Background: FHF (fibroblast growth factor homologous factor) variants associate with arrhythmias. Although FHFs are best characterized as regulators of voltage-gated sodium channel (VGSC) gating, recent studies suggest broader, non-VGSC-related functions, including regulation of Cx43 (connexin 43) gap junctions and hemichannels, mechanisms that have generally been understudied or disregarded.
Methods: We assessed cardiac conduction and cardiomyocyte action potentials in mice with constitutive cardiac-specific Fgf13 ablation (cFgf13KO) while targeting Cx43 gap junctions and hemichannels pharmacologically. We characterized FGF13 regulation of Cx43 abundance and subcellular distribution. With proximity labeling proteomics, we investigated novel candidate mechanisms underlying FGF13 regulation of Cx43. …

In the 2003 documentary Woman of Heart and Mind, Joni Mitchell states, “Chords are depictions of emotions. These chords that I was getting by twisting the knobs on the guitar until I could get these chords that I heard inside that suited me, they feel like my feelings. You know, I called them, not knowing, chords of inquiry.” In investigating Mitchell’s music, we address the integration of guitar, harmony, and expression of feeling exemplified by “chords of inquiry.” We take as a corpus her early songs from 1968 to 1972 in which she accompanies herself on guitar. Background: The classification of periodontal disease published in 2017 by the European Federation of Periodontology (EFP) and American Academy of Periodontology (AAP), provides a framework for diagnosis and treatment. The aim of this study was to evaluate the diagnostic skills and self-perceived confidence of dentists and dental students based in Asian countries in the use of this classification. Methods: A cross-sectional analytic study design was employed. An online questionnaire encompassing four periodontitis cases was used for data collection. This paper provides a comprehensive analytical examination of the Global Technological Maturity Index (GTMI), with a focus on assessing the determinants of national digital maturity and structurally modeling GTMI performance. Using rigorous data preparation methods including imputation, feature transformation, and exploratory statistical analysis the study applies multiple regression models (Decision Tree, Random Forest, Gradient Boosting, and MLP) to replicate the GTMI structure and identify its primary drivers. The UAE demonstrates a strong maturity profile, ranking in the Very High category globally. BACKGROUND: Mitochondrial dysfunction caused by abnormally high RyR2 (ryanodine receptor) activity is a common finding in cardiovascular diseases. Mechanisms linking RyR2 gain of function with mitochondrial remodeling remain elusive. We hypothesized that RyR2 hyperactivity in cardiac disease increases [Ca 2+ ] in the mitochondrial intermembrane space (IMS) and activates the Ca 2+ -sensitive protease calpain, driving remodeling of mitochondrial cristae architecture through cleavage of structural protein OPA1 (optic atrophy protein 1).

The cerebral cortex provides the main input to the striatum, constituting the first step in cortico-basal ganglia loops. Decades of careful anatomical tract-tracing research have established the exquisite topography of each cortical region's projection to the striatum in nonhuman primates. In parallel, neuroimaging research has demonstrated the relationship between cortico-striatal resting-state functional connectivity and specific cognitive, behavioral, psychiatric, and neurological states in humans. However, still unclear is the extent to which functional connectivity recapitulates the specific topographies of cortico-striatal anatomical connectivity. When rivers achieve a great enough flow rate, sediment along their riverbed becomes mobilized and transported downstream. This process is known as bedload transport. As bedload transport initiates, micro-collisions of various sediments and gravel generate underwater acoustic signals that can be detected by hydrophones. While there is extensive research on underwater acoustic theory and surrogate methods for monitoring bedload transport, there is a lack of heuristic work that analyzes collected bedload samples in conjunction with underwater acoustic recordings. White adipose tissue (WAT) performs vital metabolic and endocrine functions, but roles in female reproduction remain understudied and poorly understood. Here, we report that female mice experiencing progressive lipoatrophy after knockout of Ubc9 in adipocytes (Ubc9fKO) displayed disrupted estrous cycles, reduced ovarian reserve, and subfertility. During aging, female Ubc9fKO mice lose subcutaneous WAT more quickly than their male counterparts and weigh less than littermate controls. Subcutaneous WAT excised from female Ubc9fKO mice strongly enriched for thermogenesis genes generally associated with metabolic benefits. Female Ubc9fKO mice exhibited hypermetabolism and accumulated thermogenic, Uncoupling Protein 1-expressing beige fat cells in residual subcutaneous WAT …

Clinical trials of immunotherapy in metastatic castration-resistant prostate cancer (mCRPC) have largely been unsuccessful despite promising preclinical studies and proven efficacy in other solid tumors. These disappointing clinical outcomes have been attributed to an immunosuppressive tumor microenvironment, a relative lack of infiltrating immune effector cells, and tumor-related and host-related factors, which collectively render prostate cancer a relatively immunologically "cold" tumor. Sipuleucel-T (Provenge), an autologous cellular immunotherapy, induces an immune response targeted against prostatic acid phosphatase. It received approval from the US Food and Drug Administration in 2010, marking the first immunotherapy to show an overall survival benefit in patients with …

BACKGROUND: Chronic obstructive pulmonary disease (COPD) affects approximately 16 million Americans and often results in avoidable readmissions due, in part, to medication errors and lack of education. Telehealth interventions can support medication reconciliation and inhaler education following hospital discharge for patients with COPD.
OBJECTIVE: This study aimed to design and prototype TELE-TOC (Telehealth Education: Leveraging Electronic Transitions of Care), a post-discharge, in-home, televisit intervention, and to map its workflow to ensure integration into the routine discharge care transition process for patients with COPD.

Educational technology (EdTech) interoperability throughout cyberspace provides the financial opportunity to collect and sell student privacy information in digital learning environments, challenging school leaders to govern schools and keep children safe. School leaders provide the resources, funding, planning, decision making, and administration for EdTech cybersecurity practices and policies, yet little is known about what public school leaders understand. The following article addresses a conundrum: What if child assent in research were a fully informed, stand-alone process that empowered all children to discuss meaningfully, negotiate, and decide how and what the research experience would look like? Past research and practices suggest that child assent, when children (youths to 17-year-olds who have not been emancipated) agree to participate in research before, during, and after parents’ or guardians’ consent, is rife with problems that minimize informed participation and prize procedural ethics over true understanding of the processes and autonomous decision-making. Enrolled Deeds As Records And Archives In Jamaica, Andrew Williams
Journal of Contemporary Archival Studies
This paper examines the history of the enrolled deeds record series in Jamaica, from its creation in the mid-seventeenth century through to the present day. It is a critical examination of a single record series through colonialism and independence. It examines the purpose of deeds and the context in which deeds registries were created before examining the history of the series through the periods of slavery and emancipation. It examines in close detail the late nineteenth century, when the Island Record Office—which has custody of the enrolled deeds—was created and several other important changes happened. Master’s students in Archives and Records Management could help low-resourced organizations address their Archives and Records Management needs while gaining practical experience. Archives and Records Management education programs could facilitate this mutually beneficial relationship by offering internships and service-learning courses. This paper analyzes the strengths and limitations of these two experiential learning mechanisms and discusses solutions to overcome the limitations. It argues that service-learning is especially suitable for addressing the Archives and Records Management needs of low-resourced organizations and proposes a service-learning model for that purpose.

Student-Designed Archival Pedagogy: A Workshop-As-Research Approach To Pluralizing Community Archives Education, Magdalena Wiśniewska-Drewniak

Community archives are becoming an increasingly important component of the heritage landscape, as well as an observable part of university archival education. This article is part of a research project titled “Teaching about Community Archives in the US and Poland--A Comparative Study” conducted by the author between March and May 2024. Using the student-centered learning approach, the article analyzes a workshop conducted with US-based students on the design of university courses on community archives. The designs included course audience, learning outcomes, course content, grading methods, and possible challenges.
