Developing A High-Throughput Machine Learning Screening Platform For Genetic And Pharmacological Modulators On Caenorhabditis Elegans Seizure Behavior, Paul A. Scarpinato

Epilepsy is a chronic neurological disorder characterized by recurrent seizures that affect over 50 million people worldwide, including more than 3.5 million in the United States. Seizures arise from a disrupted balance between excitation and inhibition. Although antiepileptic drugs (AEDs) remain the primary treatment to restore the excitation and inhibition balance, approximately 30% of patients experience drug-resistant epilepsy, highlighting the need for additional antiseizure medications. The Caenorhabditis elegans electroshock assay was developed in relative alignment with the rodent maximal electroshock seizure (MES) test, the gold standard for preclinical AED screening, provides a cost-effective platform for prescreening candidate AEDs.

The success of commercial laying hen farms depends on the possibility of managing environmental conditions properly and performing strict biosecurity measures, which would increase productivity, enhance the performance of the flock, and save money, especially in regions characterized by challenging climatic conditions, such as the sub-arid and arid areas. Untargeted Metabolomics Reveals Organ-Specific And Extraction-Dependent Metabolite Profiles In Endemic Tajik Species Ferula Violacea Korovin, Sylhiya Mavlonazarova, Kenneth Acosta, Rinat Abzalimov, Saidbeg Satorov, Vyacheslav Dushenkov

Ferula violacea Korovin, an endemic Tajikistani plant with purported medicinal properties, remains understudied. This study employs untargeted metabolomics to characterize the metabolite profiles of ethanol extracts and juices from F. violacea roots and seeds. In total, 540 distinct metabolites are putatively identified, 419 of which are previously unreported in the Ferula genus, representing a substantial expansion of its known chemical diversity. The most abundant metabolites are terpenoids, amino acid derivatives, and alkaloids. A particularly abundant group of daucane sesquiterpenoids, sharing a common (6-methyl-azulen-4-yl) cyclohexanecarboxylate substructure, is identified, including known metabolites such as ferutidin and ferutinin. We investigated Wedge-tailed Shearwater Ardenna pacifica and Barau's Petrel Pterodroma baraui exposure to infectious agents circulating in the western Indian Ocean. Using polymerase chain reaction and serological analyses, we screened for influenza viruses, flaviviruses, coronaviruses, and blood parasites (Haemoproteus, Leucocytozoon, and Plasmodium). No active infections were detected and serological results suggested limited past exposure, with only two Wedge-tailed Shearwaters testing positive for antibodies to the influenza A virus. These findings indicate the limited role of these species as epidemiological reservoirs. Updating The Status Of The Peruvian Pelican Pelecanus Thagus Following Avian Influenza And El Niño Events In Peru, Liliana Ayala, Gabriela Sandoval-Lazaro

The Peruvian Pelican Pelecanus thagus is an endemic species of the Humboldt Current System. Between November 2022 and March 2024, the avian influenza epidemic and El Niño conditions significantly affected the pelican population. This study provides an update on the population size and breeding status of the species in Peru. Pelican abundance decreased by 31.5% to 37.6% within marine protected areas. The number of live pelicans ranged from 1,652 to 46,046 in 2023 and from 14,505 to 52,540 in 2024, based on counts conducted in natural protected areas by Peruvian government officials. Brain abscess in neonates is rare but remains a severe, life-threatening complication of meningitis with significant morbidity and mortality. While Citrobacter koseri and Citrobacter freundii are commonly implicated in brain-abscess formation, Citrobacter sedlakii is an uncommon causative pathogen. To date, only two cases of neonatal brain abscess caused by C. sedlakii have been reported in the literature. Early diagnosis, timely surgical intervention and prompt antibiotic therapy are critical to achieving a favourable outcome. Background: The effect size estimated from a pilot trial is often an inaccurate reflection of the true effect size observed in a large trial, leading to either underestimation or overestimation. Published data suggest that effect sizes from large trials are typically smaller than those reported in their corresponding pilot trials. To address this discrepancy, conservative or discount adjustment methods are widely recommended to modify pilot trial effect sizes when calculating sample sizes, thereby maintaining adequate statistical power. This study aims to assess effect sizes from both pilot and large trials and to evaluate the performance of existing adjustment methods.

BACKGROUND: The 2023-2024 US influenza season was characterized by a predominance of A(H1N1)pdm09 virus circulation with cocirculation of A(H3N2) and B/Victoria viruses. We estimated vaccine effectiveness (VE) in the United States against mild-to-moderate medically attended influenza illness in the 2023-2024 season.

METHODS: We enrolled outpatients aged ≥8 months with acute respiratory illness in 7 states. Respiratory specimens were tested for influenza type/subtype by reverse-transcriptase polymerase chain reaction. Influenza VE was estimated with a test-negative design comparing odds of testing positive for influenza among vaccinated versus unvaccinated participants. We estimated VE by virus subtype/lineage and A(H1N1)pdm09 genetic subclades.

BACKGROUND: Children with central nervous system malaria (CNS-M) have gene expression profiles and plasma biomarkers of malaria pathogenesis that differ from children with uncomplicated malaria (UM, malaria infection with fever and no systemic organ dysfunction) or acute febrile non-malaria illness (AFN). However, there is scant knowledge of the temporal dynamics of gene expression and plasma biomarkers during recovery from CNS-M.

More than 2.8 million infections due to antibiotic-resistant bacteria occur annually in the United States, and more than 35,000 people die as a result. Domestic wastewater (sewage), which contains antibiotic-resistant bacteria and antibiotic resistance genes, is collected at treatment plants for removal of contaminants, e.g. pathogenic microorganisms and nutrients. While wastewater treatment is designed to remove antibiotic-resistant bacteria and other microorganisms through treatment and disinfection, some bacteria may escape treatment. Treated wastewater effluent is generally discharged into surface waters; however, some fraction of the effluent may be used as recycled water. Guillain-Barré syndrome is an acute polyradiculoneuropathy in which preceding infections often elicit the production of antibodies that target peripheral nerve antigens, principally gangliosides. Anti-ganglioside antibodies are thought to play a key role in the clinical diversity of the disease and can be helpful in clinical practice. Extensive research into clinical associations of individual anti-ganglioside antibody specificities has been performed. Recent research has highlighted glycolipid complexes, glycolipid combinations that may alter antibody binding, as targets. In this study, we investigated antibody reactivity patterns to glycolipids and glycolipid complexes using combinatorial array, in relation to clinical features in Guillain-Barré syndrome.
