to upskill so they can guide students in using generative artificial intelligence (GenAI) responsibly, rather than dismissing it. He argued that universities should move beyond prohibition and invest in AI literacy to safeguard academic integrity. Prof Menkhoff mentioned that combining the use of GenAI tools with effective prompting and Socratic questioning transforms students’ use of technology from passive consumption to active, reflective and critical engagement. To achieve this, he said that schools must set clear guidelines and design AI-compatible assessments that …

Scientists studying culture typically focus on a small number of theoretical constructs, such as individualism–collectivism, when seeking to explain cultural differences in psychological tendencies and behaviors. However, existing theories of culture could have missed out on important constructs that are useful for explaining cross-cultural differences. We used an abductive approach combining prediction and explanation to uncover important cultural values. In the prediction phase, based on 594 attitudes, values, and beliefs included in the World Values Survey, a neural network could classify respondents’ nationalities with 90% accuracy in out-of-sample data. In the explanation phase, a feature importance analysis identified the values …

Many firms are attempting to reduce greenhouse gas emissions across their value chain. However, this requires convincing suppliers to adopt relevant decarbonization practices, which is challenging when the suppliers perceive such practices as risky or detrimental for their economic well-being. We employ a field experiment to examine relational investments (i.e., investments intended to promote mutual benefit for the exchange partners) as a tool for overcoming this challenge. In a research collaboration with a global firm pursuing decarbonization of its agricultural supply chain in India, we investigated the effectiveness of complementing training their supplier farmers on climate-friendly agricultural practices with also …

ESI Working Papers
We study how three widely discussed cues—source of income (merit vs luck), own performance, and information about others’ work status and performance—shape redistribution. Prior to the dictator game, all 306 dictators and most of the 306 recipients complete the same real-effort matrix task in an online experiment that would yield either $4 or $1 for the dictators. In the Performance treatment the dictator’s payoff is based on their performance; in Luck, it is assigned by a 50-50 draw. We find that: (i) Earned entitlement is prevalent for high-performing dictators: they keep more for themselves; (ii) Earned entitlement is conditional …

Listening As An Adaptive Practice To Transform Philanthropy, Nitya Venkateswaran, Kate Szczerbacki, Jay Feldman
The Foundation Review
A growing number of grantmaking institutions (GMIs) recognize that listening to communities in which they invest—especially those to, and most affected by, the challenges being addressed—is crucial to achieve their equity goals. Despite investments and intent to listen, there is a gap in implementation.
Genuine listening for equity requires that foundation leaders view the shift as adaptive and not merely technical. A technical change entails adoption of specified methods whereas adaptive changes require a fundamental shift in values and beliefs. Solely implementing new “listening” techniques without shifting values and beliefs could leave gaps between intention and action. But current resources …
