Language And Social Justice In Us Climate Movements: Barriers And Ways Forward, Julia Coombs Fine Jun 2022

Language And Social Justice In Us Climate Movements: Barriers And Ways Forward, Julia Coombs Fine

Environmental Studies Faculty Publications

Climate movements increasingly conceptualize the climate crisis as an issue of social injustice, both in terms of its root causes and its present and future effects. Climate justice calls for participatory decision-making within climate movements, which, as communication scholars have pointed out, necessitates inclusive and accessible communicative practices. Within sociocultural linguistics, a growing body of research has explored sociolinguistic justice, or marginalized groups' struggle for self-determined language use. This analysis interweaves these two research areas, applying the theory of sociolinguistic justice to climate communication in organizing contexts. Drawing on 67 semi-structured interviews and 112 online surveys with climate activists from …

Cell Origin-Dependent Cooperativity Of Mutant Dnmt3a And Npm1 In Clonal Hematopoiesis And Myeloid Malignancy., Jennifer M Sanmiguel, Elizabeth Eudy, Matthew A Loberg, Linde A Miles, Timothy M Stearns, Jayna J Mistry, Michael J Rauh, Ross L Levine, Jennifer J. Trowbridge

Faculty Research 2022

In adult acute myeloid leukemia (AML), the acquisition of driver somatic mutations may be preceded by a benign state termed clonal hematopoiesis (CH). To develop therapeutic strategies to prevent leukemia development from CH, it is important to understand the mechanisms by which CH-driving and AML-driving mutations cooperate. Here, we use mice with inducible mutant alleles common in human CH (DNMT3AR882; mouse Dnmt3aR878H) and AML (NPM1c; mouse Npm1cA). We find that Dnmt3aR878H/+ hematopoietic stem cells (HSCs), but not multipotent progenitor cell (MPP) subsets, have reduced cytokine expression and proinflammatory transcriptional signatures and a functional competitive advantage over their wild-type counterparts. Dnmt3aR878H/+ …

Commemorating The Past: Nebraska Museum Practices In Interpreting, Memorializing, And Mythologizing History, Carissa Dowden Jun 2022

Commemorating The Past: Nebraska Museum Practices In Interpreting, Memorializing, And Mythologizing History, Carissa Dowden

Department of Geography: Dissertations, Theses, and Student Research

Commemorative landscapes are spaces that have a symbolic meaning to a group of people and are often identified by a government or by a local community. These landscapes act as “symbolic conduits” to both express and legitimize interpretations of the past, though geographic interpretations are largely limited to the American South and Europe (Alderman and Dywer 2012). This research seeks to better understand landscapes of commemoration and memorialization in Nebraska, specifically how memories of the West and pioneers are constructed and represented within heritage and history institutions. Applying methods in geography, public history and digital humanities, this research considers both …

Acinetobacter baumannii is an opportunistic pathogen of growing concern, as isolates are commonly multidrug resistant. While A. baumannii is most frequently associated with pulmonary infections, a significant proportion of clinical isolates come from urinary sources, highlighting its uropathogenic potential. The type II secretion system (T2SS) of commonly used model Acinetobacter strains is important for virulence in various animal models, but the potential role of the T2SS in urinary tract infection (UTI) remains unknown. Here, we used a catheter-associated UTI (CAUTI) model to demonstrate that a modern urinary isolate, UPAB1, requires the T2SS for full virulence. A proteomic screen to identify …

Since the development of acid copper plating technology, the role of additives is indispensable. In addition to the main salt copper sulfate and supporting electrolyte sulfuric acid, suppressors, accelerators, levelers and chlorine ions (Cl-) are also required to be added into the plating solution. Appropriate additive system can have a significant impact on the coating or the plating solution, which can help improve the quality of the coating and increase the brightness of the coating. Through electrochemical measurement, vibrational spectroscopy, scanning probe microscopy, molecular dynamics simulation and other methods, researches have a deeper understanding in the adsorption configuration …

The Dark Annulus Of A Drop In A Hele-Shaw Cell Is Caused By The Refraction Of Light Through Its Meniscus, Sangjin Ryu, Haipeng Zhang, Carson Emeigh Jun 2022

The Dark Annulus Of A Drop In A Hele-Shaw Cell Is Caused By The Refraction Of Light Through Its Meniscus, Sangjin Ryu, Haipeng Zhang, Carson Emeigh

Department of Mechanical and Materials Engineering: Faculty Publications

Knowing the meniscus shape of confined drops is important for understanding how they make first contact and then coalesce. When imaged from the top view by brightfield microscopy, a liquid drop (e.g., corn syrup) confined in a Hele-Shaw cell, surrounded by immiscible liquid (e.g., mineral oil), had a dark annulus, and the width of the annulus decreased with increasing concentration of corn syrup. Since the difference in the annulus width was presumed to be related to the meniscus shape of the drops, three-dimensional images of the drops with different concentrations were obtained using confocal fluorescence microscopy, and their cross-sectional meniscus …

Explainable And Cooperative Autonomy Across Networks Of Distributed Systems, Peter Joseph Jorgensen

USF Tampa Graduate Theses and Dissertations

Large networks of complex systems-of-systems are commonplace and evermore present in both mundane and extraordinary facets of human existence. From the exponential growth of connectivity via the internet and other information networks, to the miniaturization of computers and sensors, to cross-domain sensor and communication networks, these networks of distributed systems-of-systems (NDSS) present incredible benefits and challenges. Autonomy is perhaps the most important and most difficult to achieve enabling technology for efficient performance of the NDSS. Giving each individual agent in a network the ability to manage its internal state in dynamic operating environments and in pursuit of multiple complex and …

Investigating The Role Of Amyloid Beta Peptides In Inflammation And Alzheimer’S Disease Pathogenesis, Nguyen Le Jun 2022

Investigating The Role Of Amyloid Beta Peptides In Inflammation And Alzheimer’S Disease Pathogenesis, Nguyen Le

Theses and Dissertations

Alzheimer’s Disease (AD) is a neurodegenerative disease characterized by severe memory decline and cognitive impairments. In AD patients’ brains, aggregates of amyloid beta (Aβ) peptides, called amyloid plaques, are hypothesized to trigger innate immune responses that contribute to AD pathogenesis. Interestingly, recent studies demonstrated that an increased level of Aβ protected the host against pathogen infections. Using Drosophila as a model organism, our preliminary data showed that the loss of the Drosophila APP homolog, APPL, led to immune deficits against parasite infection, and that overexpression of Drosophila Aβ produced an inflammatory phenotype. These findings suggested that Aβ might be required …

Classification Of Teleseismic Shear Wave Splitting Measurements: A Convolutional Neural Network Approach, Yanwei Zhang, Stephen S. Gao Jun 2022

Classification Of Teleseismic Shear Wave Splitting Measurements: A Convolutional Neural Network Approach, Yanwei Zhang, Stephen S. Gao

Geosciences and Geological and Petroleum Engineering Faculty Research & Creative Works

Shear wave splitting (SWS) analysis is widely used to provide critical constraints on crustal and mantle structure and dynamic models. In order to obtain reliable splitting measurements, an essential step is to visually verify all the measurements to reject problematic measurements, a task that is increasingly time consuming due to the exponential increase in the amount of data. In this study, we utilized a convolutional neural network (CNN) based method to automatically select reliable SWS measurements. The CNN was trained by human-verified teleseismic SWS measurements and tested using synthetic SWS measurements. Application of the trained CNN to broadband seismic data …

Impacts Of Population Level Environmental Contaminants, Sex Hormones, And Fibroblast Cell Subsets On Systemic Sclerosis (Ssc), Noelle N. Kosarek

Dartmouth College Ph.D Dissertations

Systemic sclerosis (SSc) is a rare autoimmune disease characterized by fibrosis of the skin and internal organs, vascular abnormalities, and autoantibody formation. The etiology of SSc is unknown, though the disease is thought to arise in genetically predisposed individuals after exposure to an environmental factor. There are few FDA approved disease modifying medications available to treat SSc.

The express aims of this dissertation are tripart. First, we aimed to validate a 3D tissue model known as the self-assembled skin equivalent (saSE) model. Second, we sought to describe the geographic distribution of SSc in a US Medicare population to better understand …

Tannic Acid Exhibits Antiangiogenesis Activity In Nonsmall-Cell Lung Cancer Cells, Elham Hatami, Prashanth K.B. Nagesh, Mohammed Sikander, Anupam Dhasmana, Subhash C. Chauhan, Meena Jaggi, Murali M. Yallapu

School of Medicine Publications

Nonsmall-cell lung cancer (NSCLC) is the most common type of lung cancer, with a dismal prognosis. NSCLC is a highly vascularized tumor, and chemotherapy is often hampered by the development of angiogenesis. Therefore, suppression of angiogenesis is considered a potential treatment approach. Tannic acid (TA), a natural polyphenol, has been demonstrated to have anticancer properties in a variety of cancers; however, its angiogenic properties have yet to be studied. Hence, in the current study, we investigated the antiproliferative and antiangiogenic effects of TA on NSCLC cells. The (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium) (MTS) assay revealed that TA induced a dose- and time-dependent …

Epigenetic abnormalities in DNA hydroxymethylation (5hmC) have been detected in patients with myeloid neoplasms, suggesting that 5hmC might act as a valuable epigenetic mark to reflect the disease status of myeloid neoplasms. Here, we report systematic genome-wide mapping of the DNA hydroxymethylomes in over 70 patients with myeloid neoplasms. Our integrative analysis leads to the identification of distinct 5hmC signatures that can sensitively discriminate patients from healthy individuals. At the molecular level, we unveiled dynamic 5hmC changes within key transcription factor (e.g., the CEBP family) binding motifs that are essential for hematopoiesis and myeloid lineage specification. 5hmC redistribution was found …

WUSCHEL-related homeobox (WOX) genes are plant-specific transcription factors (TFs) involved in multiple processes of plant development. However, there have hitherto no studies on the WOX TFs involved in secondary cell wall (SCW) formation been reported. In this study, we identified a Populus trichocarpa WOX gene, PtrWOX13A, which was predominantly expressed in SCW, and then characterized its functions through generating PtrWOX13A overexpression poplar transgenic lines; these lines exhibited not only significantly enhanced growth potential, but also remarkably increased SCW thicknesses, fiber lengths, and lignin and hemicellulose contents. However, no obvious change in cellulose content was observed. We revealed that PtrWOX13A directly …

Robust Explainability: A Tutorial On Gradient-Based Attribution Methods For Deep Neural Networks, Ian E. Nielsen, Dimah Dera, Ghulam Rasool, Nidhal Bouaynaya, Ravi P. Ramachandran

Electrical and Computer Engineering Faculty Publications

With the rise of deep neural networks, the challenge of explaining the predictions of these networks has become increasingly recognized. While many methods for explaining the decisions of deep neural networks exist, there is currently no consensus on how to evaluate them. On the other hand, robustness is a popular topic for deep learning research; however, it is hardly talked about in explainability until very recently. In this tutorial paper, we start by presenting gradient-based interpretability methods. These techniques use gradient signals to assign the burden of the decision on the input features. Later, we discuss how gradient-based methods can …

Organizing Economies: Narrative Sensemaking And Communciative Resilience During Economic Disruption, Timothy Betts Jun 2022

Organizing Economies: Narrative Sensemaking And Communciative Resilience During Economic Disruption, Timothy Betts

USF Tampa Graduate Theses and Dissertations

This dissertation unites organizational communication, and economic theory to understand how individuals make sense of economic crises, imbed power and logic in those understandings, and construct new economic realities in the aftermath of crisis. Contra economic orthodoxy, this project conceives of economy and economics as a social construct. As a social construct, individuals organize economy and economics through discourse, make sense of through narrative, and rebuild through communication. This dissertation combines different theoretical perspectives—actor-network theory, antenarrative organization theory, and the communication theory of resilience—to recenter social scientific accounts of economic reality around communication, story, and power.

Specifically, I focus on …

The main protease, Mpro, of SARS-CoV-2 is required to cleave the viral polyprotein into precise functional units for virus replication and pathogenesis. Here, we report quantitative reporters for Mpro function in living cells in which protease inhibition by genetic or chemical methods results in robust signal readouts by fluorescence (enhanced green fluorescent protein [eGFP]) or bioluminescence (firefly luciferase). These gain-of-signal systems are scalable to high-throughput platforms for quantitative discrimination between Mpro mutants and/or inhibitor potencies as evidenced by validation of several reported inhibitors. Additional utility is shown by single Mpro amino acid variants and structural information combining to demonstrate that …

Neuroblastoma (NB) is a childhood cancer arising from sympatho-adrenal neural crest cells. MYCN amplification is found in half of high-risk NB patients; however, no available therapies directly target MYCN. Using multi-dimensional metabolic profiling in MYCN expression systems and primary patient tumors, we comprehensively characterized the metabolic landscape driven by MYCN in NB. MYCN amplification leads to glycerolipid accumulation by promoting fatty acid (FA) uptake and biosynthesis. We found that cells expressing amplified MYCN depend highly on FA uptake for survival. Mechanistically, MYCN directly upregulates FA transport protein 2 (FATP2), encoded by SLC27A2. Genetic depletion of SLC27A2 impairs NB survival, and …
