Since the FDA approval of IL-2 in cancer treatment in the 1980s, immunotherapy has emerged as the standard of care for several tumor types (e.g., melanoma, lung, bladder, colorectal). However, approximately 70 percent of cancer patients, independent of prior treatment or staging, administered immunotherapy will not achieve complete responses and will exhibit disease progression. Several factors have been identified to contribute to treatment failure, including the accumulation of intratumoral immunosuppressive cell populations, insufficient tumor-infiltrating anti-tumor immune populations, or the occurrence of immunoediting on tumor cells. Chronic lymphocytic leukemia (CLL) is a predominant B-cell malignancy impacting elderly populations in Western societies. Targeted therapies have revolutionized the CLL treatment landscape. Still, patients often require long-term therapy and are subject to drug-induced resistance and severe toxicities, for which CLL is still considered an incurable disease. T-cell-based immunotherapy, particularly chimeric antigen receptor (CAR) T cells, has shown remarkable success, leading to various FDA approvals in hematological malignancies. Nonetheless, CAR T-cell therapy success rate is limited in CLL because of the dysfunctional nature of T cells. Nutrition, cellular metabolism, and inflammatory regulation of the immune system are interconnected, and advancing our understanding of this will greatly improve treatment of metabolic and inflammatory disease. Specifically, dietary saturated fats are becoming increasingly appreciated for their ability to modify innate immune cell function in mammals following absorption into the blood and lymphatic circulation systems. The work herein focuses on the capacity for lipids in the diet to modulate innate immune memory. Symbionts, or organisms that form close and persistent associations with hosts, are ubiquitous across the tree of life, and symbioses are among the most common interspecific interactions in the natural world. Symbionts play a fundamental ecological and evolutionary role across multiple levels of biological organization – from individuals to ecosystems – and can have strong effects on global biodiversity. However, many aspects of these relationships are poorly understood including symbiont genetic diversity (inter- and intraspecific), host specificity, and the mechanisms underlying their functional nature or any context-dependent patterns. Humanized mice, defined as mice with human immune systems, have become an emerging model to study human hematopoiesis, infectious disease, and cancer. Here, we describe the techniques to generate humanized NSGF6 mice using adult human CD34 + hematopoietic stem and progenitor cells (HSPCs). We describe steps for constructing and monitoring the engraftment of humanized mice. We then detail procedures for tissue processing and immunophenotyping by flow cytometry to evaluate the multilineage hematopoietic differentiation. In this review article, it is emphasized that adopting advanced molecular diagnostics is essential for modern UTI management, offering a more comprehensive, accurate, and rapid approach compared to traditional methods. Standard urine culture’s limitations include low sensitivity and failure to culture certain microorganisms, leading to undiagnosed cases and increased morbidity. Advanced molecular techniques, like multiplex-PCR and pooled antibiotic susceptibility testing have been shown to reduce empiric treatments and negative outcomes significantly. And advanced molecular methods like metagenomics (mNGS) offer comprehensive pathogen detection without prior knowledge of target organisms, improving diagnostic yield. These methods also detect antibiotic resistance genes, aiding in …

Background: The human immunodeficiency virus (HIV) prevalence among transport workers in sub-Saharan Africa remains high, estimated at as high as 9.9% in western Uganda compared with the national prevalence of 5.4%. The prevalence of HIV among transport workers has been partly attributed to the level of knowledge regarding HIV prevention, perceived HIV risk, and stigma. Accordingly, these have been linked to high-risk HIV transmission behaviours that increase the chances of acquiring HIV among adults. Therefore, this study investigated the predictors of HIV knowledge, perceived HIV risk, and stigma among transport workers in Mbarara city in southwestern Uganda.

Group B Streptococcus (Streptococcus agalactiae; GBS) is a leading cause of neonatal sepsis worldwide. Intrapartum antibiotics in GBS-colonized mothers significantly reduced the incidence of early-onset sepsis occurring in the first week of life; however, there are currently no prophylactic interventions for late-onset sepsis, occurring within the first 3 months of life. GBS is a pathobiont that primarily colonizes the gastrointestinal tract and can cross the intestinal barrier via both the paracellular and transcellular routes, causing invasive disease. Neonatal susceptibility to GBS intestinal translocation is mediated by inadequate epithelial barrier function and microbiome immaturity; however, the mechanisms by which GBS exploits …

Approximately 22 percent of the world population is estimated to be infected with Mycobacterium tuberculosis. The largest number of cases are seen in Sub-Saharan Africa, India, and the islands of Southeast Asia, while North America and Western Europe are “low incidence” regions.1 In the US, most cases are seen in foreign-born individuals who emigrate from high-incidence countries. Despite the low incidence in the US, prevention and control of Tuberculosis (TB) remain an important component of Public Health.2 Here we present a case of a patient of Vietnamese origin with reactivation TB and rare manifestations of TB.

Multidrug resistant Escherichia Coli (MDR E. Coli) is a less common cause of bacterial community-acquired pneumonia as compared to Streptococcus pneumoniae and Staphylococcus aureus. We present a case of MDR E. Coli community-acquired pneumonia diagnosed on culture of bronchoalveolar lavage specimens (BAL) after bronchoscopic debulking of a metastatic melanoma mass completely obstructing the left main stem bronchus (LMSB).

The practicum site observed that education on the needs of the transgender community was lacking, discussing a discrepancy in the information offered and the scant attention paid to the unique requirements of transgender individuals. Infant nutrition and immune protection require the consumption of human milk, with macrophages mainly contributing to immune defense and infant development. This study was performed to investigate the correlation between macrophage cell numbers in human milk and various maternal factors. Thirteen human milk samples were collected from lactating mothers and were evaluated for macrophage cell numbers using a hemacytometer. The recorded questionnaire containing several questions, including age, multivitamin consumption, coronavirus disease 2019 vaccination status, allergy history, and lactation duration, was used to determine the maternal factors. Pearson’s correlation and Mann-Whitney test was employed in statistical analysis, with p-values < 0.05 considered significant. The macrophage cell number reached 54.236 ± 7.456 cells per mL. Acute liver injury may be very difficult to treat, especially when the etiology is unknown. We describe a case of a patient who presented with signs and symptoms of acute liver injury and went through an extensive work-up that did not reveal a definitive cause. The lack of diagnosis can be distressing for the patient, so it is important to regularly communicate the current plan and potential future necessary steps (such as a transplant surgery) to facilitate smooth, informed decision-making throughout the whole process.

This dissertation examines Indigenous women’s access to and beliefs regarding gender-based violence (GBV) services and the Guatemalan legal system. It illuminates the legal, structural, and institutional challenges and power dynamics associated with seeking support and justice for women in Guatemala, as well as why and how women decide to seek formal assistance. This research also investigates changes in attitudes and beliefs regarding women’s rights, gender roles, and formal and informal forms of support for GBV survivors. This paper investigates the application of immersive multimedia tools as an effective means of distraction in Emergency Departments (ED). With a focus on ageing populations exhibiting Acute Behavioural Disturbance (ABD) and the aim of reducing the reliance on psychotropic medications and sedatives. The study explores the use of immersive multimedia for distraction using readily available technology (VR headsets, curved displays, and 360-degree video on tablets). Consumer devices were customised through co-design collaboration with experienced ED clinicians to adapt to the unique challenges of the ED environment. Healthcare systems are complex social systems wherein improvements related to staff practice and behavior can be difficult to implement. Knowledge and practice can be uneven between various specializations, hierarchical imbalances limit the degree of agency of different professionals, and evidence-based guidelines may be interpreted or implemented according to context-specific factors. Engaging communities and stakeholders in developing user-centred urban health solutions, whilst linking the research to their own development, pose major challenges for design researchers working in the Global South. In a number of circular sanitation projects in a community school in Ghana, we co-designed and installed an anaerobic digester delivering electricity and sanitation improvements. To enhance impact we developed and pilot-tested a Living and physical Lab design approach. One project focussed on hand hygiene.
