This paper examines the effects of a teacher-focused school reform program — combining performance pay with teacher observation and feedback — implemented in high-need schools on students’ longer-run educational, criminal justice, and economic self-sufficiency outcomes. Using linked administrative data from a Southern state, we leverage the quasi-randomness of the timing of program adoption across schools to show that the school reform improved educational attainment and reduced both felony criminal activity before age 19 and dependence on government assistance in early adulthood (ages 18–22). We find little scope for student sorting or changes in the composition of teacher workforce to explain …

Calorie restriction (CR) extends maximal lifespan and maintains cellular homeostasis in various animal models. We have previously shown that CR induces a global reduction of protein fractional synthesis rates (FSRs) across the hepatic proteome in mice, but the timing and regulatory mechanisms remain unclear. Nitric oxide (NO), a bioactive molecule upregulated during CR, is a potential regulator of protein synthesis. To explore the role of NO in hepatic proteome fluxes during CR, we used in vivo deuterium labeling from heavy water and liquid chromatography/mass spectrometry-based (LC/MS-based) flux proteomics in WT and NO-deficient (NO-) mice. We observed a transition to reduced …

Pancreatic ductal adenocarcinoma (PDAC) occurs as a complex, multifaceted event driven by the interplay of tumor-permissive genetic mutations, the nature of the cellular origin, and microenvironmental stress. In this study, using primary human pancreatic acinar 3D organoids, we performed a CRISPR-KO screen targeting 199 potential tumor suppressors curated from clinical PDAC samples. Our data revealed significant enrichment of a list of candidate genes, with neurofibromatosis type 2 associated gene (NF2) emerging as the top target. Functional validation confirmed that loss of NF2 promoted the transition of PDAC to an invasive state, potentially through extracellular matrix modulation. NF2 inactivation …

Demyelination associated microglia (DMAM) orchestrate the regenerative response to demyelination by clearing myelin debris and promoting oligodendrocyte maturation. Peroxisomal metabolism has emerged as a candidate regulator of DMAMs, though the cell-intrinsic contribution in microglia remains undefined. Here we elucidate the role of peroxisome integrity in DMAMs, using cuprizone-mediated demyelination coupled with conditional KO of peroxisome biogenesis factor 5 (PEX5) in microglia. Absent demyelination, PEX5 conditional KO (PEX5cKO) had minimal impact on homeostatic microglia. However, during cuprizone-induced demyelination, the emergence of DMAMs unmasked a critical requirement for peroxisome integrity. At peak demyelination, PEX5cKO DMAMs exhibited increased lipid droplet burden and reduced …

Breast and other solid tumors respond poorly to immune therapy. Myeloid cells (MCs) such as macrophages contribute to resistance. Established clinical evidence links cholesterol to cancer outcomes, with MC function being regulated by cholesterol metabolism. We screened MC-expressed regulators of cholesterol homeostasis linked to survival and identified the cholesterol efflux protein ABCA1. ABCA1 activity increases anticancer functions of macrophages: enhancing tumor infiltration, decreasing angiogenic potential, reducing efferocytosis, and improving support of CD8+ T cell activity. Mechanistically, different AKT isoforms are involved, through both PI3K-dependent and PI3K-independent mechanisms. Highlighting the clinical relevance of our findings are correlations between ABCA1 in macrophages …

Rhabdomyolysis is a clinical syndrome characterised by skeletal muscle necrosis with subsequent release of intracellular constituents into the circulation. Although multifactorial in aetiology, chemotherapeutic agents, including etoposide, are rarely implicated. We report a 62-year-old male who presented to a tertiary care hospital in Muscat, Oman, in 2023 with severe rhabdomyolysis and advanced-stage diffuse large B-cell lymphoma following the first cycle of R-CEOP chemotherapy. Extensive evaluation excluded infectious, inflammatory and structural neuromuscular aetiologies. Work-up for metabolic myopathy suggested, but did not confirm, multiple acyl-CoA dehydrogenase deficiency. The patient’s clinical course was complicated by acute kidney injury necessitating dialysis, followed by gradual …

Disease risk and severity are influenced by genetics, epigenetics, and environmental factors. However, immune responses vary even among genetically similar or related individuals, shaped by inherited and noninherited factors. Using Caenorhabditis elegans, we found that pathogen susceptibility can be predicted by preinfection biomarkers. Individuals with high-basal expression of irg-5, an infection response gene regulated by the p38 mitogen-activated protein kinase-1 (PMK-1) pathway, were more susceptible to Pseudomonas aeruginosa infection. A genome-wide screen identified the myeloid ecotropic viral integration site-1 (MEIS) homeobox protein UNC-62 as a regulator of irg-5 expression, acting through PMK-1 and GATA binding erythroid-like transcription factor …

A greater understanding of chronic lung allograft dysfunction (CLAD) pathobiology, the primary cause of death after lung transplantation (LTx), is needed to improve outcomes. The complement system links innate to adaptive immune responses and is activated early after lung transplantation to form C3 convertase, a critical enzyme that cleaves the central complement component C3. We hypothesized that LTx recipients with a genetic predisposition to enhanced complement activation have worse CLAD-free survival mediated through increased adaptive alloimmunity. We interrogated a known functional C3 polymorphism (C3 R102G) that increases complement activation through impaired C3 convertase inactivation in 2 independent LTx recipient cohorts. …

In short-term mine planning for deposits characterized by multiple variables, sequential diglines are generated with an emphasis on maintaining consistent grade distributions across mining periods. This research integrates an array of mineral variables, including deleterious elements, to facilitate the establishment of excavation geometries that enable precise grade and volume control within operational production zones. Through the systematic application of geostatistical analysis coupled with process optimization, geological risks associated with grade uncertainties are minimized, and an optimized operational sequence for the extraction of mineral blocks is pursued. The main point of this approach is maintaining grade distributions aligned with the historical …

The location of an orebody known to show regional variability in space is determined only through 3D geological modeling. A reliable 3D orebody model must be obtained to ensure resource efficiency and sustainability in the mining industry. In addition, 3D geological modeling has become indispensable in intelligent mining, smart mining, and mining 4.0. In previous years, explicit methods were widely used for 3D geological modeling. However, the validity of geological models produced with these methods has been debated. It has been understood that they are inadequate in modeling vein-type mineralizations with complex geometry in particular. For this reason, implicit modeling …

This article explores the technical principles of pneumatic seismic sources, focusing on their application in underground mining operations, including environments prone to fire hazards. It introduces an intrinsically safe pneumatic seismic source as a cost-effective solution for quasi-continuous seismic excitation, useful for detecting stress concentrations in mining settings. These stress concentrations are identified through anomalies in seismic wave velocity, which may indicate potentially hazardous deformations in the rock mass. The developed system enables fully remote operation from the surface and integrates with a seismic monitoring system to capture high-resolution waveforms. The study provides an overview of the pneumatic system’s technical …

The mining industry faces evolving challenges in adapting to the impacts of climate change. Among these challenges is the management of tailings. This study focuses on investigating surface disposal of non-cemented paste tailings, particularly the effect of water content, freeze-thaw cycles (FTC, defined as freezing the sample to –20°C and thawing it to +20°C), and ambient temperature on the mechanical characteristics and thermophysical properties of paste tailings. The potential for maintaining non-cemented tailings in a frozen state year-round is also investigated through field-scale numerical simulations. Experiment results of thermophysical properties of tailings with water content, freeze-thaw cycles and ambient temperatures …
