Lung cancer remains the leading cause of cancer-related death globally, with metastasis driven by circulating tumor cells (CTCs)-particularly clusters-being a major treatment challenge. Despite their critical role, the biological differences between single CTCs and CTC clusters remain unclear. Here, we comprehensively compared their behavioral, transcriptomic, and proteomic profiles in lung cancer models. Compared with single cells, CTC clusters present enhanced metastatic potential, greater survival in the bloodstream and increased resistance to microenvironment. Mechanistically, the Src/FN1 pathway is centrally activated in clusters, promoting intercellular cohesion and protecting against immune clearance and stress in circulation. Pharmacological inhibition of Src with the clinical …

Soil-transmitted helminths (STHs) are intestinal parasites that affect over a billion people worldwide. STH control relies on microscopy-based diagnostics to monitor parasite prevalence and enable post-treatment surveillance; however, molecular diagnostics are rapidly being developed due to increased sensitivity, particularly in low-STH-prevalence settings. The genetic diversity of helminths and its potential impact on molecular diagnostics remain unclear. Using low-coverage genome sequencing, we assess the genetics of STHs within worm, faecal, and purified egg samples from 27 countries, identifying differences in the genetic connectivity and diversity of STH-positive samples across regions and cryptic diversity between closely related human- and pig-infective species. We …

Social Forestry As Solution To Agrarian Conflicts? A Case Study Between Oil Palm Smallholders And Industrial Plantations In Jambi, Indonesia, Muhammad Reza Halomoan, Arya Hadi Dharmawan, Satyawan Sunito
Forest and Society
Extraction activities across Indonesia have caused drastic changes to forest ecologies. The Indonesian government responded to this through a policy of Industrial Plantation Forest concessions, which were given to the private sector in the hopes of overcoming damages from forest extraction activities while at the same time utilizing unproductive forest land. However, this policy does not pay attention to the existence of people who de facto control and use the land and has triggered new conflicts around agrarian claims based on different interpretations of land rights. In an effort to resolve conflicts, the government has implemented a community-based forest area …

The Impact Of The Covid-19 Pandemic And Race On Access To Maternal Healthcare Services For Patients Based On Insurance Status, Trishana Monlyn
Walden Dissertations and Doctoral Studies
Due to insurance status, COVID-19 has restricted how and when patients can access healthcare providers. In the health services community, examining obstetric care services during the coronavirus outbreak was important because it increased awareness about how changes to maternal healthcare access affect the health of the mother and unborn child during a pandemic. The theories and/or concepts that grounded this study include the intersectionality theory, which examined the connections between social groups and systems as individuals try to access health services based on health insurance reform, depending on their race. The purpose of this study was to determine if there …

Alfalfa (Medicago sativa L.) is a globally vital forage crop valued for its perennial growth and multiple annual harvests. A breeding effort is underway to improve the crop for productivity and persistence against biotic and abiotic stresses using “creeping rootedness,” a trait where plants exhibit horizontal root growth, similar to rhizomes, with increased vegetative ground surface area. In this study, we genotyped a breeding population of 648 alfalfa lines segregating for creeping rootedness using the 3K DArTag marker panel to identify trait-associated genomic loci and evaluate the feasibility of genomic prediction to accelerate breeding cycles. Using genome-wide association studies …

Weaning Beef Calves, David Secrist
All Current Publications
Weaning a beef calf is likely the most stressful event in the animal’s life. Future growth and reproductive performance can be compromised by health challenges encountered through the weaning process. Consequently, proper management aimed at reducing stress and preparing the beef animal for the next stage of production is critical to reach its genetic potential. Proper management involves applying good animal husbandry techniques, using sound science in developing health and feeding protocols, and implementing low-stress animal handling practices.

Both neuronal and peripheral tissues become disrupted in Alzheimer's disease (AD). However, a comprehensive understanding of how AD impacts different tissues across the whole organism is lacking. Using Drosophila, we generated an AD Fly Cell Atlas (AD-FCA) based on whole-organism single-nucleus transcriptomes of 219 cell types from flies expressing AD-associated proteins, either human amyloid-β 42 peptide (Aβ42) or Tau, in neurons. We found that Aβ42 primarily affects the nervous system, including sensory neurons, while Tau induces accelerated aging in peripheral tissues. We identified a neuronal cluster enriched in Aβ42 flies, which has high lactate dehydrogenase (LDH) expression. This LDH-high cluster …

A Single High-Zinc Activation Enhancer Can Control Two Genes Oriented Head-To-Head In Caenorhabditis Elegans, Hanwenheng Liu, Brian Earley, Adelita Mendoza, Patrick Hunt, Sean Teng, Daniel Luke Schneider, Kerry Kornfeld
2020-Current year OA Pubs
Enhancers play critical roles in gene expression, but a full understanding of their complex functions has yet to be defined. The cellular response to excess zinc levels in Caenorhabditis elegans requires the HIZR-1 transcription factor, which binds the high-zinc activation (HZA) enhancer in the promoters of multiple target genes. Cadmium hijacks the excess zinc response by binding and activating HIZR-1. By analyzing the genome-wide transcriptional response to excess zinc and cadmium, we identified two positions in the genome where head-to-head oriented genes are both induced by metals. In both examples, a single predicted HZA enhancer is positioned between the two …

Psilocybin, the naturally occurring psychedelic compound produced by hallucinogenic mushrooms, has received attention due to considerable clinical evidence for its therapeutic potential to treat various psychiatric and neurodegenerative indications. However, the underlying molecular mechanisms remain enigmatic, and few studies have explored its systemic impacts. We provide the first experimental evidence that psilocin (the active metabolite of psilocybin) treatment extends cellular lifespan and psilocybin treatment promotes increased longevity in aged mice, suggesting that psilocybin may be a potent geroprotective agent.

The primary treatment for obesity involves calorie restriction (CR) to promote dietary weight loss achieved through interventions including behavioral modification, bariatric surgery, and antiobesity medications. In adults with obesity, CR-induced weight loss enhances physical function and improves quality of life, while also reducing the burden of various obesity-related chronic conditions, including hypertension, diabetes, obstructive sleep apnea, and atherosclerotic heart disease. However, it is also associated with a decline in lean mass and bone mineral density, which increases the risk of sarcopenia and osteoporosis. When performed alongside CR, progressive resistance training (RT) attenuates this loss of lean mass and bone mass, …

Background: High-intensity functional training (HIFT) varies daily workout programming, and trainees are often tasked with completing “as many repetitions as possible” (AMRAP) within a set time. Successful performance depends on fatigue management skills and maintaining a consistent expression of force and velocity when performing and transitioning between exercises over the duration of the workout. Multi-ingredient preworkout (MIPS) supplement formulations often contain ingredients shown to facilitate energy availability and mitigate fatigue, and therefore, might positively affect HIFT performance. This study aimed to assess a multi-ingredient pre-workout supplement’s acute effect on pacing and the expression of kinetics throughout a 5- and 15-minute …

The term food noise has been recently popularized by individuals managing their weight or adhering to specific diets for health reasons. Anecdotal evidence from patients and clinical observations suggests that food noise involves constant preoccupation with food-related decisions-such as which foods to eat, caloric intake, macronutrient balance, and meal timing-which can become intrusive and unpleasant. Food noise also appears to affect cognitive burden and quality of life, and is being cited as one reason weight-loss attempts fail. The increase in public discourse about food noise has highlighted its potential significance, yet a formal clinical definition and method of measurement of …

The Effect Of Temperature For Producing Thayer-Martin Agar Media Using Sheep Blood On Neisseria Gonorrhoeae Growth With Various Inoculums, Ika Ningsih, Kayla Adyapratista Sumadiyo
Indonesian Journal of Medical Chemistry and Bioinformatics
Neisseria gonorrhoeae is a Gram-negative bacterium in the form of diplococci that infects mucous membranes through direct inoculation, is fastidious, and requires special media for growth because it is susceptible to heating and dryness, so it cannot survive long outside its host. Thayer-Martin agar media is a selective culture medium for the diagnosis of Neisseria gonorrhoeae bacterial infections enriched with sheep blood. The purpose of this study was to determine the optimum temperature for various temperature variations of making Thayer-Martin agar when mixing with sheep blood, compared to the temperature according to the manufacturer's procedure. Determining this ideal temperature may …

The current study aimed to identify how manipulated sleep restriction affects dietary choices and physical activity (PA). Young adults were administered one week of well-rested sleep levels (WR: 8–9 h/night) and sleep-restriction (SR: 5–6 h/night) in their (naturalistic) at-home setting, using a randomized cross-over design. Participants made consequential bids for snacks/beverages following SR and WR in an auction task. Other primary outcome measures included daily self-reported dietary intake and actigraphy-measured PA. Multivariate regression analyses examined main and moderator impacts of SR on primary outcome measures. In total, 118 treatment participants completed the study (M = 20 years old, n = …
