SignificanceEosinophils contribute to type 2 immunity against helminths and allergens. The small intestine harbors eosinophils with incompletely understood pathophysiological roles. Here, we show that intestinal eosinophils include two subsets. One expresses the inhibitory receptor Clec4a4 and the inhibitory ligand PD-L1 and is unique to the small intestine; the other manifests a proinflammatory phenotype. Both subsets are blood derived. BACKGROUND: Epidermal growth factor receptor (EGFR) activation is associated with increased production of interleukin 6 (IL6), which is intensified by radiotherapy (RT) induced inflammatory response. Elevated IL6 levels intensifies RT-induced anemia by upregulating hepcidin causing functional iron deficiency. Cetuximab, an EGFR inhibitor, has been associated with lower rates of anemia for locally advanced head and neck squamous cell carcinoma (HNSCC). We hypothesized that concomitant cetuximab could prevent RT-induced anemia.
METHODS: We queried our institutional head and neck cancers database for non-metastatic HNSCC cases that received RT with concomitant cetuximab or RT-only between 2006 and 2018. Cetuximab was administered for some …

The lack of seasonally sustained floral resources (i.e. pollen and nectar) is considered a primary global threat to pollinator health. However, the ability to predict the abundance of flowering resources for pollinators based upon climate, weather, and land cover is difficult due to insufficient monitoring over adequate spatial and temporal scales. Here we use spatiotemporally distributed honey bee hive scales that continuously measure hive weights as a standardized method to assess nectar intake. Background: Epidemiologic observations suggest increased potato consumption correlates with weight gain, adiposity, and diabetes risk, whereas nut consumption is associated with weight control and metabolic health. Randomized controlled trial (RCT) data indicate humans respond to changes in energy intake in single dietary components and compensate for extra energy consumed.
Objectives: We completed an RCT testing whether increased daily potato consumption influences energy balance [specifically, fat mass (FM)] compared with calorie-matched almond consumption.

Fungal infections are a major health problem that often begin in the gastrointestinal tract. Gut microbe interactions in early childhood are critical for proper immune responses, yet there is little known about the development of the fungal population from infancy into childhood. Here, as part of the TEDDY (The Environmental Determinants of Diabetes in the Young) study, we examine stool samples of 888 children from 3 to 48 months and find considerable differences between fungi and bacteria. The metagenomic relative abundance of fungi was extremely low but increased while weaning from milk and formula. Overall fungal diversity remained constant over …

C-type lectin domain family 4, member a4 (Clec4a4) is a C-type lectin inhibitory receptor specific for glycans thought to be exclusively expressed on murine CD8α− conventional dendritic cells. Using newly generated Clec4a4-mCherry knock-in mice, we identify a subset of Clec4a4-expressing eosinophils uniquely localized in the small intestine lamina propria. Clec4a4+ eosinophils evinced an immunomodulatory signature, whereas Clec4a4− eosinophils manifested a proinflammatory profile. Clec4a4+ eosinophils expressed high levels of aryl hydrocarbon receptor (Ahr), which drove the expression of Clec4a4 as well as other immunomodulatory features, such as PD-L1. Vitamin D supplementation has numerous effects on athletic performance and plays a significant role in preventing an athlete’s risk of getting injuries. Vitamin D has an impact on numerous physiological functions such as: bone health, muscle function, inflammatory response, and immune function. An athlete’s bone and muscle health are essential for maximum performance and career success. A bone fracture due to vitamin D deficiency can delay an athlete’s training and ultimately inhibit obtaining a collegiate scholarship and/or contract. The prevalence of gestational diabetes mellitus (GDM) has increased with the increasing rate of obesity. However, national data on the prevalence and secular trends of GDM during the past decade in the United States are lacking. This study included 37,357 women aged more than 18 years and who had ever been pregnant from the National Health Interview Survey (NHIS). We examined GDM prevalence in 2006, 2016, and 2017, with age-standardized to the US population in 2000. We found that the prevalence of GDM per 100 people increased from 4.6 (95% CI, 4.1–5.1) in 2006 to 8.2 (95% CI, 7.5–8.9) in …

Undergraduate curriculum is not representative of all students. Course content, language, images, and textbooks often reinforce societal power relations and hierarchies that tend to center white, male, hetero, middle-class, able-bodied identities. When students' varied cultural and linguistic identities are not represented in the curriculum, inadequately represented students are less likely to actively participate, persist, and continue their education. Emerging scholarship indicates that student-instructor cocreation of course syllabi, materials, and/or classroom experiences is a promising practice for increasing representation and responsiveness to student voices, although researchers do not know how the process of cocreation unfolds in asynchronous spaces. This thesis explores the life of one woman, Patricia Carpio Whiting, during the second half of the twentieth century, in an effort to expand the genre of women and environmental studies. It provides context for Carpio Whiting's accomplishments as an elected official in Oregon by describing her childhood in Chicago and her formative years in California, particularly how growing up Filipino American shaped her as an adult. As such, this thesis engages themes of gender, race, and class in historical scholarship. Chenopodium quinoa Willd., an ancient Andean pseudocereal almost exclusively consumed in South America, jumped onto the global stage when Western cultures noted quinoa's advantageous nutritional profile. Quinoa seed's high protein content, nutritionally balanced amino acid profile, low glycemic index, and high fiber, vitamin, and mineral content, make it a highly sought-after 'superfood'. Pitseed goosefoot (C. berlandieri Moq.), a closely related North American species sharing quinoa's genome composition (AABB), grows across the North American continent, inhabiting diverse environments including the saline coastal soils of the Gulf of Texas and the drought-prone regions of the Southwest. Identifying Colorectal Cancer Caused By Biallelic Mutyh Pathogenic Variants Using Tumor Mutational Signatures, Peter Georgeson, Yin Cao, Et Al

Carriers of germline biallelic pathogenic variants in the MUTYH gene have a high risk of colorectal cancer. We test 5649 colorectal cancers to evaluate the discriminatory potential of a tumor mutational signature specific to MUTYH for identifying biallelic carriers and classifying variants of uncertain clinical significance (VUS). Using a tumor and matched germline targeted multi-gene panel approach, our classifier identifies all biallelic MUTYH carriers and all known non-carriers in an independent test set of 3019 colorectal cancers (accuracy = 100% (95% confidence interval 99.87-100%)). Federal Work-Study (FWS) is a form of student employment in postsecondary education funded by a student’s financial aid. At the University of South Florida (Tampa campus), in the Community-Based Federal Work-Study (CBFWS) program managed by the Office of Community Engagement and Partnerships (OCEP), there has been a decreasing rate of participation among its male students compared to female students. Introduction Obese men with prostate cancer have an increased risk of biochemical recurrence, metastatic disease and mortality. For those undergoing androgen deprivation therapy (ADT), substantial increases in fat mass are observed in the first year of treatment. Recently, we showed that a targeted supervised clinic-based exercise and nutrition intervention can result in a substantial reduction in fat mass with muscle mass preserved in ADT-treated patients. However, the intervention needs to be accessible to all patients and not just those who can access a supervised clinic-based programme. Flies are implicated in carrying and mechanically transmitting many primate pathogens. We investigated how fly associations vary across six monkey species (Cercopithecus ascanius, Cercopithecus mitis, Colobus guereza, Lophocebus albigena, Papio anubis, and Piliocolobus tephrosceles) and whether monkey group size impacts fly densities. Fly densities were generally higher inside groups than outside them, and considering data from these primate species together revealed that larger groups harbored more flies. Within species, this pattern was strongest for colobine monkeys, and we speculate this might be due to their smaller home ranges, suggesting that movement patterns may influence fly–primate associations. Changing in sentencing laws and policies have created a cycle of imprisonment, racially isolating and oppressing BIPOC communities. Reintegration and reentry programs are an avenue to break the cycle of recidivism. Through my work, I have developed hands-on, structured opportunities for justice-involved adults to rebuild a life for themselves by increasing the accessibility of gardening reentry programs. Commercial honey bees are transported between pollinated crops and play an essential role in our agricultural system. This thesis proposes a network of bee stations and corridors that could improve the health of transported bee populations, increase local beekeeping, and help educate the public about the importance of bees and pollinators. The bee stations have at least 100 acres of diversified high-quality wildflower species where trucks that transport bees can stop to “refuel” their hives. This project investigates the physical and narrative barriers constructed by the phosphate mining and processing industry to dissuade public engagement with its phosphogypsum sites in Central Florida, USA. The barriers around PG utilize decades of misinformation, racial geographies, and misrepresentations of scale, origin, and risk to create powerful narratives about how we should imagine and relate to these materials. With a background in Human Factors Engineering, user engagement remains a central focus in my design. Whether reducing barriers to scientific information or untangling deeply complex data, design must contain intuitive concepts without the need for extended explanation.
