Since the early 1990s, there has been a dramatic rise in gastrointestinal cancers diagnosed in patients under age 50 for reasons that remain poorly understood. The most significant change has been the increase in incidence rates of early-onset colorectal cancer, especially rates of left-sided colon and rectal cancers. Increases in gastric, pancreatic, and other gastrointestinal cancer diagnoses have further contributed to this trend. We formed a multidisciplinary Think Tank to develop a strategic, coordinated approach to studying early-onset gastrointestinal cancers. This area of research is challenging given multifactorial etiologies. We focused on epidemiology and the environment, the microbiome, and survivorship …

BACKGROUND: Antiretroviral therapy (ART) is recommended for all individuals with HIV infection, including those with acute HIV-1 infection (AHI). While recommendations are similar to those for chronic infection, efficacy data regarding treatment of acute HIV is limited.

METHODS: This was a single arm, 96-week study of a once-daily integrase inhibitor (INSTI)-based regimen using elvitegravir/cobicistat/emtricitabine/tenofovir disoproxil fumarate (EVG/COBI/FTC/TDF) in AHI. Primary endpoint was proportion of participants with HIV-1 RNA <200 copies/mL and <50 copies/mL by treatment weeks 24 and 48, respectively. We also examined time to viral suppression and weight gain after treatment initiation. Outcomes and characteristics were compared with a historical AHI cohort using a non-nucleoside reverse transcriptase inhibitor (NNRTI)-based regimen with efavirenz/emtricitabine/tenofovir disoproxil fumarate (EFV/FTC/TDF).

RESULTS: Thirty-three participants with AHI were enrolled with 31 available for analyses. Most were African American (61%) and men who have sex with men (73%). Median age was 26 (IQR 22-42). Demographics …

Georgia Journal of Science

Flinders Technology Associates (FTA) cards are increasingly accepted as an accurate method for nucleic acid detection, including testing for the presence of various avian diseases. However, the accuracy of FTA cards for detection of Mycoplasma gallisepticum (MG) in wild birds has not been published. Traditional techniques for testing MG in birds include serum plate agglutanation (SPA), hemagglutination inhibition (HI), polymerase chain reaction (PCR), and enzyme linked immunosorbent assay (ELISA) all of which are time consuming and require significant financial investment into specialized equipment. We tested the efficacy of using FTA cards to detect MG in wild bird carcasses collected in …

Optimizing Biofilm Assay Reproducibility And Characterizing The 12-Hydroxystearic Acid On Escherichia Coli, Khilola Khusanova

Theses and Dissertations

Biofilm formation in Escherichia coli plays a critical role in microbial chemical communication and surface-associated persistence. This study investigates the antibiofilm activity of 12-hydroxystearic acid (12-HSA), evaluating its R and S enantiomers as well as a racemic mixture. E. coli BL21, biofilms were grown under standardized laboratory conditions and quantified using the Crystal Violet staining assay. Treatment with 12-HSA resulted in a measurable reduction in biofilm biomass, with differences observed between the individual enantiomers and the mixture. These findings suggest that 12-HSA influences early adhesion and biofilm development, supporting its potential as a small-molecule modulator of bacterial surface behavior. This …

Mouse models are vital tools for discerning the relative contributions of host and microbial genetics to disease, often requiring the transfer of microbiota between different mouse strains. Transfer methods include antibiotic treatment of recipients and colonization using either co-housing with donors or the transplantation of faecal or caecal donor material. However, the efficiency and dynamics of these methods in reconstituting recipients with donor microbes is not well understood. We thus directly compared co-housing, faecal transplantation, and caecal transplantation methods. Donor mice from Taconic Biosciences, possessing distinct microbial communities, served as the microbial source for recipient mice from Jackson Laboratories, which …

Lusaka's Colonial Legacy: Town Planning, Public Health, And The Making Of A Segregated City, 1913–1963. A Study Of Power, Inequality, And Urban Development, Wamburakwao J. Sapao

Zambia Social Science Journal

This paper explores the intersection of town planning and public health policy in Lusaka, Northern Rhodesia (now Zambia), during the colonial period (1913–1963), which was a time of significant urban growth and transformation. British colonial authorities adapted Ebenezer Howard's Garden City principles to design and manage Lusaka, but these measures perpetuated spatial control, segregation, and social engineering, thus exacerbating racial and socio-economic disparities. By analysing secondary and primary sources, including planning ordinances, zoning regulations, and public health policies, this study reveals how these measures shaped the city's development and reinforced inequalities. Despite colonial intentions for a white settler-city, Lusaka evolved …

Measuring Spatial And Temporal Variation In Species Composition Of Ephemeral Pond Amphibian Assemblages Using Metabarcoding, Elizabeth Strasko

Masters Theses & Specialist Projects

Amphibian populations worldwide face severe threats, heightening the need for reliable monitoring methods. In this thesis, I explored and tested environmental DNA (eDNA) metabarcoding protocols to survey amphibian assemblages in ephemeral ponds of the the Green River Preserve in central Kentucky, USA. First, I evaluated whether the commonly used mitochondrial cytochrome c oxidase subunit I (COI) gene could effectively capture species-level variation in local amphibians for eDNA metabarcoding. Extensive primer design and testing revealed significant challenges in amplifying fragmented eDNA from water samples. In a second study, I applied a metabarcoding approach using the mitochondrial 12S ribosomal RNA (12S) gene …

The Effect Of Covid-19 Lockdowns And Social Isolation On Adolescence, Madeleine J. Bakken

All Electronic Theses and Dissertations

Adolescents around the world were impacted by social isolation resulting from the COVID-19 pandemic. Students experienced sudden school closures and isolation from classmates resulting in increased screen time, the erosion of social skills, lower academic motivation, and a period of unstructured and unfamiliar learning (Hong et al., 2022; Kannan & Veazie, 2023; Leventhal & Morishta, 2024; Menzies et al., 2024; Warford, 2019). Current research is working to measure the impact and severity of these factors. Early findings suggest that students are academically and emotionally behind where their predecessors were pre-COVID (Menzies et al. 2023; NWEA Connection, 2024). Educators and mental …
