Autoregulation Of Rpl7b By Inhibition Of A Structural Splicing Enhancer, David Granas, Ishan Gammadde Hewa, Michael A White, Gary D Stormo

Yeast ribosomal protein gene RPL7B is autoregulated by inhibition of splicing. The first intron has a "zipper stem" that brings the 5' splice site near the branch point (BP) and serves as an enhancer of splicing that is required for efficient splicing because the intron has a nonconsensus BP sequence of UGCUAAC. The intron also contains an alternative, and mutually exclusive, structure that is conserved across many yeast species. That conserved structure is a binding site for the Rpl7 protein so that when the protein is in excess over what is required for ribosomes, the protein binds to the conserved …

Epigenetic modifications directly regulate the patterns of gene expression by altering DNA accessibility and chromatin structure. A knowledge gap is presented by the need to directly measure these modifications, especially for unannotated organisms with unknown primary histone sequences. In the present work, we developed and applied a novel workflow for identifying and annotating histone proteoforms directly from mass spectrometry-based measurements for the endangered Caribbean coral Acropora cervicornis. Unlocking Efficiency In Real-World Collaborative Studies: A Multi-Site International Study With One-Shot Lossless Glmm Algorithm, Jiayi Tong, Chongliang Luo, Et Al.

The widespread adoption of real-world data has given rise to numerous healthcare-distributed research networks, but multi-site analyses still face administrative burdens and data privacy challenges. In response, we developed a Collaborative One-shot Lossless Algorithm for Generalized Linear Mixed Models (COLA-GLMM), the first-ever algorithm that achieves both lossless and one-shot properties. COLA-GLMM ensures accuracy against the gold standard of pooled data while requiring only summary statistics and completes within a single communication round, eliminating the usual back-and-forth overhead. We further introduced an enhanced version that employs homomorphic encryption to reduce the risks of summary statistics misuse at the coordinating center. The …

BACKGROUND: Osteoarthritis (OA) is a debilitating joint disease characterized by cartilage degeneration, synovial inflammation, and bone remodeling, with limited therapeutic options targeting the underlying pathophysiology. Mechanosensitive ion channels Piezo1 and Piezo2 play crucial roles in chondrocyte responses to mechanical stress, mediating mechanotransduction pathways that influence chondrocyte survival, matrix production, and inflammatory signaling, but their distinct contributions to OA pathogenesis remain unclear.

BACKGROUND: Risk stratification is a population health management approach that classifies patients according to their health risks and projected care needs. This strategy is especially valuable in primary care, where timely interventions for high-risk individuals can lead to better health outcomes, reduced healthcare expenditures, and a more sustainable healthcare system. PURPOSE: Previous literature suggests a relationship between carpal tunnel syndrome (CTS) and heart failure (HF), indicating that patients with CTS are more likely to experience cardiomyopathy. Amyloid deposition leading to CTS may be prodromal for the development of amyloid cardiomyopathy. We hypothesized that patients undergoing surgery for CTS would have an increased risk of HF compared to those who did not undergo surgery.

BACKGROUND: Advanced stage ovarian carcinoma has a poor prognosis with recurrence rates of over 80%, 5-year survival of 36-45%, and limited response to standard therapy. Pembrolizumab and lenvatinib are FDA approved for treatment of microsatellite stable (MSS)/mismatch repair proficient (pMMR) endometrial and renal cell cancers. Early phase II studies have shown promising results in a variety of advanced solid tumors, including ovarian cancer. We report on the clinical outcome of recurrent MSS/pMMR ovarian cancer patients treated with this therapy.

Some authors have argued that liberalism cannot support lockdowns because it cannot engage in collective welfare-type reasoning. Others have thought that liberalism can justify lockdown measures, but that legitimate lockdown measures must be based on pre-legal moral principles that set acceptable limits to the imposition of risk or forbid contributing to collective harm. In this essay, I assess the overall relationship between liberalism and lockdowns. Against the first interpretation of liberalism, I argue in favor of the idea that lockdowns can be justified by liberalism. Liberalism protects the liberty of each and so can presumably protect persons from some kinds …

INTRODUCTION: In Sub-Saharan Africa, most people with dementia are cared for in the home setting (dementia self-care) with hardly any support from the formal health and social care system. The study explored family caregivers' perceptions of quality of dementia self-care. METHODS: A descriptive-exploratory qualitative study design was used. Data were collected through four focus group discussions with 48 family caregivers of older people with dementia, recruited from four villages of the Wakiso district in Uganda. Background/Objectives: Prostate cancer (PCa) is diagnosed at an earlier median age, more advanced stage, and has worse clinical outcomes in African American (AA) men compared to European Americans (EA).
Methods: To investigate the role of aberrant DNA methylation in tumor-adjacent stroma (TAS), methyl binding domain sequencing (MBD-seq) was performed on AA (n = 17) and EA (n = 15) PCa patients. This was independently confirmed using the long interspersed nuclear element-1 (LINE-1) assay. Metastable epialleles (MEs) are genomic loci at which epigenetic marks are established stochastically during early embryonic development and maintained during subsequent differentiation and throughout life, leading to stable epigenetic and phenotypic variation among genetically identical individuals. Although MEs were first described in mice over 20 years ago, the extent of epigenetic metastability in the mouse genome remains unknown. We present the first unbiased genome-wide screen for MEs in mice. Using deep whole-genome bisulfite sequencing across tissues derived from the three embryonic germ layers in isogenic C57BL/6J mice, we identified only 29 MEs, precisely localizing them and documenting their rarity. Diabetic foot ulcers (DFUs) are a serious complication of diabetes, associated with high recurrence and amputation rates. Adherence to offloading devices is critical for wound healing but remains inadequately monitored in real-world settings. This study evaluates the SmartBoot edge-computing system-a wearable, real-time remote monitoring solution integrating an inertial measurement unit (Sensoria Core) and smartwatch-for its validity in quantifying cadence and step count as digital biomarkers of frailty, and for detecting adherence. Pancreatic cancer (PC) is aggressive and deadly; most PC cells overexpress an epidermal growth factor receptor (EGFR). EGFR-targeting peptide (GE11) was conjugated to gold nanoparticles (AuNPs) for delivering rapamycin (Rap), an mTOR inhibitor that reduces proliferation and induces apoptosis. Biophysical techniques confirmed the conjugation. Rap loading efficiency was 41.5%, with sustained release at a lower pH. Conjugates showed higher uptake in PANC-1 cells than unmodified AuNPs. It also inhibited proliferation and induced apoptosis via pathways involving reactive oxygen species, mitochondrial damage, and caspase activation. This approach represents a promising nanoparticle-based therapeutic strategy targeting PC cells, utilizing GE11-EGFR interactions to minimize …

Transgender people may opt for the use of sex steroids and, in some cases, hormone blockers, which can lead to changes in body fat distribution. The aim of this study was to verify the accuracy of anthropometric equations to estimate the body density (BD) of transgender men who are undergoing Gender-Affirming Hormone Therapy (GAHT). In the western U.S., climate change is exacerbating already difficult situations in many lake systems, including Bear Lake, which spans the Idaho-Utah border and faces several environmental issues and a new wave of recreational development. In this research note, we discuss preliminary findings from community-engaged research aimed at assessing different concerns that scientists, residents, and community organizations have for Bear Lake and the surrounding communities. Existing misleading information studies mostly focus on either the message content or social network structures, overlooking the association between the content and structural factors. This research examines the role of brokers, the key network actors straddling between misleading and true/regular information groups. Content-wise, morality expressions are examined, which signal social media users’ expressions toward what is right and wrong. This research provides insights into the understudied relationship between network brokerage and moral expressions and identifies connections between moral foundation and network brokerage theories. This research develops and validates a measure of the emerging concept of informal safety leadership (ISL), which integrates elements of safety leadership and informal leadership to address a significant gap in organizational safety literature. Despite extensive research on formal safety leadership, there remains limited understanding of how non-managerial employees influence safety practices and culture within organizations. Obesity significantly impacts women’s health, particularly among middle-aged women, by increasing the risk of hormone-sensitive cancers such as breast, endometrial, and reproductive system cancers. This study examines the educational needs of this demographic group regarding obesity-related cancer risks and explores effective intervention strategies. Obesity-induced mechanisms – hormonal imbalances, chronic inflammation, and insulin resistance – drive cancer susceptibility, emphasizing the need for targeted health education. Gender disparities can be seen throughout the criminal justice system, where females are seen as weak and incapable of causing harm. This quantitative study sought to add to the existing research regarding physical characteristics linked to criminality. The labeling theory explains how physical characteristics labeled as deviant can alter a person’s perception when deciding whether an individual is violent. Physical activity is essential for children’s health. Primary schools offer an opportunity to equitably promote physical activity. However, school-based interventions have been shown to have little to no effect, potentially due to a lack of consideration of school heterogeneity. This study reports on a rapid ethnography study that was used to capture insights into English primary schools physical activity. As offshore wind gains momentum within US (United States) renewable energy goals, New Jersey's ambitious targets for offshore wind development represent a significant opportunity to reduce emissions and transition towards cleaner energy sources. This study presents a life cycle assessment (LCA) of a planned offshore wind farm off of New Jersey's coast, emphasizing the implications of a domestic supply chain. Key findings suggest that the offshore wind farm is projected to produce 0.013 kg CO2 kWh−1 of electricity generated, reflecting a 98 % decrease in carbon emissions compared to natural-gas-derived electricity. This study is aimed to compare two different planning constraints for post-lumpectomy whole breast radiation treatments to evaluate whether or not the mean dose to the contralateral breast can be significantly reduced while maintaining plan quality for treatment. A total of 10 patients who had first undergone a lumpectomy were included into this study. Each patient’s initial treatment plan was created using a conventional approach with a mean dose constraint of < 600 cGy (Plan A) and then was replanned using the Import High protocol of a mean dose constraint, aiming to further push for a mean dose < 150 cGy (Plan B) to the contralateral breast. Both of these plans were analyzed for PTV coverage and dose to the surrounding critical structures. The results have shown that the contralateral mean dose was significantly reduced from an average of 216.32 in Plan A to 126.53 cGy in Plan B. All while meeting the target coverage of > 95% isodose line. Statistical testing showed a significant difference between the two planning techniques of lowering the contralateral dose (p < 0.0001), and normality testing supported the validity of the data analysis. In addition to a substantial dose reduction to the contralateral breast, Plan B maintained equal or improved dose constraint to other surrounding critical organs at risk, including the heart, lungs, spinal cord, and thyroid. The Import High protocol demonstrated consistent outcomes across diverse patient anatomies. These findings suggest that by using the Import High mean dose constraint, it is possible to maintain plan quality while reducing dose to the contralateral breast. This approach may help reduce the risk of long-term side effects and improve the patient’s overall outcomes in whole breast radiation treatments.
