Breast cancer is the most frequently diagnosed cancer globally and the second leading cause of cancer-related deaths in American women. Triple-negative breast cancer (TNBC) lacks estrogen receptor, progesterone receptor, and human epidermal growth factor receptor 2. Thus, fewer targeting therapies are available for this most aggressive subtype. The A Disintegrin and Metalloproteinase (ADAM) family plays a vital role in cancer pathophysiology. Previous studies focused on single ADAM members. However, none of these have entered into the clinical arena as diagnostics or therapeutics for breast cancer. In this study, we demonstrate the upregulation of a panel of ADAM members in TNBC, …
Safe Nurse Staffing For Scotland: A Cross-Sectional Observational Study, Eileen T. Lake
Safe Nurse Staffing For Scotland: A Cross-Sectional Observational Study, Eileen T. Lake
Sigma Foundation for Nursing Research Grant Reports
Amid a global nurse staffing crisis, the project is designed to describe the baseline nursing standards adherence, nurse staffing, and work environments in hospitals before the April 2024 implementation of Scotland’s Safe Staffing Act 2019. Quantifying the variation is important to evaluate the Law’s subsequent implementation (whether hospitals meet standards) and impact (whether standards improve quality and safety). The results will aid policymakers by providing baseline data from Scotland before the Law is implemented and an informative exemplar for states and countries globally as many government authorities consider nurse staffing legislation.

Electricity And The Environment, Praveen Kumar
Electricity And The Environment, Praveen Kumar
Doctoral Theses
This thesis explores the intersection of electricity and the environment across three studies. The first study examines the 2018 policy in Telangana that removed agricultural electricity rationing, providing farmers with 24-hour power. Using detailed panel data and a difference-in-differences approach, the study finds a 53% rise in agricultural electricity consumption and an expansion of water-intensive rice cultivation relative to boundary districts in neighboring states. However, no statistically significant change is detected in groundwater depth measured by government monitoring wells, suggesting that current monitoring systems may not adequately capture water availability in fragmented hard-rock aquifers. The second study evaluates a subsidized …

Recent advances in large language models and retrieval-augmented generation, a method that enhances language models by integrating retrieved external documents, have created opportunities to deploy AI in secure, offline environments. This study explores the feasibility of using locally hosted, open-weight large language models with integrated retrieval-augmented generation capabilities on CPU-only hardware for tasks such as question answering and summarization. The evaluation reflects typical constraints in environments like government offices, where internet access and GPU acceleration may be restricted. Four models were tested using LocalGPT, a privacy-focused retrieval-augmented generation framework, on two consumer-grade systems: a laptop and a workstation. A technical …

Ratios between two metabolites are sensitive indicators of metabolic changes. Lipidomic profiling studies have revealed that plasma ether lipids, a class of glycero- and glycerophospho-lipids with reported health benefits, are negatively associated with obesity. Here, we utilized lipid ratios as surrogate markers of lipid metabolism to explore the processes underlying the inverse relationship between ether lipid metabolism and obesity. Plasma lipidomics data from two independent human cohorts (n = 10,339 and n = 4,492) were integrated to assess the associations between 82 lipid ratios and obesity-related markers in males and females. Results were externally validated using mouse transcriptomics data …

Acute myeloid leukemia (AML) is an aggressive hematologic malignancy characterized by the clonal proliferation of myeloid precursors and rapid progression. Historically consisting of intensive chemotherapy, AML management has evolved significantly due to advances in molecular diagnostics and risk stratification. This review discusses current therapeutic paradigms in AML, emphasizing the growing role of personalized medicine across age and risk groups. For younger, fit patients, intensive regimens such as the "7 + 3" protocol remain the standard, often enhanced by targeted agents like FMS-like tyrosine kinase 3 (FLT3) and IDH inhibitors. Older or unfit individuals benefit from low-intensity treatments such as hypomethylating …

Pressure And Force Dynamics In Artificial Muscle Actuators: A State-Space And Optimization-Based Approach, Mohammad Elzein
Dissertations and Theses
This two-part investigation explores the dynamic behavior of braided pneumatic actuators (BPAs) under bio-inspired pulse modulation, with the aim of improving their biomimetic force output and control. The first study examines the effect of pulse length and inter-pulse timing on BPA performance, revealing that force output is highly sensitive to the temporal structure of input pulses mirroring biological muscle behavior. Using dual-pulse actuation schemes, the results demonstrate that force responses exceed the additive contributions of individual pulses, with peak amplification occurring consistently at a 27 ms inter-pulse gap. Shorter pulse lengths (10–20 ms) yielded the highest normalized force increases, up …

Patients with myeloid neoplasms with loss-of-function TP53 mutations and erythroid differentiation have poor outcomes, and a better understanding of disease biology is required. Upregulation of interferon-γ (IFN-γ) signaling has been associated with acute myeloid leukemia (AML) progression, selection of TP53 mutated clones and chemotherapy resistance, but its drivers remain unclear. In this study, we found that the surface receptor C-C motif chemokine receptor-like 2 (CCRL2) is overexpressed in AML with erythroid differentiation and TP53 mutations compared to other AML subtypes and healthy hematopoietic cells. CCRL2 knockout (KO) suppressed erythroleukemia growth in vitro and in vivo. Further proteomics and transcriptomics analysis …

Background/objectives: High-quality prognostic communication in pediatric oncology is essential to support informed decision making, foster trust, and honor goals of care. While families differ in their preferences for how prognosis is discussed, clinicians often rely on assumptions rather than directly eliciting these preferences, risking misalignment and distress. To address this gap, we aimed to characterize patient, parent, and oncologist perspectives on key variables, experiences, and circumstances that influence their preferences for prognostic communication.
Methods: Semi-structured interviews were conducted with 85 participants (25 patients aged 12-25, 40 parents, and 20 oncologists) to elicit their preferences for prognostic communication and the reasons …

Introduction: Endovascular thrombectomy (EVT) is an effective treatment for basilar artery occlusion (BAO) stroke in select patients. While there is a growing body of literature suggesting that advanced imaging modalities such as computed tomography perfusion (CTP) and magnetic resonance (MR) may not be necessary for selecting anterior circulation large vessel occlusion stroke patients for EVT, whether advanced imaging may be superior to conventional imaging (non-contrast CT and CT angiography) in identifying good treatment candidates among BAO patients is less clear.
Patients and methods: This was a multicenter retrospective cohort study of BAO EVT patients treated from 2013 to 2022 in …

Cannabinoid receptor 1 (CB1) has been implicated in multiple inflammatory diseases by regulating pro-inflammatory mediators or altering immune cell polarization. However, the expression and direct functional role of CB1 in T cells remain largely unexplored. Here, we demonstrate that primary murine T cells express CB1 and that its novel agonist, BI-5756, directly increases the frequencies of regulatory T cells (Tregs) in primary murine pan T cells after activation. In addition, BI-5756 exhibits an in vivo protective effect against graft-versus-host disease (GvHD), an allogeneic T cell-mediated inflammatory complication after allogeneic hematopoietic cell transplantation (allo-HCT), resulting in an improved overall survival with …

Creating Healthy Work Environments (CHWE)
Background: Previous studies report that the healthcare industry significantly contributes to environmental degradation and intensive care units (ICUs) are a major generator of waste (Barbariol, 2023). Many ICU staff and support staff personnels have expressed frustration over extra supplies being brought into the room and later discarded. The “GreenICU” aims to reduce the million-pound contribution of medical waste to our landfills.
Method: "GreenICU” focused on four key areas. First, an ICU green team was formed, consisting of PCAs and nurses from day and night shifts. Forming a team is crucial for gathering input, making recommendations, and ensuring the sustainability of …

Addressing Job Satisfaction In Emergency Services: An Exploratory Study, Jacqueline Christianson, Tiffany Warthon, Tala Fuad Abuzahra
Creating Healthy Work Environments (CHWE)
Purpose: Emergency services professionals (ESPs), like emergency medical technicians, paramedics, and registered nurses, work in stressful, low-resource environments and are vulnerable to work-related burnout (Crowe et al., 2020; Hinderer et al., 2014). While many ESPs are motivated by compassion for others, most ESPs never learn what happened to the patients they cared for (Ntatamala & Adams, 2022; Patterson et al., 2005). Disconnection from patient outcomes may limit compassion satisfaction among ESPs and make it more difficult to actualize their role in patient care and contribute to burnout, which negatively affects patient care (Georgakakos et al., 2022). The purpose of this …

Creating Healthy Work Environments (CHWE)
With escalating nursing faculty shortages, faculty retention is important. Developing an infrastructure to support work-life balance and create programs that address wellness is needed to enhance nursing faculty satisfaction by improving well-being and career satisfaction. Following the Healthy Nurse Healthy Nation initiative launched in 2017 to improve the health of nurses within the United States, educational settings must promote and maintain healthy work environments on a more personal level. Within a midwestern College of Nursing, faculty and instructional academic staff satisfaction surveys are deployed every three years.
