The aminoacyl-tRNA synthetases (AaRSs) are an ancient family of structurally diverse enzymes that are divided into two major classes. The functionalities of most AaRSs are inextricably linked to their oligomeric states. While GluRSs were previously classified as monomers, the current investigation reveals that the form expressed in Pseudomonas aeruginosa is a rotationally pseudosymmetrical homodimer featuring intersubunit tRNA binding sites. Both subunits display a highly bent, “pipe strap” conformation, with the anticodon binding domain directed toward the active site. The tRNA binding sites are similar in shape to those of the monomeric GluRSs, but are formed through an approximately 180-degree rotation …

Teachers' Multiculturalism And Digital Learning Choices For Students With Limited Or Interrupted Formal Education, Jitka Nelsonova Brychtova
Teachers' Multiculturalism And Digital Learning Choices For Students With Limited Or Interrupted Formal Education, Jitka Nelsonova Brychtova
Walden Dissertations and Doctoral Studies
The English language learner environment in U.S. secondary schools is increasingly complex due to the growing numbers of students with limited or interrupted formal education (SLIFE) who require a culturally responsive approach. Teachers are not prepared to meet the needs of SLIFE, especially in the technology-enhanced classrooms. A gap in the literature exists regarding the culturally responsive use of digital learning resources (DLRs) with high school SLIFE. The purpose of this quantitative study was to examine whether the teachers’ self-reported multicultural teaching competencies (skills and knowledge) predict the choice of DLRs used with SLIFE. The study was grounded in Vygotsky’s …

Mass Shooting Perceptions, Public Mental Illness Stigma, And Social Media Use: A Regression Analysis, Kayla Cross
Walden Dissertations and Doctoral Studies
Mental illness is frequently associated with mass shootings in news media, which can result in negative stigma among individuals diagnosed with mental illness. While mental illness and mass shootings have been explored previously, social media perceptions have not yet been examined in relation to mental illness stigma and mass shootings. The purpose of this study was to examine public mental illness stigma, mass shootings, and perceptions through social media in a correlational study subjected to regression analysis. The theoretical framework for the study was based on Tajfel and Turner et al.’s theory of social identity and intergroup behaviors. The analysis …

Understanding mechanisms of resistance to active therapies is crucial for developing more effective treatments. Here, we investigate resistance to anti-EGFR and anti-VEGF plus chemotherapy treatment in colorectal cancer (CRC) patients from the IMblaze370 trial (NCT02788279). While anti-VEGF does not select for secondary mutations, anti-EGFR leads to simultaneous mutations in EGFR and MAPK, but not PI3K pathway genes. Notably, we observe frequent acquired mutations in the EGFR extracellular but not intracellular domain and that patients with higher baseline expression of EGFR-ligands are prone to acquire resistant mutations. This data reveals a ligand-activated EGFR/MAPK-signaling dependency in CRC. We also observe enrichment for …

Osteoarthritis As A Multi-Factorial Disease: Exploring Piezo Ion Channels And Adipose Tissue As Therapeutic Targets, Erica Valentine Ely
McKelvey School of Engineering Graduate Student Theses & Dissertations
Osteoarthritis (OA) is a complex disease involving mechanical and inflammatory factors that affect both the joint and systemic environments. Despite its prevalence as a leading cause of pain and disability worldwide, no disease-modifying OA drugs (DMOADs) are currently available. This dissertation focuses on exploring key targets and developing innovative tools to combat OA, addressing its mechanical and inflammatory drivers both independently and in combination. First, we investigated the role of mechanosensitive ion channels Piezo1 and Piezo2 in OA pathogenesis using a murine destabilization of the medial meniscus (DMM) model. Our findings demonstrated that knocking out Piezo1, Piezo2, or both in …
The Crosstalk Between Muscle And Adipose Tissue In Health And Disease, Chang Gui
The Crosstalk Between Muscle And Adipose Tissue In Health And Disease, Chang Gui
McKelvey School of Engineering Graduate Student Theses & Dissertations
The aim of this dissertation is to investigate the crosstalk between adipose and muscle tissues in physiology and pathophysiology, particularly focusing on the therapeutic potential of muscle-associated adipose tissue in muscle regeneration. Adipose tissue, an endocrine organ, secretes cytokines (a.k.a. adipokines) that significantly and negatively impact muscle function in conditions like diabetes, obesity, and aging. Whether similar signals mediate communication between muscle and its adjacent adipose tissue through paracrine signaling is unknown, as is whether this communication is therapeutically modifiable. Understanding the interactions between adipose and muscle tissues could lead to novel therapeutic strategies for improving muscle regeneration and function. …

Investigating Neural, Neurovascular, And Vascular Function: Insights From Imaging 5-Ht2ar Compounds, Optogenetic Motor Mapping, And Cross-Modal Functional Mapping, Jonah Aryeh Padawer-Curry
McKelvey School of Engineering Graduate Student Theses & Dissertations
Understanding how brain activity relates to behavior and cognition remains a central challenge in neuroscience. Brain function emerges from interactions between electrical, chemical, and blood dynamics across multiple spatial and temporal scales. Capturing this complexity requires methods that assess both large-scale neurophysiological activity and precise, cell-type-specific processes. Widefield optical imaging (WFOI) enables high-resolution monitoring of cortical function by simultaneously measuring hemodynamic and calcium dynamics. WFOI is particularly valuable for studying psychedelics, which show therapeutic potential but may alter neurovascular coupling, complicating the interpretation of hemodynamic signals. Using WFOI, we examined the hallucinogenic 5-HT2A receptor agonist DOI in awake mice, revealing …

Lewy Body Disease (LBD) and Multiple System Atrophy (MSA) are synucleinopathies with distinct prognoses and neuropathologies, however, with overlapping clinical symptoms. Different disease characteristics are proposed to be determined by distinct conformations of alpha-synuclein (α-Syn) aggregates, which can self-propagate and spread between cells via a prion-like mechanism. The goal of this study is to investigate whether α-syn aggregates amplified from brain and CSF samples of LBD and MSA patients using the Seed Amplification Assay (SAA) maintain α-Syn seeding properties similar to those of α-syn aggregates derived from patients' brains. To address this, SAA-amplified and un-amplified α-Syn aggregates from LBD and …

This chapter aims to help readers prepare for going to 'the field'- the location(s) where data will be collected. We discuss starting a new field site, collaborating at established sites, and practical strategies for building and maintaining ties to the field site longitudinally. Throughout, we emphasize that developing trusting relationships with the community is critical to ethical research practice and essential for good science. Starting from this principle, we review the practical strategies for relationship-building and establishing ethical research practices, especially in regard to work with children and in settings with little to no infrastructure for research oversight. Also, personal …

Study On Metal-Drug Coordination And Development Of Hydrogel Drug Delivery Systems, Tamanna Ferdous
Electronic Theses and Dissertations Archive
This dissertation outlines a novel triple-networked hydrogel for a hydrophobic drug delivery system. The drug Simvastatin is used as a cardiovascular medication, but it has also been reported to function as an osteoinductive agent. Therefore, to deliver the drug locally to the affected area, a mechanically stable and tunable hydrogel has been designed using modified chitosan, PEGDMA, and PLA-PEG micelles. This hydrogel exhibited a steady release of simvastatin over a long period of 14 weeks. Additionally, the hydrogel is biocompatible and biodegradable, as demonstrated by the cytotoxicity test. This dissertation presents a comprehensive exploration of a novel triple-networked hydrogel specifically …
Understanding The Behavioral Health Of Children In Child Protective Services: Prevalence, Community Context, And System Involvement Using Administrative Data, Tzu-Hsin Huang
Brown School Graduate Student Theses and Dissertations
This dissertation investigates the behavioral health of children involved in the U.S. child welfare system through a three-paper approach. Amid rising concerns over youth mental health, suicide rates, and ADHD diagnoses, this study explores how these issues manifest among children reported for maltreatment – a population at elevated risk and often underserved. Paper 1 estimates the prevalence of behavioral health problems using national administrative datasets (NCANDS and AFCARS), providing insight into trends over time and disparities by age, race/ethnicity, sex, geographic context (urban vs. rural), and system involvement (CPS investigation vs. foster care). Paper 2 leverages county-level data to examine …

A Novel Method For Information Hiding In Images, Ali Al-Nuaimi, Shaymaa Al-Juboori
School of Engineering, Computing and Mathematics
Information security is a fundamental principle during data transmission. The main threat to information security is hacking. They are focusing on finding new techniques to breach encryption methods. Therefore, developing new approaches for hiding information against hacker attacks is a potential solution. Different steganography methods are available for hiding information, such as in texts, disk space, network packets, audio and images. In contrast to cryptography, which is concerned with data encryption. Steganography is the art of hiding transmitted information by using seemingly harmless carriers to hide the presence of information. Steganography is cost-effective and unrelated to hardware components. This study …

Theses
Peach Street connects to the universal phenomenon of losing innocence of childhood and navigating the perils and disappointments of the adult world and visualizes my ongoing experience with anxiety and what psychiatrists call derealization, the feeling of being detached from reality. Set in the same location as where I first felt immense panic, my photos create a fictional narrative where a small town, its residents, and the environment are being controlled by a force emanating from the town’s lake. The lake’s aura blinds the residents to its control, forcing them to live in a set way without personal expression or …

Optical Thickness Determination Of Graphene By Color, Caleb A. Woodcock, Michael S. Crosser
Linfield University Student Symposium: A Celebration of Scholarship and Creative Achievement
Graphene, and multi-layered graphene, are single atomic layers of carbon, with remarkable electrical properties. This makes it a highly valuable material across scientific and technological fields. Accurate thickness determination is essential, as many of the electrical properties are layer dependent; however, typical methods are slow. This thesis investigates a non-invasive optical method for estimating graphene thickness by correlating RGB color values from microscope images with step height measurements obtained via atomic force microscopy (AFM). Graphene flakes were exfoliated onto a SiO₂/Si substrate, imaged under white light illumination, and analyzed using ImageJ to extract color values. Results show that red-channel contrast …
Televangelism And The Religious Uses Of Television, William E. Biernatzki S.J.
Televangelism And The Religious Uses Of Television, William E. Biernatzki S.J.
Communication Research Trends
The end of the policy of networks giving free broadcast time to 'mainstream' religious denomination--Catholics, Lutheran, Baptists, etc--has led to radical changes in religious broadcasting in the United States. The organized churches have almost disappeared from most TV channels, while the so-called 'televangelists'--non-demoninational preachers who finance their broadcast by appeals to their viewers--have purchased time on local stations and the networks, as well as vigorously exploiting cable. Some observers even feel that televangelists have become the chief spokespersons for the 'religious viewpoint' in America.
Televangelism has begun to spread outside the United States, to Europe, Latin America, Africa and East …

Popularising Semiotics, Keyan G. Tomaselli, Arnold Shepperson
Communication Research Trends
Semiotics, deconstructionism, structuralism and postmodernism are words which lurk on boundaries of the consciousness of most of us. But they remain shadowy presences except on the rare occasions when we need to wrestle out of them an explanation of just what they are all about.
In this issue of Trends we grapple with one of them, semiotics. C. S. Peirce, the American pragmatist philosopher who coined the term, saw semiotics as a 'method of methods', useful in many disciplines to clarify their own theory and practice. Everyone uses signs and symbols. Everyone thinks they know the meanings of the signs …

Legislative Discouragement Of Maine's Marine Industrial Growth, John H. Pursel
Legislative Discouragement Of Maine's Marine Industrial Growth, John H. Pursel
Maine Law Review
At common law the public rights to navigation and fishing could not be alienated by the King. Dry land and the land beneath the sea could be alienated but only so long as the public rights to navigation and fishing were preserved. This common law doctrine was rejected in 1842 by the United States Supreme Court as binding precedent for limiting the alienation of the public domain by the legislatures of the various states. In Martin v. Wadell the Court reasoned that this common law principle was not applicable because the people of the United States through their act of …
Maine's Educational Television Network: Legal Difficulties, Charles L. Cragin Iii
Maine's Educational Television Network: Legal Difficulties, Charles L. Cragin Iii
Maine Law Review
Maine's educational television network (ETV) cannot pride itself in achieving broad goals and, in fact, cannot attempt to achieve them because of legislation of questionable legal validity which has become an anachronistic barrier to innovations consistent with a keener awareness of the utility of educational television. This note focuses on the state-controlled system servicing northern Maine and points out the conflicts between the system's enabling legislation and the manner in which the system currently operates, and between the enabling legislation and federal laws and regulations. Additionally, it recommends, in a statutory format, the manner in which the system should be …
