Precision oncology has transformed the treatment landscape for patients with advanced solid tumors. Tumor-agnostic therapies, those that have been approved based on genetic mutations or biomarkers across tumor histology types, are important examples of how the implementation of precision oncology can expand therapeutic options for patients, especially those with rare cancer types and treatment-refractory disease. In this review, we first discuss how advances in next-generation sequencing and molecular profiling have enabled the identification of shared actionable alterations. Recent research advances have resulted in an experimental benchtop X-ray fluorescence computed tomography (XFCT) system that likely meets the imaging dose/scan time constraints for benchtop XFCT imaging of live mice injected with gold nanoparticles (GNPs). For routine in vivo benchtop XFCT imaging, however, additional challenges, most notably the need for rapid/near-real-time handling of X-ray fluorescence (XRF) signal extraction and XFCT image reconstruction, must be successfully addressed. Microvascular proliferation (MVP) is a disease-defining hallmark of glioblastoma and other World Health Organization grade 4 gliomas. MVP also serves as a poor prognostic marker in various solid tumors. Despite its clinical significance, the mechanisms and biological consequences of MVP are controversial and remain unclear. In this study, we performed single-cell RNA sequencing on paired CD45-CD105+ vascular/perivascular stromal cells (PVSC) and CD45+CD105± immune cells from 16 primary glioma patient samples, both with and without MVP. This analysis revealed the presence of developmentally related mesenchymal stem cells alongside cancer-associated fibroblasts, pericytes, fibromyocytes, and smooth muscle cells within the CD45-CD105+ compartment. BACKGROUND: Poorly healing wounds represent the primary health-related burden for hereditary recessive dystrophic epidermolysis bullosa (RDEB) patients. Contribution of wound-associated soluble constituents to wound progression remains not well defined.

OBJECTIVE: To conduct cross-sectional analysis of cytokine, chemokine, and growth factor in exudates from RDEB wounds and define changes associated with wound progression.

METHODS: Concentrations of selected cytokines, chemokines, and growth factors were evaluated by multiplex ELISA in eight blister fluids and 66 exudates from early, established, and chronic RDEB and five chronic venous ulcers (VU). A cross-sectional analysis was performed.

Digital mental health interventions (DMHIs) offer scalable solutions for addressing mental health needs, but low adherence remains a significant challenge. Single session DMHIs may offer a feasible alternative to address this issue, providing an accessible and low-effort option for users. The present study examines whether adding the choice of a single session DMHI in addition to a multi-session DMHI provides a feasible alternative for users who might otherwise disengage. This study examined naturalistic data from 509 university students who registered for a 12-session DMHI, single session DMHI, or both. BACKGROUND: The diagnosis of atrial fibrillation (AF) is increasing among elite athletes. This case highlights the novel use of an implantable loop recorder to guide treatment decisions for AF in an elite soccer player.

CASE SUMMARY: A 25-year-old professional soccer player who had received a diagnosis of asymptomatic AF during routine screening underwent cardioversion and continuous monitoring with an implantable loop recorder. A rising AF burden prompted ablation.

DISCUSSION: This case demonstrates the potential utility of advanced monitoring technologies for managing AF in athletes, particularly when symptoms are absent or minimal. Additional studies are needed to confirm whether continuous monitoring …

California's Imperial Valley, with lithium-rich geothermal brines, extensive flat agricultural fields with abundant desert sunshine, and access to transmission lines, is a leading case to explore interconnected themes around just energy transitions. Despite being the poorest county in California and one of the smallest, Imperial already provides 15% of the state’s solar electricity, and the region as a whole represents on the order of 25% of the state’s electric power capacity. The REnal cancer consortium for Focused Investigation of Novel biomarkers and Expression (REFINE) consortium represents an important initiative in integrating clinical data with molecular sequencing in patients with advanced renal cell carcinoma (RCC) treated with immunotherapy-based approaches. While the incidence and survival associated with lymphoma have improved in recent years, outcome disparities related to sociodemographic factors such as age, sex, race, ethnicity, socioeconomic status (SES), and other social determinants of health (SDOH) remain prevalent in the modern era. Clinical trials are crucial for further improving outcomes, yet not all sociodemographic groups are equally represented in contemporary lymphoma trials. We conducted a systematic review of the literature evaluating the impact of SDOH on enrollment into clinical trials in the United States and identified 21 relevant studies. During the COVID-19 pandemic, African American adolescents experienced reduced access to needed resources and may have suffered poorer mental health outcomes as a result. The purpose of this quantitative study was to examine the association between race and the availability of mental health resources, food, medical health resources and mental health outcomes among African American adolescents in the United States during the COVID-19 pandemic compared to their White counterparts. The problem this quantitative study addressed was police officers’ willingness to use organizational help-seeking interventions and rely on organizational support to mitigate operational and organizational stress. Previous literature discussed how officers engaged in different maladaptive coping behaviors as a result of work related stressor or low well-being, divorce, depression, prescription drug abuse, alcoholism, and suicide. Background: Methotrexate is a critical component of pediatric acute lymphoblastic leukemia (ALL) therapy that can result in neurotoxicity which has been associated with an increased risk of relapse. We leveraged machine learning to develop a neurotoxicity risk prediction model in a diverse cohort of children with ALL.

Methods: We included children (age 2-20 years) diagnosed with ALL (2005-2019) and treated in Texas without pre-existing neurologic disease. Clinical information was obtained by medical record review. Neurotoxicity occurring post-induction and prior to maintenance therapy was defined as neurologic episodes occurring within 21 days of methotrexate. Kaiso (ZBTB33) is a transcription factor involved in mitotic clonal expansion and tumorigenesis in association with Adenomatous Polyposis Coli (APC) loss of heterozygosity. ENCODE data show strong overlap of the Kaiso promoter-binding site—encode-derived Kaiso-binding site (eKBS) and many other transcription factors, including BRCA1. Here we sought to determine whether BRCA1 is a component of the Kaiso enhanceosome that regulates gene transcription. Using proximal ligation assays, immunoprecipitation followed by mass spectrometry, luciferase assays and ChIP-seq experiments, we evaluated the association between BRCA1 and Kaiso. Water quality assessments are critical for ensuring timely responses to water-related concerns, particularly in low-resource areas with limited water, sanitation, and hygiene (WASH) infrastructure. In collaboration with the Belize Ministry of Health and Wellness and the Ministry of Education, Culture, Science and Technology, we conducted a survey on WASH infrastructure and resources among 221 schools. We identified 65 schools across all six districts of Belize for water quality testing. Among these 65 schools, 83% had at least one water sample that did not meet the WHO's recommended free chlorine residual level for drinking water. Nonstructural Roles Of Junctophilin-2 In The Heart, Daniel F Hutchison, Satadru K Lahiri, Kurt W Prins, Xander H T Wehrens

Junctophilin-2 (JPH2) is an essential structural protein that maintains the integrity of junctional membrane complexes (JMCs) in cardiomyocytes, thereby playing its canonical role in excitation-contraction (E-C) coupling. Recent studies reveal novel, nonstructural functions of JPH2 in cardiomyocytes, including regulation of mitochondrial function, lipid metabolism, nuclear signaling, and innate immune responses. Calpain-mediated cleavage of JPH2 generates nuclear fragments, which differentially modulate stress responses. Moreover, full-length JPH2 also localizes to the nucleus, where it forms phase-separated nuclear droplets during oxidative stress. Collectively, these findings highlight JPH2 as a multifunctional protein with critical roles beyond its canonical structural function in E-C coupling.

Oxygen (O2) is vital for cellular development, function, proliferation, and repair, underscoring its critical role in organogenesis. Both hypoxia (reduced tissue O2) and hyperoxia (excess tissue O2), when prolonged, can trigger inflammation and oxidative stress, contributing to acute and long-term cardiopulmonary and neurodevelopmental morbidity. In sick neonates, immature defense mechanisms and coexisting morbidities demand nuanced and sometimes opposing strategies for O2 saturation targets and therapeutic titration. Most current neonatal O2 targeting guidelines are based on animal models or small clinical studies, resulting in recommendations with limited evidence. Neuromuscular priming and post-activation potentiation (PAP) are techniques utilized to increase nervous system excitation, muscle force, and rate of force development during subsequent muscle contractions in sport. PAP is a commonly used approach that has the potential to influence neuromuscular status monitoring and flagging, and optimize power training. School leadership is often described in terms of vision, strategy, and efficiency. However, the lived experiences of school leaders reveal a far more complex reality—one shaped by relationships, ethical dilemmas, and the daily challenges of guiding a school community. Research has documented the positive impact of the nurse practitioner workforce on patient and system outcomes in multiple countries. Achieving these outcomes relies on successfully integrating the workforce into the health system. Research has reported negative effects of integration barriers on the ability for nurse practitioners to improve outcomes. Barriers include scope of practice restrictions, organizational climates inconducive to nurse practitioner care, and lacking mentorship. This paper applies corpus linguistic discourse analysis to approach the social media discourse of a recent political movement in Iran, known as #WomanLifeFreedom and Mahsa Amini. The movement included vast protests in different parts of Iran and went viral on an international scale. Protesters relied heavily on social media, particularly Twitter, to determine strategies and support each other during the expansive calls. To address the discourse shaped in that platform, a corpus was compiled from the Twitter posts, named Mahsa Amini Tweets Corpus (MATC). The management of the herdsmen-farmers conflict-induced internal displacement in Nigeria was characterised by remote factors that affected the protection and assistance of internally displaced persons (IDPs). This qualitative study was conducted on 12 participants comprising humanitarian workers, IDPs, and lawmakers who were conversant with the persistent herdsmen-farmers conflict-induced internal displacement. The study explored the inter-connected relationship among stakeholders within the humanitarian ecosystem. It found that though the herdsmen-farmers conflict led to mass displacement, remote factors such as unwillingness of government, data challenges, the undiplomatic approach of government in the management of the displacement failed to win support of other stakeholders …

In the last decade we have witnessed rising protests in the United States associated with issues that form part of society’s social fabric that can either facilitate or break down collective behaviour. Rising social inequalities can cause people to no longer share the same values and force individuals to work against each other.
