Background. – BK polyomavirus (BKPyV) is a common opportunistic infection in kidney transplant recipients, typically reactivating in the context of immunosuppression. Although asymptomatic in immunocompetent individuals, reactivation in transplant recipients can cause BKPyV-associated nephropathy (BKPyVAN), a leading cause of graft dysfunction and loss. BKPyV viremia affects approximately 10%–15% of transplant recipients, and once BKPyVAN is established, the risk of graft failure can exceed 50%. The current standard of care involves immunosuppression reduction to restore virus-specific immunity, but this approach increases the risk of acute and chronic rejection. Intravenous immunoglobulin (IVIg), which contains antiviral and immunomodulatory antibodies, has been proposed as …

Improved Process For The Preparation Of Calcium Gluconate By Phase Transfer Catalysis, Anonymous
Defensive Publications Series
Calcium gluconate is an essential pharmaceutical and nutritional compound, widely employed as a calcium supplement and therapeutic agent. Conventional aqueous synthesis methods, while effective, often suffer from kinetic inefficiencies and purification challenges. Phase transfer catalysis (PTC) offers a promising alternative by enabling ionic substrates to cross phase boundaries, thereby enhancing reaction rates and selectivity. Building on CN110256234B, which demonstrates the use of selected quaternary ammonium salts, this work discloses an expanded spectrum of PTCs applicable to calcium gluconate synthesis. The improved process achieves higher efficiency, reproducibility, and product purity.

Background: Vamorolone is a dissociative glucocorticoid receptor agonist for treating Duchenne muscular dystrophy (DMD). The VBP15-006 study assessed vamorolone safety, tolerability, and pharmacokinetics in 2- to < 4 and 7- to < 18-year-old boys with DMD. Results for 7- to < 18-year-old boys, either corticosteroid (CS)-untreated or switching from prior CS treatment, are reported here. Exploratory objectives included efficacy and pharmacodynamic biomarkers related to safety. Methods: In this phase II, open-label, multiple-dose study, participants received 2 or 6 mg/kg/day vamorolone for 12 weeks, followed by treatment in an expanded access protocol (EAP Canada). Results: 34 participants (12 CS-untreated, 22 CS-treated) aged 7- to < 18 years were enrolled. Most treatment-emergent adverse events were mild. All 34 participants completed VBP15-006 and entered EAP Canada. During EAP Canada, CS-untreated participants maintained stable linear growth, while CS-treated participants exhibited catch-up growth consistent with serum bone biomarkers (median total vamorolone exposure 1.3 and 1.7 years, respectively). Some weight gain occurred, especially in CS-untreated participants receiving 6 mg/kg/day. Dose-dependent adrenal suppression occurred in CS-untreated and CS-treated participants; 2 individuals who switched from deflazacort to vamorolone 2 mg/kg/day had generalized weakness consistent with adrenal insufficiency. Vamorolone showed dose-dependent pharmacokinetics, rapid clearance, and no accumulation. There were no relevant efficacy changes in CS-treated and CS-untreated groups at either dose. Conclusions: Vamorolone demonstrated a consistent safety profile in 7- to < 18-year-old boys with DMD. Switching to 6 mg/kg/day vamorolone appeared to mitigate adrenal insufficiency risk. There was no negative effect on growth, and catch-up growth occurred in previously CS-treated individuals switching to vamorolone. Trial registration: ClinicalTrials.gov: NCT05185622, NCT03863119. First submitted 09-11-2021.

Purpose: Visceral white adipose tissue (vWAT) accumulation causes systemic inflammation, insulin resistance, metabolic syndrome, and senescent cell accumulation that are risk factors for Alzheimer’s disease (AD). Visceral fat removal (VFR) improves metabolism and reduces pro-inflammatory cytokines. We hypothesized that VFR removal in AD mice would improve metabolism and cognition.
Methods: Male and female APPNL-F mice underwent sham or vWAT surgical resection (periovarian or epididymal and perirenal) at 4 (pre-symptomatic) and 16 (symptomatic) months of age to understand interventional and therapeutic effects, respectively. At 18 months of age, glucose metabolism and novel object recognition (NOR) memory were assayed followed by …
Force–Power Profiles And Countermovement Utilization Ratios In U.S. Ski & Snowboard National Team Athletes, Joshua Bullock
Force–Power Profiles And Countermovement Utilization Ratios In U.S. Ski & Snowboard National Team Athletes, Joshua Bullock
Doctorate in Kinesiology
Mogul skiing imposes high neuromuscular and lower-extremity loading athlete demands, yet mogul-specific reference values for jump-derived force–power characteristics and differential utilization metrics have remained limited. The specific aims of this study were to: 1) establish descriptive statistics, including means, percentiles, and variability for jump height (JH), peak force (PF), and peak power (PP) during the squat jump (SJ) and countermovement jump (CMJ) in elite mogul skiers, 2) determine corresponding countermovement utilization ratio (CUR) values for each measure (JH-CUR, PF-CUR, and PP-CUR), and 3) report descriptive outcomes independently for both male and female athletes to provide sex-specific reference profiles. Archived testing …

Jackfruit Peel Valorization Through An Integrated Biorefinery Approach, Rohil Kumar K Mr
Jackfruit Peel Valorization Through An Integrated Biorefinery Approach, Rohil Kumar K Mr
Theses and Dissertations
Jackfruit (Artocarpus heterophyllus) peel (JFP), an abundantly available lignocellulosic biomass, remains largely underutilized despite being rich in cellulose, hemicellulose, lignin, and pectin. In India, approximately 13,460 hectares are under jackfruit cultivation, with Panruti in Tamil Nadu as a major production zone. The discarded peel often contributes to environmental pollution and greenhouse gas emissions. The present study emphasizes sustainable valorization of JFP through an integrated biorefinery framework for generating value-added bioproducts.
Major biopolymers, cellulose, hemicellulose, lignin, and pectin were extracted and fractionated, followed by detailed physicochemical characterization using FTIR, NMR, XRD, SEM, TGA, DSC, GPC, and BET analyses. Pectin extraction with …

Cultivation Of Arthrospira Platensis (Spirulina) In Food-Based Industrial Wastewater For High-Yield Phycocyanin Extraction, Pavithira Sathinathan
Student Works (2020-2029)
Arthrospira platensis (Spirulina) is a filamentous cyanobacterium valued for its nutritional profile, particularly the pigment–protein phycocyanin, with potent antioxidant and anti-inflammatory properties. Despite its industrial significance, large-scale commercialization remains constrained by high cultivation costs and inefficient extraction methods. This study investigated the use of food-based industrial wastewater as an alternative nutrient medium for Spirulina cultivation, aiming to reduce production costs, enhance phycocyanin yield, and support sustainable wastewater remediation. Three types of food-based wastewater with different nutrient composition (WW1, WW2, WW3) was diluted with Kosaric medium (1:1) and used for Spirulina cultivation. The highest biomass concentration was achieved in WW2 (1.93 …

Handgrip strength is a proxy for muscular fitness, an indicator for general health status, and is associated with cardiometabolic health. The mechanisms connecting handgrip strength to skeletal muscle function are incompletely understood. We applied integrated linkage-disequilibrium-adjusted colocalization analysis of genome-wide association study summary statistics for handgrip strength, combined with expression and splicing quantitative trait loci from skeletal muscle, and identified glycogen branching enzyme 1 (GBE1) as a candidate gene for handgrip strength. CRISPR-interference knockdown of GBE1 in immortalized human skeletal muscle cells (HMCL-7304) demonstrated decreased glycogen content and accumulation of polyglucosan bodies. Knockdown of GBE1 led to increased oxygen consumption …
