Spaceflight associated neuro-ocular syndrome (SANS) is one of the potential barriers to human long-duration spaceflight (LDSF), including a manned mission to Mars. While a large barrier, the pathophysiology of SANS is not well understood, and functional and structural findings from SANS continue to be further characterized. Currently on the International Space Station (ISS), scheduled visual assessments are static visual acuity, Amsler grid, and a self-reported survey. Additional visual assessments may help the understanding of this neuro-ophthalmic phenomenon, as well as the effects of spaceflight of overall ocular health. In this paper, a case is made for expanding scheduled visual assessments …

Major achievements in bone research have always relied on animal models and in vitro systems derived from patient and animal material. However, the use of animals in research has drawn intense ethical debate and the complete abolition of animal experimentation is demanded by fractions of the population. This phenomenon is enhanced by the reproducibility crisis in science and the advance of in vitro and in silico techniques. 3D culture, organ-on-a-chip, and computer models have improved enormously over the last years. Nevertheless, the overall complexity of bone tissue-cross talk and the systemic and local regulation of bone physiology can often only …

Noncanonical Binding Of Transcription Factors: Time To Revisit Specificity?, Md Abul Hassan Samee Aug 2023

Noncanonical Binding Of Transcription Factors: Time To Revisit Specificity?, Md Abul Hassan Samee

Faculty, Staff and Students Publications

Transcription factors (TFs) are one of the most studied classes of DNA-binding proteins that have a direct functional impact on gene transcription and thus, on human physiology and disease. The mechanisms that TFs use for recognizing target DNA binding sites have been studied for nearly five decades, yet they remain poorly understood. It is classically assumed that a TF recognizes a specific sequence pattern, or motif, as its binding sites. However, recent studies are consistently finding examples of noncanonical binding, that is, TFs binding at sites that do not resemble their sequence motifs. Here we review the current literature on …

VEGF inhibitors are one of the most successful antiangiogenic drugs in the treatment of many solid tumors. Nevertheless, pancreatic adenocarcinoma (PAAD) cells can reinstate tumor angiogenesis via activation of VEGF-independent pathways, thereby conferring resistance to VEGF inhibitors. Bioinformatic analysis showed that BICC1 was one of the top genes involved in the specific angiogenesis process of PAAD. The analysis of our own cohort confirmed that BICC1 was overexpressed in human PAAD tissues and was correlated to increased microvessel density and tumor growth, and worse prognosis. In cells and mice with xenograft tumors, BICC1 facilitated angiogenesis in pancreatic cancer in a VEGF-independent …

To account for athletes of different sizes, kinetic values are commonly ‘normalized’ by dividing by mass and/or height. However, the creation of a ratio variable requires certain statistical assumptions to be met. The purpose of this study was to determine if elbow valgus torque predicted by pitching velocity is influenced by the normalization method using regression model comparison with normalized torque values. Both mass and mass*height normalization satisfied the correlation and zero intercept assumptions. Results did not agree between analysis methods that elbow valgus torque could be predicted with pitching velocity at the α = 0.05 level, indicating caution should …

An epidemic of obesity has affected large portions of the world, increasing the risk of developing many different age-associated diseases, including cancer, cardiovascular disease, and diabetes. In contrast with the prevailing notion that "a calorie is just a calorie," there are clear differences, within and between individuals, in the metabolic response to different macronutrient sources. Recent findings challenge this oversimplification; calories from different macronutrient sources or consumed at different times of day have metabolic effects beyond their value as fuel. Here, we summarize discussions conducted at a recent NIH workshop that brought together experts in calorie restriction, macronutrient composition, and …

The Effects Of Ketones On Brain Metabolism And Cognition, Erin Reiko Saito Jul 2023

The Effects Of Ketones On Brain Metabolism And Cognition, Erin Reiko Saito

Theses and Dissertations

The brain is one of the most energetically demanding organs within the human body and is cognitively susceptible to energetic deficits such that the rise in obesity, insulin resistance, and Alzheimer’s disease in recent decades pose a substantial threat to cognitive longevity. The therapeutic efficacy of ketones are well-established in epilepsy and are currently being applied to other disease states. Alzheimer’s disease is characterized by impairments in brain glucose uptake and metabolism in regions relevant to learning, memory, and cognition that progress with the disease. While brain glucose uptake is impaired, ketone uptake is unaltered, potentially enabling ketones to fuel …

Cardiac Output And Stroke Volume From The Pneumocardiogram: A New Mechanistic Insight And Practical Calculation, Charles F. Babbs Jul 2023

Cardiac Output And Stroke Volume From The Pneumocardiogram: A New Mechanistic Insight And Practical Calculation, Charles F. Babbs

Weldon School of Biomedical Engineering Faculty Working Papers

This paper presents a new and more complete mechanism for the genesis of the pneumocardiogram—a record of the small oscillations in airflow in the trachea and upper airways accompanying each heartbeat. This signal may be the basis of a simple, inexpensive and noninvasive measure of cardiac stroke volume; however, research into its practical use has languished because of poor understanding of its underlying cause and meaning. This paper explains the genesis of the pneumocardiogram, based upon net changes in intrathoracic blood volume during the cardiac cycle, together with low pass filtering of the volume signal by airway resistance and lung-chest …

Nitric oxide (NO) is a gaseous molecule that has a central role in signaling pathways involved in numerous physiological processes (e.g., vasodilation, neurotransmission, inflammation, apoptosis, and tumor growth). Due to its gaseous form, NO has a short half-life, and its physiology role is concentration dependent, often restricting its function to a target site. Providing NO from an external source is beneficial in promoting cellular functions and treatment of different pathological conditions. Hence, the multifaceted role of NO in physiology and pathology has garnered massive interest in developing strategies to deliver exogenous NO for the treatment of various regenerative and biomedical …

Identifying Factors Contributing To Under-Five Mortality In Nigeria, Olateju Alao Bamigbala, Ayodeji Oluwatobi * Ojetunde

Tanzania Journal of Science

The under-5 mortality rate (U5MR) is the probability that a child born in a specified year will die before turning 5 years of age. U5MR is still high in Nigeria. Therefore, this study aimed to identify factors contributing to under-5 mortality in Nigeria. The data used in this study were from the 2018 NDHS, encompassing 2013–2018. The methods of analysis used for this study were frequency, percentage, and Zero Inflated Negative Binomial (ZINB) regression. Data were analyzed using R programming v.4.1.2 and p < 0.05 was considered to be statistically significant. The result showed 35.4% under-5 mortality out of the 33,924 infant mortality sample data that were collected. The findings revealed maternal age, regions (North East and North West), maternal education (no education), wealth index (poorest households), and size of child at birth (very small birth size) as significant factors associated with under-5 mortality in Nigeria. The results also showed that the odds of under-5 mortality increase as the age of the mother increases. Therefore, the Nigerian government should understand that poverty is not just an economic problem but also a significant factor in health; as a result, the battle against poverty needs to receive the necessary attention.  Keywords:    Under-5 mortality; World Health Organization; Zero inflated distribution; Risk factors; Nigeria.

Investigation Of Permanent Tattoos To Increase Myoelectric Signal Connectivity In Prosthetics, Guy Pugh Jun 2023

Investigation Of Permanent Tattoos To Increase Myoelectric Signal Connectivity In Prosthetics, Guy Pugh

Master of Science in Kinesiology

Myoelectric prosthetics offer users increased functionality in many facits of their lives. However, maintaining reliable connectivity between sEMG sensors and targeted muscle locations can be problematic. Volume fluctuation of the residual limb, dirt, sweat, and movement of the socket and sensor over the limb can all because of signal disruption leading some users to find the myoelectric prostheses, costing thousands of dollars, to be unreliable, and stop using it. To increase connectivity between the optimal myoelectric sites and sEMG sensors, this paper proposes the use of permanent ink tattoos to create a stable location which a sensor can move over …

Articles

Metabolic dysfunction increases with age and is a contributing factor to Alzheimer's disease (AD) development. We have previously observed impaired insulin sensitivity and glucose homeostasis in the APP/PS1 model of AD. To improve these parameters, we chronically exposed male and female mice to mild hypothermic environmental temperature (eT), which positively modulates metabolism. Although a hypothermic eT normalized insulin sensitivity, glucose tolerance was still impaired in both sexes of AD mice. We observed increased plasma glucagon and B-cell activating factor in both sexes, but additional sexually dimorphic mechanisms may explain the impaired glucose homeostasis in AD mice. Hepatic Glut2 was decreased …

The melanocortin pathway is well established to be critical for body-weight regulation in both rodents and humans. Despite extensive studies focusing on this pathway, the downstream brain sites that mediate its action are not clear. Here, we found that, among the known paraventricular hypothalamic (PVH) neuron groups, those expressing melanocortin receptors 4 (PVHMc4R) preferably project to the ventral part of the lateral septum (LSv), a brain region known to be involved in emotional behaviors. Photostimulation of PVHMc4R neuron terminals in the LSv reduces feeding and causes aversion, whereas deletion of Mc4Rs or disruption of glutamate release from LSv-projecting PVH neurons …
